  • Typhula incarnata grew over a temperature range of -5 to $20^{\circ}C$ with maximum growth at 10 to $15^{\circ}C$. Sclerotial production for T. incarnata was greatest at the higher temperature. Maximum mycelial growth of this pathogen occurred from pH 5.4 to 6.2. When carbon sources were added to a basal salt medium (Czapek's dox agar) at 5 g carbon sources/l, inulin, soluble starch, galactose, glucose, mannose, manitol, sucrose, maltose, cellobirose, trehalose, raffinose, and dextrin supported growth better than other carbon sources did. Of the twenty-three nitrogen sources tested, glycine, serine, ammonium sulfate, asparagine, asparatic acid, and ${\beta}-alanine$ were the most favorable for mycelial growth of T. incarnata. Cystine and cysteine were poor nitrogen sources. Ammonium salt of nitrogen sources supported growth better than nitrate salt of nitrogen sources. Potato dextrose agar, oat meal agar, and V-8 juice agar were the most favorable for mycelial growth and sclerotial formation. Appropriate addition of pepton to PDA decreased mycelial dry weight, but sucrose supported good growth of T. incarnata. Percent viable sclerotia of T. incarnate buried in bentgrass soil decreased from 2 months after treatment remarkably. Trichoderma riride and bacteria were isolated from non-germinated sclerotia. Live orchard grass leaf pieces within the soil were colonized by T. incarnata better than sterile and unsterile dead leaf pieces at $0^{\circ}C$. Saprophytic ability of T. incarnate on sterile leaf sheath occurred better at $0^{\circ}C$ than at $10^{\circ}C$. Saprophytic microflora consisting of Cladosporium sp., Fusarium sp., Mucor sp., Pythium sp., and unidentified fungi were the competitors for the sterilized and unsterilized substrate, but their colonization was not find on live leaf sheath buried in the soil at $0^{\circ}C$. In the effect of fungicides to Typhula snow mold disease of creeping bentgrass, mixture of polyoxin and thiram was the most effective, followed by iprodione, mixture of iprodione and oxine copper, thiophanate-methyl, myclobutanil, and tolclofos-methyl.

  • Several antagonistic bacteria, SD-1, 4, 10, 11, 14, 15, and 16, which have strong CMCase and amylase activities, were isolated from the fermented mushroom media. Among them, SD-1, 10, 11, and 15 have strong antibacterial activities against the mushroom pathogenic bacteria Pseudomonas sp., and SD-1, 10, 11, 14, and 16 have strong antifungal activities against the mushroom pathogenic fungi, Trichoderma sp. SD-14, 15, and 16 did not inhibit the growth of mushroom Pleurotus eryngii ASI-2302, and Pleurotus ostreatus ASI-2042 and ASI-2180. When the culture broth mixture of the seven bacterial strains was applied to the mushroom media, the growths of pathogens, Pseudomonas sp. and Trichoderma sp., were inhibited.

  • Neozygites fresenii (Zygomycetes: Entomophthorales), aphid-attacking fungus, was found in June 1998 for the first time in Korea. The fungus produces the globose primary conidia with a truncate papillar and two types of the secondary conidiophores and conidia. Resting spores were not found in our specimens, but the fungal structures observed clearly distinguish the fungus from other aphid-attacking fungi, allowing inclusion in the species N. fresenii.

  • In the course of screening for specific inhibitors against the mycelial form of Candida albicans from natural resources, we have isolated and identified A6792-1 from Streptomyces sp. A6792 by using several chromatographies. By spectral analyses, this compound was determined as 7-oxostaurosporine, having a structure of staurosporine aglycon noiety. 7-Oxostaurosporine exhibited a selective growth inhibitory activity against the mycelial form of Candida spp. up to $100\mu\textrm{g}/disc$ in bioassay. It also exhibited a specific antifungal activity against the mycelial form of Candida spp. including C. krusei, C. albicans, C. tropicalis, and C. lusitaniae with MICs ranging from 3.1 to $25\mu\textrm{g}/ml$ 7-Oxostaurosporine demonstrated no in vivo toxicity in SPF ICR mice. Therefore, this compound may have a considerable potential as an antifungal agent based on the preferential inhibition against growth of the mycelial form of Candida spp., dimorphic fungi.

  • The anaerobically digestion and agricultural application of organic wastes was conducted using food wastes and cow dung. Twenty kilograms each of the feed stocks was added into two 30 liters-capacity batch digesters. The anaerobic digestion was carried out within a temperature range of $25-31^{\circ}C$ for a retention time of 51 days. The results showed a cumulative gas yield of 5.0 bars for food waste and no gas production for cow dung within the retention time. Bacteria such as Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Proteus vulgaris and Clostridium sp were isolated. Fungi isolated included Aspergillus niger, Aspergillus nidulan, Trichophyton rubrum and Epidermophyton flocossum. The non-dispersive infrared (NDIR) analysis of the biogas produced confirmed that the gas consisted of $CH_4$, $CO_2$ and $H_2$. Statistical analysis revealed there was no significant correlation between temperature and biogas produced from the organic wastes (r= 0.177, p = 0.483).The organic wastes from the biogas production process stimulated maize growth when compared to control (soil without organic waste) and indicated maximum height. The study therefore reveals that food waste as potential substrates for biogas production has a moderate bio-fertilizer potential for improving plant growth and yield when added to soil.

  • The infection rate of soybean black root rot disease caused by Calonectria crotalariae was about $14\%$. The isolated fungi from the infected soybean roots and stems were Calonectria crotalariae, Fusarium solani, F. roseum, Phomopsis sojae, Pythium aphanidermatum, Rhizoctonia solani and Macrophomina sp. Among them, C. crotalariae was the most virulent pathogen under the laboratory conditions. Mycelial growth and microsclerotial formation were good on PSA containing 1000cc of water, 100g of potato and 20g of sugar. Mycelial growth, sporulation and microsclerotial formation were good on sterilized root. Perithecial formation was better in the dark condition than in the light. Survival of macroconidia was not available between $0\~25\%$ soil water content. Microsclerotia and mycelium in infected plant debris were survived for 4 months at to $8\%\;50\%$ soil water content. The plant height, when inoculated with $1.2\%$ inoculum density, reached approximately half of uninoculated plants. Disease severity was much higher at nonsterilized soil than completely sterilized soil. It was determined that the host range of this pathogen includes soybean, peanut, green bean and red bean.

  • Ginseng is major medicinal plant in Korea. Because of its long cultivation period the yield losses of 5 years of ginseng is 50% due to various diseases. The objective of this study is to select potential biocontrol agents. As the result of research so far achieved to contribute to rational prevention of ginseag plant disease for the stable cultivation of ginseng, three bacterial strains, Streptomyces lauretii strain B8180, Bacillus subtilis strain 8856, and Burkholderia cepacia strain 7944 were isolated from oak leaf compost. The strains showed antagonistic activities against five ginseng pathogenic fungi (Cylindrocarpon destructans, Rhizoctonia solani, Phytophthora cactorum, Botrytis cinerea, Fusarium solani f. sp. panacis) and control effects on Phytophthora blight.

  • Two peptides, MA-inv AND MA-ME, with the sequence 10-17 of maganin 2 at their-N-termini were designed and synthesized. The peptides had higher antifungal activities against Aspergilus fumigatus without hemolytic activities. The minimal inhibition concentratory (MIC) values of both peptides against A. fumigatus were 5 .mu.g/ml, whereas those of the native peptides, magainin 2 and melittin, were 10.mu.g/ml. At 3 .mu.g/ml, MA-inv and MA-ME inhibited the mycelium growth of A. fumigatus by 94.6% and 97.3% respectively, whereas magainin 2 and melittin inhibited by 62.2% and 32.4, respectively. MA-inv showed up to 80% inhibition of (1, 3)-.betha.-D-glucan synthase activity of A. fumigatus. The peptides also showed up to 80% inhibition of (1, 3)-.betha.-D glucan synthase activity of A. fumigatus. The peptides also showed antifungal activities for other fungi of Aspergillus sp. However, the antibiotic activities of MA-ME against Escherichia coli, Bacillus subtilis and Fusarium oxysporum were more effective than those of MA-inv, suggesting that the C-terminal sequences of MA-inv and MA-ME may also influence their antibiotic activities. These results suggest that the N-terminal sequence of the designed peptides, KKFGKAFV, is important for their antifungal activities against A. fumigatus and their C- terminal sequences are related to the organism selectivity.

  • The actinomycete strain PK-A41 was isolated from a soil sample from pepper fields in Ko-yang, Korea. The strain PK-A41 inhibited the mycelial growth of some plant pathogenic fungi and oomycete, Alternaria mali, Colletotrichum orbiculare, Fusarium oxysporum f.sp. lycopersici, Magnaporthe grisea, Rhizoctonia solani, and Phytophthora capsici. The presence of LL-diaminopi-melic acid in the cell wall extract and the nucleotide sequence of the 16S rDNA region of the strain PK-A41 was assigned to Streptomyces scabiei. Further morpho-logical, biochemical, and pathological analyses also confirmed the strain PK-A41 to be S. scabiei, which is pathogenic to potato tubers. The maximum antibiotic production of the strain PK-A41 was achieved when grown on the glycerol peptone broth (GPB) medium for 9 days.
