저장온도 및 포장조건이 건멸치의 이화학적 품질에 미치는 영향 (Effects of Storage Temperature and Packaging Methods on the Physicochemicl Quality of Boiled-Dried Anchovies)

  • For improved preservation of boiled-dried anchovies, the current preservation method of corrugated-cardboard box packaging and freezing below -18$^{\circ}C$ was compared to the nylon/polyethylene(NY/PE) packaging along with cooling temperature ranging from 5$^{\circ}C$ to 1$0^{\circ}C$ as well as ambient condition for eight months by determining physicochemical quality of stored samples. Lipid oxidation of stored anchovies and their browning increased with storage time. The phenomenon was delayed under the conditions of lower temperature and air-tight packaging. As quality-indicative criteria of stored anchovies, carbonyl value(r=-0.989), browning (r=-0.949) and color a value(r=-0.989), browning(r=-0.965), Hunter's color b value(r=-0.949) and color a value(r=-0.940) showed a highly-negative correlations with organoleptic qualities of the samples. Based on the above results, air-tight packaging in a laminated film and subsequent storage at cooling temperature was found to be a possible alternative to the current freezing-storage of boiled-dried anchovies from the physicochemical point of view.

  • Salmonella typhi Ty21a is an attenuated strain of S. typhimurium and used for oral typhoid vaccine. In an attempt to increase the stability of Ty21a manufacturing typhoid vaccine, we studied about the stability of freeze-dried Th21a including additives at various temperature conditions. In order to investigate the freeze-drying rate of Ty21a according to various absorbance, we lyophilized Ty21a by using 8% sucrose as a stabilizer. The optimal freeze-drying rate of Ty21a was appeared when OD (optical density) value of the growth was between 2.5 and 3.0. To investigate the stability of Ty21a at various temperature, the viability was measured after storaging the freeze-dried Ty21a at the room temperature, cold and freezing condition for 1 week. The viability of Ty21a in cold and freezing storage condition was 5 times more stable than in room temperature. To search the most stable additives for the freeze-dried Ty21a, the viability of Ty21a including additives at the various storage condition was estimated. Mannitol and loctose were the most stable additives. Theses results suggest that the OD value of Ty21a growth, low temperature, mannitol and lactose are important factors for the optimal freeze-drying rate, the stable storage and the most stable additives, respectively.

Effects of High-Pressure, Microbial Transglutaminase and Glucono-δ-Lactone on the Aggregation Properties of Skim Milk

  • The object in this study is to investigate the effects of high pressure and freezing processes on the curdling of skim milk depending on the presence of transglutaminase (TGase) and glucono-δ-lactone (GdL). Skim milk was treated with atmospheric freezing (AF), high pressure (HP), pressure-shift freezing (PSF) and high pressure sub-zero temperature (HPST) processing conditions. After freezing and pressure processing, these processed milk samples were treated with curdling agents, such as TGase and GdL. Pressurized samples (HP, PSF and HPST) had lower lightness than that of the control. In particular, PSF had the lowest lightness (p<0.05). Likewise, the PSF proteins were the most insoluble regardless of whether they were activated by TGase and GdL, indicating the highest rate of protein aggregation (p<0.05). Furthermore, the TGase/GdL reaction resulted in thick bands corresponding to masses larger than 69 kDa, indicating curdling. Casein bands were the weakest in PSF-treated milk, revealing that casein was prone to protein aggregation. PSF also had the highest G' value among all treatments after activation by TGase, implying that PSF formed the hardest curd. However, adding GdL decreased the G' values of the samples except HPST-treated samples. Synthetically, the PSF process was advantageous for curdling of skim milk.

  • A sliding accident on the road have a high percentage by road freezing, especially, it is often appeared at bridges and Tunnel of freezing areas. Thus, the stability of road operations is enhanced by preventing a partial freezing phenomenon. According to the geothermal snow melting system analysis, a pattern of thermal conductivity is found out about pavement materials of concrete and asphalt when it is buried. The thermal conductivity study is essential that be applied the geothermal snow melting system according to heating exchanger pipe laying of lower pavements. The model tests are conducted on low temperature in freezer using the manufactured test model which is equal to pavement materials. And Many variables are discovered from numerical analyzes of the same conditions with model test.

Variation of Antifreeze Proteins during Cold Acclimation among Winter Cereals and Their Relationship with Freezing Resistance

  • Freezing-resistant plants can survive subzero temperatures by withstanding extracellular ice formation. During cold acclimation, their leaves accumulate antifreeze proteins (AFPs) that are secreted into the apoplast and have the ability to modify the normal growth of ice crystals. Three barley, two wheat and two rye cultivars were grown under two different temperature regimes (20/16$^{\circ}C$ and 5/2$^{\circ}C$, day/night). Apoplastic proteins from winter cereals were separated by SDS-PAGE and detected with antisera to AFPs from winter rye. Apoplastic proteins accumulated to much higher levels in cold-acclimated (CA) leaves compared with nonacclimated (NA) ones in winter cereals. After cold acclimation, the protein concentration of apoplastic extracts increased significantly from 0.088 $mgmL^{-1}$ to 0.448 $mgmL^{-1}$, with about 5-fold increment. Also, the apoplastic protein content per gram leaf fresh weight in CA leaves ranged from 31 $\mu\textrm{g}$ $(gFW)^{-1}$ to 120 $\mu\textrm{g}$ $(gFW)^{-1}$ with an averaged value of 77 $\mu\textrm{g}$ $(gFW)^{-1}$, and coefficients of variation of 54.9%. The CA leaves in Musketeer (a Canadian winter rye cultivar) showed the greatest AFPs and antifreeze activity followed by 'Geurumil' (a Korean winter wheat cultivar), and 'Dongbori l' (Korean facultative barley cultivar). The proteins secreted into the wheat leaf apoplast at CA condition were more numerous than those observed in winter rye, where two $\beta$-1,3-glucanase-like proteins (GLPs), two chitinase-like proteins (CLPs) and two thaumatin-like proteins (TLPs) accumulated during cold acclimation. The proteins in barley leaf apoplast at CA conditions were a little different from those in wheat leaves. The AFPs were various among and within species. More freezing-resistant cultivars had more clear and numerous bands than less freezing-resistant ones. The high determination coefficient ($R^2$ =91 %) between freezing resistance and AFPs per gram leaf fresh weight indicated that the amount of AFPs was highly related to freezing resistance in winter cereal crops.

동결토의 일축압축강도와 전단강도 상관관계 분석에 관한 연구 (Analysis of the Relationship between Unconfined Compression Strength and Shear Strength of Frozen Soils)

  • 동결토는 물과 얼음의 상변화에 따라 일반적인 흙과는 다른 거동을 보이며, 동결토의 강도는 온도, 함수비, 시간, 토질 특성 등 다양한 인자에 의해 영향을 받는다. 특히 지반이 동결되는 경우, 얼음과 흙 입자의 결합력에 의해 높은 강도 특성을 보이며, 이때의 전단강도는 흙의 구조적 결합에 영향을 미치는 얼음과 세립분의 함량에 따라 달라지며, 이로 인해 점착력이 동결토의 전단강도에 미치는 영향은 증가한다. 본 논문에서는 동결조건에서 직접전단시험과 일축압축강도시험을 실시하여 온도에 따른 흙의 강도 특성을 확인하였으며, 점착력과 일축압축강도와의 상관관계를 확인하였다.

냉동 온도에 따른 블루베리의 품질 특성 비교 (Effect of Freezing Temperature on Blueberry Quality)

  • 고품질의 냉동 블루베리 생산 유통을 위한 과학적 자료 확보를 위한 냉동 온도별 블루베리 품질 비교 연구를 수행하였다. 수확한 블루베리를 각각 $-20^{\circ}C$, $-45^{\circ}C$, $-70^{\circ}C$로 냉동 후 $-20^{\circ}C$에 2개월 보관한 다음 drip율, 산도, 고형분 함량, 색도, hardness, 유리당 함량, 미생물 오염 정도, 단면 미세구조 등을 관찰하여 냉동 및 해동 조건에 따른 블루베리의 품질 차이를 비교하였다. 냉동 블루베리의 pH와 유리당 함량은 냉동조건에 따른 차이가 거의 나타나지 않았다. 초기 $-20^{\circ}C$로 냉동된 블루베리의 drip율은 4.09%로 가장 많았으며 냉동 온도가 낮아질수록 drip율이 감소하는 경향을 보였다. $25^{\circ}C$로 해동한 경우 저온 해동 시료에 비하여 유의적으로 낮은 drip율을 나타냈다. 냉동 및 해동을 거친 후 블루베리의 hardness는 생과에 비해 절반가량으로 감소하였으며 냉동 온도에 따른 유의적인 차이는 나타나지 않았다. 생과 상태에 비해 냉동 후 해동된 블루베리의 미생물 수는 전체적으로 100배 이상 감소했으며, $4^{\circ}C$보다 $25^{\circ}C$에서 해동한 경우가 상대적으로 더 높은 미생물 정도를 보였다. 냉동된 블루베리의 단면 미세구조를 관찰한 결과, 전체적으로 냉동 온도 차이에 따른 뚜렷한 경향은 나타내지 않았다. 생산비용과 품질을 고려한 냉동 블루베리의 생산 및 유통을 위한 최적조건 설정을 위해서 추가적으로 관능평가 등의 연구가 필요할 것으로 사료된다.

유리화 및 완만동결법에 의한 토끼 전핵배의 동결보존 후 배발달율 (Post-thaw Development of Rabbit Zygotes Following Vitrification or Slow Freezing)

  • This study was carried out to investigate the effect of vitrification and slow freezing methods on the post-thaw developmental rate of rabbit zygotes. After exposing rabbit zygotes in EFS solution for 0.5, 1, 2, 3 and S min at room temperature, they were washed with 0.5 M sucrose solution, D-PBS and TCM-199 and then cultured in TCM-199 plus 10% FBS with bovine oviduct epithelial cells(BOEC) to examine whether the cryoprotectant induced injury during the various exposure periods. The embryo development rates to hatched blastocyst after exposing in EFS solution for 3 and 5 min(40.0 and 16.7%) were significantly lower than in 0.5, 1 and 2 min(63.0, 72.0 and 54.5%), respectively. The post-thaw development rates to hatched blastocyst were significantly(P<0.05) higher in in vivo morula with intact mucin coat(85.2%) and mucin seperated morula(77.8%) than those of in vitro morula(58.5%) and zygote(5.9%), hut no difference was shown between in vitro morulae and mucin separated morula. The cryoprotectant dilution procedures showed no effects on the post-thaw development rates to hatched blastocyst under the present culture conditions. The post-thaw development to hatched blastocyst in the rabbit zygotes was not significantly different between the slow freezing(12.8%) and vitrification(5.9%). These results indicated that the rabbit frozen zygotes could he successfully developed in vitro to hatched blastocysts, though their developmental rate was very low, compared with morula stage embryos, in either vitrification or slow freezing procedure under the present conditions.

동결온도와 수직구속응력 변화에 따른 모래와 알루미늄 재료의 접촉면에서 작용하는 동착강도 실험 연구 (Experimental Study on Adfreeze Bond Strength Between Frozen Sand and Aluminium with Varying Freezing Temperature and Vertical Confining Pressure)

  • 동토지역 말뚝기초의 지지력은 말뚝과 토사의 접촉면에서 작용하는 동착강도에 지배된다. 말뚝주변 토사 내 간극수의 동결로 인해 발현되는 동착강도는 동토지반 기초설계에 있어 가장 주요한 설계정수로 고려되고 있다. 지난 50년간 동착강도에 대한 연구가 다각도로 수행되어 왔으나, 대부분 동결온도와 지중온도를 고정조건으로 그 영향력을 고려하지 않은 채 토사종류, 말뚝종류, 재하속도 등의 영향인자를 분석하기 위한 목적으로 수행되었다. 본 연구에서는 동결온도와 마찰면에 작용하는 수직구속응력을 주요 변수로 적용하고, 토사종류, 말뚝종류, 재하속도 등은 고정조건으로 적용하여 직접전단방식의 동착강도 측정실험을 수행하였다. 실험재료로는 표면 가공이 용이하여 거칠기를 정밀하게 조절할 수 있는 알루미늄 모형과 주문진표준사를 활용하였다. 실험은 상온(> $0^{\circ}C$), $-1^{\circ}C$, $-2^{\circ}C$, $-5^{\circ}C$, $-10^{\circ}C$의 동결온도및 1atm, 2atm, 3atm의 수직구속응력 조건에서 수행되었으며, 그 결과를 바탕으로 동결온도와 수직구속응력이 동착강도에 미치는 영향을 분석하였다. 전반적으로 동착강도는 동결온도가 낮아질수록, 혹은 수직구속응력이 커질수록 증가하는 경향을 보였으며, 특히 단위온도차에 따른 동착강도의 증가율이 1)급증하는 구간과 2)점진적으로 감소하는 구간을 뚜렷하게 나타내며 변화하는 특성을 보였다. 또한, 동결온도의 저하에 따라 동착강도의 변화를 지배하는 요소가 마찰각에서 부착력으로 변화하며 수렴구간을 형성하는 경향을 나타냈다.

저 에너지형 축냉식 저온유통 시스템 개발 (Development of Cold Chain System Using Thermal Storage with Low-Energy Type)

  • The purpose of this study is to find the optimal conditions of PCM slurry manufacturing equipment for saving the marketing cost and keeping the original quality of products. In addition, the characteristics of the movable container for shipping or distributing products is analysed. The major results are as follows. 1. PCM thermal storage system is designed with the conditions of temperature($-5{\sim}10^{\circ}C$), cold chain time(30 minutes), and one time usage(50 liter). This system includes tank, freezer, circulating pump, cycle type heat exchanger, swelling tank, equipment of supplying PCM supplying unit includes cold tank, cycle type heat exchanger, suction unit and control equipments, etc. 2. After ability test of PCM thermal storage system, it shows that the required freezing time of PCM thermal storage system is less than one of the previous system. The reason is that churn (top and bottom) and compulsion circulation are occurred simultaneously and unit cooler type method is better than chiller type method. 3. By the experiment of transportation latent heat container, it is decided that the best container is $K_1$ with latent heat temperature($0{\sim}5^{\circ}C$) and density(0.15%). However, for $K_l\;and\;K_2$, it is necessary more studies on latent heat thermal conditions and conditions of making method.
