Performance of MiniPCRTM mini8, a portable thermal cycler (휴대용 DNA증폭기 MiniPCRTM mini8 Thermal Cycler의 성능 검토)

  • A small and inexpensive thermal cycler (PCR machine), known as the MiniPCRTM Mini8 Thermal Cycler (Amplyus, Cambridge, MA, USA), was developed. In this study, the performance of this PCR machine was compared with the GeneAmp® PCR system 9700 (Applied Biosystems) using four autosomal short tandem repeat (STR) kits, a Y-chromosome STR kit, and a mitochondrial DNA HV1/HV2 sequence analysis. The sensitivity and stochastic effects of the STR multiplex kits and the quality of the DNA sequence analysis were similar between the two PCR machines. The MiniPCRTM Mini8 Thermal Cycler could be used for analyses at forensic DNA laboratories and crime scenes. The cost of the PCR is so economical that school laboratories and individuals could use the machines.

Comparative study of serological detection methods on old bloodstain samples (오래된 혈흔 시료에 대한 혈청학적 식별법의 비교 연구)

  • Blood is a commonly found body fluid at crime scenes, and plays an important role in identifying suspects and in the reconstruction of crime scenes. Although serological detection of blood has been widely used in the field of forensic science, research on the detection of old bloodstains is scarce. This work aimed to compare various methods for the detection of old bloodstains and validate the reliability of their results. Four presumptive tests-Tetramethylbenzidine, $Bluestar^{(R)}$, Leucomalachite Green, Kastle-Meyer tests-and two confirmatory tests-Fecal Occult Blood (FOB) and Rapid Stain $Identification^{(TM)}-Blood$ ($RSID^{TM}-Blood$) tests-were compared. Bloodstain samples from post-mortem cases were collected on gauzes and then stored at room temperature for periods from 7 to 30 years. All the presumptive tests were positive, even for the 30-year-old sample. However, FOB and $RSID^{TM}-Blood$ provided false negative results for some samples stored for 17 years or more (1988 to 2001). The results indicate that FOB and $RSID^{TM}-Blood$ are not reliable for the detection of old bloodstains. These findings can be useful in the selection of an appropriate detection method for serological testing of old bloodstains. In addition, the information will be useful background knowledge when applied in the field of forensic practice.

A Survey of Radiologic Science Students' Awareness and Educational Needs of Forensic Medicine (방사선학과 전공 학생들의 법의학에 대한 인식과 교육 요구도 조사)

  • Due to the development of the domestic economy and science, the people's standard of living has increased. Accordingly, we want to improve the quality of life. In other words, we guarantee human rights and pursue dignity and value as human beings. Therefore, the medical field extends human life and helps maintain a healthy life. The social medicine that protects human rights is forensic medicine. Forensic medicine identifies deaths and analyzes the cause using forensic radiology images. Forensic radiology is the acquisition and provision of medical images by the radiographer. Therefore, the radiographer must have expertise by completing forensic science-related courses. Recently, medical and nursing schools have opened and operated various subjects such as forensic medicine and forensic nursing. However, the Department of Radiology science is the only school that offers courses related to forensic science. For the future development and exploration of the radiographer and department of radiology science, forensic education should be considered. For this purpose, we investigated the necessity and demand for forensic education in the department of radiology science undergraduate and graduate schools. The department of radiology science students' awareness of forensic science was found to be 2.977 points, but the need for forensic science education for the radiographer was high at 3.759 points. In addition, current students' demand for forensic science courses was high at 84.1%, with the majority responding that it was necessary to open and operate the course. This study was able to determine the demand for forensic science-related subjects among the department of radiology science undergraduate and graduate students, and there is a need to explore diversity and expertise in education. We hope that it will be used as basic data for the development of forensic medicine and radiology science.

  • Embezzlement incident of staff at the casino occurred. Staff of some casinos have lower job satisfaction, moral gap is seriously considering a change jobs. In addition, cash lure of large amounts and a lack of money management system causes embezzlement incident. In this paper, the uniqueness of the casino industry and that employee job satisfaction is investigated. Content analysis of occurrence for casino embezzlement incident and tracking that bank account and bank check, suspect's call list, and so on that digital forensic investigation technology will be studied. Problems and solutions suggest that conducted a loss prevention program, a digital forensics technology and introduce of investigator. Through this study, the computerization of the casino business to embezzlement accident prevent will contribute to that give back profits of property to society, the develop of forensic investigation technology.

  • The chemical element and structural characterization of different types of cements and its brick stones are been investigated under forensic aspects using X-ray florescence (XRF) and X-ray diffraction (XRD) spectrometer. The XRF provides rapid compositional data for controlling almost all stages of raw materials, clinker and cement. The decisive advantage of XRD methods is based on the unique character of the diffraction patterns of crystalline substances, the ability to distinguish between elements and their oxides, and the possibility to identify chemical compounds, polymeric forms, and mixed crystals by non-destructive examination. Therefore, combination of these examinations is useful and able to apply for the forensic analysis in comparison of cements and brick stones. There are more study remained to determine the viability of method for forensic analysis of brick stones and the limits of the discrimination that can be achieved.

  • In the past, the domestic research on mobile forensics has been limited to cell phones. Increasing use of smart phones, studies on smart phone forensic will be conducted actively in the future. In particular, the study on Android forensic is very important because Android smart phone market share is increasing rapidly. In this paper, we describe an implementation of an Android smart phone forensic tool based on logical analysis. Compared with Oxygen Forensic Suite 2010, this tool saves time it takes to perform Android smart phone forensic because this tool provides search feature and resource links for extracted media information. So far, no smart phone forensic tool is introduced in Korea. Accordingly, this tool would contribute to the advancement of the technology on smart phone forensic.

  • Starvation, diabetes, alcoholism and hypothermia cause ketoacidosis in the human body; therefore, the cause of death can be determined by analyzing ketone bodies in the blood of the deceased. In the case of decomposition of the cadaver, however, since collecting intact blood is impossible, ketone body analysis is performed using the spleen. However, the index for diagnosing ketoacidosis is based on blood concentration, and its correlation with ketone bodies present in the spleen remains unknown. In particular, since decomposition proceeds rapidly during summer, when temperature and humidity are high, understanding the correlation between ketone bodies in the blood and spleen is important to estimate the state at the time of death from a decaying body. Therefore, in the present study, the correlation between ketone bodies in the blood and spleen of the deceased was explored. Ketone bodies (beta-hydroxybutyric acid [BHB] and acetone) in the blood and spleen were analyzed and compared from autopsies (>100 mg·L-1 BHB, blood basis) conducted at the Daejeon Forensic Research Institute from June to December 2021. Moreover, the concentration of ketone bodies in the spleen juice and tissues was compared assuming the scenario of extreme decomposition. Ketone retention concentration in the blood and spleen was positively correlated, and the ratio of BHB concentration in the spleen to BHB concentration in the blood ranged from 0.52 to 1.08 (mean = 0.85 ± 0.12), although the ratio may vary depending on the degree of decomposition of the corpse.

Effect of novel luminol-based blood detection reagents on DNA stability (새로운 루미놀 기반 혈흔 탐지 시약이 디엔에이에 미치는 영향에 대한 연구)

  • Detection of bloodstains is a very important process in scientific investigations, and luminol is often used for the detection of bloodstains that are not visible. Recently, new preparation methods of blood detection reagents based on luminol (BloodFlareA, B) were developed and reported to have higher active persistence and to be more economical than conventional blood detection reagent, BlueStar forensic. In this paper, we tested the specificity and effect of the BloodFlares (A and B) on DNA and compared them with those of BlueStar forensic. False positive results for the BloodFlares were not observed in semen, saliva, vaginal fluids, urine, sweat, and nasal discharge, but were observed in $CuSO_4$, $FeSO_4$, and bleach solutions, and the observed patterns were similar to those of BlueStar forensic. The effect on DNA was determined by analyzing the DNA yield, degradation index, and DNA profiling. Based on these results, we concluded that the BloodFlares based on luminol do not affect DNA stability and are applicable in forensics.

  • Gloves are very important evidence at a crime scene; specifically, rubber gloves can be found easily at homes. Therefore, crime scene investigators attempt to develop fingerprints inside the rubber gloves that are discovered, for identifying unknown suspects. This study compared the effectiveness of three different methods that are used for developing latent prints on gloves with aging time. These were the powder, cyanoacrylate fuming, and multi-metal deposition I methods. The powder method achieved good results for 1-3 days of aged prints, and the cyanoacrylate fuming method worked well on 2-week-old prints. In comparison, multi-metal deposition I method developed good quality fingerprints for 6 weeks of aging time.

Application and evaluation of machine-learning model for fire accelerant classification from GC-MS data of fire residue

  • Detection of fire accelerants from fire residues is critical to determine whether the case was arson or accidental fire. However, to develop a standardized model for determining the presence or absence of fire accelerants was not easy because of high temperature which cause disappearance or combustion of components of fire accelerants. In this study, logistic regression, random forest, and support vector machine models were trained and evaluated from a total of 728 GC-MS analysis data obtained from actual fire residues. Mean classification accuracies of the three models were 63 %, 81 %, and 84 %, respectively, and in particular, mean AU-PR values of the three models were evaluated as 0.68, 0.86, and 0.86, respectively, showing fine performances of random forest and support vector machine models.
