Effects of Glycerol Concentration on Viability of Frozen-thawed Canine Spermatozoa

  • Glycerol is the cryoprotectant most frequently used to freeze semen in several of species. The objective of the present study was to compare the effect of three different glycerol concentrations (4, 6 or 8%, v/v) on frozen-thawed dog sperm survival rate. Ejaculates from 9 dogs collected by digital manipulation were pooled and assessed by macroscopic and microscopic criteria. Semen was divided into 3 aliquots, which were centrifuged and the sperm pellets rediluted with first Tris-glucose-citric acid extender. After 1 h cooling at $4^{\circ}C$, second extender containing 4, 6 or 8% glycerol was added, respectively. The semen was loaded into 0.25 ml straws and frozen and stored in liquid nitrogen and thawed. Sperm vigor, live:dead spermatozoa ratio using HOS test, and sperm morphology using $Spermac^{(R)}$ stain were evaluated. After thawing, there were no significant differences among groups in vigor, viability and morphology. In conclusion, the three glycerol concentrations (4, 6 or 8%) can be used successfully in cryopreservation of canine semen. Therefore the use of 4% glycerol in the extender has less toxic effect and reduces of freezing injuries.

Effects of Antioxidants Supplement in Porcine Sperm Freezing on in vitro Fertilization and the Glutathione and Reactive Oxygen Species Level of Presumptive Zygotes

  • The present study was aimed to determine the effects of green tea extract (GTE) and beta-mercaptoethanol (${\beta}-ME$) supplementation in boar sperm freezing extender on in vitro fertilization (IVF) and reactive oxygen species (ROS) and glutathione (GSH) levels of presumptive zygotes (PZs). Experimental groups were allocated into lactose egg yolk (LEY) without antioxidant (control), GTE (1,000 mg/l in LEY) and ${\beta}-ME$ ($50{\mu}M$ in LEY). In freezing, spermatozoa extended with LEY were cooled to $5^{\circ}C$ for 3 h and then kept at $5^{\circ}C$ for 30 min following dilution with LEY containing 9% glycerol and 1.5% Equex STM. The final sperm concentration was $1{\times}10^8/ml$. Spermatozoa were loaded into straws and frozen in nitrogen vapor for 20 min. For IVF, oocytes were matured in NCSU-23 medium and co-cultured with spermatozoa following thawing at $37^{\circ}C$ for 25 sec. At 12 h following IVF, IVF parameters (sperm penetration and monospermy) were evaluated. In addition, GSH and ROS levels of PZs were determined by Cell Tracker Blue CMF2HC and DCHFDA, respectively. IVF parameters did not show any significant difference among the experimental groups. GSH and ROS levels of PZs were not significantly different between groups. In conclusion, antioxidant supplementation in boar sperm freezing could not influence IVF parameters, ROS and GSH levels of PZs.

진도개 정액 동결 시 단당류 및 동결 속도가 생존율에 미치는 영향 (Factors Affecting the Survival Rates of Frozen-Thawed Spermatozoa in Jindo Dog by Monosaccharides and Freezing Rate)

  • This studies were conducted to investigate the survival rate of frozen-thawed spermatozoa of Jindo Dog by monosaccharide and freezing rates. Experimental animals were prepared 12 males within 1~8 year's old and collected once in a couple of weeks by digital manuplation methods. Collected semen was diluted 1:1 with Tris-egg yolk extender and added 4, 6 or 8% of glycerol and none, 4 mM glucose or 4 mM fructose as cryoprotectant and was equilibrated for 2 hrs in $4^{\circ}C$. In monosaccharide groups, the freezing rate was 5 cm-5 min. above $LN_2$. The survival rates without monosaccharide were $50.7{\pm}19.0%$, $58.6{\pm}18.0%$, $40.0{\pm}10.0%$ in 4, 6 or 8% glycerol, respectively. In addition of glucose, the survival rates were $43.1{\pm}14.7%$, $38.1{\pm}16.5%$, $33.3{\pm}4.0%$ in 4, 6 or 8% glycerol, respectively and in fructose, were $47.9{\pm}21.1%$, $61.3{\pm}6.2%$, $34.3{\pm}12.6%$ in 4, 6 or 8% glycerol, respectively. There showed significantly different between glycerol groups and monosaccharides groups (p<0.05). The survival rates of freezing rate in 5 cm-5 min. group was $64.5{\pm}15.8%$, $51.9{\pm}27.6%$, $29.7{\pm}24.8%$ and in 10 cm-10 min. group was $62.5{\pm}20.3%$, $64.9{\pm}23.6%$, $34.5{\pm}27.4%$ in 4, 6 or 8% glycerol, respectively. There were significantly different between freezing rates (p<0.05). These results suggest that the addition of fructose with 6%-glycerol and slow freezing improve the survival of frozen-thawed sperm in Jindo Dog.

Effect of Cholesterol-loaded-cyclodextrin in Presence and Absence of Egg Yolk during Freezing Step on Quality of Markhoz Buck's Spermatozoa

  • Cryopreservation protocols induce partially irreversible damage to mammalian sperm plasma membranes. Previous studies have indicated that adding cholesterol to the plasma membrane, as cholesterol-loaded-cyclodextrins, improves cryosurvival of sperm. Therefore, the purpose of this study was to determine if treating sperm of Markhoz bucks with cholesterol-loaded-cyclodextrins (CLC) (0, 0.75, 1.5, 2.25 and 3 mg/ml diluted $240{\times}10^6$ sperm/ml) in Tris-citric acid-glucose diluents with and without egg yolk (containing 5% glycerol) would improve the post-thaw sperm quality. The motion characteristics were evaluated with a Computer Assisted System Analyzer (CASA); acrosome integrity and vitality were measured with the triple-stain technique. Samples were recovered before and after freezing by means of putting straws into $37^{\circ}C$ water for 30 sec and then parameters were assessed. The results showed that the treatments significantly affected motility, progressive motility, recovery rate, curvilinear velocity, beat cross frequency, live sperm with reacted acrosome, live sperm with unreacted acrosome, dead sperm with reacted acrosorne, and dead sperm with unreacted acrosome during freezing (p<0.05). However; no significant differences were found for average path velocity, straight line velocity, amplitude of lateral head displacement, straightness and linearity (p>0.05). The best results were observed for extender containing 2.25 mg/ml ($240{\times}10^6$ sperm/ml) CLC supplemented with 2.6% egg yolk. In conclusion, the findings of this study indicate improved Markhoz sperm viability and motility following treatment in the presence of egg yolk.

Spermatozoa motility, viability, acrosome integrity, mitochondrial membrane potential and plasma membrane integrity in 0.25 mL and 0.5 mL straw after frozen-thawing in Hanwoo bull

  • In the present study, we examined the effect of straw size on spermatozoa motility, viability, acrosome integrity, mitochondrial membrane potential, and plasma membrane integrity after freezing-thawing. Hanwoo semen was collected from three bulls and diluted with an animal protein-free extender, divided into two groups, namely, 10 million spermatozoa in 0.25 mL and 20 million spermatozoa in 0.5 mL straw, and cryopreserved. In Experiment 1, the motility and motility parameters of the frozen-thawed spermatozoa were evaluated. After freezing-thawing, the spermatozoa motility parameters fast progressive, straight line velocity, and average path velocity were compared between the 0.25 mL straw and 0.5 mL straw groups. They were 35.2 ± 1.0 and 32.3 ± 0.7%, 34.6 ± 0.7 and 31.8 ± 0.5 μm/s, 51.4 ± 1.3 and 47.1 ± 1.1 μm/s, 0.25 mL straw and 0.5 mL straw groups, respectively. In Experiment 2, the viability, acrosome membrane integrity, and mitochondrial membrane potential of the frozen-thawed spermatozoa were assessed. After freezing-thawing, the percentages of spermatozoa with live, intact acrosomes and high mitochondrial membrane potential were compared between the in 0.25 mL straw and 0.5 mL straw groups. They were 48.0 ± 2.6% and 35.6 ± 2.8% between the 0.25 mL straw and 0.5 mL straw groups. In Experiment 3, the plasma membrane integrity of frozen-thawed spermatozoa was compared. After freezing-thawing, the plasma membrane integrity was higher for the in 0.25 mL straw group than the 0.5 mL straw group. They were 62.0 ± 2.2 and 54.1 ± 1.3% between the 0.25 mL straw and 0.5 mL straw groups. In conclusion, our results suggest that freezing semen in 0.25 mL straw improves the relative motility, viability, and acrosomal, mitochondrial membrane potential, and plasma membrane integrity of Hanwoo bull spermatozoa.

  • This experiment was carried out with the purpose to get some information about the properties of Simmental bull semen during summer season. The results obtained were summarized as follows: 1. Semen volume per ejaculation, sperm concentration and sperm viability were averaged 5.16ml, 6.6billion and 65%, respectively. 2. Percentage of motile sperm after dilution in skmmilk solution and trisbuffer for 5 days were 34.16% and 35.0%, respectively. 3. Viability of spermatozoa frozen in skimmilk extender and trisbuffer for 5 days were 20. 83% and 25.66%, respectively. 4. Percentage of live sperm, MRT and pH value were 71.8∼72.1%, 8.40∼8.21 minutes and 6.78, respectively. 5. Diluted semen showed strong resistance to the cold shock than that of fresh semen. 6. Rscovery of sperm motility after freezing for 24 hours was relatively weak.

Effects of Dimethyl-sulfoxide on Sperm Cryopreservation of Grass Carp (Ctenopharyngodon idellus)

  • This study was conducted to investigate the effects of three different concentrations (6%, 8% and 10% final volume) of dimethyl sulfoxide (DMSO) on cryopreserved sperm of grass carp (Ctenopharyngodon idellus). Grass carp sperm was suspended in Kurokura extender #2 and equilibrated at $4^{\circ}C$ for 10 min. French straws (0.25 ml) of sperm were frozen from $4^{\circ}C\;to\;-4^{\circ}C$ at a rate of $4^{\circ}C\;min^{-1}$ and then ken $-4^{\circ}C\;to\;-80^{\circ}C$ at a rate of $11^{\circ}C\;min^{-1}$. The straws were kept at $-80^{\circ}C$ for 10 min and finally stored in liquid nitrogen $(-196^{\circ}C)$. The cryopreserved sperm was thawed in a water bath at $40^{\circ}C$ for 30 sec and fertilization, hatching rate and larval malformation were compared with fresh sperm (control). The fertilization rate of post-thawed sperm was comparable (from 88.21% to 94.30%) to that of fresh sperm. However, hatching rate of all frozen sperm were significantly lower (P<0.05) than that of control. Additionally, the larval abnormality rate of frozen sperm was significantly higher than that of fresh sperm. The results indicate that DMSO could affect the quality of cryopreserved sperm of grass carp, and a freezing program and a proper extender composition should be further studied.

Effect of Extenders with TCG and DMSO on the Viability of Rabbit Sperm

  • The purpose of this study was to evaluate the effect of addition of ethylene glycol, glycerol and sucrose to TCG (Tris, Citric Acid, Glucose, Egg Yolk) and DMSO Frozen. The extender containing Egg yolk concentration (10%, 20%) affects viability and acrosome morphology of rabbit sperm. Sperm viability was then assessed for the freezing extenders TCGD (Tris + Citricacid + Glucose + DMSO), TCGED (Tris + Citricacid + Glucose + Egg yolk + DMSO), TCGGD (Tris + Citricacid + Glucose + Glycerol + DMSO) and TCGSD Tris + Citricacid + Glucose + Sucrose + DMSO) during thawing at 38℃. for 20 seconds, respectively. TCG + 10% egg yolk (viability: 77.0 ± 0.8, NAI: 73.3 ± 0.9) was significantly (sperm viability and normal acrosome interaction (NAI)) higher than TCG + 20% egg yolk (70.7 ± 1.1, 70.0 ± 0.9) in the sperm normalcy analysis according to the yolk concentration. TCGGD (53.4 ± 0.1, 62.3 ± 0.4), TCGSD (61.3 ± 0.0, 67.1 ± 0.1) sperm viability and normal acrosome interaction (NAI) in frozen spermatozoa are TCGD (46.4 ± 2.8 and 56.3 ± 1. 4) and TCGED (23.0 ± 1.1 and 54.6 ± 1.4) extenders was thawed at 38℃ for 20 seconds. According to the results from each frozen bulking agent, sperm membrane integrity by hypotonic swelling test (HOST) analysis in TCGGD (59.8 ± 0.7), TCGSD (59.3 ± 0.5) was significantly high compared to other experimental groups (p < 0.05). In conclusion, these results suggested that TCGGD and TCGSD extenders enhance survivability of rabbit sperm after frozen-thawing.

돼지 정액의 동결시 Taurine과 $\alpha$-Tocopherol 첨가가 동결$\cdot$융해 정자의 성상과 기능에 미치는 영향 (Effects of Taurine and $\alpha$-Tocopherol Treatment during freezing on Sperm Characteristics and Function in Frozen-Thawed Porcine Semen)

  • 본 연구는 돼지 정자 동결시 taurine과 $\alpha$-tocopherol의 첨가가 융해후 정자 성상과 정자 기능 활성산소계(reactive oxygen species; ROS)의 발생 정도 및 지질 산화(lipid peroxidation, LPO)에 미치는 영향을 구명하기 위하여 시행하였다. 1차 및 2차 희석액내 taurine과 $\alpha$-tocopherol이 첨가된 돼지 동결정액의 융해 후 정자 운동성 양상, 정자 생존성, 정자 기법을 적용하여 다음과 같은 결과를 얻었다. Taurine(25mM, 50mM), $\alpha$-tocopherol($500{\mu}M,\;1,000{\mu}M$ 단일처리군 그리고 taurine과 $\alpha$-tocopherol의 혼합처리군($25mM\~500{\mu}M\~1,000{\mu}M$)은 동결$\cdot$융해 후 대조군과 비해 정자 운동성과 생존성은 유의적인 차이를 나타내지 않았다. Taurine과 $\alpha$-tocopherol의 혼합처리군 $50mM\~1,000{\mu}M$에서 HOST, 점체 반응이 대조군과 비교하여 유의차는 인정되지 않았으나 다소 증가하였다. 동결$\cdot$융해 정자의 ROS 발생 억제를 위한 taurine과 $\alpha$-tocopherol의 모든 처리군은 대조군과 비교하여$\cdot\{O_2}{^-}$ 발생을 유의적으로 완화시키지 못하였다. 그러나 taurine 단일처리군(25mM), $\alpha$-tocopherol 단일처리군($50mM,\;1,000{\mu}M$)과 혼합처리군($25mM\~500mM,\;50mM\~1,000{\mu}M$$H_{2}O_{2}$의 발생을 대조군과 비교하여 유의적으로 완화시켰다(P<0.05). 동결$\cdot$융해 정자의 malondialdehyde의 생산은 taurine과 $\alpha$-tocopherol의 모든 처리군에서 대조군과 비교하여 유의적인 감소를 보였다(P<0.05). 이상의 결과를 종합해 보면 돼지 정액의 동결보존시 taurine와 $\alpha$-tocopherol 같은 항산화제의 처리는 ROS 발생과 LPO을 효과적으로 완화시킴에 따라 돼지 정액의 동결보존 효율을 증진시킬 수 있는 유용한 방법이라 사료된다.

  • 본 실험은 실험견의 정액 동결 시 희석 액에 첨가되는 Glycerol 농도, 동결속도, 동결보존액 첨가후 평형시간, 융해온도와 시간에 따라 정자의 생존성과 운동성을 조사하여 최적의 동결조건을 확립하기 위해 실시하였다. 1. 각기 다른 Glycerol 농도를 함유한 동결보존액에서 동결보존 후 융해하였을 때 4%의 Gly-cerol 농도에서 각각 68.8$\pm$7.4%, 73.2$\pm$8.3%로서 다른 군보다 유의하게 높은 생존율과 운동성 나타냈다(P<0.05). 2. 희석 액 I 을 첨가한 정액을 상온에서 4$^{\circ}C$까지 각각 30, 60, 120분간 속도로 하강 시켜 동결보존 후 융해하였을 때 60분간의 속도로 하강시킨 군에서 생존성은 70.6$\pm$6.8%, 운동성은 72.8$\pm$8.4%로서 다른 군보다 유의하게 높은 생존율과 운동성을 나타냈다(P<0.05). 3. 희석 액 I 을 상온에서 4$^{\circ}C$까지 60분의 속도로 하강시킨 후 희석 액 II를 첨가하여 각각 30, 60, 120분 동안 평형을 유지시킨 후 동결보존 하였을 때 60분 동안 평형을 유지시킨 군에서 생존성은 71.4$\pm$7.3%, 운동성은 74.6$\pm$7.5%로서 다른 군보다 유의하게 높은 생존율 및 운동성을 나타냈다(P<0.05). 4. 정액을 동결함에 있어 액체질소의 표면에서 각각 3, 5, 7, 9cm의 높이에서 정치시킨 후 동결을 실시하였을 때 액체질소의 표면 5cm의 높이에서 동결을 실시한 군에서 융해 후 생존성과 운동성은 각각 77.0$\pm$7.0%, 80.2$\pm$6.3% 로서 다른 군보다 유의하게 높은 생존율과 운동성을 나타냈다(P <0.05). 5. 일정기간 보존된 동결정액을 융해 시 융해온도에 따른 정자의 생존성과 운동성은 37$^{\circ}C$의 온도에서 120초 동안의 기간에 녹였을 때 각각 77.0$\pm$7.0%, 80.2$\pm$6.3%로서 다른 군보다 유의하게 높은 생존율을 나타냈다(P<0.05).
