LPD vectors are non-viral vehicles for gene delivery comprised of polycation-condensed plasmid DNA and liposomes. Here, we described a novel anionic LPD formulation containing protamine-DNA complexes and pH sensitive liposomes composed of DOPE and cholesteryl hemisuccinate (Chems). Central composite design (CCD) was employed to optimize stable LPD formulation with small particle size. A three factor, five-level CCD design was used for the optimization procedure, with the weight ratio of protamine/DNA ($X_1$), the weight ratio of Chems/DNA ($X_2$) and the molar ratio of Chems/DOPE in the anionic liposomes ($X_3$) as the independent variables. LPD size ($Y_1$) and LPD protection efficiency against nuclease ($Y_2$) were response variables. Zeta potential determination was utilized to define the experimental design region. Based on experimental design, responses for the 15 formulations were obtained. Mathematical equations and response surface plots were used to relate the dependent and independent variables. The mathematical model predicted optimized $X_1-X_3$ levels that achieve the desired particle size and the protection efficiency against nuclease. According to these levels, an optimized LPD formulation was prepared, resulting in a particle size of 185.3 nm and protection efficiency of 80.22%.
A static linear programming formulation for minimum weight design of frames that is based on a matrix displacement method is presented in this paper. According to elementary theory of plasticity, minimum weight design of frames can be carried out by using only the equilibrium equations, because the system is statically determinate when at an incipient collapse state. In the present formulation, a statically determinate released frame is defined by introducing hinges into the real frame and the bending moments in yield constraints are expressed in terms of unit hinge rotations and the external loads respectively, by utilizing the matrix displacement method. Conventional Simplex algorithm with some modifications is utilized for the solution of linear programming problem. As the formulation is based on matrix displacement method, it may be easily adopted to the weight optimization of frames with displacement and deformation limitations. Four illustrative examples are also given for comparing the results to those obtained in previous studies.
This study proposes an empirical formulation to predict the maximum deformation of offshore blast wall structure that is subjected to impact loading caused by hydrocarbon explosion. The blast wall model is assumed to be supported by a simply-supported boundary condition and corrugated panel is modelled. In total, 1,620 cases of LS-DYNA simulations were conducted to predict the maximum deformation of blast wall, and they were then used as input data for the development of the empirical formulation by regression analysis. Stainless steel was employed as materials and the strain rate effect was also taken into account. For the development of empirical formulation, a wide range of parametric studies were conducted by considering the main design parameters for corrugated panel, such as geometric properties (corrugation angle, breadth, height and thickness) and load profiles (peak pressure and time). In the case of the blast profile, idealised triangular shape is assumed. It is expected that the obtained empirical formulation will be useful for structural designers to predict maximum deformation of blast wall installed in offshore topside structures in the early design stage.
This study set out to improve the physical and pharmaceutical characteristics of the present formulation using an appropriate experimental design. The work described here concerns the formulation of the dispersible tablet applying direct compression method containing roxithromycin in the form of coated granules. In this study $2^3$ factorial design was used as screening test model and Central Composite Design (CCC) associated with response surface methodology was used as optimization study model to develop and to optimize the proper formulation of roxithromycin dispersible tablet. The three independent variables investigated were functional excipients like binder (X1), disintegrant (X2) and lubricant (X3). The effects of these variables were investigated on the following responses: hardness (Y1), friability (Y2) and disintegration time (Y3) of tablet. Three replicates at the center levels of the each design were used to independently calculate the experimental error and to detect any curvature in the response surface. This enabled the best formulations to be selected objectively. The effect order of each term to all response variable was X3> X2> Xl> X1*X2> X2*X2> X2*X3> X3*X3> Xl*X3> Xl*Xl and model equations on each response variables were generated. Optimized compositions of formula were accordingly computed using those model equations and confirmed by following demonstration study. As a result, this study has demonstrated the efficiency and effectiveness of using a systematic formulation optimization process to develop the tablet formulation of roxithromycin dispersible tablet with limited experiment.
Many researchers have recently studied multi-level formulation strategies to solve the MDO problems and they basically distributed the coupling compatibilities across all disciplines, while single-level formulations concentrate all the controls at the system-level. In addition, approximation techniques became remedies for computationally expensive analyses and simulations. This paper studies comparisons of the MDO methods with respect to computing performance considering both conventional sequential and modem distributed/parallel processing environments. The comparisons show Individual Disciplinary Feasible (IDF) formulation is the most efficient for sequential processing and IDF with approximation (IDFa) is the most efficient for parallel processing. Results incorporating to popular design examples show this finding. The author suggests design engineers should firstly choose IDF formulation to solve MDO problems because of its simplicity of implementation and not-bad performance. A single drawback of IDF is requiring more memory for local design variables and coupling variables. Adding cheap memories can save engineers valuable time and effort for complicated multi-level formulations and let them free out of no solution headache of Multi-Disciplinary Analysis (MDA) of the Multi-Disciplinary Feasible (MDF) formulation.

This paper deals with the reliability analysis of design formulations derived for predicting the punching shear capacity of FRP-reinforced two-way slabs. Firstly, a new design code formulation was derived by means of gene expression programming. This formulation differs from the existing ones as the slab length (L) was introduced in the equation. Next, the proposed formulation was tested for its generalization capability by a parametric study. Then, the stochastic analyses of derived and existing formulations were performed by Monte Carlo simulation. Finally, the reliability analyses of these equations were carried out based on the results of stochastic analysis and the ultimate state function of ASCE-7 and ACI-318 (2011). The results indicate that the prediction performance of new formulation is significantly higher as compared to available design equations and its reliability index is within acceptable limits.
A continuum-based shape design sensitivity analysis (DSA) method is presented for the acoustic radiation from thin body. The normal derivative integral formulation is employed as an analysis formulation and differentiated directly by using material derivative to get the acoustic shape design sensitivity. In the acoustic sensitivity formulation, derivative coefficients of the structural normal velocities on the surface are required as the input. Thus, the shape design sensitivities of structural velocities on the surface with respect to the shape change are also calculated with continuum approach. A simple disk is considered as a numerical example to validate the accuracy and efficiency of the analytical shape design sensitivity equations derived in this research. This research should be very helpful to design an application involving thin body and to change its acoustic characteristics.

This paper presents a generalized formulation for optimizing the design of concrete beam reinforced with glass fiber reinforced polymer bar. The optimization method is formulated to find the design variables leading to the minimum weight of concrete beam with constraints imposed based on ACI code provisions. A simple genetic algorithm is utilized to solve the optimization task. The weights of concrete and glass fiber reinforced polymer bar are included in the formulation of the objective function. The ultimate limit states and the serviceability limit states are included in formulation of constraints. The results of illustrated example demonstrate the efficiency of the proposed method to reduce the weight of beam as well as to satisfy the above requirement. The application of the optimization based on the most economical design concept have led to significant savings in the amount of the component materials to be used in comparison to classical design solutions.
The design analysis of axisymmetric, multi-stage deep drawing dies was performed using the rigid-viscoplastic finite element formulation. In the formulation, the axisymmetric CFS algorithm was employed. Hill's non-quadratic normal anisotropic yield criterion and isotropic hardening rule were considered. For trial initial displacements and tool contact points, the geometric force equilibrium method was adopted. In order to see the validity of the formulation, the multi-stage deep drawing processes of shell-cylinder front part of hydraulic booster were simulated. The simulation showed good agreements with measurements and PAM-STAMP.
