Antimicrobial activity of Specific IgY against Streptococcus mutans (Streptococcus mutans에 대한 specific IgY의 항균력)

  • Antimicrobial effects of the specific IgY separated from eggs which were laid by hens vaccinated with Streptococcus mutans were investigated. The comparison tests of vaccination, addition levels of crude specific IgY, and innoculation concentration were applied by microscopic observation and turbidity test. Ten% addition of crude specific IgY obtained from vaccinated hens showed agglomerative clusters of S. mutans cells in supernatants and sediments, while crude IgY produced by non-vaccinated hens showed no cluster. IgY addition above 5% showed agglutinating clusters of most S. mutans cells and there was definite difference between IgY addition below 2.5% and above 5%. Concentration tests of crude IgY revealed that antimicrobial effects were differentiated by addition level and addition over 10% produced satisfactory results with turbidity test. The cluster size was dependent upon concentration of crude IgY addition. $10^5\;cfu/mL$ inoculation showed agglutinated cells and extent of agglomeration was proportional to cell numbers. Study of inoculation levels showed that 10% addition of crude IgY decreased turbidity effectively regardless of number of S. mutans cells. Plaque formation decreased to 75% with 15% addition of specific IgY concentration. These results implied that IgY separated from eggs laid by S. mutans-vaccinated hens might prevent dental caries caused by S. mutans.

Dietary Patterns among the Elderly in Jeollanam-do Area based on Their Physical and Mental Function State (전라남도 일부 지역 노인들의 신체적·정신적 기능 원활 정도에 따른 식생활 패턴의 차이)

  • This study investigated dietary patterns among the elderly over 75 years old living in Jeollanam-do area in May 2012. Although structured interviews were conducted with 236 consenting subjects, only 194 who completed the ADL, IADL, and K-MMSE tests were used for statistical data analysis. Using ADL, IADL, K-MMSE scores, cluster analysis was first performed and resulted in two groups: IFG (Insufficiently Functioning Group) and SFG (Sufficiently Functioning Group). Chi-square tests for nominal scales, Mann-Whitney tests for ordinal scales, and ANOVAs and t-tests for interval and ratio scales were conducted to compare two groups. More than 70% of IFG were illiterates compared to 28.1% of SFG. 'Excessive eating', 'appetite', 'digestion', and 'balanced diet' did not differ between groups. SFG more frequently had snacks and ate out and were more likely to take health supplements than IFG. Among the 100 major food items, consumption frequencies of several foods differed between groups. Study implications and limitations were discussed.

The Effects of Food Safety Accident on the Consumption of Eggs : Focusing on the Pesticide-related Accident (식품안전사고가 계란 소비에 미치는 영향: 계란의 살충제검출사고를 중심으로)

  • The objective of this study is to investigate the effect of pesticide-related accident on prices and sales of eggs and the perception of food safety accidents among consumers. For this, we analyzed the impact of the pesticide incident on consumers' purchases by separating large discount stores and eco-friendly specialty stores with econometrics methods. In addition, the value changes for each egg certification were analyzed after the accident. Perception of food safety accidents was conducted through a survey to investigate the awareness of the pesticide-related accidents, changes in purchases, and the causes of the pesticide accidents. Furthermore, the risk analysis was conducted. This results show the importance of trust and communication in food safety accidents between distributors, consumers and concerned authorities. Also, after the accident, consumers' interest and premium exist in the breeding process such as animal welfare, not only in the final product. Therefore in order to actively respond to food safety accidents such as pesticides-related accident, response and improvement are necessary considering various aspects such as risk assessment, risk management, and risk communication.

Urease Characteristics and Phylogenetic Status of Bacillus paralicheniformis

  • In 2015, Bacillus paralicheniformis was separated from B. licheniformis on the basis of phylogenomic and phylogenetic studies, and urease activity was reported as a phenotypic property that differentiates between the two species. Subsequently, we have found that the urease activity of B. paralicheniformis is strain-specific, and does not reliably discriminate between species, as strains having the same urease gene cluster were identified in B. licheniformis and B. sonorensis, the closest relatives of B. paralicheniformis. We developed a multilocus sequence typing scheme using eight housekeeping genes, adk, ccpA, glpF, gmk, ilvD, pur, spo0A, and tpi to clearly identify B. paralicheniformis from closely related Bacillus species and to find a molecular marker for the rapid identification of B. paralicheniformis. The scheme differentiated 33 B. paralicheniformis strains from 90 strains formerly identified as B. licheniformis. Among the eight housekeeping genes, spo0A possesses appropriate polymorphic sites for the design of a B. paralichenofomis-specific PCR primer set. The primer set designed in this study perfectly separated B. paralicheniformis from B. licheniformis and B. sonorensis.

Dietary Assessment Using Dietary Pattern Analysis of Middle School Students in Seoul (서울 지역 일부 중학생의 식이 패턴에 따른 식생활 평가)

  • The objectives of this study were to identify dietary patterns of Korean middle school students and to investigate the characteristics of dietary intake of subjects with different dietary pattern. Three-day diet records were obtained from 163 male and 155 female 7th graders in Seoul, Korea. Food items from the diet records were aggregated into 22 food groups before subjected to factor analysis. Four dietary patterns emerged from factor analysis with different factor score. Cluster analysis using factor score classified subjects into three groups named 'Traditional' (n = 42), 'Westernized' (n = 135), and 'Intermediate' (n = 145). Major nutrient intake and dietary quality assessed by NAR (Nutrients Adequacy Ratio), MAR (Mean Adequacy Ratio), DDS (Dietary Diversity Score), and DVS (Dietary Variety Score) of the three groups were compared. Mean energy intakes of three groups were 1783, 1916, 1578 kcal in Traditional, Westernized, and Intermediate diet group respectively. Differences in nutrient intake of the groups were significant in all nutrients except vitamin B$_1$. Percent energy from fat was significantly higher in Westernized and Intermediate diet group, and cholesterol intake of Westernized diet group was higher than 300mg. NARs of most nutrients were higher in Traditional and Westernized than Intermediate diet group except vitamin E. Traditional and Westernized diet groups had the highest MAR of fourteen nutrients. DDS was the highest in Westernized and DVS was the highest in Traditional and Westernized diet group. Traditional diet groups had 22% of energy consumption from breakfast, significantly higher than other diet groups. In conclusion, these results suggest that Korean teenagers with Traditional diet pattern have lower diet in % energy from fat, diversity of food and regularity of meals. Future studies need to focus on the relationship between dietary patterns and health status of Korean teenagers.

A Study of Singles' Food Consumption Behavior based on Food-related Lifestyle -On the Adults between 25 to 54 years old in the Seoul metropolitan area- (싱글족의 식생활 라이프스타일에 따른 식품구매행동 연구 - 서울 및 수도권지역 25세 이상 54세 이하 성인을 대상으로 -)

  • The purpose of this study was to investigate the differences is food consumption behavior based on singles' food-related lifestyle. A survey was conducted with singles(age 25~54) in the Seoul metropolitan area. Out of 2,051 questionnaires distributed, 300 were analyzed(15.21% response rate). The data was analyzed using SPSS windows(ver. 17.0). Singles' food-related lifestyles were categorized into three groups. In addition, the respondents were divided into three groups by cluster analysis: convenience oriented group, wellbeing dining-out oriented group, and uninvolved group. The food consumption behavior of each group was significantly different in terms of considerations of food consumption, preference of food product types, and breakfast types. The convenience oriented group prefered to buy ready to cook food, ready to heat food and ready to eat food. Moreover, they often eat instant food. The wellbeing dining-out group is more interested in foods and their health than other groups. The uninvolved group was not concerned about food. This study suggests that the food industry the characteristics of singles' needs to be analyzed more systematically. By analyzing the characteristics of singles', the food industry can use the data to establish a marketing strategy for them.

Diversity of Leuconostocs on Garlic Surface, an Extreme Environment

  • Thirty-nine strains of Leuconostocs found to be tolerant to $10\%$ or more garlic were selected for further identification, by comparing their whole-cell protein pattern, 16S rRNA gene (first 530 bases) sequence, cellular fatty acid composition, and carbon source metabolism. Two isolates were Identified as Leuconostoc mesenteroides and 32 others as Leuconostoc citreum. Five other strains belonging to a cluster could not be allocated to the existing species. 16S rRNA gene sequence and cellular fatty acid composition of the unidentified bacteria exhibited close similarity with Leuconostoc argentinum. The unidentified isolates were not allocated to L. argentinum, because they formed polysaccharide from sucrose, while L. argentinum strains do not. Leuconostocs tolerant to high concentration of garlic were found predominantly on garlic surface, an extreme environment which is unfit for most of other microorganisms.

Isolation and Identification of Urease-Positive Photobacterium sp. Strain HA-2 from Sea Water (해수에서 urease 양성 Photobacterium sp. Strain HA-2의 분리 및 동정)

  • A urease-positive bacterium isolated from sea water was identified as Photobacterium sp. by morphological, biochemical, and 16s rRNA gene analyses and named Photobacterium sp. strain HA-2. 2.0-fold increase enzyme activity was observed in LB medium containing 3% NaCl and 0.1% urea or not and the enzyme activity was 16.0-fold lower compared to urease-positive Vibrio parahaemolyticus AQ4673 strain when grown in the LB medium containing 3% NaCl with 0.1% urea. The cloning and sequencing of Photobacterium sp. strain HA-2 urease gene cluster is currently being analyzed in our laboratory.

Comparison of Level of Consumption Deprivation by the Patterns of Consumption Deprivation of Urban Poor Households in Inchon, Korea (도시빈곤가계의 소비박탈유형에 따른 소비박탈수준의 비교 : 인천시를 중심으로)

  • The purpose of this study is to classify the patterns of consumption deprivation and to compare the level of consumption deprivation by them. For empirical analysis, this study used the data on 563 households in the city of Inchon collected by the researchers through interviews. The statistical methods used in this study were cluster analysis, oneway ANOVA, and crosstabs. four clusters were classified: 'Overall satisfied Pattern (OSP)', 'Food satisfid with Others deprived Pattern (FS-ODP)', Food Extremely deprived with Food Deprive Pattern (HED-FDP)'. Those I FED-HSP tended to be old aged, and those in GED-FDP tended to be urban unstable workers. The results imply that welfare policy for urban poor households should be differentiated by the characteristics of consumption of each pattern of consumption deprivation.

A Study on the Food Service Selection Attributes and Consumption Behaviors based on Lifestyle Market Segments: Empirical Evidences from Luoyang (라이프스타일에 따른 세분시장별 외식 선택속성과 소비행동에 관한 연구: 중국 낙양지역을 대상으로)

  • The purpose of this study was to examine the market segments of Chinese dining-out customers based on their lifestyle. This study focused on the selection and consumption behavior of dining-out customers. The subjects of this study were 20 years old or older diners in Luoyang, China, and the data were collected for 11 days from April 5, 2016. 400 questionnaires were distributed, and 390 copies were collected. After excluding 9 inadequate questionnaires, 381 responses were used for data analysis by using IBM SPSS 23.0, and Data analysis included frequency analysis, cluster analysis, one-way ANOVA, and cross tabulation. The results of empirical analysis showed that there was a significant difference in selection attributes, consumption behavior and demographic characteristics in terms of lifestyle market segments.
