Fungal Community Analyses of Endophytic Fungi from Two Oak Species, Quercus mongolica and Quercus serrata, in Korea

  • Fungal endophytes have been recorded in various plant species with a richness of diversity, and their presence plays an essential role in host plant protection against biotic and abiotic stresses. This study applied the Illumina MiSeq sequencing platform based on the amplification of fungal ribosomal ITS2 region to analyze fungal endophytic communities of two oak species (Quercus mongolica and Q. serrata) with different oak wilt disease susceptibilities in Korea. The results showed a total of 230,768 sequencing reads were obtained and clustered at a 97% similarity threshold into 709 operational taxonomic units (OTUs). The OTUs of Q. serrata were higher than that of Q. mongolica with the number of 617 OTUs and 512 OTUs, respectively. Shannon index also showed that Q. serrata had a significantly higher level of fungal diversity than Q. mongolica. Total of OTUs were assigned into 5 fungal phyla, 17 classes, 60 orders, 133 families, 195 genera, and 280 species. Ascomycota was the dominant phylum with 75.11% relative abundance, followed by Basidiomycota with 5.28%. Leptosillia, Aureobasidium and Acanthostigma were the most abundant genera detected in Q. serrata with the average relative abundance of 2.85, 2.76, and 2.19%, respectively. On the other hand, Peltaster, Cladosporium and Monochaetia were the most common genera detected in Q. mongolica with the average relative abundance of 4.83, 3.03, and 2.87%, respectively. Our results indicated that fungal endophytic communities were significantly different between two oak species and these differences could influence responses of host trees to oak wilt disease caused by Raffaelea quercus-mongolicae.

제주 수생식물에서 분리한 내생균류의 다양성 (Diversity of Endophytic Fungal Strains from Jeju Aquatic Plants)

  • 내생균류는 숙주식물 내에서 생물학적 그리고 비생물학적 스트레스로부터 저항성을 갖는데 도움을 준다. 수생식물은 수생환경에 서식하며 육상식물보다 수분스트레스에 더 많이 노출되어있다. 본 연구에서는 제주에 있는 남생이못, 수장동습지, 용수저수지, 강정천 4개의 습지에서 11개의 수생 수변식물을 채집하여 연구하였다. 전처리를 통해 식물 외생균을 제거하고 뿌리에서 내생균류를 분리하였다. 남생이못에서 126균주, 수장동습지에서 22균주, 용수저수지에서 44균주, 강정천에서 32균주가 분리되어 총 224개의 내생균류를 분리하였다. 분리된 균은 ITS를 이용해 동정한 후, 다양성 분석을 하였다. 남생이못에서 분리된 내생균류는 4개 문(Phylum), 7개 강(class), 12개 목(order), 19개 과(family), 30개 속(genus)로 구분되었다. 수장동습지에서 분리된 내생균류는 4개 문, 5개 강, 6개 목, 11개 과, 11개 속으로 구분되었으며, 용수저수지에서 분리된 내생균류는 4개 문, 5개 강, 7개 목, 12개 과, 13개 속으로 구분되었다. 강정천에서 분리된 내생균류는 1개 문, 2개 강, 5개 목, 7개 과, 9개 속으로 구분되었다. 4개의 습지에서 공통으로 분리된 속은 Alternaria속, Colletotrichum속, Fusarium속이었다. 향후 내생균류의 다양한 생태학적 분포와 다양성 규명에 대한 연구에 기초자료로 이용될 것이다.

Endophytic fungi harbored in Panax notoginseng: diversity and potential as biological control agents against host plant pathogens of root-rot disease

  • Background: Endophytic fungi play an important role in balancing the ecosystem and boosting host growth. In the present study, we investigated the endophytic fungal diversity of healthy Panax notoginseng and evaluated its potential antimicrobial activity against five major phytopathogens causing root-rot of P. notoginseng. Methods: A culture-dependent technique, combining morphological and molecular methods, was used to analyze endophytic fungal diversity. A double-layer agar technique was used to challenge the phytopathogens of P. notoginseng. Results: A total of 89 fungi were obtained from the roots, stems, leaves, and seeds of P. notoginseng, and 41 isolates representing different morphotypes were selected for taxonomic characterization. The fungal isolates belonged to Ascomycota (96.6%) and Zygomycota (3.4%). All isolates were classified to 23 genera and an unknown taxon belonging to Sordariomycetes. The number of isolates obtained from different tissues ranged from 12 to 42 for leaves and roots, respectively. The selected endophytic fungal isolates were challenged by the root-rot pathogens Alternaria panax, Fusarium oxysporum, Fusarium solani, Phoma herbarum, and Mycocentrospora acerina. Twenty-six of the 41 isolates (63.4%) exhibited activity against at least one of the pathogens tested. Conclusion: Our results suggested that P. notoginseng harbors diversified endophytic fungi that would provide a basis for the identification of new bioactive compounds, and for effective biocontrol of notoginseng root rot.

무제치늪에 자생하는 식물의 뿌리에서 분리한 내생진균의 군집분석 및 다양성 분석 (Community Analysis of Endophytic Fungal strains Isolated from the Roots of Plants Inhabiting Mujechi-neup)

  • 이 논문은 무제치늪에서 채집한 기장대풀(Isachne globosa Kuntze), 솔방울고랭이(Scirpus karuisawensis Makino), 이삭귀개(Utricularia racemosa Wall.), 좀개수염(Eriocaulon decemflorum Maxim.)등 4종의 식물 뿌리에서 서식하는 내생곰팡이의 분포 및 다양성을 확인하기 위해 조사하였다. 총 226 균주가 분리되었고, 3문(Phyla), 7강(Class), 10목(Order), 22과(Family), 31(genera)속으로 분리되었다. 내생진균에서 가장 많이 분리된 속은 Acephala (19.9%), Tolypocladium (16.3%), Neopestalotiopsis (11.5%), Perenniporia (7.1%) 순으로 확인하였다. 다양성지수 분석에 있어서, 이삭귀개(Ur)가 다른 식물 종에 비해 종 풍부도 (Menhinick's index = 2.37), (Margalef's index = 4.46)와, 종 균등도 (Simpson's index diversity = 0.91), 그리고 종 다양성(Shannon's indx = 2.57)로 높게 나왔다. 이러한 분석 결과를 봤을 때, 이삭귀개(Ur)에서 내생진균이 가장 다양하게 서식할 수 있는 것으로 보여진다.

Fungal Clusters and Their Uniqueness in Geographically Segregated Wetlands: A Step Forward to Marsh Conservation for a Wealth of Future Fungal Resources

  • Here, we investigated fungal microbiota in the understory root layer of representative well-conserved geographically segregated natural wetlands in the Korean Peninsula. We obtained 574,143 quality fungal sequences in total from soil samples in three wetlands, which were classified into 563 operational taxonomic units (OTU), 5 phyla, 84 genera. Soil texture, total nitrogen, organic carbon, pH, and electrical conductivity of soil were variable between geographical sites. We found significant differences in fungal phyla distribution and ratio, as well as genera variation and richness between the wetlands. Diversity was greater in the Jangdo islands wetland than in the other sites (Chao richness/Shannon/Simpson's for wetland of the Jangdo islands: 283/6.45/0.97 > wetland of the Mt. Gariwang primeval forest: 169/1.17/0.22 > wetland of the Hanbando geology: 145/4.85/0.91), and this variance corresponded to the confirmed number of fungal genera or OTUs (wetlands of Jangdo islands: 42/283> of Mt. Gariwang primeval forest: 32/169> of the Hanbando geology: 25/145). To assess the uniqueness of the understory root layer fungus taxa, we analyzed fungal genera distribution. We found that the percentage of fungal genera common to two or three wetland sites was relatively low at 32.3%, while fungal genera unique to each wetland site was 67.7% of the total number of identified fungal species. The Jangdo island wetland had higher fungal diversity than did the other sites and showed the highest level of uniqueness among fungal genera (Is. Jangdo wetland: 34.5% > wetland of Mt. Gariwang primeval forest: 28.6% > wetland of the Hanbando geology: 16.7%).

Nine New Records of Ascomycetes from Different Niches in Korea

  • We isolated nine fungal strains from different environmental materials collected from different locations during a survey of fungal diversity in Korea. Using molecular phylogenetic analyses and morphological characteristics, nine previously undescribed strains were identified and assigned to the species Collariella robusta, Fusicolla acetilerea, Hongkongmyces pedis, Hongkongmyces snookiorum, Mariannaea fusiformis, Metarhizium pemphigi, Pallidocercospora crystallina, Scopulariopsis candida, and Volutella citrinella. Diverse environmental samples may thus be a promising source for isolating and investigating novel fungal species, thus sampling efforts should be increased in future studies. This study also reports identification of some rare fungal species belonging to the genera Hongkongmyces and Pallidocercospora from Korea.

Endolichenic Fungal Community Analysis by Pure Culture Isolation and Metabarcoding: A Case Study of Parmotrema tinctorum

  • Lichen is a symbiotic mutualism of mycobiont and photobiont that harbors diverse organisms including endolichenic fungi (ELF). Despite the taxonomic and ecological significance of ELF, no comparative investigation of an ELF community involving isolation of a pure culture and high-throughput sequencing has been conducted. Thus, we analyzed the ELF community in Parmotrema tinctorum by culture and metabarcoding. Alpha diversity of the ELF community was notably greater in metabarcoding than in culture-based analysis. Taxonomic proportions of the ELF community estimated by metabarcoding and by culture analyses showed remarkable differences: Sordariomycetes was the most dominant fungal class in culture-based analysis, while Dothideomycetes was the most abundant in metabarcoding analysis. Thirty-seven operational taxonomic units (OTUs) were commonly observed by culture-and metabarcoding-based analyses but relative abundances differed: most of common OTUs were underrepresented in metabarcoding. The ELF community differed in lichen segments and thalli in metabarcoding analysis. Dissimilarity of ELF community intra lichen thallus increased with thallus segment distance; inter-thallus ELF community dissimilarity was significantly greater than intra-thallus ELF community dissimilarity. Finally, we tested how many fungal sequence reads would be needed to ELF diversity with relationship assays between numbers of lichen segments and saturation patterns of OTU richness and sample coverage. At least 6000 sequence reads per lichen thallus were sufficient for prediction of overall ELF community diversity and 50,000 reads per thallus were enough to observe rare taxa of ELF.

Guild Patterns of Basidiomycetes Community Associated With Quercus mongolica in Mt. Jeombong, Republic of Korea

  • Depending on the mode of nutrition exploitation, major fungal guilds are distinguished as ectomycorrhizal and saprotrophic fungi. It is generally known that diverse environmental factors influence fungal communities; however, it is unclear how fungal communities respond differently to environment factors depend on fungal guilds. In this study, we investigated basidiomycetes communities associated with Quercus mongolica using 454 pyrosequencing. We attempted to detect guild pattern (ectomycorrhizal or saprotrophic fungal communities) by comparing the influence of geography and source (root and surrounding soil). A total of 515 mOTUs were detected from root (321) and soil (394) of Q. mongolica at three sites of Mt. Jeombong in Inje County. We found that patterns of diversity and community structure were different depending on the guilds. In terms of alpha diversity, only ectomycorrhizal fungi showed significant differences between sources. In terms of community structure, however, geography significantly influenced the ectomycorrhizal community, while source appeared to have a greater influence on the saprotrophic community. Therefore, a guildbased view will help to elucidates novel features of the relationship between environmental factors and fungal communities.

Rhizospheric fungi of Panax notoginseng: diversity and antagonism to host phytopathogens

  • Background: Rhizospheric fungi play an essential role in the plantesoil ecosystem, affecting plant growth and health. In this study, we evaluated the fungal diversity in the rhizosphere soil of 2-yr-old healthy Panax notoginseng cultivated in Wenshan, China. Methods: Culture-independent Illumina MiSeq and culture-dependent techniques, combining molecular and morphological characteristics, were used to analyze the rhizospheric fungal diversity. A diffusion test was used to challenge the phytopathogens of P. notoginseng. Results: A total of 16,130 paired-end reads of the nuclear ribosomal internal transcribed spacer 2 were generated and clustered into 860 operational taxonomic units at 97% sequence similarity. All the operational taxonomic units were assigned to five phyla and 79 genera. Zygomycota (46.2%) and Ascomycota (37.8%) were the dominant taxa; Mortierella and unclassified Mortierellales accounted for a large proportion (44.9%) at genus level. The relative abundance of Fusarium and Phoma sequenceswas high, accounting for 12.9% and 5.5%, respectively. In total,113 fungal isolates were isolated from rhizosphere soil. They were assigned to five classes, eight orders (except for an Incertae sedis), 26 genera, and 43 species based on morphological characteristics and phylogenetic analysis of the internal transcribed spacer. Fusarium was the most isolated genus with six species (24 isolates, 21.2%). The abundance of Phoma was also relatively high (8.0%). Thirteen isolates displayed antimicrobial activity against at least one test fungus. Conclusion: Our results suggest that diverse fungi including potential pathogenic ones exist in the rhizosphere soil of 2-yr-old P. notoginseng and that antagonistic isolates may be useful for biological control of pathogens.

Diversity and Geographic Distribution of Anti-cancer Higher Fungi in Korea

  • Many higher fungi were collected at Korea from 1976 to 1998. They were identified and surveyed on resources with many reference books. According to the results, fungal fungi were 40 families, 90 general and 215 species. Among them , anti-cancer resources used in Korea were Ganoderma lucidum, Phellinus linteus, Agaricus brazei and Cordyceps militaris. Three species exception Agaricus brazei were distributed in Korea. All these are cultivated in Korea.
