Successful Use of Bortezomib for Recurrent Progressive Familial Intrahepatic Cholestasis Type II After Liver Transplantation: A Pediatric Case with a 9-Year Follow-Up

  • Recurrence of progressive familial intrahepatic cholestasis (PFIC) type II poses challenges during postoperative liver transplant care. Posttransplant patients with PFIC type II risk developing recurrent cholestasis with normal gamma-glutamyl transferase activity, which mimics the original bile salt export pump (BSEP) protein deficiency and is related to a form of immunoglobulin G antibody (anti-BSEP)-mediated rejection. Bortezomib effectively induces apoptosis of actively antibody-producing plasma cells that may have a role in antibodymediated rejection. In this case, we used bortezomib to treat PFIC type II recurrence after liver transplantation in a child.

감마나이프의 물리적 특성 및 그의 임상적 적용 (Physical Aspect of The Gamma Knife and Its Clinical Application)

  • 정위 방사선 수술 전용의 감마나이프 (LGU-type B)가 아시아에서 처음으로 본원에 설치되었다. 인수 시험을 위하여 기계적 정밀도, 선량률, 각 콜리메이터 크기에 따른 선량 분포등의 특성을 조사하였다. 1990년 5월 7일부터 1990년 9월 10일까지 총 68명 09예의 환자에게 방사선수술을 시행하였다. 환자별로는 AVM 25예 $(35\%)$, Acoustic tumor가 10예 $(14\%)$, Pituitary adenoma가 4예 $(6\%)$ metastaic tumor가 7예$(10\%)$ meningioma가 6예$(9\%)$ 기타가 18예$(26\%)$등이었다. 질병 및 병소, 크기에 따라 $25\;Gy\sim100\;Gy$를 1회에 조사하였다.

Phosphoinositides Signaling and Epithelial-to-Mesenchymal Transition: Putative Topic for Basic Toxicological Research

  • Ptdlns(4,5)$P_2$ is a key cellular phosphoinositide that localizes in separate and distinctive pools in subcellular membrane and vesicular compartments. In membranes, Ptdlns(4,5)$P_2$ acts as a precursor to second messengers and is itself a main signaling and targeting molecule. Specific subcellular localization of type I PIP kinases directed by interacting with specific targeting module differentiates Ptdlns(4,5)$P_2$ production in a spatial and temporal manner. Several lines of evidences support the idea that Ptdlns(4,5)$P_2$ is generated in very specific pools in a spatial and temporal manner or by feeding Ptdlns(4,5)$P_2$ directly to effectors. In this concept, the interaction of PIPKI isoforms with a specific targeting module to allow precise subcellular targeting modulates highly specific Ptdlns(4,5)$P_2$ synthesis and channeling overall effectors. For instance, localization of PIPKI${\gamma}$661 to focal adhesions by an interaction with talin results in spatial and temporal production of Ptdlns(4,5)$P_2$, which regulates EGF-stimulated directional cell migration. In addition, Type $I{\gamma}$ PIPK is targeted to E-cadherin in cell adherence junction and plays a role in controlling dynamics of cell adherence junction and endocytosis of E-cadherin. Characterizing how PIP kinase isoforms are regulated by interactions with their targeting modules, as well as the mechanisms by which their product, Ptdlns(4,5)$P_2$, exerts its effects on cellular signaling processes, is crucial to understand the harmonized control of numerous cellular signaling pathways. Thus, in this review the roles of the Ptdlns(4)P(5) kinases and Ptdlns(4,5)$P_2$ were described and critically reviewed in terms of regulation of the E-cadherin trafficking, cell migration, and formation of cell adherence junction which is indispensable and is tightly controlled in epithelial-to-mesenchymal transition process.

감마형 삼수소 알루미늄 습식합성반응의 공정변수 연구 (Process variables of gamma-type aluminum trihydride in wet chemical synthesis)

  • Alane(aluminum trihydride, $AlH_3$)으로 명명되는 고에너지 물질인 삼수소알루미늄은 수소저장물질로서 뿐만 아니라 우주항공분야의 고체 추진제나 방위산업의 화약제조용으로도 사용될 수 있다. 본 연구는 습식공정을 통하여 합성하고, 에테르를 세밀하게 분리하는 결정화 공정을 통하여 최종 수소화물을 추출하였다. 결정화 공정에서 삼수소알루미늄-에테레이트($AlH_3{\cdot}(C_2H_5)_2O$)가 alane으로 상변이하면서 입자가 성장하고, $85^{\circ}C$에서 2 시간의 결정화 시간이 이루어졌을 때 가장 안정된 결정상이 나타나는 모습을 확인하였다. 최종적으로 추출된 고체상 삼수소알루미늄은 막대모양의 ${\gamma}$-형태가 가장 많은 양을 차지하는 것으로 나타났으며, 크기는 $50-100{\mu}m$ 수준이었다.

Immunomodulatory Effect of a New Herbal Preparation (HemoHIM) in Cyclophosphamide-treated Mice

  • The immunomodulatory effect of a new herbal preparation, HemoHIM, on the recovery from leukopenia induced by cyclophosphamide treatment was investigated. The HemoHIM was made up with an addition of the ethanol-insoluble fraction to the total water extract of Angelica Radix, Cnidii Rhizoma and Paeonia Radix. Daily oral administration of 100 mg/kg BW or 500 mg/kg BW HemoHIM accelerated the recovery from cyclophosphamide-induced leukopenia. HemoHIM increased the number of leukocytes and lymphocytes in the peripheral blood when compared with the cyclophosphamide-treated control. Moreover, the suppressed natural killer (NK) cell activity and interferon $(IFN)-{\gamma}$ secretion in the cyclophosphamide-treated mice were restored by the administration of HemoHIM. HemoHIM significantly reduced the abnormally heightened ratio of interleukin $(IL)-4/IFN-{\gamma}$ and immunoglobulin (Ig)E/IgG2a in the cyclophosphamide-treated mice. These results suggest that HemoHIM accelerates the recovery from leukopenia and alleviates the imbalanced T helper (Th)l/Th2 responses in the cyclophosphamide-treated mice. Additionally, HemoHIM was found to stimulate normal splenocytes to secrete not only Thl type cytokines such as $IFN-{\gamma}$ and IL-2, but also Th2 type cytokine IL-4. In conclusion, our results show that HemoHIM certainly has an influence on the balanced recovery of immune cells and the activation of their activities in the cyclophosphamide-treated mice.

Changes of Cytokine and Chemokine mRNA Expression in Whole Blood Cells from Active Pulmonary Tuberculosis Patients after T-Cell Mitogen and Mycobacterium tuberculosis Specific Antigen Stimulation

  • Tuberculosis (TB) is one of the major global health problems and it has been estimated that in 5~10% of Mycobacterium tuberculosis (MTB)-infected individuals, the infection progresses to an active disease. Numerous cytokines and chemokines regulate immunological responses at cellular level including stimulation and recruitment of wide range of cells in immunity and inflammation. In the present study, the mRNA expression levels of eight host immune markers containing of IFN-${\gamma}$, TNF-${\alpha}$, IL-2R, IL-4, IL-10, CXCL9, CXCL10, and CXCL11 in whole blood cells from active pulmonary TB patients were measured after T-cell mitogen (PHA) and MTB specific antigens (ESAT-6, CFP-10, and TB7.7). Among the TH1-type factors, IFN-${\gamma}$ mRNA expression was peaked at 4 h, TNF-${\alpha}$ and IL-2R mRNA expression was significantly high at the late time points (24 h) in active TB patients, TH2-type cytokine (IL4 and IL10) mRNA expression levels in both active TB and healthy controls samples did not changed significantly, and the mRNA expression of the three IFN-${\gamma}$-induced chemokines (CXCL9, CXCL10, and CXCL11) were peaked at the late time points (24 h) in active TB patients after MTB specific antigen stimulation. In conclusion, the mRNA expression patterns of the TB-related immune markers in response to the T-cell mitogen (PHA) differed from those in response to MTB specific antigens and these findings may helpful for understanding the relationship between MTB infection and host immune markers in a transcripts level.

Runaway Replication Plasmid를 이용한$\gamma$Phage $cI_{857}$ Repressor 단백질의 생산 및 정제 (Production and Purification of $\gamma$ Phage $cI_{857}$ Repressor Protein by the Use of a Runaway Replication Plasmid Vector)

  • Runaway replciation plasmid pSY35AT를 이용하여 이 plasmid가 가지고 있는 $cI_{857}$ repressor를 숙주 MC1065를 이용하여 $30^{\circ}C$에서 $37^{\circ}C$로 온도를 올려 shift-up 법으로 생산하였다. $cI_{857}$은 wild type cI repressor 정제법을 수정하여 사용하였으며 정제된 $cI_{857}$ repressor 농도는 0.11mg/ml이었다. 그리고 정제된 $cI_{857}$ repressor와 $^3H-CTP$로 labelinf한 PrOr과의 결합활성은 cell 파쇄액보다 약 23배 높았으며, 온도가 올라갈수록 결합활성은 떨어졌다.

접촉연소식 가스센서의 탄화수소계 가스 감응 특성 (Hydrocarbon Gas-sensing Properties of Catalytic Combustion Type Gas Sensor)

  • 비표면적이 약 $200\;m^2/g$${\gamma}-Al_2O_3$를 합성하여 얻은 ${\gamma}-Al_2O_3$ 담체에 Pd, Pt와 같은 귀금속 촉매를 첨가하여 접촉연소식 가스감지소자를 제작하였다. 제작된 센서는 최적동작을 위한 인가전압 1.75 V에서 소모전력은 약 500 mV였고, 부탄, 메탄 및 프로판 각각 100 %LEL에 대해서 약 120 mV의 높은 감도 특성을 보였다. 제작된 센서는 탄화수소 가스 농도에 대한 선형성, 재현성, 그리고 상대 습도변화에 대해서 안정된 감도 특성을 보여 주었다. 그리고 약 100일 동안 부탄 가스에 대해서 안정된 센서 감도 특성을 보여 주었다.

Effect of Ulmus davidiana Planch (Ulmaceae) on T-lymphocyte-producing cytokines such as IL-2, IL-6, and $IFN-{\gamma}$ production in collagen-induced arthritis of rats

  • Objective : The effect of Ulmus davidiana Planch(UD), which has long been known to have anti-inflammation and protective effects on damaged tissue, inflammation and bone among other functions, on the development of type II collagen (CII)-induced arthritis (CIA) in rats was studied. Methods : Male rats were immunized with an emulsion of $200\;{\mu}g$ of CII and complete Freund's adjuvant (CFA). The rats were then given intraperitoneal stimulation of Ulmus davidiana Planch herbal acupuncture(UDHA)or saline during the experiment. When compared with rats treated with saline as control, UDHA at doses of more than $20{\mu}g/100\;g$ rat once a day for 7 days inhibited the ability of inguinal lymph node cells to produce T cell cytokines interleukin-2, interleukin-6, $IFN-{\gamma}$ when the cells were obtained from rats 14 days after immunization and cultured in vitro with CII. Results : When rats were injected intraperitoneally, UD -treated group and control group rats did not differ significantly when low doses of UD was given to rats. Conclusion : The recommended dose of UD in the management and treatment of rat CIA will be more than $20{\mu}g/100\;g$, which is two-firth of human therapeutic dose. From the results, it was concluded that the effect of UDHA is dependent of dosage.

방사능 수치 오염 지도 작성을 위한 방사선 계측 시스템 연구 (Study of Radiation Mapping System for Water Contamination in Water System)

  • As nuclear industry has been developed, a various types of radiological contamination has occurred. After 9.11 terror in U.S.A., it has been concerned that terrorists' active area has been enlarged to use nuclear or radioactive substance. Recently, the most powerful earth-quake stroke, which triggered a massive tsunami in Japan and then Fukushima nuclear power plant reactor has suffered from a serious accident in history. The Fukushima reactor accident has occurred an anxiety of radiation leaks and about 170,000 people have been evacuated from the accidental area near the nuclear power plant. For these reasons, a social chaos can be occurred if radiological contamination occurs to the supply system for the drinking water. As such, the establishment of the radiation monitoring system for the city main water system is compelling for the national security. In this study, a feasibility test of radiation monitoring system which consists of unified hybrid-type radiation detectors was experimented for multi detection system by using gamma-ray imaging. The hybrid-type radiation sensors were fabricated with CsI(Tl) scintillators and photodiodes. A preamplifier and amplifier was also fabricated and assembled with the sensor in the shielding case. For the preliminary test of detection of radiological contamination in the river, multi CsI(Tl)-PIN photodiode radiation detectors and $^{137}Cs$ gamma-ray source were used. The DAQ was done by Linux based ROOT program and NI DAQ system with Labview program. The simulated contamination was assumed to be occurred at Gapcheon river in Daejeon city. Multi CsI(Tl)-PIN photodiode radiation detectors were positioned at the Gapcheon river side. Assuming that the radiological contaminations flows in the river the $^{137}Cs$ gamma-ray source has been moved and then, the contamination region was reconstructed.
