Synthesis of Side-Chain Nonlinear Optical Polymers with Carbazolylnitrostilbene Chromophores

  • Two different carbazolylnitrostilbene chromophores with second-order nonlinear optical (NLO) activity were newly synthesized by the reaction of 9-(2-hydroxyethyl)-9H-carbazol-3-carbaldehyde with 4-nitrophenylacetonitrile or 4-nitrophenylacetic acid. The NLO monomers were obtained by reaction of these chromophores with methacryloyl chloride. The side-chain nonlinear optical polymers were synthesized by the copolymerization of NLO monomer with methylmethacrylate using a free radical initiator. The chemical structures of the polymers were identified by spectroscopic means and the polymer properties such as molecular weight, Tg, solubility, UV-visible absorption, and second-harmonic generation (SHG) coefficients were investigated.

  • Various alkoxyamine initiators for nitroxide mediated radical polymerization (NMRP) were prepared in high yields by a simple substitution reaction of nitroxide anions with benzyl bromide. The required nitroxide anions were easily generated by treating either nitroxide free radicals or hydroxy amine with an alkali metal such as sodium or potassium in THF. This method is both practical and efficient, since the ionic conditions prevent other side reactions from occurring, such as the self-coupling or oligomerization reactions that are observed in the case of radical trapping conditions. To demonstrate the utility of the resulting alkoxyamine initiators, end- and telechelic-alkoxyamine PEG macroinitiators derived from the alkoxyamines were synthesized by a simple chemical modification, and used for the preparation of PEG-b-PS and PS-b-PEG-b-PS block copolymers by NMRP.

Effect of PTMGDA-PEGMA dopant on PVDF ultrafiltration membrane

  • As a novel hydrophobic monomer, polytetrahydrofuran diacrylate (PTMGDA) was synthesized by the esterification reaction between polyethylene tetrahydrofuran (PTMG) and acryloyl chloride (AC). In situ free radical polymerization reaction method was utilized to fabricate poly (vinylidene fluoride) (PVDF)-PTMGDA-poly(ethylene oxide) dimethacrylate (PEGMA) ulrafiltration (UF) membranes. The performances of PVDF-PTMGDA-PEGMA UF membranes in terms of morphologies, mechanical properties, separation properties and hydrophilicities were investigated. The introduction of the PTMGDA-PEGMA dopants not only increased the membranes' pure water flux, but also improved their mechanical properties and the dynamic contact angles. The addition of the PTMGDA/PEGMA dopants led to the formation of the finger-like structure in the membrane bulk. With the increase concentration of PTMGDA/PEGMA dopants, the porosity and the mean effective pore size increased. Those performances were coincide with the physicochemical properties of the casting solutions.

  • 자유라디칼 telomerization 반응을 이용하여 스티렌-메타크릴산메틸 block 혼성중합체 (PS-b-PMMA)를 합성하였다. AIBN을 개시제로 사용하여 CCI$_4$로 스티렌을 telomerization시킨 다음 생성된 중합체의 $CCI_3$ 말단기를 macrotelogen으로 사용하여 AIBN 개시에 의해 메타크릴산 메틸을 다시 telomerization하여 스티렌-메타크릴산메틸 block 혼성중합체를 얻었다. Macrotelogen의 농도, 단위체의 농도, macrotelogen의 분자량, 반응온도 및 용매의 농도가 block 혼성중합체생성에 미치는 영향을 조사하였다. 반응조건을 조절하여, PMMA가 약 10무게%까지 포함된 block 혼성중합체를 얻었다.

싸리꽃 에탄올 추출물의 피부(皮膚) 생리활성(生理活性) 효과 (Dermal Bioactive Properties of the Ethanol Extract from Flowers of Lespedeza bicolor)

  • Objective : In this study, the ethanol extract from flowers of Lespedeza bicolor was investigated for their dermal bioactive properties related to cosmeceuticals such as depigmentation and radical scavenging effect. Results : The ethanol extract from flowers of Lespedeza bicolor showed considerable radical scavenging activity ($SC_{50}:\;10\;{\mu}g/ml$) and inhibited the production of nitric oxide (NO) in Raw 264.7 macrophages activated with the endotoxin lipopolysaccharide (LPS). Although the proliferation of B16/F10 cells was slightly decreased by the ethanol extract from flowers of Lespedeza bicolor at the concentration of $100\;{\mu}g/ml$, it did not appear necrosis. The ethanol extract from flowers of Lespedeza bicolor down-regulated melanin formation effectively. Methods : The free radical scavenging activity of Lespedeza bicolor was assayed in cell free systems using a stable free radical, 1,l-diphenyl-2-picrylhydrazyl (DPPH). Nitrite accumulated in culture medium was measured as an indicator of NO production using a Griess reaction. Cell viability was measured by MTT assay and melanin content was assessed using the method of Hosei with some modifications. Conclusions : These results suggest that the ethanol extract from flowers of Lespedeza bicolor is a potent depigmetation agent and it may be a candidate for antioxidant and anti-inflammatory agent.

전기화학적 공정에 의한 클로레이트의 생성메커니즘 (Formation Mechanism of Chlorate ($ClO_3\;^-$) by Electrochemical Process)

  • 본 연구는 염화 이온 ($Cl^-$)을 전해질로 이용하는 전기화학적 공정에서 생성되는 염소산화부산물인 클로레이트 ($ClO_3\;^-$, 염소산염)의 생성 메커니즘을 알아보기 위해 수행되었다. 우선, pH 및 초기농도에 따른 생성 특성을 살펴보았으며, 유리염소 생성과의 관련성 및 오존, OH 라디칼 등의 혼합산화제의 영향을 간접 평가하여 클로레이트의 생성 메커니즘을 구체화하였다. 그 결과, 클로레이트의 생성은 유리염소 (HOCl/$OCl^-$)의 전기화학적 반응을 주된 반응으로 하며, 염화 이온의 직접 양극산화 반응 및 OH 라디칼에 의한 경로가 있음을 확인하였다. 이어서 생성된 클로레이트가 퍼클로레이트로 산화되는 반응도 볼 수 있었다. 또한, 전극 간격에 따른 생성 농도를 유리염소 생성과 함께 평가하여, 유리염소 생성 효율은 극대화 시키되 클로레이트의 발생을 최소화 할 수 있는 최적조건을 찾는 방안을 제시하였다.

Protective effects of carnosine and homocarnosine on ferritin and hydrogen peroxide-mediated DNA damage

  • Previous studies have shown that one of the primary causes of increased iron content in the brain may be the release of excess iron from intracellular iron storage molecules such as ferritin. Free iron generates ROS that cause oxidative cell damage. Carnosine and related compounds such as endogenous histidine dipetides have antioxidant activities. We have investigated the protective effects of carnosine and homocarnosine against oxidative damage of DNA induced by reaction of ferritin with $H_2O_2$. The results show that carnosine and homocarnosine prevented ferritin/$H_2O_2$-mediated DNA strand breakage. These compounds effectively inhibited ferritin/$H_2O_2$-mediated hydroxyl radical generation and decreased the mutagenicity of DNA induced by the ferritin/$H_2O_2$ reaction. Our results suggest that carnosine and related compounds might have antioxidant effects on DNA under pathophysiological conditions leading to degenerative damage such as neurodegenerative disorders.

Olefin Polymerization Activity and Crystal Structure of Alkyliron(Ⅲ) Porphyrin Complexes

  • Alkyliron(Ⅲ) porphyrins, n-butyliron(Ⅲ) tetraphenylporphyrin, (TPP)Fe-Bu and n-butyliron(Ⅲ) tetrakis-(pentafluorophenyl)porphyrin, $(F_{20}TPP)Fe-Bu$ have been evaluated as suitable for olefin free-radical polymerization. Butyl radicals dissociated from n-butyliron(Ⅲ) porphyrin initiated the polymerization reaction, but the ratio of the propagation was low. The GCMS analysis of the reaction mixture of nbutyliron(Ⅲ) porphyrin and styrene has revealed several products containing two butyl groups, while traces of b-hydrogen-abstracted products were observed. The crystal structure of (TPP)Fe-Bu has been determined. The structure of the n-butyliron(Ⅲ) porphyrin reveals the compound containing five-coordinated iron with the average Fe-N distance of 1.973(1) Å and Fe-C of 2.030(2) Å. The iron atom is displaced by 0.137Å from a four nitrogen mean plane. Crystal system is triclinic, and space group is P-1.

Fumonisin B1에 의한 세균바이러스 DNA손상 (Bacterial Virus DNA Damage Caused by Fumonisin B1)

  • Fumonisin B1 is a secondary metabolite of Fusarium moniliforme, a contaminant of corn and corn product. Fumonisin B1 has been shown to be responsible for major toxicological effects of the fungus in rats, horses, and pigs. Fumonisin B1 induced λ DNA fragmentation, which was increased with incubation time, reducing agent NADPH and metal ion (Cu2+). The DNA damage was inhibited by dimethyl sulfoxide (DMSO) or mannitol as radical scavenger for free radicals. DNA fragmentation, induced by fumonisin B1 in the presence of 1 mM NADPH and 0.1 mM CuCl2, was inhibited by 100 mM DMSO. By the in vitro reaction of fumonisin B1 with supercoiled plasmid pBR322 DNA, plasmid DNA was relaxed, eventually linearized in the agarose gel electrphoresis. From rifampicin sensitive E. coli CSH138 in bacterial mutagenesis system, the rifampicin resistant E. coli mutants were obtained by fumonisin B1. These results suggest that fumonisin B1 may be a possible environmental mutagen in bacterial mutagen assay system.

Kinetic Approaches to Measuring Peroxiredoxin Reactivity

  • Peroxiredoxins are ubiquitous thiol proteins that catalyse the breakdown of peroxides and regulate redox activity in the cell. Kinetic analysis of their reactions is required in order to identify substrate preferences, to understand how molecular structure affects activity and to establish their physiological functions. Various approaches can be taken, including the measurement of rates of individual steps in the reaction pathway by stopped flow or competitive kinetics, classical enzymatic analysis and measurement of peroxidase activity. Each methodology has its strengths and they can often give complementary information. However, it is important to understand the experimental conditions of the assay so as to interpret correctly what parameter is being measured. This brief review discusses different kinetic approaches and the information that can be obtained from them.
