치자(Gardenia jasminoides Ellis) 과피의 용매별 추출물의 Flavonoid 함량 및 항산화 활성 비교 (Comparison of Flavonoid Content and Antioxidant Activities of Peel Extracts from Gardenia jasminoides Ellis by Various Solvents)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides fructus Ellis (GJE) in Namhae, Korea, following some established methods. CM (Chloroform:Methanol, 2:1, v/v), 70% ethanol, and n-butanol extracts were collected. Flavonoid content and value as a functional food ingredient of GJE peel was investigated through assessing antioxidant [DPPH (1,1'-diphenyl-2-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid)], and hydroxyl radical scavenging activities; superoxide dismutase like ability; ferrous ion-chelating capacity; and tannin content by solvent extraction. Solvent extract antioxidant activities significantly increased (p<0.05) at increasing concentrations (0.2, 0.4, 0.6 mg/mL). GJE peel extracts were less active than the positive control [ascorbic acid, BHA (butylated hydroxyanisole), and EDTA (ethylenediaminetetraacetic acid disodium salt dihydrate)]. Based on the results of this study, GJE peel could be used as a natural antioxidant source due to its high antioxidant activity and bioactive compound content.

치자(Gardenia jasminoides Ellis fructus) 종자 추출물의 항산화 활성 및 지질과산화 저해능 (Antioxidant Activities and Lipid Peroxidation Inhibition Ability of Gardenia jasminoides Ellis Fructus Seed Extracts)

  • The purpose of this study was to investigate the bioactivity of extracts from the seeds of Gardenia jasminoides Ellis fructus (GJE) found in Namhae, Korea. Extraction was performed using three solvents, 70% methanol, Distilled Water (DW), and Ethyl Acetate (EA). We determined the total phenol and phytic acid contents of the extracts to evaluate their nitrogen oxide scavenging activity, antioxidant activity, reducing power, and lipid peroxidation inhibition ability. The phytic acid content of GJE was found to be 1.157 mg PAE (Phytic Acid Equivalent) /g DW. The yields of the three extraction processes were as follows: DW, 36.61%; 70% methanol, 30.10%; and EA, 20.40%. The physiological activities of the extract solvents increased significantly with increasing concentrations (0.2, 0.4, and 0.6 mg/mL) (p<0.05), but were lower than those of ascorbic acid, BHA, and trolox. Total phenol content was the highest in the 70% methanol extract, followed by DW and EA extracts. Further, nitrogen oxide scavenging activity and antioxidant activity were the highest for the 70% methanol extract followed by DW and EA extracts. Based on these results, the bioactivities of the 70% methanol and DW extracts of GJ seeds were excellent. These extracts can be used as natural antioxidants.

Comparative Evaluation of Hepatoprotective Activities of Geniposide, Crocins and Crocetin by CCl4-Induced liver Injury in Mice

  • Iridoid glycosides (mainly geniposide) and crocetin derivatives (crocins) are the two major active constituents in Gardenia jasminoides Ellis. In the present study, geniposide, crocins, crocin-1 and crocetin were separated from gardenia chromatographically. Then, mice were orally administrated with geniposide (400 mg/kg b.w.), crocins (400 mg/kg b.w.), crocin-1 (400 mg/kg b.w.) and crocetin (140 mg/kg b.w.) once daily for 7 days with $CCl_4$. Hepatoprotective properties were evaluated by biochemical parameters: Administration of geniposide, crocins, crocin-1and crocetin significantly lowered serum alanine transaminase (ALT), aspartate transaminase (AST) and alkaline phosphatase (ALP) levels in $CCl_4$-treated mice. The reduced glutathione (GSH) levels and antioxidant enzymes (SOD and CAT) activities were also increased by geniposide, crocins, crocin-1 and crocetin. Histopathological examination of livers showed that these components reduced deformability, irregular arrangement and rupture of hepatocyte in $CCl_4$-treated mice. These biochemical results and liver histopathological assessment demonstrated that geniposide, crocetin derivatives and crocetin show comparative beneficial effects on $CCl_4$-induced liver damage via induction of antioxidant defense. Therefore, contents of geniposide and crocetin derivatives should be both considered for hepatoprotective efficacy of Gardenia jasminoides Ellis.

Ameliorating Effect of Gardenia jasminoides Extract on Amyloid Beta Peptide-induced Neuronal Cell Deficit

  • The brains of Alzheimer's disease (AD) patients are characterized by large deposits of amyloid beta peptide ($A{\beta}$). $A{\beta}$ is known to increase free radical production in nerve cells, leading to cell death that is characterized by lipid peroxidation, free radical formation, protein oxidation, and DNA/RNA oxidation. In this study, we selected an extract of Gardenia jasminoides by screening, and investigated its ameliorating effects on $A{\beta}$-induced oxidative stress using PC12 cells. The effects of the extract were evaluated using the 2',7'-dichlorofluorescein diacetate (DCF-DA) assay and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction assay. To find the active component, the ethanol extract was partitioned with hexane, chloroform, and ethyl acetate, respectively, and the active component was purified by silica-gel column chromatography and HPLC. The results suggested that Gardenia jasminoides extract can reduce the cytotoxicity of $A{\beta}$ in PC 12 cells, possibly by reducing oxidative stress.

소목, 치자, 쑥으로부터 추출된 천연염료의 색도분석 (Chromaticity Analysis of Natural Dyes Extracted from Sappan Wood, Gardenia, and Mugwort)

  • 환경 친화적이고 질병을 유발시키지 않은 천연색소의 사용이 증가함에 따라 천연색소를 추출하는 다양한 방법이 연구되고 있다. 본 연구에서는 천연염료인 brazilein, crocin, chlorophyll을 포함하고 있는 식물성 원료인 소목, 치자, 쑥을 대상으로 천연염료를 추출하였다. 추출용매의 pH와 추출시간을 변수로 추출된 천연염료의 적색, 황색, 녹색계열의 목표색을 설정하고 추출된 천연염료의 명도와 색의 좌표(L, a, b)의 정량적 수치로부터 색도분석을 수행하였다. 소목의 적색소인 brazilein과 치자의 황색소인 crocin의 경우 목표색인 카드뮴 레드와 카드뮴 옐로에 근접한 색상을 추출할 수 있었다. 추출시간 20 min을 기준으로 brazilein의 경우 pH 12, crocin의 경우 pH 9에서 색차를 나타내는 ${\Delta}E^*$가 각각 18.2와 18.4로 목표색과 유사한 색의 색소가 추출되었다. 그러나 쑥의 녹색색소인 chlorophyll의 경우 목표색인 크롬 그린과는 차이를 보였다.

Jar Fermentor에서 생산된 치자 생물변환 색소의 특성 및 안정성 (Characterization and Stability of Gardenia Jasminoides Biotransformed Pigment Produced in Jar Fermentor)

  • 치자황색소로부터 생물변환색소를 생산하기 위하여 Gram 양성 세균 7종, Gram 음성세균 4종, 효모 2종 등 총 13종의 미생물을 jar fermentor에서 배양하고 치자황색소를 첨가한 후 7일간 생물변환된 색소의 색가에 대해 조사한 결과,S. mutans MK-34에 의해 변환된 색소의 색가가 가장 높았다. 치자황색소가 jar fermentor 안에서 S. mutans MK-34에 의한 변환의 특징은 588 nm에서 새로운 흡수극대가 나타나며, 흡광도값은 배양시간이 경과함에 따라 증가하였다 S. mutans에 의한 생물변환 색소를 대상으로 광 및 온도에 대한 안정성 조사에서 치자황색소에 비해 생물변환 색소가 일광 조건 및 $60^{\circ}C$ 온도조건에서 안정성을 나타내었다.

치자와 복분자분말을 첨가한 연근차의 품질특성 (Quality Characteristics of Lotus Root Tea added with Gardenia jasminoides Powder and Rubus coreanus Miquel Powder)

  • This study was performed to investigate the quality characteristics of lotus root tea (LT) prepared with Gardenia jasminoides powder (GLT) and Rubus coreanus Miquel powder (RLT). The proximate compositions, physicochemical properties, mineral contents, and sensory evaluations of samples were measured. GLT sample showed higher contents of crude protein, crude fat, crude ash, and moisture (p<0.001) as well as pH (p<0.001) compared to LT and RLT. In terms of color, lightness (L) and yellowness (b) of GLT were the highest among the samples, whereas redness (a) of GLT was the lowest. The results of mineral analysis showed that the mineral contents of K, Ca, and Fe of LT were the highest, whereas the content of Mg of GLT was the highest. The results of browning color intensity of all samples showed that LT had the lowest. GLT showed higher contents of water soluble solids than LT and RLT. In the sensory evaluation, the scores of color, flavor, aftertaste, feeling of throat, and overall preference of LT and GLT were significantly higher than those of RLT. From the findings, this study suggests that addition of Gardenia jasminoides powder was effective for preparation of lotus root tea in the aspects of consumer acceptability.

Effects of the Constituents of Gardenia Fructus on Prostaglandin and NO Production

  • The fruits of Gardenia jasminoides Ellis have been previously reported to possess anti-inflammatory activity. In this study, the constituents including geniposide, geniposidic acid, genipin and crocin were evaluated for their effects on prostaglandin and NO production in an attempt to establish anti-inflammatory cellular mechanisms. Among the constituents tested, only genipin significantly inhibited cyclooxygenase-2-mediated $PGE_2$ and inducible nitric oxide synthase-mediated NO production from lipopolysaccharide-treated RAW 264.7 cells at 10-100 ${\mu}$M. Genipin also inhibited nuclear transcription factor-${\kappa}B$ activation. Moreover, genipin showed in vivo antiinflammatory activity on ${\lambda}$-carrageenan-induced paw edema in mice (10.4-29.9% inhibition at 20-100 mg/kg, i.p.). All of these results suggest that genipin may contribute to anti-inflammatory activity of the fruits of G. jasminoides and an inhibitory action on prostaglandin and NO production is, at least, the part of anti-inflammatory mechanism of genipin.

치자의 염색성, 항균성, 소취성에 관한 연구 (The Dyeability, Antibacterial Activity and Deodorization of Gardenia)

  • The purpose of this study is to investigate the dyeability, antibacterial activity and deodorization on cotton and silk fabrics dyed with extracted solution from gardenia. The results are as follows: 1. The K/S value of silk fabrics was higher than that of cotton fabrics, and the K/S value was slightly increased by mordanting with $SnCl_2.2H_2O$ and $K_2Cr_2O_{7}$ on cotton and silk fabrics. 2. Cotton fabrics showed yellow from GY to Y and silk fabrics showed yellow or reddish yellow from GY to Y-YR. 3. The fastness to washing of cotton fabrics was very poor and that of silk fabrics was middle or lower, excluding good fastness to wool contamination of both fabrics. The fastness to dry cleaning and rubbing of both fabrics was very good. The fastness to perspiration of cotton fabrics was middle or higher, but that of silk fabrics was much better than that of cotton fabrics. The fastness to the light of both fabrics showed very poor with 1 grade. Dyeing fastness was slightly improved by mordanting on both fabrics. 4. Silk and cotton fabrics did not show antibacterial activity, but the activity of silk fabrics was higher than that of cotton fabrics. The antibacterial activity was increased by mordanting. 5. The deodorization of dyed cotton and silk fabrics was excellent, and that of silk fabrics showed better than that of cotton fabrics.

Hepatoprotective Effects of Gardenia jasminoides Ellis Extract in Nonalcoholic Fatty Liver Disease Induced by a High Fat Diet in C57BL/6 Mice

  • This study was carried out to investigate the potential effects of Gardenia jasminoides (GJ) extracts, on hepatic steatosis and lipid metabolism in mice fed with high-fat diet (HFD). GJ extracts (100 mg/kg, ${\times}10$ weeks) fed mice showed reduced body weight, adipose tissue weight, reduced aminotransferase level in plasma and hepatic lipid (triglyceride, total cholesterol) content. These effects were accompanied by decreased expression of lipogenic genes, sterol regulatory element binding protein-1c (SREBP-1c), liver X receptor (LXR), fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC), cluster of differentiation 36 (CD36), lipoprotein lipase (LPL) and decreased lipogenic enzyme FAS and HMG-CoAR enzyme activities while elevating carnitine palmitoyltrasferase-1 (CPT) activity. Based on these results, we speculated that the inhibitory effect on hepatic steatosis of GJ extract containing geniposide is the result of suppression of lipid synthesis in mice fed with HFD, suggesting that GJ extract may be beneficial in preventing hepatic steatosis.
