The occurrence of HHS was confirmed for the first time in Korea from chickens submitted for diagnosis to our laboratory from broiler and baeksemi farms. Clinical signs included depression, inappetence, ruffled feathers and a increase in mortality. At necropsy, severe hydropericardium and hepatic necrosis was founded characteristically and the most remarkable microscopic changes were seen in the liver. These included basophilic intranuclear inclusion bodies in the hepatocytes, massive hemorrhages and necrosis in the liver parenchyma. We could also identify fowl adeno-virus(FAV) by polymerase chain reaction(PCR) and electro-microscopic confirmation. Abbreviation: HHS=hydropericardium hepatitis syndrome, EM=electron microscopy, FAV=fowl adenovirus, PCR=polymerase chain reaction.
This survey was performed to investigate avian salmonellosis from broiler farms in Namwon area for 4 years. The whole samples from farms were 541 and chicken samples were 294. Out of 294, 83 cases were identified with avian salmonellosis for 4 years by various serological and biochemical tests. There was no outbreak of pullorum disease, but fowl typhoid occurred in 36 farms. The outbreak of avian salmonellosis occurred continuously regardless of seasons throughout the year. Avian salmonellosis mostly happened within 10 days of age which were 52 cases(63%). The selection of susceptible antibiotics has changed from Quinoline group in 2000 to amoxacillin + clavulanic acid(AmC).
Concept and establishment of essential amino acids in animals and human beings rendered immeasurable contributions to animal production and human health. In ruminant animals, however, essential amino acids have never been completely established. The present review proposes a hypothesis that histidine may not be an essential amino acid for normal growing cattle (Japanese black) at least at the growing stage after about 450 kg of body weight on the basis of the experimental results of histidinol dehydrogenase activities in some tissues of the cattle together with hints from which the hypothesis was derived. At the same time, histidinol dehydrogenase activities in liver, kidney and muscle of swine, mouse, fowl and wild duck will be shown and the essentiality of histidine in these animals will be discussed. Finally, the essentiality of histidine for adult human will briefly be discussed.
The results of application of the organic manure obtained from livestocks fed with BLCS in the Tomato under structure were as follow. The No. of Fruit weight, Fruit width, Fruit length and Sugar content of Tomato in the Dung of fowl(BLCS) were high in the other treatment, second was cattle dropping(BLCS), third was Pig dropping(BLCS). The contents of Total N, P2O5, K2O, CaO, and MgO in the Tomato fruit of the cattle dropping(BLCS), the Pig dropping(BLCS), and the Dung of fowl(BLCS) were high than animal dropping plots.
Streptomyces (S.) fradiae is a microbe with broad-spectrum antimicrobial activity, isolated from soil. In the present study, antibacterial effects of S. fradiaea against Salmonella (S.) gallinarum was determined. S. fradiae inhibited growing of S. gallinarum in Luria-Bertani media agar. Moreover, ingestion of S. fradiae markedly inhibited mortality of chickens experimentally infected with S. gallinarum. There is no side effect by S. fradiaeon, in safety of chickens and antibiotic material residues in chicken meat. Taken together, S. fradiae have the antibacterial effects against S. gallinarum. Therefore, we concluded that S. fradiae might be a good microbial candidate for treatment or control of fowl typhoid in chickens.

In this study, the characterization of minor livestock's excretions in terms of unit load generation and discharge was conducted by investigation and analysis of urine, manure and wastewater from stall of fowl, duck, horse, deer, goat and sheep. The results are summarized as follows: The unit load generation of fowl estimated by discharged amount and concentration analysis increases in egg layers due to the difference of planting head numbers, feed stuffs and manure disposal. In case of deer unit load generation by herbivora were calculated to be higher than data from existing references because of the gap between weight per livestock and the generation amounts of manure and urine. In case of sheep unit load generation by urine were analyzed two times higher than by manure but unit load generation by manure were reported higher than by urine in references, so large differences between this and previous study resulted.
Fowl adenovirus (FAV) is an important cause of several diseases, which result in considerable economic losses to the poultry farm. An outer capsid protein of FAV, fiber 2 is essential for virus growth, assembly or spread. This study was performed to produce about 22 kDa of recombinant fiber 2 protein and to immunize in laying hens to acquire the specific IgY antibody against the recombinant fiber 2. Laying hens were immunized with the recombinant fiber 2 intramuscularly in the breast muscle by injection 4 times at intervals of three weeks. At 12 weeks, serum- and egg yolk-antibody titers of hens against fiber 2 were increased up to 430,000 and 414,000, respectively. The recombinant fiber 2 could be recognized be the anti-His monoclonal antibody. Anti-fiber 2-IgY antibody could recognize the fiber 2 specifically in western blot analysis. These results suggested that the recombinant fiber 2 antigen could be used as an immunogen to elicit IgY antibody against fiber 2 and the anti-fiber 2-IgY could neutralize fowl adenovirus fiber 2 effectively.
The objective of this study was to determine appropriate sample size that simulated different assumptions for diagnostic test characteristics and true prevalences when designing serological surveillance plan for pullorum disease and fowl typhoid in domestic poultry production. The number of flocks and total number of chickens to be sampled was obtained to provide 95% confidence of detecting at least one infected flock, taking imperfect diagnostic tests into account. Due to lack of reliable data, within infected flock prevalence (WFP) was assumed to follow minimum 1%, most likely 5% and maximum 9% and true flock prevalence of 0.1%, 0.5% and 1% in order. Sensitivity were modeled using the Pert distribution: minimum 75%, most likely 80% and maximum 90% for plate agglutination test and 80%, 85%, and 90% for ELISA test. Similarly, the specificity was modeled 85%, 90%, 95% for plate agglutination test and 90%, 95%, 99% for ELISA test. In accordance with the current regulation, flock-level test characteristics calculated assuming that 30 samples are taken from per flock. The model showed that the current 112,000 annual number of testing plan which is based on random selection of flocks is far beyond the sample size estimated in this study. The sample size was further reduced with increased sensitivity and specificity of the test and decreased WFP. The effect of increasing samples per flock on total sample size to be sampled and optimal combination of sensitivity and specificity of the test for the purpose of the surveillance is discussed regarding cost.

The objective of this study was to examine the binding potential of sperm to the epithelium of the sperm storage tubules (SST) in vitro and in vivo to assess the functional maturation of fowl sperm. Sperm from the testis, epididymis, as well as the proximal, middle and distal vas deferens were incubated in vitro with either the uterovaginal junction (UVJ)- or infundibular tissue containing SST at $39^{\circ}C$ for 30 min. Aliquots of sperm were also artificially inseminated into the uteri of hens, and the UVJ and infundibulum were collected 24 h post artificial insemination (AI). After incubation and AI, tissues were washed to remove loosely adhered sperm and subjected to fluorescence staining with 4', 6-diamidino-2-phenylindole, dihydrochloride (DAPI) for counting the number of bound sperm per 0.25 mm2 of surface area. Sperm from the testis, epididymis, and the three segments of the vas deferens exhibited their differential (p<0.05) binding capacity, which increased gradually from the testicular to distal vas deferens sperm under both in vitro and in vivo conditions. Existing similar trend, sperm, regardless of their source had a lesser affinity to bind to the epithelium of the infundibular SST than to the UVJ-SST. These experimental results suggested that fowl sperm may undergo gradual changes in the process of functional maturation, whereby they gain the ability to bind to the epithelium of the SST during their passage through the male reproductive tract (MRT).

Blood samples were collected from eight native Japanese breeds of chickens (Miyadi-dori, Ohiki, Onaga-dori, Shoukoku, Tosa-Jidori, Tosa-Kukin, Toutenkou and Uzurao) and two foreign breeds of chickens (White Leghorn and Rhode Island Red) to examine the genetic variability and relationships among the breeds by using a microsatellite DNA technique. Except for the Shoukoku breed, the other Japanese chicken breeds all originate from Kochi Prefecture. Ohiki, Onaga-dori, Tosa-Jidori, Toutenkou and Uzurao are fancy fowl, and Miyadi-dori and Tosa-Kukin are utility fowl. Among the fancy fowl, Ohiki, Onaga-dori, and Toutenkou males have thick and long feathers in the saddle and tail. Genetic variabilities of the 20 microsatellites examined, varied depending on the breed: the mean number of alleles per locus ranged from 2.05 (Miyadi-dori) to 3.90 (Rhode Island Red); proportion of polymorphic loci ranged from 0.75 (Miyadi-dori) to 1.00 (Rhode Island Red, Shoukoku and Uzurao); and mean expected heterozygosity ranged from 0.330 (Miyadi-dori) to 0.607 (Rhode Island Red). Unique microsatellite alleles were detected in each breed. Using the neighbour-joining method, phylogenetic trees were constructed based on the genetic distances of D$_{A}$ and D$_{ST}$. Among the breeds originating from Kochi Prefecture, fancy and utility breeds belonged to different clusters. Among the fancy breeds, those having thick and long feathers in the tail and saddle showed a close genetic relationship to the Shoukoku breed, which also has thick and long feathers in the tail and saddle.
