PB-Overexpression of OsZn15, a CCCH-tandem zinc finger protein, increases drought tolerance in rice

  • Zinc finger proteins constitute a large family which has been studied to have various functions in different organisms. Tandem CCCH zinc finger proteins (TZFs), members of the zinc finger protein family, are known to participate as post-transcriptional regulators of gene expression in eukaryotes. Here, we showed that the OsZn15, a gene for tandem CCCH zinc finger protein, is induced by abiotic stress and its overexpression in transgenic rice plants (PGD1:OsZn15) gains higher drought tolerance. Gene expression analysis of promoter:GFP plants revealed that OsZn15 is specifically expressed in anther and embryo, but not in vegetative organs. In-field evaluation, grain yield was higher in the PGD1:OsZn15 than nontransgenic plants under drought conditions. Interestingly, OsZn15 is shown to not only localize at nucleus but also co-localize with both processing bodies (PB) and stress granules (SG), two messenger ribo-nucleoprotein complexes which are known to activate by forming cytoplasmic foci under stress conditions. In sum, these results suggest that OsZn15 increases drought stress tolerance of rice probably by participating in RNA turnover in PB and SG.

  • As many systems depend on electronics in an intelligent vehicle, concern for fault tolerance is growing rapidly. For example, a car with its braking controlled by electronics and no mechanical linkage from brake pedal to calipers of front tires(brake-by-wire system) should be fault tolerant because a failure can come without any warning and its effect is devastating. In general, fault tolerance is usually designed by placing redundant components that duplicate the functions of the original module. In this way a fault can be isolated, and safe operation is guaranteed by replacing the faulty module with its redundant and normal module within a predefined interval. In order to make in-vehicle network fault tolerant, this paper presents the concept and design methodology of an IEEE 1451 based dual CAN module. In addition, feasibility of the dual CAN network was evaluated by implementing the dual CAN module.

A Fault-Tolerant Scheme Based on Message Passing for Mission-Critical Computers (임무지향 컴퓨터를 위한 메시지패싱 고장감내 기법)

  • Fault tolerance is a crucial design for a mission-critical computer such as engagement control computer that has to maintain its operation for long mission time. In recent years, software fault-tolerant design is becoming important in terms of cost-effectiveness and high-efficiency. In this paper, we propose MPCMCC which is a model-based software component to implement fault tolerance in mission-critical computers. MPCMCC is a fault tolerance design that synchronizes shared data between two computers by using the one-way message-passing scheme which is easy to use and more stable than the shared memory scheme. In addition, MPCMCC can be easily reused for future work by employing the model based development methodology. We verified the functions of the software component and analyzed its performance in the simulation environment by using two mission-critical computers. The results show that MPCMCC is a suitable software component for fault tolerance in mission-critical computers.

A dominant negative OsKAT2 mutant delays light-induced stomatal opening and improves drought tolerance without yield penalty in rice

  • Stomata are the main gateways for water and air transport between leaves and the environment. Inward-rectifying potassium channels regulate photo-induced stomatal opening. Rice contains three inward rectifying shaker-like potassium channel proteins, OsKAT1, OsKAT2 and OsKAT3. Among these, only OsKAT2 is specifically expressed in guard cells. Here, we investigated the functions of OsKAT2 in stomatal regulation using three dominant negative mutant proteins, OsKAT2(T235R), OsKAT2(T285A) and OsKAT2(T285D), which are altered in amino acids in the channel pore and at a phosphorylation site. Yeast complementation and patch clamp assays showed that all three mutant proteins lost channel activity. However, among plants overexpressing these mutant proteins, only plants overexpressing OsKAT2(T235R) showed significantly less water loss than the control. Moreover, overexpression of this mutant protein led to delayed photo-induced stomatal opening and increased drought tolerance. Our results indicate that OsKAT2 is an inward-rectifying shaker-like potassium channel that mainly functions in stomatal opening. Interestingly, overexpression of OsKAT2(T235R) did not cause serious defects in growth or yield in rice, suggesting that OsKAT2 is a potential target for engineering plants with improved drought tolerance without yield penalty.

  • One of the challenges facing precision manufacturers is the increasing feature complexity of tight tolerance parts. All engineering drawings must account for the size, form, orientation, and location of all features to ensure manufacturability, measurability, and design intent. Geometric controls per ASME Y14.5 are typically applied to specify dimensional tolerances on engineering drawings and define size, form, orientation, and location of features. Many engineering drawings lack the necessary geometric dimensioning and tolerancing to allow for timely and accurate inspection and verification. Plus-minus tolerancing is typically ambiguous and requires extra time by engineering, programming, machining, and inspection functions to debate and agree on a single conclusion. Complex geometry can result in long inspection and verification times and put even the most sophisticated measurement equipment and processes to the test. In addition, design, manufacturing and quality engineers are often frustrated by communication errors over these features. However, an approach called profile tolerancing offers optimal definition of design intent by explicitly defining uniform boundaries around the physical geometry. It is an efficient and effective method for measurement and quality control. There are several advantages for product designers who use position and profile tolerancing instead of linear dimensioning. When design intent is conveyed unambiguously, manufacturers don't have to field multiple question from suppliers as they design and build a process for manufacturing and inspection. Profile tolerancing, when it is applied correctly, provides manufacturing and inspection functions with unambiguously defined tolerancing. Those data are manufacturable and measurable. Customers can see cost and lead time reductions with parts that consistently meet the design intent. Components can function properly-eliminating costly rework, redesign, and missed market opportunities. However a supplier that is poised to embrace profile tolerancing will no doubt run into resistance from those who would prefer the way things have always been done. It is not just internal naysayers, but also suppliers that might fight the change. In addition, the investment for suppliers can be steep in terms of training, equipment, and software.

Adaptive Intrusion Tolerance Model and Application for Distributed Security System (분산보안시스템을 위한 적응형 침입감내 모델 및 응용)

  • While security traditionally has been an important issue in information systems, the problem of the greatest concern today is related to the availability of information and continuity of services. Since people and organizations now rely on distributed systems in accessing and processing critical services and mission, the availability of information and continuity of services are becoming more important. Therefore the importance of implementing systems that continue to function in the presence of security breaches cannot be overemphasized. One of the solutions to provide the availability and continuity of information system applications is introducing an intrusion tolerance system. Security mechanism and adaptation mechanism can ensure intrusion tolerance by protecting the application from accidental or malicious changes to the system and by adapting the application to the changing conditions. In this paper we propose an intrusion tolerance model that improves the developmental structure while assuring security level. We also design and implement an adaptive intrusion tolerance system to verify the efficiency of our model by integrating proper functions extracted from CORBA security modules.

The Effect of Gamma Irradiation on the Acid Tolerance and Bile Tolerance of Lactic Acid Bacteria (감마선 조사가 젖산균의 내산성 및 내담즙성에 미치는 영향)

  • The acid and bile tolerance changes of 5 different lactic acid bacteria (LAB; Lactobacillus paracasei, Lactobacillus casei, Lactobacillus acidophilus, Lactobacillus plantarum, and Lactobacillus debruekii) with gamma irradiation were evaluated. The abilities of LAB to survive in the acidic conditions at the stomach and the bile acidic conditions at the beginning of the small intestine are the key functions for applying LAB to probiotics. In the results, all of LAB survived more than 50% after incubation in PBS (pH 2.5) for 2 hr, which indicated more than half of LAB are possible to pass through the stomach. However, gamma irradiation decreased the acid tolerances of LAB. The bile tolerances of all bacteria except Lactobacillus acidophilus were observed to survive at a 3% oxgall concentration in MRS, and 1 kGy of gamma irradiation to LAB did not affect any bile tolerances changes. But gamma irradiated Lactobacillus casei and Lactobacillus casei (3 kGy) showed decreasing survival rate with oxgall added MAS agar. In conclusion, gamma irradiation should be applied to yogurt or fermented foods with care because LAB could be changes their properties on acid and bile tolerances.

Reliability Analysis for Train Control System by Software Fault Tolerance Techniques (소프트웨어 결함허용 기법에 의한 열차제어시스템 신뢰도 분석)

  • PES (Programmable Electronic System) is used by software development for the train control system. PES has been widely used in real world and consists of hardware, firmware and application software. The PES are easily apply to many applications because its implementation has high flexibility. Many safety critical functions are realized through software in safety critical system. Normally, it is difficult to detect failures for PES system because the PES is too sophisticated to identify sources of the failure. So, the reliability analysis is needed by using software fault tolerance techniques. Currently, there are the recovery block, distributed recovery block, N-version programming, N self-checking programming in fault tolerance techniques. In this paper, the models of recovery block and N-version programming in software fault tolerance techniques are suggested by using the Markov model. Also, the reliability in the train control system is analyzed through changing time. The fault occupancy rates of the program, adjustment test and voter are stationary. So, the relation between time and reliability is presented by using Matlab program. In the result of reliability, the reliability of recovery block is more high than N-version programming in case of the same number of substitution block.

Developed 3-axis Educational CNC Machine Tool (3축 CNC 교육용 공작기계 개발)

  • In this study, we developed for processing complex features using CAM software that satisfies precision for example practice and related qualification tests suiTable for CNC training purposes. In addition, functions such as location control, speed control, and processing path generation, which are the main functions of CNC machining machines, were constructed using small equipment parts, servo motors, inverters, general purpose PCs, and commercial NC software and researched with the goal of developing low-cost education equipment. In the static accuracy inspection, the degree of machine when measuring the parallelism of the X, Y and Z axes and the vibration of the main shaft did not reach the allowable value. However, we have obtained a finished product that satisfies the CNC machine book sample shape machining, detailed functions of the position control function of the CNC machine tool, linear interpolation function, circular interpolation function, and tool offset function. In the qualification test shape processing, a shape with a degree of 1/100 mm was processed to obtain position accuracy that satisfied the tolerance.

New Weighting Functions for the Modified CIELAB Colour-Difference Formulae (수정 CIELAB 색차식을 위한 새로운 색차 가중 함수)

  • The lightness, chroma and hue tolerances with respect to the standard colour position in the CIELAB space have been studied in detail using the various existing data sets and the set form this study. The lightness tolerance showed a clear dependency upon the metric lightness for medium to light colour, but in the case of dark colours there was a discrepancy between the data sets. Both the chroma and hue tolerances showed dependency upon both the chroma and hue-angle and not the single dependency upon the metric chroma, as assumed in the CIE94 formula. New weighting functions were derived from the above experimental evidence, and finally a new formula, LCD(Leeds Colour Difference) was proposed. The LCD formula is nearly as simple and flexible as CIE94 but smoothes the individual weighting functions, especially for lightness tolerances for light colours and chromaticity discrimination near the blue region.
