일반과 자색 건조 돼지감자의 식품 성분 및 항산화 활성 (Food Components and Antioxidant Activities of Dried Jerusalem Artichoke with White and Purple Colors)

  • 본 연구는 일반과 자색 건조 돼지감자의 일반성분, 무기질 함량, 유리아미노산 함량, 색도, 항산화 활성을 측정하였다. 돼지감자의 일반성분은 품종과 관계없이 탄수화물, 조단백, 회분, 수분 함량 순으로 높았으며, 색의 차이는 나타나지 않았다. 돼지감자의 품종과 관계없이 가장 높은 무기질은 칼륨이었고, 다음으로 칼슘, 마그네슘, 나트륨, 철분 순으로 나타났다. 일반시료가 자색시료보다 높은 무기질은 칼슘, 마그네슘, 철분, 아연이었고 자색이 일반보다 높은 것은 칼륨, 나트륨, 구리, 망간으로 나타났으나 품종 간의 유의적인 차이는 없었다. 돼지감자에 가장 많이 함유된 아미노산은 품종과 관계없이 arginine이었고, 다음으로 asparagine, aspartic acid, ${\gamma}-amino-n-butyric$ acid 순으로 나타났다. 또한, 일반시료보다 자색시료에서 유의적으로(P<0.05) 높게 나타난 아미노산은 cystine, leucine, tyrosine이었고, 일반시료가 자색시료보다 유의적으로(P<0.05) 높게 나타난 아미노산은 phosphoethanolamine으로 나타났다. 색도의 경우 L, a, b 모두 일반시료보다 자색시료에서 높게 나타났으나 유의적 차이는 없었다. 항산화 활성에서는 방법론 중 ABTS assay를 제외하고는 모든 활성에서 자색시료가 일반시료에 비해 항산화 활성이 높게 나타났다. 일부 FRAP와 ORAC assay의 경우 추출물의 낮은 농도에서는 일반시료와 자색시료 간 차이가 없는 것으로 나타났으며, 자색시료의 경우 높은 농도에서 항산화 활성이 있는 것으로 나타났다.

GABA-enriched Fermented Laminaria japonica Protects against Alcoholic Hepatotoxicity in Sprague-Dawley Rats

  • The sea tangle, Laminaria japonica has long been used in Korea as a folk remedy to promote health. Gamma-amino butyric acid-enriched (5.56% of dry weight) sea tangle was obtained by fermentation with Lactobacillus brevis BJ-20 (FLJ). A suppressive effect of FLJ on carbon tetrachloride-induced hepatotoxicity has been shown previously. Alcohol administration to Sprague-Dawley rats leads to hepatotoxicity, as demonstrated by heightened levels of hepatic marker enzymes as well as increases in both the number and volume of lipid droplets as fatty liver progresses. However, FLJ attenuated alcohol-induced hepatotoxicity and the accumulation of lipid droplets following ethanol administration. Additionally, FLJ increased the activities and transcript levels of major alcoholmetabolizing enzymes, such as alcohol dehydrogenase and aldehyde dehydrogenase, and reduced blood concentrations of alcohol and acetaldehyde. These data suggest that FLJ protects against alcohol-induced hepatotoxicity and that FLJ could be used as an ingredient in functional foods to ameliorate the effects of excessive alcohol consumption.

척추동물 뇌조직의 Energy 대사에 관한 계통학적 비교연구 (A Comparative Study on the Energy Metabolism of Brains of Several Vertebtrates with Respect to Their Phyogeny)

  • The present paper deals with the comparative study on phylogenic difference in the patterns of energy metabolism of brain slices of several vertebrate species by measuring oxygen consumptionwith glucose-6-phosphate, glucose-1-phosphate, glyceraldehyde-3-phosphate or glutamate as respiratory substrate employing Warburg's manometric method, by determination of the utilization rate of glucose using glucose-1-C14 by analyzing patterns of free amino acid distribution , and by histochemical determination using glucose-1-C14 by analyzing patterns of free amino acid distribution acid distribution , and by histochemical determination of glycogen contents. 1. Glucose enhances the oxygen consumption of brain slices of animals belinging to reptile, aves and mammalia while it shows a tendency to decrease that of animals belonging to pisces and amphibia. 2. Glucose-6--phosphate increase oxygen consumption more than glucose in every species examined, while glucose-1-phosphate and glyceraldehyde-3-phosphate increase that of Rana nigromaculata only . In general m, it appears that phosphosugars are more effective as a respiratory substrate to those species which have less endogenous respiration than to those having larger endogenous respiration. 3. Similar patterns of free amino acid distribution and the relative amount are found among the species and in every species examined glutamic acid is detected in the larges amount . ${\gamma}$-Amino butyric acid, glycine, alanine and aspartic acid are found in every species. 4. Ophicephalus showed less oxygen consumption than endogenous respiration when glutamate was added to the medium. When sodium fluoride was added, the oxygen consumption was some what increased . Such phenomenon wasnot found in the frog. 5. The result of histochemcial analysis of the brain showed that glycogen was abundantly present in the fish , amphibia , and especially in the reptile and that no distinctive grains of glycogen were found in the bird and mammal . From these facts, it may be supposed that anaerobic glycolysis as energy source dominates in fish and amphibia and aerobic respiration through the oxidation of glucose dominates in bird and mamal , the reptile occupying transitional position between these two categories. The way of obtaining energy for brain activity by the oxidation of glucose supplied from the circulating blood is seemed to be first acquired by reptile and the function is completed both in aves and mammal.

  • 종간, 속간의 잡종에서는 흔히 불임현상이 일어나는데 인삼의 경우도 예외가 아니어서 고려인삼(panax ginseng: P.G.)과 미국인삼(panax quinquefolium: P. Q.) 그리고 죽절인삼(Panax japonicus: P.J) 간의 교잡육종에서 P.G.${\times}$P.J.에서만 아주 희귀하게 종자를 얻을수 있고 다른 종합에서는 전혀 잡종 제이세대를 얻을 수가 없었다고 하므로 본실험에서는 P.G.${\times}$P.Q.와 P.G.${\times}$P.J.의 교잡종의 화기 및 종자형성중의 유리아미노산의 소장을 추구함으로서 불임현상과 물질대사와의 관련성의 일단을 밝히고저 한다. 1. 두 교잡종의 시료에서 검출된 유리아미노산의 Chromatogram은 서로 유사한 pattern을 나타냈으며 그들의 양친이 나타내는 pattern과 유사하나 spot의 종류나 크기 정색도는 여러 가지 점에서 특이한 변화가 있다. P.G.${\times}$P.Q.에서 19종, P.G.${\times}$P.J.에서는 21종의 spot을 검출하였는데 전자에서 Alanine, Valine, Leucine, Phenyl alanine, Proline, Hydroxy proline, Serine, Threonine, Tyrosine, Aspartic acid, Glutamic acid, Lysine, Arginine, ${\beta}$-Alanine, Cysteic acid, Tryptophan, Asparagine, Glutamine, ${\gamma}$-Amino butyric acid를 확인하고 후자의 경우는 상기한 것 외에 Methionine과 한 개의 미지 spot를 얻었다. 2. Alanine, Aspartic acid, Glutamic acid, Cysteic acid와 Asparagine이 전체적으로 보아 주체를 이루고 있는 것은 P.G. P.Q. P.J에서와 같으나 Asparagine이 소포자기와 화분성숙기에 최다량으로 나타나는 것은 이들의 양친의 경우에는 보지 못했던 특이한 일이다. 3. 홍숙기에 Cysteic acid가 감소되는 것은 P.Q. P.J.의 경우와는 유사하나 P.G의 경우와는 반대되는 현상이며 P.G.${\times}$P.J.에서 Methionine이 검출된 것도 특이하다. 4. Proline은 그들의 양친의 경우와는 현저한 차이가 있었는데 소포자기에서 아주 미약한 정색을 나타낼뿐이며 이 때에 Asparagine의 spot가 가장 크고 강한 정색을 나타내었는데 이제까지의 여러 연구 결과를 종합해 보면 Proline의 양과 화분의 임성과는 아주 밀접한 관계가 있으며 이런 관계가 인삼 종자 형성에서도 나타나고 있다. 또한 Proline이 결핍될 때 Asparagine의 축적이 있다는 많은 보고도 있다. 5. 교잡종에서의 결실불능의 원인을 Proline의 과소에 의한 것으로 보며 Proline이 약의 화분퇴화와 중요한 관계를 가지고 있을 것으로 추찰된다. 6. 그밖에 여러 아미노산의 소장이 P.G. P.Q. P.J. 등의 자식계와 그들과의 잡종사이에 있어서 상당한 차이가 생기는 것은 교잡에 의한 Gene-action system의 변혁에 따른 결과라고 추찰되는 것으로 Proline의 대사 및 생리적 영향과 더불어 보다 깊은 조사가 필요한 것으로 생각된다.

추출용매에 따른 함초 추출물의 피부미용 효과 (Effect of Salicornia bigelovii Extract on the Activities of Whitening and Anti-wrinkle)

  • 본 연구는 함초를 이용하여 기능성 화장품 개발을 위한 기초자료를 얻기 위해 추출용매에 따른 함초 추출물의 항산화 활성, tyrosinase 저해활성, elastase 저해활성, collagenase 저해활성을 검토 하였다. 여러 아미노산 중에서 glutamic acid이 가장 높았고, 그 다음으로는 proline, alanine, $\gamma$-amino-n-butyric acid, arginine 순이였다. 폴리페놀 함량 및 항산화 효과는 ethyl acetate extract > n-butanol extract > hexane extract > hot water extract > ethanol extract 순이였다. 특히 ethyl acetate를 추출용매로 사용 할 경우 폴리페놀 함량이 31.6 mg/mL였고, 항산화 활성은 82.6%였다. Tyrosinase활성 저해도는 ethyl acetate 추출물의 농도에 의존적으로 증가하였다. 특히 추출물 농도가 20에서 100 mg/mL로 증가 할 경우 tyrosinase활성 저해도는 15.2에서 98.2%로 증가 했다. Ethylacetate 추출물 농도가 20에서 100 ug/mL 증가 할 경우 elastase 저해활성은 2.9에서 32.5%로 증가 되었고, collagenase 저해 활성은 3.8에서 29.7%로 증가 하였다. 이상의 결과는 함초 ethylacetate추출물이 화장품 기능성 소재로서 응용 가능성을 나타내었다.

Increased Production of γ-Aminobutyric Acid from Brewer's Spent Grain through Bacillus Fermentation

  • Brewer's spent grain (BSG) is a waste product of the beer industry, and γ-aminobutyric acid (GABA) is a physiologically active substance important for brain and neuron physiology. In this study, we used the bacterial strains Bacillus velezensis DMB06 and B. licheniformis 0DA23-1, respectively, to ferment BSG and produce GABA. The GABA biosynthesis pathways were identified through genomic analysis of the genomes of both strains. We then inoculated the strains into BSG to determine changes in pH, acidity, reducing sugar content, amino-type nitrogen content, and GABA production, which was approximately doubled in BSG inoculated with Bacillus compared to that in uninoculated BSG; however, no significant difference was observed in GABA production between the two bacterial strains. These results provide the experimental basis for expanding the use of BSG by demonstrating the potential gain in increasing GABA production from a waste resource.

Immediate Decrease in γ-AminoButyric Acid after Caffeine Intake in Adolescents: a Preliminary MRS Study

  • In adolescents, sleep deprivation problem is getting worse, and increased caffeine consumption is considered to relieve the stress caused by sleep deprivation and academic burden. In this study, immediate neurologic effects of caffeine intake on adolescents were evaluated in three high school students using the ${\gamma}-aminobutyric$ acid (GABA)/creatine ratio on magnetic resonance spectroscopy (MRS). MEGA-PRESS MRS and TE 135 ms single voxel MRS were performed in the anterior cingulate cortex before and after drinking a cup of coffee, which contained 104 mg of caffeine. GABA and creatine were measured on LCModel 6.3, respectively. In all three students, GABA/creatine ratios were decreased after caffeine intake. The GABA/creatine ratios obtained before caffeine intake were decreased after caffeine intake in all the three adolescents. In this preliminary study, caffeine intake caused an immediate decrease in the GABA/creatine ratio in the brain and it may be related to the neurologic effects of caffeine on an adolescent's brain.

Development and Metabolite Profiling of Elephant Garlic Vinegar

  • Elephant garlic (Allium ampeloprasum var. ampeloprasum), which belongs to the Alliaceae family along with onion and garlic, has a flavor and shape similar to those of normal garlic but is not true garlic. Additionally, its properties are largely unknown, and its processing and product development have not been reported. In this study, we focused on using elephant garlic to produce a new type of vinegar, for which the market is rapidly growing because of its health benefits. First, we evaluated the effects of elephant garlic addition on acetic acid fermentation of rice wine by Acetobacter pasteurianus. In contrast to normal garlic, for which 2% (w/v) addition completely halted fermentation, addition of elephant garlic enabled slow but successful fermentation of ethanol to acetic acid. Metabolite analysis suggested that sulfur-containing volatile compounds were less abundant in elephant garlic than in normal garlic; these volatile compounds may be responsible for inhibiting acetic acid fermentation. After acetic acid fermentation, vinegar with elephant garlic did not have any sulfur-containing volatile compounds, which could positively contribute to the vinegar flavor. Moreover, the amino acid profile of the vinegar suggested that nutritional and sensory properties were more enhanced following addition of elephant garlic. Thus, elephant garlic may have applications in the development of a new vinegar product with improved flavor and quality and potential health benefits.

NMDA Receptor-dependent Inhibition of Synaptic Transmission by Acute Ethanol Treatment in Rat Corticostriatal Slices

  • The effects of ethanol on corticostriatal synaptic transmission were examined, using extracellular recording and analysis of population spike amplitudes in rat brain slices, to study how acute ethanol intoxication impairs striatal function. Ethanol caused a decrease in population spike amplitudes in a dose dependent manner ($50{\sim}200mM$). Pretreatment with picrotoxin, a ${\gamma}-amino$ butyric acid $(GABA)_{A}$ receptor antagonist, increased the population spikes but ethanol (100 mM) was still effective in decreasing the population spikes under this condition. In the presence of $_{(DL)}-2-amino-5-phosphonovaleric$ acid (APV), N-methyl-D-aspartate (NMDA) receptor antagonist, the inhibitory action of ethanol on population spikes was not shown. These results suggest that ethanol inhibits the glutamatergic corticostriatal synaptic transmission through blockade of NMDA receptors.

Methanol Extract of Zizyphi Spinosi Semen Augments Pentobarbital-Induced Sleep through the Modification of GABAergic Systems

  • Zizyphi Spinosi Semen (ZSS) have been widely used for the treatment of insomnia in Asia. This experiment was performed to investigate whether methanol extract of ZSS (MEZSS) has hypnotic effects through the ${\gamma}$-amino butyric acid (GABA)ergic systems. MEZSS inhibited the locomotor activity. MEZSS enhanced pentobarbital-induced sleep behaviors. However, MEZSS itself did not induce sleep at higher dose, similar to muscimol. On the other hand, both pentobarbital and MEZSS increased the non rapid eye move (NREM) sleep, especially reducing the -wave electroencephalogram (EEG) activity in REM sleep. MEZSS showed similar effects with muscimol on potentiating chloride influx induced by pentobarbital. MEZSS significantly increased GABAA receptors ${\gamma}$-subunit expression and slightly decreased ${\beta}$-subunit expression in hypothalamus and thalamus, showing that subunit-expression was similar to diazepam. In addition, MEZSS enhanced the expression of glutamic acid decarboxylase (GAD). In conclusion, it is suggested that MEZSS might augment pentobarbital-induced sleep behaviors through the modification of GABAergic systems.
