Response of Mulberry Brown Leaf Spot Fungus Myrothecium roridum to Different Plant Extracts

  • Anti-fungal potential of 5 plant extracts viz., Eucalyptus citriodora, Allium sativum, Cassia sophera, Chromolaena odorata and Datura metel on the growth of mulberry brown leaf spot pathogen Myrothecium roridum were examined. Except fur the aqueous extract of Allium bulb, ethanolic leaf extract of all other plants more efficiently reduced the colony growth of the fungus on potato-dextrose-agar, Of which, Allium and Eucalyptus extracts were more effective. Initiation of radial growth of M. roridum on solid media was deferred maximum 6 days by ethanolic Eucalyptus extract and 4 days by aqueous Allium extract at $0.4 mg.ml^{-1}$. In the liquid media amended with Eucalyptus extract ($0.4 mg.ml^{-1}$) complete inhibition of sporulation was noticed upto 8 days, and initial inhibition of mycelial bio-mass generation was considerably diminished with time and reduction was 1.3 fold 14 days after application. While, complete inhibition of mycelial growth for 6-14 days was recorded with $\geq$0.1 mg.ml$^{-1}$ commercial eucalyptus oil. However, rejuvenation of growth appeared when fungus was re-inoculated in fresh media. Post-inoculate application of different doses Of Eucalyptus and Allium extracts significantly (p < 0.05) reduced the disease severity in pot-ted mulberry. However, persistence of the effect up to 28 days was apparent at $\geq$ 1.0 mg.ml$^{-1}$ and effectively was on par with carbendazim (1 mg.ml$^{-1}$ ). Almost equal control ability of 1.0 mg.ml$^{-1}$ Eucalyptus extracts can be achieved by ca. 10 times lowered dose of commercial eucalyptus oil. It seems, the toxic principle of E. citrodora to M. roridum is fungistatic in nature and may have essential oil based origin.

  • Amaranth(Amaranthus spp. L.) and quinoa (Chenpodium quinoa Willd.) are old crops from South, Central America and Central Asia and their grains have been identified as very promising food crops because of their exceptional nutritive value. Squalene is an important ingredient in skin cosmetics and computer disc lubricants as well as bioactive materials such as inhibition of fungal and mammalian sterol biosynthesis, antitumor, anticancer, and immunomodulation. Amaranth has a component called squalene (2,6,10,15,19,23-hexamethyl-2,6,10,14,22-tetraco-sahexaene) about 1/300 of the seed and $5\~8\%$ of its seed oil. Oil and squalene content in amaranth seed were different for the species investigated. Squalene content in seed oil also increased by $15.5\%$ due to puffing and from 6.96 to $8.01\%$ by refining and bleaching. Saponin concentrations in quinoa seed ranged 0.01 to $5.6\%$. Saponins are located in the outer layers of quinoa grain. These layers include the perianth, pericarp, a seed coat layer, and a cuticle like structure. Oleanane-type triterpenes saponins are of great interest because of their diverse pharmacological properties, for instance, anti-inflammatory, antibiotic, contraceptive, and cholesterol-lowering effects. It is known that quinoa contains a number of structurally diverse saponins including the aglycones, oleanolic acid, hederagenin, and phytolaccagenic acid, which are new potential in gredient for pharmacological properties. It is likely that these saponin levels will be considerably affected by genetic, agronomic and environmental factors as well as by processing. With the current enhanced public interest in health and nutrition amaranth and quinoa will most likely remain in the immediate future within the realm of exotic health foods until such time as agricultural production meets the quantities and qualify required by industrial food manufacturers.

Analysis of Bacterial Diversity and Community Structure in Forest Soils Contaminated with Fuel Hydrocarbon

  • Oil spill was found in 1999 from a diesel storage facility located near the top of Baekun Mountain in Uiwang City. Application of bioremediation techniques was very relevant in removing oil spills in this site, because the geological condition was not amenable for other onsite remediation techniques. For efficient bioremediation, bacterial communities of the contaminated site and the uncontaminated control site were compared using both molecular and cultivation techniques. Soil bacterial populations were observed to be stimulated to grow in the soils contaminated with diesel hydrocarbon, whereas fungal and actinomycetes populations were decreased by diesel contamination. Most of the dieseldegrading bacteria isolated from contaminated forest soils were strains of Pseudomonas, Ralstonia, and Rhodococcus species. Denaturing gradient gel electrophoresis (DGGE) analysis revealed that the profiles were different among the three contaminated sites, whereas those of the control sites were identical to each other. Analysis of 16S rDNA sequences of dominant isolates and clones showed that the bacterial community was less diverse in the oil-contaminated site than at the control site. Sequence analysis of the alkane hydroxylase genes cloned from soil microbial DNAs indicated that their diversity and distribution were different between the contaminated site and the control site. The results indicated that diesel contamination exerted a strong selection on the indigenous microbial community in the contaminated site, leading to predominance of well-adapted microorganisms in concurrence with decrease of microbial diversity.

Antimicrobial Activity of Some Essential Oils Against Microorganisms Deteriorating Fruit Juices

  • Seventeen microbial species including 10 fungal taxa, two yeasts and five bacteria, were isolated from freshly prepared orange, guava and banana juices kept in open bottles at room temperature for 7 days. Eight different essential oils, from local herbs, were tested for their antimicrobial activity against these test organisms. The essential oils of Cymbopogon citratus, Ocimum basilicum and Origanum majorana were found to be highly effective against these microorganisms. Aspergillus niger, A. flavus and Saccharomyces cerevisiae, the most prevalent microorganisms in juice, showed the highest resistance against these essential oils. GC-MS analysis showed that while e-citral, a'-myrcene, and z-citral represent the major components (75.1 %) of the essential oil of Cymbopogon citratus; bezynen,l-methyl-4-(2-propenyl), 1,8-cineole and trans-a'-bisabolene were the main components (90.6%) of Ocimum basilicum; whereas 3-cyclohexen-l-0l,4-methyl-l(l-methylethyl)-(CAS), c-terpinene and trans-caryophyllene represent the major components (65.1%) of Origanum majorana. These three essential oils were introduced into juices by two techniques namely, fumigation and direct contact. The former technique showed more fungicidal effect than the latter one against A. flavus, A. niger, and S. cerevisiae. The essential oil of Cymbopogon citratus by comparison to other test oils showed the strongest effect against these fungi with a minimum inhibitory concentration of $1.5\;{\mu}l/ml$ medium and a sublethal concentration of $1.0\;{\mu}l/ml$. The antimicrobial activity of this oil is thermostable at $121^{\circ}C$ for 30 min.

Suppression of melon powdery mildew and tomato leaf mold disease by the antifungal activity of tea tree (Melaleuca alternifolia) essential oil

  • Essential oils (EOs) have been shown to be plant-extracted antimicrobial agents. However, there are limited studies investigating the efficacy of EOs against pathogens. Among them, tea tree oil (TTO) is extracted from Melaleuca alternifolia, which is also used as an antifungal agent. In this study, the effect of TTO was investigated on the suppression of melon powdery mildew caused by Podosphaera xanthii and tomato leaf mold disease caused by Passalora fulva. Both powdery mildew and leaf mold diseases were significantly suppressed by a spray of TTO. Eighty percent of powdery mildew and 81% of leaf mold disease of the control value were suppressed by 0.5% TTO liquid, when sprayed 3 times every 7 days on the melon and tomato leaves. Inhibition of mycelial growth was also greatly affected by different concentrations of TTO against four different fungal pathogens. Ninety-eight percent of Pseudocercospora fuligena, 97% of P. fulva, 95% of Botrytis cinerea, and 94% of Phytophthora infestans mycelial growth were inhibited by 0.2% to 1.0% of TTO contained in plate media, respectively. However, phytotoxicity in plants by the TTO treatments was revealed when melon and tomato leaves were sprayed with a 1% and 2% concentration of TTO, respectively. Therefore, our findings show that TTO has high antifungal effects against various plant pathogens that occur during crop cultivation. We also suggest that when applying TTO to plant leaves, it is necessary to establish an accurate treatment concentration for different crops.

전나무 정유의 항진균 효과와 유효성분의 시너지효과 평가 (Evaluation on Anti-fungal Activity and Synergy Effects of Essential Oil and Their Constituents from Abies holophylla)

  • 본 연구는 전나무 정유의 Epidermophyton floccosum, Trichophyton mentagrophytes, Trichophyton rubrum과 같은 피부사상균에 대한 항진균 활성을 구명하고 피부염 치료제로서의 전나무 정유의 잠재력을 평가하고자 하였다. 전나무 정유와 분획물, 유효성분의 표준시약을 이용하여 한천희석법, paper disc 확산법, TEM을 이용한 세포 변화 관찰 등을 통해 항진균 활성을 평가하였고, 주성분은 GC-MS로 분석하였다. 유효성분들의 시너지효과는 체커보드법을 통해 평가하였다. 그 결과, 가장 높은 활성을 나타낸 물질은 ${\alpha}$-bisabolol을 함유하고 있는 분획 G4였으며 다른 분획 보다 뛰어나게 높은 활성을 나타냈다. 체커보드법을 통해 ${\alpha}$-bisabolol과 bornyl acetate를 병용시 뛰어난시너지 효과를 나타냈고 세포 변화 관찰을 통해 분획 G4에 노출된 E. floccosum과 T. rubrum 균사는 정상세포와 달리 세포막과 세포소기관이 파괴됨이 관찰되었다. 따라서 전나무 정유와 그 구성성분들은 높은 항진균 효과를 가지고 있기 때문에 피부염 치료제의 원료로 이용이 가능할 것이라 기대되며, 특히 ${\alpha}$-bisabolol의 우수한 항진균 활성에 주목할 필요가 있다고 사료된다.

  • The growing interest in the application of arachidonic acid (ARA) in various fields of health and dietary requirements has elicited much attention on the industrial production of ARA-containing oil by the cultivation of Mortierella fungi. For the industrial production of ARA, various studies, such as isolation of a high-potential strain and optimization of culture conditions, have been conducted. Studies including the investigation of morphology are important because ARA is accumulated in the mycelia, and thus cultivation with high biomass concentration is essential for obtaining a high ARA yield. Combining the results derived from various studies, a high ARA yield was attained in an industrial fermentor. These ARA production techniques are applicable to the production of other polyunsaturated fatty acids (PUFAs), and will contribute to the improvement of fermentation technology especially in the field of fungal cultivation.

  • Curcuma aeruginosa Roxb. (FamilyZingeberaceae), commonly known as 'pink and blue giner' is widely used in Indian system of medicine since time immemorial. The plant is found well in wild habitat and cultivated in southern India. The plant is distinguished by red corolla lobes and ferruginous or greenish-blue rhizome. Rhizoma is used medicinally as anti-diarrheal, anti-fungal; external use as astringent for wounds. A detailed pharmacognostic evaluation of its rhizome showed total ash 6.1%, acid insoluble ash 1.20%, alcohol soluble extractives 3.70% , water soluble extractives 14.50%, sugar 20.93%, starch 41.85%, and tannins 0.68%. Oh hydro distillation, the rhizomes and sessile tubers yielded 0.17% oil.

  • Cinnamaldehyde is the main component of cinnamon bark oil and show several biological activities such as anti-tumor, anti-fungal, anti-mutagenic and anti-inflammatory effects. A couple of studies have investigated how the natural compound exerts its anti-inflammatory effect. In despite of numerous investigations, the biological mechanism of effects belong to cinnamaldehyde remain unclear. We isolated 2-hydroxycinnamaldehyde(HCA) from the bark of Cinnamomun cassia Blume and reported a various of biological activities of HCA. (omitted)

오동나무상자의 항균활성 분석 및 활성 증진을 위한 천연 살생물제 적용연구 (Analysis on Antifungal Activity of Paulownia-Wood Storage Box and Application of Natural Biocide for the Activity Enhancement)

  • 고문서 보관을 위해 사용되고 있는 오동나무상자의 항균활성을 측정하기 위해 오동나무로부터 방출된 휘발성 물질에 대한 항균활성과 휘발성 유기물질의 주성분을 정성분석 하였다. 항균활성은 상자 내부에 TSA 배지를 장착한 미생물 포집기를 설치하고 포집 후 배지에 성장한 미생물의 집락수를 관찰하였다. 그 결과 실외에서 포집한 배지의 집락수 85개 대비 14.82%가 감소한 72개 집락수가 관찰되었다. 포집된 공기질을 열탈착시스템 및 가스크로마토그래피/질량분석법을 통해 분석한 결과, 오동나무 상자 내 VOCs에서 limonene이 특징적으로 검출되었다. Eugenol과 anethole을 주성분으로 하는 천연 살생물제 I과 II를 상자 내에 첨가하여 훈증에 의한 항균활성을 측정한 결과, 약제 모두 실외와 오동나무 상자 내부에서 포집된 미생물 개체수 대비 각각 92.6%, 99.9% 이상 감소율을 보였다. 위의 결과로부터 사용된 오동나무 상자로부터 방출된 휘발성 유기화합물에 의한 항균활성 및 그 유효성분은 분석되지 않았으며, 생물피해로부터 고서적을 장기보관하기 위한 유물 보관함의 효율을 증진시키기 위해 천연 살생물제의 적용 가능성을 확인할 수 있었다.
