Isolation of Gamma-Induced Rice Mutants with Increased Tolerance to Salt by Anther Culture

  • Doubled haploids have long been recognized as a valuable tool in plant breeding since it not only offers the quickest method of advancing heterozygous breeding lines to homozygosity, but also increased the selection efficiency over conventional procedures due to better discrimination between genotypes within any one generation. Salt tolerant mutants were obtained in rice the variety, 'Hawsungbyeo', through in vitro mutagenesis of in vitro cultured anther-derived calli. Various doses (30, 50, 70 and 90 Gy) of gamma ray were applied to investigate the effect of radiation on callus formation on medium containing 1% NaCl, green plant regeneration, frequency of selected doubled haploid mutants and of the salt tolerant screen. It was demonstrated that the dose of 30 and 50 Gy gamma rays had significant effects on callus formation, regeneration and selection of salt tolerance. No tolerant lines were obtained from non-mutagenized cultures. From gamma ray irradiated cultures, five tolerant lines ($M_2$generation) at germination stage and 13 tolerant lines ($M_3$genoration) at seedling stage were obtained. The frequency of salt tolerant mutants indicates that anther culture applied in connection with gamma rays is an effective way to improve salt tolerance.

  • Various thermoluminescent properties of Korean natural quartz for possible use in ${\gamma}$-ray dosimetry has been studied. If the heating is exactly linear, ${\gamma}$-irradiated radiation sensitive (type 1) $\alpha$-quartz can yield a glow curve of single peak, hence glow peak height could be taken as a ${\gamma}$-dose for its dosimetry. Quartz crystal dosimeter exhibited the linearity of thermoluminescent intensity in the range from about 2$\times$10$^{3}$R to 2$\times$10$^{6}$ R, and also had an advantage of low fading because of the high peak temperature (300$\pm$4$0^{\circ}C$). The pulverized quartz sample having the grain size of 0.3<ø<0.9mm showed the linearity of T. L. intensity in the range from 50R to 2$\times$10$^3$R. Therapeutic application of the pulverized sample on the correct measurement of the absorbed dose in a body region of a cancer patient seems to be successful.

  • In this paper, It is studied that the variation of the dielectric absorption of the specimen according to the change with $^{60}$ Co-${\gamma}$ ray irradiation dose of the influence of temperature and applied voltage. In order to investigate the effect of irradiated oriented polypropylene film, we have observed dielectric properties within the temporature range of 30~130 ($^{\circ}C$) and voltage range of 100~250 (V). As for the dependency of temperature by tan $\delta$, the $\alpha$ peak which appears at high temperature increases accordingly to the increment of irradiation dose which is contributed by the crystal region and moves towards the high temperature. The $\beta$ peak which appears the orientation by dipoles and molecular motions in the amorphous region at low temperature. For the dependency of voltage by tan $\delta$ at low temperature, the peak of the tan $\delta$ shifts accordingly to the increment of irradiation dose towards the high temperature region.

High Dose $^{60}Co\;{\gamma}$-Ray Irradiation of W/GaN Schottky Diodes

  • W/n-GaN Schottky diodes were irradiated with $^{60}Co\;{\gamma}-rays$ to doses up to 315Mrad. The barrier height obtained from current-voltage (I-V) measurements showed minimal change from its estimated initial value of ${\sim}0.4eV$ over this dose range, though both forward and reverse I-V characteristics show evidence of defect center introduction at doses as low as 150 Mrad. Post irradiation annealing at $500^{\circ}C$ increased the reverse leakage current, suggesting migration and complexing of defects. The W/GaN interface is stable to high dose of ${\gamma}-rays$, but Au/Ti overlayers employed for reducing contact sheet resistance suffer from adhesion problems at the highest doses.

Natural Beryl as a Thermoluminescent Dosimeter

  • The possibility of using natural beryl thermoluminesence for gamma-ray dose measurement was investigated through the analysis of glow curves obtained with Co-60 gamma-ray irrediation. The natural beryl powder of 80-200 mesh has a good gamma-ray thermoluminescent response and stability at room temperature. The thermoluminescent response is linear from 10mR to 10$^3$R and can be measured up to 10$^{6}$ R.

  • Various mechanical test were conducted on conventional and crosslinked ultra-high molecular weight polyethylene(UHMWPE) all prepared from the same lot of medical grade GUR 1050 for artificial joint. The conventional materials were not irradiated and treated by heating. The cosslinked materials were irradiated with $25kGy{\sim}200kGy$ by gamma-ray andthen annealed or remelted. Gamma-ray irradiation and heat treatment process were found to significantly impact the crystallinity, and hence the mechanical behavior, of the highly crosslinked UHMWPE. The radiation dose and heating conditions were key predictors of the uniaxial yielding, plastic flow, and failure properties of conventional and highly crosslinked UHMWPE. The correlation model from experiments would be the basic information to enhance the were resistance of artificial joint liner.

  • This study was aimed to investigate the effect of oxidants on biodegradability and decomposition of acetaminophen (ACT) by gamma ray. Three kinds of chemical, potassium persulfate, hydrogen peroxide and ferrous sulfate were selected as an oxidant. The absorbed dose was ranged from 0.2 to 10 kGy and the concentration of oxidants was from 0.1 to 10 mM and the initial concentration of acetaminophen was $30mg\;l^{-1}$ in this study. The concentration of ACT was gradually decreased corresponding to the increase of the absorbed dose. However, mineralization of ACT was not occurred by the increased of the absorbed dose. When the 10 mM of oxidants applied to the ACT aqueous solution, the concentration of ACT was rapidly decreased according to absorbed dose and the mineralization was observed in potassium persulfate. Biodegradability of ACT with potassium persulfate was higher than that of ACT without potassium persulfate in lower absorbed dose and decreased according to higher absorbed dose.

  • Adaptive response induced by low dose ${\gamma}-ray$ irradiation in human peripheral lymphocytes was examimed. Human lymphocytes were exposured to low dose of ${\gamma}-ray$ (priming dose, 0.01Gy) followed by high dose (challenging dose, 1.5Gy) after various time intervals (4, 7, 20 hours). Frequencies of micronuclei were enumerated in both primed and unprimed groups. Maximum reduction in frequency of micronuclei was observed when challenging dose irradiation was followed by priming dose after 4hr incubation period. When challenging doses were irradiated 7 or 20hr after priming dose, frequencies of micronuclei were reduced slighty. However, these reduction were not statistically significant. In this study, human peripheral lymphocytes were irradiated at Go phase and they showed adaptive response induced by low dose radiation. Since micronucleus assay is relatively simpler and faster than other methods, it may be a good tool for evaluating radiation-induced adaptive responses.

  • The threshold voltage shift observed in TDE (Total Dose Experiment) on board the KITAT-1 is converted into dose (rad($SiO_2$)) using the result of laboratory calibration with Co-60 gamma ray source in KAERI (Korea Atomic Energy Research Institute). Simulation using the NASA radiation model of geomagnetosphere verifies that the dose difference between RADFET1 and RADFET3 observed on KITSAT-1 comes from the difference in shielding thickness at the position of these RADFETs.

  • Silver nano-particles (AgNPs) have attracted much attention for centuries due to their unique optical properties, electrical conductivities, oxidative catalysis, and antibacterial effect. In this study, AgNPs have been prepared by using aqueous $AgNO_3$ solution in the poly(vinyl alcohol) (PVA)/poly(ethylene glycol) (PEG) hydrogels. PVA and PEG powders were dissolved in deionized water, and then irradiated by a gamma-ray with a radiation dose of 50 kGy to make hydrogels. PVA/PEG hydrogels were dipped into $1.0{\times}10^{-2}M$ $AgNO_3$ solution for 1 hour. After that, the swollen hydrogels were irradiated by gamma-ray for the formation of AgNPs. FE-SEM is used to observe the formation of AgNPs as a function of the content of PEG and the irradiation dose. Also, AgNPs in the PVA/PEG hydrogels were monitored by UV-Vis. It is observed that the content of PEG and gamma-ray irradiation in the hydrogel is crucial to the formation of AgNPs. Finally, antibacterial tests indiacted that the hydrogel containing silver nanoparticle has antibacterial activity.
