Comparison for enzymic activity of Nuruk and quality properties of Yakju by different fungi (곰팡이 균주에 따른 누룩의 효소활성 및 약주 품질특성 비교)

  • The enzymatic activity of Nuruk and the quality properties of Yakju were investigated according to different fungi. The fungi that were used in this study were Aspergillus kawachii KCCM 32819, Aspergillus niger KCCM 32005, Rhizopus japonicus KCCM 11604, Rhizopus oryzae KCCM 11272, Rhizopus oryzae KCCM 11273, Rhizopus oryzae KCCM 11276, and Mucor rouxii KCCM 60148. The study results are as follows. The saccharogenic power of Rhizopus oryzae KCCM 11272 Nuruk was the highest (3,647.72 SP/g) among all the samples. The ${\alpha}$-amylase production and protease activities were highest (3.76 DU and 4.7 tyrosine mg/min, respectively) in the Rhizopus japonicus KCCM 11604 Nuruk. The pH levels of the Yakju made with commercial Nuruk and Rhizopus japonicus KCCM 11604 Nuruk were 4.14 and 4.07, respectively. The total titratable acid content of the Yakju made with Rhizopus oryzae KCCM 11273 Nuruk was the highest (0.56%) among all the samples. Rhizopus japonicus KCCM 11604 and Rhizopus oryzae KCCM 11272 had the highest ethanol yields (15.18% and 15.10%, respectively). In the sensory evaluation carried out in this study, the panel preferred the Yakju made with Rhizopus japonicus KCCM 11604 Nuruk. Overall, however, the panel did not like the Yakju made with Aspergillus niger KCCM 32005 Nuruk.

  • An antifungal antibiotic was produced from Bacillus sp. YJ-63, and antibiotic was purified to a homogeneity by butanol extraction, Silica-gel column chromatography, Sephadex LH-20 gel filtration column chromatography and HPLC. The purified antibiotic showed one spot on Thin Layer Chromatography, which was negative to ninhydrin and positive to iodine. This finding indicated that the antibiotic was a cyclopeptide structure. UV absorption spectrum of the antibiotic in methanol was maximal at 277 nm, which corresponded to the spectrum for all of the Iturin antibiotic. The antibiotic was stable up to $121^{cdot}C$ and in the range of pH $3.O{\sim}l2.0$ with a stable of ${\alpha}-amino$ acid and ${beta}-amino$ acid, and showed high activity for fungi and yeasts,

A Study on Isolation of Penicillium expansum Link of Apple (한국산 Penicillium expansum Link의 분이에 관한 연구)

  • The isolations of fungi were observed, the isolations which were of rotting apples, pears, pine-seeds and dates cultivated in Korea, while they were living upon them. As a result of such observation. Penicillium sp. which rotted apples and pears could be isolated from the apples and pears. When the Penicillium sp. was inoculated into some fresh apples and pears, they were all rotted away. This fungus was identified as Penicillium expansum. Some fresh apples and pears inoculated with a mass of mycelium and conidia of P. janthinellun or some others inoculated with those of P. frequentans were not rotted. Though the P. expansum has been known to us a destructive not of pomaceous fruits, this fungus strain also caused the putrefaction of pears. It was found that P. expansum could be properly grown up at the temperature of $25^{\circ}C$ and pH 3~5 of media.

  • Antagonistic effect of bacterial strains isolated from phylloplane of strawberry plants grown In greenhouse was tested on Botrytis cinerea Among the promising bacterial strains, Bacillus sp. S1-0210 showed highest inhibition of mycelial growth of B. cinerea and a broad spectrum of antifungal activities against many plant pathogenic fungi in vitro. Bacillus sp. S1-0210 was identified as Bacillus subtilis based on the analysis of 185 rDNA as well as its biochemical characteristics. Application of wettable powder formulation of B. subtiiis S1-0210 significantly reduced the incidence of gray mold on trawberry fruits during storage. Results showed that treatment of B. subtilis S1-0210 decreased the incidence of gray mold by 4.8% whereas the incidence in control was 77.9%, indicating that the formulation of B. subtilis S1-0210 will be practically applied on strawberry fruits as a biocontrol agent of gray mold during storage.

  • About 1, 300 strains isolated from soil were evaluated for acyl-CoA:cholesterol acyltransferase (ACAT) inhibition activity. About 4.0% of actinomycetes and 3.6% of fungi showed greater than 50% inhibition activity, respectively. However, none of the isolated bacteria exhibited inhibition activity more than 50%. Among them, one Streptomyces sp. A-3 showed a higher ACAT inhibition activity in culture broth. Isolation of the ACAT inhibitor (AI-3) was achieved by Amberlite XAD-7 column, silica-gel column, Sephadex LH-20 gel-filtration and reverse phase HPLC.

A Novel Helicosporium Isolate and Its Antimicrobial and Cytotoxic Pigment

  • One Helicosporium strain, isolated from a wilted chestnut tree, evidenced in vitro antimicrobial activity against various types of bacteria and fungi, and generated a diffusible pigment. The antimicrobial compounds and the diffusible pigment of the Helicosporium sp. isolate were purified via solvent fractionation, column chromatography, and recycling preparative chromatography. Both the major antimicrobial compound and the diffusible pigment were identified as 2-methylresorcinol via nuclear magnetic resonance spectroscopy. Therefore, 2-methylresorcinol, a diffusible pigment generated by Helicosporium sp., appears to be an active antimicrobial principle. This pigment also exhibited considerable cytotoxicity against mammalian cells.

New Cyclic Peroxides from a Sponge, Plakortis sp.

  • Two new cyclic peroxides were isolated from an undescribed sponge of the genus Plakortis sp. collected at Discovery Bay, Jamaica. The molecular structures were elucidated by interpreting ID and 2D NMR and HRMS data. The cyclic peroxides, Compound 1 and 2, exhibited significant antimicrobial activity against pathogenic bacteria and fungi with IC$_${50} values of 0.9-5.0$\mug/mL and 0.7-8.0$\mug/mL, respectively.

  • Pitch canker of Pinus rigida, caused by Fusarium subglutinans f. sp. pini, was first noticed in Inchon, Korea, and is now being spread into other plantations of pines. Typical symptoms re resin flows from canders on the shoots, branches, and trunks, and resin-soaking of the sap wood under the bark. Pathogenic fungi were isolated from the infected shoots, branches, and sees on Fusarium-selective medium. Mycelial growth and microscopic characteristics were examined. Pathogenicity test was carried out by inoculating four common species of pines (P. rigida, P. densiflora, P. thunbergii, and P. koraiensis) in Korea. P. rigida and P. thunbergii showed symptoms identical to those of naturally infected trees, while P. densiflora and P. koraiensis remained free symptomless.

  • Bioactivity guided fractionation of the cultured filtrates of Aspergillus versicolor, which was derived from a marine sponge Petrosia sp., yielded three polyketides: decumbenones A (1),B (2), and versiol (3). These compounds were identified on the basis of 1D and 2D NMR spectroscopic and MS analysis. The absolute configuration was defined by the modified Mosher's method. The isolated compounds were tested for cytotoxicity against a panel of five human solid tumor cell lines and antibacterial activity against twenty clinically isolated methicillin-resistant strains. This is the first report on the isolation of these compounds from a marine source.

  • To confirm the antifungal activity to plant pathogenic fungi, Rhizoctonia solani, Rhizoctonia solani AG2-2, Fusarium oxysporium, Phytophthora dreschler, Alternaria sp. were selected. Helicosporium has the antifungal activity to Rhizoctonia solani, Rhizoctonia solani AG2-2, while Fusarium oxysporium, Phytophthora dreschler did not affected even though Alternaria made a feeble response to that antifungal compounds. From $^1H-NMR$ spectrum of antifungal compound, this compound was guessed to be a structure corresponding to cholesterol.
