Effects of Ursolic Acid on Oncogene Expression Detected by In Situ Hybridization in Mice (생쥐에서 종양세포의 암유전자발현에 대한 울솔산의 효과)

  • To investigate the effect of ursolic acid on the expression of oncogenes in tumor cells of mice, sarcoma 180 ascites tumor cells were implanted into the left groin of ICR mice and the tumor bearing mice were treated with ursolic acid. The expression of oncogenes were measured by in situ hybridization method. Ursolic acid significantly reduced the expression of oncogenes in the tumor cells. Therefore, it can be said that the prestated anticarcinogenic effect of ursolic acid could be partly ascribed to the mechanism included in the oncogene´s transcription level.

  • Fiber Bra99 grating (FBG) sensor, one of the fiber optic sensor (FOS) offers lots of advantages for structural health monitoring due to its multiplexing capability. Also, it is proper to measure the structural vibration with no mass concentration effect. In this paper, we constructed two sensor arrays composed of 9 FBG sensors for the vibration and mode sensing of a composites beam. For an accurate measurement of wavelength shift, a signal processing board with an electric circuit based on time-interval counting was developed. This sensor system showed a good resolution of dynamic strain (<10${\mu}{\varepsilon}$). Using this sensor system, dynamic strains at 9 points of composite beam was measured and strain measured mode shape of the beam was calculated from the acquired strains and compared with numerical results by ABAQUS.

  • Cancer-associated antigen (CAGE) induces the expression of matrix metalloproteinase-2 (MMP-2) by activating Akt, which in turn interacts with inhibitory kappa kinase $\beta$ ($I{\kappa}K{\beta}$) to activate nuclear factor ${\kappa}B$ (NF-${\kappa}B$). Akt and p38 mitogen activated protein kinase (p38 MAPK) are necessary for CAGE-mediated induction of the AP-1 subunit JunB, whereas extracellular regulated kinase (ERK) is necessary for the induction of fos-related antigen-1 (Fra-1). Induction of MMP-2 by CAGE requires activator of protein-1 (AP-1) to be bound. Specific binding of JunB to MMP-2 promoter sequences was shown by chromatin immunoprecipitation (ChIP) analysis.

  • 설탕을 원료로 제조한 액상 프락토올리고당 제품의 품질 유지기간을 향상시키기위하여 pH, 고형분 농도, 과당 농도가 프락토올리고당(FOS)의 가수분해에 미치는 영향을 조사하였다. 효소 반응액의 pH 6.0 이상과 최종 제품의 고형분 농도 80%에서 FOS의 잔존율이 크게 향상되었다. 또한 과당 농도가 증가할수록 FOS의 잔존율은 감소하였다. 효소 반응액의 pH 6.0, 고형분 농도 80%, 과당 농도 1.1%의 시료를 $60^{\circ}C$에 16 일간 보관한 결과 FOS의 잔존율은 95%로 높게 유지되었다.

The Heterochromatin-1 Phosphorylation Contributes to TPA-Induced AP-1 Expression

  • Activator protein-1 (AP-1) is an inducible transcription factor that contributes to the generation of chronic inflammation in response to oxidative and electrophilic stress. Previous studies have demonstrated that the PI3K/Akt1 pathway plays an important role in the transcriptional regulation of AP-1 expression. Although the histone post-translational modifications (PTMs) are assumed to affect the AP-1 transcriptional regulation by the PI3K/Akt pathway, the detailed mechanisms are completely unknown. In the present study, we show that heterochromatin 1 gamma ($HP1{\gamma}$) plays a negative role in TPA-induced c-Jun and c-Fos expression. We show that TPA-induced Akt1 directly phosphorylates $HP1{\gamma}$, abrogates its suppressive function and increases the interaction between histone H3 and 14-3-$3{\varepsilon}$. Collectively, these our data illustrate that the activation of PI3K/Akt pathway may play a permissive role in the recruitment of histone readers or other coactivators on the chromatin, thereby affecting the degree of AP-1 transcription.

Development and Verification of an Optimum Composition Model for a Synbiotic Fermented Milk Using Sequential Quadratic Programming Techniques

  • The purpose of this research was to develop an optimum composition model for a new synbiotic fermented dairy product with high probiotic cell counts, and to experimentally verify this model. The optimum composition model indicated the growth promoter ratio that could provide the highest growth rate for probiotics in this fermented product. Different levels of growth promoters were first blended with milk to improve the growth rates of probiotics, and the optimum composition model was determined. The probiotic viabilities and chemical properties were analyzed for the samples made using the optimal formula. The optimal combination of the growth promoters for the synbiotic fermented milk product was 1.12% peptides, 3% fructooligosaccharides (FOS), and 1.87% isomaltooligosaccharides (IMO). A product manufactured according to the formula of the optimum model was analyzed, showing that the model was effective in improving the viability of both Lactobacillus spp. and Bifidobacterium spp.

  • This paper deals with the applicability of long-gauge deformation fiber optic sensors (FOS) to prestressed concrete structures. A main motivation is the desire to monitor the deflection of the railway bridges without intervenes of the signal intensity fluctuations. A 25 m long, 1.8 m deep PSC girder was fabricated compositely with 22 cm thick reinforced concrete deck. Two pairs of 3 m long-gauge sensors are attached to the prestressed concrete girder with parallel topology. Using the relationship between curvature and vortical deflection and the quadratic regression of curvatures at the discrete point, it is possible to extrapolate the deflection curve of the girder. The estimated deflection based on the developed method is compared with the results using conventional strain gauges and LVDTS. It has been demonstrated that the proposed instrumentation technique is capable of estimating the vertical deflection and neutral axis position of the prestressed concrete girder up to weak nonlinear region.

  • Mammalian cell based biosensor kits are expected to be in assessment of samples toxicity more sensitive and accurate. A recombinant fluorescent Chinese Hamster Ovary (CHO) cell line was known to be responsive to the various toxicants Specially. KFC- AlO cell line. which contain the c-fos SRE::GFP plasmid (pKFG). was found to be able to detect toxicants sensitively. A biosensor kit was developed by using an immobilized KFC-A10 cell line. Immobilized recombinant fluorescent cells within agarose, known as a representative hydrogel matrix, have been maintained in the matrix viably and have shown constant fluorescent levels for long time. Immobilized cells have shown the ability to detect the chemical toxicity in the keep of fluorescent level as the metabolism is inhibited under toxic conditions.

  • 실시간 운영체제 GUI 에서 가장 중점적인 것은 임베디드 시스템상에서 중요한 자원 중 하나인 메모리의 효율적인 관리이다. 특히 파일시스템이 없는 임베디드 시스템의 경우 GUI 를 사용하게 되면 폰트 이미지(Font Image)와 그림파일 등이 메모리상에 존재하여야 하기 때문에 메모리를 많이 차지하게 된다. 본 논문에서는 실시간 운영체제 $UbiFOS^{TM}$에서 파일시스템 모듈을 제거하고 GUI 를 설계 및 구현하였고 특히 폰트 이미지와 그림파일을 처리하는 방법과 폰트 이미지를 위한 메모리를 절약할 수 있는 메커니즘을 제안하였다.

  • 실시간 운영체제를 탑재한 임베디드 시스템(Embedded System)은 특성상 다른 시스템에 비해 상대적으로 저 용량의 메모리를 지닌다. 따라서 제한된 메모리를 효율적으로 사용할 수 있는 기법이 적용될 필요가 있다. 외부 단편화(External Fragmentation)로 생긴 메모리 공간을 재조정하는 기법도 효율적인 메모리 사용을 위한 방안이며, 본 논문에서는 실시간 운영체제에서 가용한 메모리를 효율적으로 활용할 수 있는 메모리 압축(Memory Compaction) 기법에 대해서 설계 및 구현하였다.
