This study evaluated the binding abilities of rabbit anti-ovalbumin (OVA) immunoglobulin G (IgG) and egg-allergic patient IgE on gamma-irradiated OVA during proteolysis using pepsin and trypsin. The concentrations of both the intact and the irradiated OVAs decreased during proteolysis when detected with IgG However, when detected by patient IgE the concentration of the intact OVA decreased up to 30 min after the trypsin treatment and increased thereafter. Irradiated OVA detected by patient IgE showed a lower initial concentration (0.16%) than that of the intact OVA, and this reduced concentration was maintained stably. The results indicate that irradiation, rather than enzymatic treatment, could reduce the binding of the irradiated and enzyme-treated OVA. Therefore, gamma irradiation has potential as an effective method to reduce OVA-induced allergy and may enhance the safety of egg-allergic individuals.

High density polyethylene (HDPE) is degraded due to a radiation-induced oxidation when it is used as a neutron moderator in a neutron counter for a nuclear material accounting of spent fuels. The HDPE exposed to the gamma-ray emitted from the fission products in a spent nuclear fuel results in a radiation-induced degradation which changes its original molecular structure to others. So a neutron moderating power variation of HDPE, irradiated by a gamma radiation, was investigated in this work. Five HDPE moderator structures were exposed to the gamma radiation emitted from a $^{60}Co$ source to a level of $10^5-10^9$ rad to compare their post-irradiation properties. As a result of the neutron measurement test with 5 irradiated HDPE structures and a neutron measuring system, it was confirmed that the neutron moderating power for the $10^5$ rad irradiated HDPE moderator revealed the largest decrease when the un-irradiated pure one was used as a reference. It implies that a neutron moderating power variation of HDPE is not directly proportional to the integrated gamma dose rate. To clarify the cause of these changes, some techniques such as a FTIR, an element analysis and a densitometry were employed. As a result of these analyses, it was confirmed that the molecular structure of the gamma irradiated HDPEs had partially changed to others, and the contents of hydrogen and oxygen had varied during the process of a radiation-induced degradation. The mechanism of these changes cannot be explained in detail at present, and thus need further study.

This study was done to investigate effects of ${\gamma}$ irradiated beef feeding on the formation of gluta thione S transferase placental form positive(GST P+) foci, lipid peroxidation, cytochrome P450 system and microsomal glucose 6 phosphate activity in diethylnitrosamine(DEN) initiated rat hepatocarci nogenesis. Weaning Sprague Dawley male rats were fed the diet containing ${\gamma}$ irradiatied ground beef at the dose of 0, 3, 5kGy as a 20% of protein source for 8 weeks. One week after feeding, rats were intraperitoneally injected twice with a dose of DEN(50mg/kg BW). As a promoter, 0.05% phenobarbital was fed in drinking water from one week after DEN treatment until the end of experiment. At the end of 8th week, rats were sacrificed and hepatic GST P+ foci, microsomal malondialdehyde(MDA) and conjugated diene contents were determined. In addition, cytochrome P450 content and the activities of NADPH cytochrome P450 reductase and glucose 6 phosphatase were also measured. There was no significant effect by gamma irradiation on microsomal MDA content, conjugated diene, cytochrome P450 content and activities of NADPH cytochrome P450 reductase and glucose 6 phosphatase. However with DEN treatment, microsomal MDA content and conjugated diene contents were significantly changed. Cytochrome P450 content was also significantly increased while microsomal glucose 6 phophatase activity was significantly decreased with DEN treatment. However activity of NADPH cytochrome P450 reductase was not affected. An interesting finding in this study was that the number and area of hepatic GST P+ foci of the rats fed gamma irradiated beef were significantly(p<0.05) lower than those of the control. Such a lowering effect on GST P+ foci formation was highest at the dose of 3kGy than others. Overall results suggest that the consumption of low dose of gamma irradiated beef does not affect the formation of lipid peroxide, cytochrome P450 system and membrane stability.
This study investigated the effect of gamma irradiation on immune enhancing activity of Chaga mushroom extract (CME). CME was prepared by hot water extraction at $70^{\circ}C$ for 4 hours and lyophilized. Lyophilized CME powder was dissolved with deionized water at $10mg\;ml^{-1}$ and then irradiated at the doses of 10, 30 and 50 kGy by cobalt 60 gamma irradiator. The gamma-irradiated and non-irradiated CME were treated into the splenocyte separated from mouse. Cell proliferation and cytokine production of the immune cells were increased by gamma-irradiated CME and these increases were more prominent when CME was irradiated at higher doses. Therefore, it is considered that gamma irradiation can be an effective method for improvement of the immunomodulating activity Chaga mushroom extract.

To establish the seed treatment method of the gamma radiation for germination enhancement of bottle gourd(Lagenaria siceraria Standl.) and pumpkin(Cucurbita ficifolia Bouche), seeds were irradiated with the dose of 1$\sim$20 Gy of the gamma radiation. The germination rate in irradiated group was higher than that of the control, depending on seed condition. In the case of bottle gourd, the germination rate of 70$\sim$80% germinative seed was increased at 4$\sim$20 Gy irradiated groups. Especially, it was the highest at 4 Gy irradiated group. The germination rate of 80$\sim$90% germinative seed was the highest at 4 Gy irradiated group. The germination rate of 70$\sim$80% and 80$\sim$90% germinative seed at 4 days after sowing was increased 66% and 26% at 4 Gy irradiated group, respectively. In the case of pumpkin, the germination rate of 70$\sim$80% and 80$\sim$90% germinative seed was increased at 2 Gy and 8 Gy irradiated group, respectively. Effect of the low-dose gamma radiation on the early growth was enhanced depending on their germination enhancement. Especially, it was the highest at 2 Gy irradiated group of 70$\sim$80% germinative seed of pumpkin. Catalase activity of bottle gourd was increased at 4 Gy irradiated group. Catalase and peroxidase activity of pumpkin were increased at 2 Gy and 8 Gy irradiated group. These results suggest that the germination, early growth and physiological activity of bottle gourd and pumpkin seeds could be promoted by the low dose gamma radiation.

In order to identify the genetic relationship analysis by acute and chronic gamma irradiation, Arabidopsis (Arabidopsis thaliana L.) were irradiated with 200 Gy of gamma-rays using gamma-irradiator (3,000 Ci; Nordion, Canada) and gamma-phytotron (400 Ci; Nordion, Canada) for acute and chronic irradiation, respectively. Genetic relationship among two acute gamma-irradiated plants (A1 and A24) and three chronic gamma-irradiated plants (C1W, C2W, C3W) were analyzed using the amplified fragment length polymorphism (AFLP) technique compared with each non-irradiated plant. A total of 28 EcoRI and MseI primer combinations were used to screen 8 treatments by the ABI3130 capillary electrophoresis system. Amplified products by 28 primer sets showed 1,679 bands with an average of 51 bands per primer combination. Out of the total bands scored, 1,164 fragments were polymorphic bands, with different alleles existing among the treatments. The cluster analysis was performed using the UPGMA (Unweighted Pair Group Method using Arithmetic) in the computer program NTSYS-pc. In clustery analysis, acute gamma-irradiation showed higher genetic variation compared with chronic gamma-irradiation.
Gamma-irradiation (0-7 kGy) of ginseng is permitted in Korea for the purpose of microbial decontamination; with strict labeling, traceability and monitoring requirements. An identification study was conducted to determine the photostimulated-luminescence (PSL) and thermoluminescence (TL) properties of gamma-irradiated fresh and white ginsengs cultivated in different areas. Dose- dependent PSL-based screening was possible for white ginseng samples; however, inappropriate results from non-irradiated fresh ginseng samples were obtained, showing intermediate (700 to 5,000) or positive ($T_2$ >5,000, irradiated) PSL counts due to the abundance of minerals on the surfaces of the samples. TL analysis of separated minerals from all non-irradiated samples gave TL glow curves of low intensity with a maximum peak after $300^{\circ}C$. However, well-defined irradiation-specific (high intensity with a maximum peak at about $200^{\circ}C$) glow curves were observed for all the irradiated samples, regardless of their type and origins. TL ratios (first glow curve /second glow curve) were also determined to confirm the irradiated (>0.1) and non-irradiated (<0.1) results. SEM-EDX (scanning electron microscope-energy dispersive X-ray) and XRD (X-ray diffraction) spectroscopic analyses showed that feldspar and quartz minerals were the main source for the typical radiation-specific luminescence properties.

Objective: To study the contribution of plant enzyme and microbial activities on protein degradation in silage, this study evaluated the nitrogen transformation dynamics during ensiling of non- and irradiated alfalfa (Medicago sativa L.) and red clover (Trifolium pratense L.). Methods: Alfalfa and red clover silages were prepared and equally divided into two groups. One group was exposed to ${\gamma}$-irradiation at a recommended dosage (25 Gky). Therefore, four types of silages were produced: i) non-irradiated alfalfa silage; ii) irradiated alfalfa silage; iii) non-irradiated red clover silage; and iv) irradiated red clover silage. These silages were opened for fermentation quality and nitrogen components analyses after 1, 4, 8, and 30 days, respectively. Results: The ${\gamma}$-irradiation successfully suppressed microbial activity, indicated by high pH and no apparent increases in fermentation end products in irradiated silages. All nitrogen components, except for peptide-N, increased throughout the ensiling process. Proteolysis less occurred in red clover silages compared with alfalfa silages, indicated by smaller (p<0.05) increment in peptide-N and free amino acid N (FAA-N) during early stage of ensiling. The ${\gamma}$-irradiation treatment increased (p<0.05) peptide-N and FAA-N in alfalfa silage at day 1, whereas not in red clover silage; these two nitrogen components were higher (p<0.05) between day 4 and day 30 in non-irradiated silages than the irradiated silages. The ammonia nitrogen and non-protein nitrogen were highest in non-irradiated alfalfa silage and lowest in irradiated red clover silage after ensiling. Conclusion: The result of this study indicate that red clover and alfalfa are two forages varying in their nitrogen transformation patterns, especially during early stages of ensiling. Microbial activity plays a certain role in the proteolysis and seems little affected by the presence of polyphenol oxidase in red clover compared with alfalfaa.
A series of experiments on the effects of ${\gamma}-irradiation$ was performed to reveal the pathogenicity of ${\gamma}-irradiated$ oocysts of E tenella from Cobalt-60 and its progeny. The SPF chickens were inoculated with differnt doses of radiation and inoculum. The level of 100 Gy ${\gamma}-irradiation$ from $^{60}Co$ and the level of inoculum with $1{\times}10^4$ oocysts were recognized more pathogenic than those of the other groups by comparison of body weight gains, blood in feces and lesion scores. The signs of blood in feces, lesion score and the number of excreted oocysts in the feces were revealed as the lowest in the group of the ${\gamma}-irradiated$ oocysts, the average in the group of the 1st and the 3rd progeny, and the highest in the group of non-irradiated oocysts of E $tenell\grave{a}$. The body weight gain of the group immunized with ${\gamma}-irradiated$ oocysts of E tenella was higher than those of the non-irradiated, the 1st and 3rd progeny groups. The body weight gain of the groups immunized with the 1st and the 3rd progeny of E tenella were higher than that of the non-irradiated group. The feed conversion ration of the group immunized with ${\gamma}-irradiated$ oocysts of E tenella was lower than those of the non-irradiated, the 1st and the 3rd progeny groups. The feed conversion ratios of the group immunized with the 1st and 3rd progeny of ${\gamma}-irradiated$ oocysts were lower than that of the group infected with non-irradiated E tenella. The anticoccidial index(ACI 190.6) in the chickens immunized with the ${\gamma}-irradiated$ oocysts of E tenella and those(ACI 142.8 and 107.4) of the 1st and the 3rd progeny groups were higher than that (ACT 87.4) of the group infected with non-irradiated E tenella. It was thought that the pathogenicity of ${\gamma}-irradiated$ E tenella would be recovered according to increase the number of generation passaged in chicken.
This study was conducted to determine the effect of gamma-ray and electron-beam irradiation on dried spices (black pepper, red pepper, parsley, and basil) using the photostimulated luminescence (PSL), thermoluminescence (TL) and electron spin resonance (ESR) methods. The spices were irradiated at 0, 1, 5, and 10 kGy. All non-irradiated spices had photon counts (PCs) less than 700 PCs. The PCs of three irradiated spices (red pepper, parsley, and basil) were clearly distinguishable from those of non-irradiated ones, exhibiting PSL signals higher than 5000 PCs. However, negative PSL counts (<700 PCs) were obtained for most irradiated black pepper, except those irradiated with 5 kGy gamma rays and 10 kGy electron-beams. TL glow curves of the irradiated spices showed a higher peak at $150-250^{\circ}C$. TL ratios were found to be less than 0.1 for non-irradiated spices and higher than 0.1 for irradiated ones. No ESR signal was observed for any irradiated spice except red pepper, which displayed cellulose-based ESR spectra. Therefore, the results suggest that the PSL, TL, and ESR methods are effective detection techniques for dried spices irradiated with electron beams as well as gamma rays.
