  • Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS)-based pathogen identification relies on the ribosomal protein spectra provided in the proprietary database. Although these mass spectra can discern various pathogens at species level, the spectra-based method still has limitations in identifying closely-related microbial species. In this study, to overcome the limits of the current MALDI-TOF MS identification method using ribosomal protein spectra, we applied MALDI-TOF MS of low-mass profiling to the identification of two genetically related Bacillus species, the food-borne pathogen Bacillus cereus, and the insect pathogen Bacillus thuringiensis. The mass spectra of small molecules from 17 type strains of two bacilli were compared to the morphological, biochemical, and genetic identification methods of pathogens. The specific mass peaks in the low-mass range (m/z 500-3,000) successfully identified various closely-related strains belonging to these two reference species. The intensity profiles of the MALDI-TOF mass spectra clearly revealed the differences between the two genetically-related species at strain level. We suggest that small molecules with low molecular weight, 714.2 and 906.5 m/z can be potential mass biomarkers used for reliable identification of B. cereus and B. thuringiensis.

  • The food-borne pathogen Listeria monocytogenes is commonly associated with meats and unpasteurized dairy products. To identify this pathogen in meats more efficiently than has been done in the past, we purchased meats from Korean markets and performed sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and serotyping analysis on Listeria organisms isolated from meat samples. Each Listeria species showed specific protein band patterns on SDS-PAGE. Whole-cell protein SDS-PAGE profiles indicated that the organisms isolated from meats sold in local Korean markets were L. monocytogenes with the serotypes 1/2a, 1/2b, 1/2c, and 4b. We suggest that it is possible to carry out molecular subtyping of L. monocytogenes using SDS-PAGE.

  • This study was performed to investigate the contamination status of microorganisms that cause food poisoning in shellfish distributed in the metropolitan area of South Korea. The analyses were conducted according to the sample type, season, and region. In particular, the genotype of food poisoning viruses that explosively cause collective food poisoning was analyzed. Total 483 each of four types of shellfish (Crassostrea gigas, Mytilus coruscus, Pectea albicans albicans, and Scapharca subcrenata) were collected from four distribution sites located in the metropolitan area. We investigated foodborne pathogen by multiplex PCR and RT/Nested PCR from shellfish. As a result, Vibrio parahaemolyticus and Bacillus cereus were detected in 13 and 21 samples and Norovirus (NoV) GI and GII were detected in 7 and 12 samples, respectively. V. parahaemolyticus and NoV GI and GII showed differences in types of shellfish and seasons, and no correlation was confirmed with regional differences. Also, as a result of genotyping for the detected NoV GI and GII, they were confirmed as NoV GI.7, GI.5 and GII.3, which are causes of collective food poisoning. Therefore, cross-infection with shellfish can cause food poisoning. In particular, attention must be paid to the handling and cooking of shellfish in organizations that implement group feeding, and it is necessary to establish a management system for microorganisms that cause food poisoning in the process of shellfish farming and distribution.

  • This study was performed to acquire the information on the foodborne pathogen risk management programs in a couple of developed countries by the expert meeting and searching the information on the web. The backgrounds, strategies and effects related to microbial hazards of the foodborne pathogen reduction programs in fresh produce (US), Escherichia coli O157:H7 in ground beef (US), Salmonella in chicken, pork and eggs (Denmark), and Vibrio parahaemolyticus in seafood (Japan) were investigated for case study. A comparison among the pathogen reduction programs was conducted to find advantages and disadvantages and implications of the policies to bring out implications of the programs. A model for foodborne pathogen reduction program was developed based on both the CODEX risk management scheme and the case studies as follows; 1) preliminary risk management activities, 2) planing a foodborne pathogen reduction program, 3) option identification and selection, 4) implementation (conducting the each stake-holders role and applying the intervention methods), 5) monitoring activities, 6) interim review, 7) continuation or amendment of implementation method by the interim review before achieving the goal, and 8) final review and additional cost-benefit analysis if necessary. This proposed model according to the role of the stake-holders can be used to conduct microbial risk management programs in Korea in the near future.

  • Objectives : The analysis of prognosis about lumbago classes is important for low back patients. This study was designed to show the clinical significance about lowback pain patients based on 10 classes lumbago ill Dong-Eui-Bo-Kham. Methods : To analyze classify 80 patients who had received the hospital treatment for low back pain, they were classified according to 10 classes lumbago in Dong-Eui-Bo-Kham. To estimate the efficacy of oriental medical treatment, we used Quardruple Visual Analog Scale (QVAS). Results & Conclusion :1. According To 10 classes lumbago in Dong-Eui-Bo-Kham, the largest group of lumbago was due to muscle strain in 30(37.5%) cases, the next was due to wind pathogen group in 18(22.5%) cases, due to blood stasis group in 16(20%), due to Kidney deficiency by 12(15.0%), due to retention of food group by 2(2.5%), the lumbago due to humidity pathogen group by 2(2.5%) in order. 2. By the results which puts out the statistics in lumbago classes on a objectivity treatment record, the Lumbago due to blood stasis, the lumbago due to kidney deficiency, the lumbago due to muscle strain, the lumbago due to wind pathogen, the lumbago due to retention of food, the lumbago due to humidity pathogen showed $3.6({\pm}0.68),\;2.83({\pm}0.84),\;3.13({\pm}0.78),\;2.67{\pm}(0.77),\;3.00,\;3.00$. 3. • Fresh sprouts such as alfalfa, mung bean, radish, broccoli, and soybean sprouts have become very popular due do their high nutritional value. However, there have been several outbreaks of illness in the last few years that have been attributed to sprout consumption. A number of methods have been used to improve the safety of seed sprouts. One promising technology is the use of ionizing radiation treatment. Irradiation with doses up to 8 kGy has been approved in the USA to control microbial pathogens in seeds intended for sprout production. This review focuses on the potential use of ionizing radiation in reducing the pathogen levels in seed sprouts. The effects of irradiation on seed germination and the nutritional quality of the sprouts are discussed.
