Cardiovascular Biofunctional Activity and Antioxidant Activity of Gugija (Lycium chinensis Mill) Species and Its Hybrids (구기자 품종과 교배종의 부위별 심혈관관련 질환 예방 기능성 및 항산화 활성비교)

  • To develop new high valuable Gugija (Lycium chinensis), biofunctionalities of Gugija standard species and its hybrids were investigated and compared with each water extracts and methanol extracts from Lycii Fructus, Lycii Folium and buds and Lycii Cortex Radicis. Among various biofunctionalities of Gugija standard species, antioxidant activity was showed the highest in methanol extracts from buds of Cheongwoon species (93%) and antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity was 84.1% in the water extracts from Lycii Cortex Radicis of Cheongyang NO.7. Futhermore, methanol extracts from Lycii Cortex Radicis of Myungan A-2 hybrid showed 93.1% of antioxidant activity and 96.9% of ACE inhibitory activity was also showed in the methanol extracts from Lycii Fructus of DO148-72(A11) hybrid. However, fibrinolytic activity and anticholesteromia HMG-CoA reductase inhibitory activity were weak or not detected in almost of Gugija standard species and its hybrids. Therefore, we finally selected Cheongwoon Gugija standard species (buds) and Myungan A-2 hybrid (Lycii Cortex Radicis) as good antioxidant sources and also DO148-72 (A11) hybrid (Lycii Fructus) as excellent antihypertensive ACE inhibitior sources for manufacturing functional food product.

Protective Effect of Corni fructus Ethanol Extracts Against Environmental Hormones in Human Prostate Cancer Cells (인체 전립선암세포에서 산수유 에탄올 추출물의 환경호르몬에 대한 방어효과)

  • Anti-proliferation effects of Corni fructus ethanol extracts were investigated in the RC58T/h/SA#4 cells treated with environmental hormones including dioxin and bisphenol A. The proliferation was decreased at the concentration over $500{\mu}g/mL$ in the RC58T/h/SA#4 cells with ethanol extracts of various concentrations (1, 10, 100, 500, and $1000{\mu}g/mL$). The environmental hormones such as dioxin and bisphenol A increased the growth of RC58T/h/SA#4 cells in the charcoal-treated FBS (cFBS) medium. The proliferation was the highest at 1 nM and $0.1{\mu}M$ for the tested dioxin and bisphenol A concentration, respectively. Ethanol extracts showed inhibition of the proliferation in a dose-dependent manner at the tested concentrations (10, 100, 300, and $500{\mu}g/mL$) in the RC58T/h/SA#4 cells treated with the environmental hormones. The anti-proliferation was the highest at $500{\mu}g/mL$ concentration among the tested ethanol extracts.

The Effects of Chaenomelis fructus,(CMF) Extract on the Alzheimer's Disease Model Induced by CT-105 and ${\beta}A$ (목과(木瓜)가 CT105 와 ${\beta}A$로 유도(誘導)된 Alzheimer's Disease병태(病態) 모델에 미치는 영향(影響))

  • This research investigates the effect of the Chaenomelis fructus(CMF) on Alzheimer's disease. Specifically, the effects of the CMF extract on (1) >$IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ mRNA of PC-12 cells treated with LPS; (2) amyloid precursor proteins(APP), acetylcholinesterase(AChE), and glial fibrillary acidic protein(GFAP) mRNA of PC-12 cells treated with CT-105; (3) the AChE activity and the APP production of PC-12 cell treated with CT-105; (4) the behavior of AD mice with ${\beta}A$; (5) expression of $IL-1{\beta}$, $TNF-{\alpha}$, MDA, $IL-1{\beta}$ mRNA, $TNF-{\alpha}$ mRNA, and ROS; (6) the infarction area of the hippocampus, and brain tissue injury in Alzheimer's diseased mice induced with ${\beta}A$ were investigated. The results were summarized as follows; 1. The CMF extract suppressed the expression of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ mRNA in THP-1 cells treated with LPS. 2. The CMF extract suppressed the expression of APP, AChE, and GFAP mRNA in PC-12 cells treated with CT-105. 3. The CMF extract suppressed the AChE activity, and the production of APP significantly in PC-12 cells treated with CT-105. 4. A significant inhibitory effect on the memory deficit was shown on the CMF extract group of the mice with Alzheimer's disease induced by ${\beta}A$ in the Morris water maze experiment, which measured stop-through latency, and distance movement-through latency. 5. The CMF extract suppressed the over-expression of $IL-1{\beta}$ protein, $TNF-{\alpha}$ protein, MDA, $IL-1{\beta}$ protein, mRNA, $TNF-{\alpha}$ mRNA, CD68/GFAP, and ROS in the mice with Alzheimer's disease induced by ${\beta}A$. 6. The CMF extract reduced the infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer’s disease induced by ${\beta}A$. These results suggest that the CMF extract may be effective for the treatment of Alzheimer’s disease. Investigation into the clinical use of the CMF extract for Alzheimer's disease is suggested for future research.

Experiment Report about the Effect on the Anti-cancer of Herbal-acupuncture with Evodiae Fructus Infusion Solution (오수유(吳茱萸)약침의 항암효과에 대한 실험적 연구)

  • This study was done to investigate the effect of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Evodiae Fructus herbal infusion solution(EF-HAS). To study the effects of anti-cancer, anti-metastasis and immune response improvement of EF-HAS, we injected EF-HAS into Chung-wan(CV12) of C57BL/6 mice implanted intravenously with Bl6-Fl0 melanoma. Then, we have examined the effect on the increasing of spleen cells, the effect on the number of CD25$^+$/CD4$^+$, CD8$^+$/CD3e$^+$, CD69$^+$/B220$^+$, NK1.1$^+$/CD3e$^+$ cells in mice's PBMCs, the effect on the pulmonary colony number, and the effect on median survival time(MST) and increase of life span(ILS) of C57BL/6 mice implanted intravenously with Bl6-Fl0 melanoma. The following results were obtained; in the experiment groups treated with EF-HAS, the spleen cell proliferation in C57BL/6 mice, the percentage of CD25$^+$/CD4$^+$, CD8$^+$/CD3e$^+$, CD69$^+$/B220$^+$, NK1.1$^+$/CD3e$^+$ cells in C57BL/6 mice's PBMCs, and MST and ILS of C57BL/6 mice implanted intravenously with Bl6-Fl0 melanoma were significantly increased compared with that of the control group. And in the experiment groups treated with EF-HAS, the pulmonary colony number of C57BL/6 mice implanted intravenously with Bl6-Fl0 melanoma was decreased significantly compared with that of the control group. These results support a role for EF-HAS might De usefully applied in treatment of cancer.

Simultaneous determination and acute toxicity study of Fructus mume extracts in ICR mice (오매(烏梅)의 다성분동시분석 및 마우스를 이용한 경구 단회투여 급성독성시험)

  • Objectives : Previous studies have shown that Fructus mume (FM) has anti-platelet effects. The present study was performed to determine the acute oral toxicity and quality control of a crude extract of FM in ICR mice. Methods : We investigated the in vivo single dose acute toxicity of FM 95% ethanol extract. This test was orally administered once by gavage to 20 male and 20 female mice at dose levels of 0 (control group), 1250, 2500 and 5000mg/kg body weight, respectively. Mortalities, clinical findings, autopsy findings and body weight changes were monitored daily for the 14 days following the administration. HPLC analysis was performed for the simultaneous determination of ursolic acid and p-hydroxycinnamic acid in FM. Reverse-phase chromatography using a C18 column and photodiode array detection at 211 nm was used for quantification of the two maker components. The mobile phase for gradient elution consists of water and acetonitrile. Results : We observed survival rates, general toxicity, change of body weight, and autopsy. The mice did not die after single oral administration of maximum dose of FM. Autopsy of animal revealed no abnormal gross finding. Therefore, $LD_{50}$ value of FM for ICR mice was more than 5000mg/kg on oral route. The HPLC analysis showed that ursolic acid and p-hydroxycinnamic acid amounts to 9.75- and 0.12% in the extract with the retention times of 47.99- and 15.38 minutes, respectively. Conclusions : These results suggest that no toxic dose level of FM in mice is considered to be more than 5000mg/kg. Consequently, it was concluded that FM have no effect on acute toxicity and side effect in ICR mice. For the quality control of FM extract, simultaneous determination of ursolic acid and p-hydroxycinnamic acid was established.

Monitoring of Sulfur Dioxide Residues in Commercial Herbal Medicines at Domestic by Geographical Origins (원산지별 국내 유통 한약재의 이산화황 잔류실태 조사)

  • This study was carried out to investigate the residual sulfur dioxide residues ($SO_2$) in commercial medicinal herbs in Korea in 2013. Among a total of 116 samples of 11 different kinds of herbs, 71 samples (11 Kinds) were domestic, and 45 samples (10 Kinds) were imported. The residual contents of $SO_2$ in the samples were determinated by a modified Monier-Williams method. Of the 116 samples, 6 samples (5.2%, 4 Kinds) failed to meet the regulations for sulfur dioxide residues of KFDA in medicinal herbs. Among 6 unsuitable samples, 1 samples (1.4%) were domestic, and 5 samples (11.1%, 3 Kinds) were imported. The highest amount of sulfur dioxide residues was 1546.3 mg/kg (Lycii Fructus) in the domestic samples. Regardless of region, $SO_2$ contents were not found at Anemarrhenae Rhizoma, Citri Unshius Pericarpium, Eucommiae Cortex, Forsythiae Fructus, Gardeniae Fructus, Leonuri Herba and Lonicerae Folium et caulis. These data will be used as a basic data for the future legislation on the regulation and control of sulfur dioxide of herbal medicines.

Antioxidative Effect of Chaenomelis Fructus Ethanol Extract (모과 에탄올 추출물의 항산화 효과)

  • This study was performed to investigate the antioxidant effect of 80% (v/v) ethanol extract from Chaenomelis Fructus (CF). Total flavonoids and total polyphenols in the extract were also measured spectrophotometrically. The extraction yield was 9.23g/100g CF. The extract was further fractionated by partition with n-hexane, chloroform, ethylacetate, butanol, and water. The water fraction showed the highest extraction yield of all fractions. The n-hexane method and compared with the properties of the commerical antioxidant BHT. The activities of the n-hexane fraction were the highest of all fractions. In addition, there was strong positive correlation between antioxidant activities and levels of antioxidative compounds, such as flavonoid and polyphenols, in CF fractions, suggesting that these antioxidative compounds may contribute to the antioxidative effect of CF.

Effects of Chaenomelis Fructus Water Extract on the Quality Characteristics of Mul-kimchi during Fermentation (모과 추출액이 물김치의 품질 특성에 미치는 영향)

  • Mul-kimchi is more watery than traditional kimchi and is prepared using large amounts of salted water, Chinese cabbage, radishes, and carrots. The quality characteristics of Mul-kimchi prepared with Chaenomelis Fructus water extract (1, 3, or 5%, w/v) (CF Mul-kimchi) or water (control) were investigated during fermentation for 21 days at $10^{\circ}C$. The initial pH values were 6.53 (control), 4.14 (1% CF Mul-kimchi), 3.61 (3% CF Mul-kimchi), and 3.54 (5% CF Mul-kimchi). The pH did not change significantly in CF Mul-kimchi but gradually decreased in the control during fermentation. Changes in titratable acidity were reflected in pH movements. Viable lactic acid bacteria in CF Mul-kimchi were at lower levels than in the control. Viable bacterial levels in Mul-kimchi decreased with increasing concentration of CF water extract. Textural features, such as hardness, cohesiveness, chewiness, and springiness, were higher in CF Mul-kimchi than in control. Anti-oxidative activity, measured by DPPH radical scavenging and nitrite scavenging, of CF Mul-kimchi, were higher than in control, and the activities rose with increasing levels of CF water extract. The sensory qualities of 1% CF Mul-kimchi showed the highest values in taste and overall acceptability among the Mul-kimchi preparations tested.

Schisandrae Fructus ethanol extract attenuates particulate matter 2.5-induced inflammatory and oxidative responses by blocking the activation of the ROS-dependent NF-κB signaling pathway

  • BACKGROUND/OBJECTIVES: Schisandrae Fructus, the fruit of Schisandra chinensis Baill., has traditionally been used as a medicinal herb for the treatment of various diseases, and has proven its various pharmacological effects, including anti-inflammatory and antioxidant activities. In this study, we investigated the inhibitory effect of Schisandrae Fructus ethanol extract (SF) on inflammatory and oxidative stress in particulate matter 2.5 (PM2.5)-treated RAW 264.7 macrophages. MATERIALS/METHODS: To investigate the anti-inflammatory and antioxidant effects of SF in PM2.5-stimulated RAW 264.7 cells, the levels of pro-inflammatory mediator such as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines including interleukin (IL)-6 and IL-1β, and reactive oxygen species (ROS) were measured. To elucidate the mechanism underlying the effect of SF, the expression of genes involved in the generation of inflammatory factors was also investigated. We further evaluated the anti-inflammatory and antioxidant efficacy of SF against PM2.5 in the zebrafish model. RESULTS: The results indicated that SF treatment significantly inhibited the PM2.5-induced release of NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. SF also attenuated the PM2.5-induced expression of IL-6 and IL-1β, reducing their extracellular secretion. Moreover, SF suppressed the PM2.5-mediated translocation of nuclear factor-kappa B (NF-κB) from the cytosol into nuclei and the degradation of inhibitor IκB-α, indicating that SF exhibited anti-inflammatory effects by inhibiting the NF-κB signaling pathway. In addition, SF abolished PM2.5-induced generation of ROS, similar to the pretreatment of a ROS scavenger, but not by an inhibitor of NF-κB activity. Furthermore, SF showed strong protective effects against NO and ROS production in PM2.5-treated zebrafish larvae. CONCLUSIONS: Our findings suggest that SF exerts anti-inflammatory and antioxidant effects against PM2.5 through ROS-dependent down-regulating the NF-κB signaling pathway, and that SF can be a potential functional substance to prevent PM2.5-mediated inflammatory and oxidative damage.

Effect of Evodiae fructus Methanol Extract on Virulence-Related Genes' Expression of Helicobacter pylori (오수유 메탄올 추출물이 Helicobacter pylori 병원성 관련 유전자 발현에 미치는 영향)

  • Helicobacter pylori, a gram-negative bacterium, is one of the risk factors that induces gastritis and gastric cancer. Therefore, much attention has been paid to the compounds that inhibit bacterial growth or eradicate bacteria. Evodiae fructus (EF), the fruit of Evodia rutaecarpa, has been used for treating diarrhea and abdominal pain. EF extract was already found to inhibit the growth of H. pylori. However, to the best of our knowledge, the effect of EF on the virulence factors of H. pylori has not been reported. In this study, when comparing the minimum inhibitory concentration (MIC) of the different methanol concentration extracts, the 95% methanol extract (EF95) showed the lowest MIC value. EF95 extract suppressed the expressions of cagA, vacA and ureB, but interestingly, it up-regulated the expression of ureA. A decrease in production of ammonia in the culture medium and the cell lysates indicated that EF95 inhibited the urease activity in H. pylori, which was the result of EF95 inhibiting the ureB expression. Although the mechanism by which EF95 extract regulates the virulence factors in H. pylori needs further study, EF95 could be used for treatment of gastric troubles induced by H. pylori.
