Effects on Reproduction Efficiency of Estrous Status in Thoroughbred Mares During the Breeding Season (더러브렛 암말의 번식기 발정상태가 번식효율에 미치는 영향)

  • The purpose of this study was to ascertain the breeding efficiency in Thoroughbred mare. A total of 106 mares were investigated for the status of follicle (462 cases), ovulation (179 cases) and pregnancy (346 cases). Of total examination, 46.8% was follicle measure to determine breeding time, and mating rate per cases examined was 39.9%. There was no correlation between reproductive results and size of follicles or endometrial edema or degrees of teasing alone. 143 cases were ovulated among 179 cases which were performed ovulation examination, and ovulation rate and fertilization rate per mating times were 79.9% and 39.0%, respectively. The use of hCG(human chorionic gonadotropin), to facilitate ovulation, presented to increase occurrence of double ovulations and twin fertilizations In conclusion, though more examination to estimate the optimal breeding time and higher mating rate was performed, fertilization rate per mating times was lower and then reproductive efficiency also became decreased. Therefore, it seemed that accurate examination of reproductive tracks, appropriate teasing programme and hCG administration before ovulation were of help to improve ovulation rate and fertilization rate.

Pattern of Ovulation in Korean Women with Premature Ovarian Failure (한국인 조기난소부전 여성의 배란양상)

  • Objective: Significant portion of premature ovarian failure patients ovulate intermittently and even can achieve pregnancy. The clinical features and reproductive potentials of Korean women with premature ovarian failure have never been elucidated. The purpose of this study is to assess ovarian follicular growth and ovulatory potential. Methods: Complete medical records of fifteen women with idiopathic premature ovarian failure were reviewed retrospectively. Subjects with more than 6 ultrasonographic scans and 36 months of follow-up period were included. Once the ovarian follicle was found by ultrasonography, the follicular growth was serially monitored. The result of ovulation induction of 6 cycles in 4 patients were also evaluated. Rate of positive ovarian follicle by ultrasound, rate of natural follicular growth and response of ovulation induction were assessed. Results: Among the 15 patients, 6 women (40%) had ovarian follicle in random ultrasonography. But the chance of positive follicle per ultrasonography scan was 7% (8 out of 115 scans). One out of 6 women eventually ovulated during the follow-up scan. In remaining 5 subjects, size of follicles remained unchanged. None of 6 ovulation induction cycles resulted successful ovulation. Conclusion: Incidence of positive ovarian follicle in Korean premature ovarian failure patients is lower than that of Western counterpart. This result, however, could not be directly compared because of the differences of study design. The ovulation rate by medical ovulation induction in this group is 0%, which is worse than the reported success rate of Western studies.

Ovarian Follicular Dynamics Monitored by Real-Time Ultrasonography during Oestrous Cycle in Buffalo (Bubalus bubalis)

  • Application of trans rectal ultrasonography to buffaloes (n=6) to follow the growth of large antral follicles individually, on each day of one interovulatory interval revealed that follicular turnover during oestrous cycle occured in waves. There was a predominance of a two-wave pattern (5/6 animals) compared to that of a three-wave pattern (1/6 animals). For two-wave pattern, the first wave emerged at Day $0.20{\pm}0.19$ (Day 0 = day of ovulation) and was marked by development of a dominant anovulatory follicle which grew in size from $5.40{\pm}0.24mm$ at the day of detection to a maximum diameter of $12.40{\pm}0.81mm$ on Day $8.60{\pm}1.57$, with a growth rate of $0.88{\pm}0.17mm/day$ and then regressed, with a mean persistence of $19.40{\pm}1.54$ days. The second wave emerged at Day $9.20{\pm}1.06$ and was marked by development of a dominant ovulatory follicle which grew in size from $4.20{\pm}0.37mm$ at the day of detection to a maximum diameter of $13.80{\pm}0.37mm$ on Day $21.00{\pm}1.38$, with a growth rate of $0.66{\pm}0.12mm/day$ and then ovulated on Day $21.60{\pm}1.25$, with a mean persistence of $11.80{\pm}1.39$ days. The maximum diameters attained and the growth rates of dominant anovulatory and dominant ovulatory follicles, and the mean number of follicles ${\geq}3mm$ diameter detected at the time of emergence of first and second waves ($11.80{\pm}1.74$ and $9.00{\pm}2.81$, respectively) were not significantly different. In the animal which showed a three-wave pattern, the first, second and third waves emerged on Days 1, 10 and 19, respectively. All animals, except one had at least one subordinate follicle in the first or second or both waves. The subordinate follicles increased in diameter over a few days and then regressed. The results indicate that in buffaloes, the follicular turnover during oestrous cycle occurs predominantly in a two-wave pattern.

Monoterpenoid Loliolide Regulates Hair Follicle Inductivity of Human Dermal Papilla Cells by Activating the AKT/β-Catenin Signaling Pathway

  • Loliolide is one of the most ubiquitous monoterpenoid compounds found in algae, and its potential therapeutic effect on various dermatological conditions via agent-induced biological functions, including anti-oxidative and anti-apoptotic properties, was demonstrated. Here, we investigated the effects of loliolide on hair growth in dermal papilla (DP) cells, the main components regulating hair growth and loss conditions. For this purpose, we used a three-dimensional (3D) DP spheroid model that mimics the in vivo hair follicle system. Biochemical assays showed that low doses of loliolide increased the viability and size of 3D DP spheroids in a dose-dependent manner. This result correlated with increases in expression levels of hair growth-related autocrine factors including VEGF, IGF-1, and KGF. Immunoblotting and luciferase-reporter assays further revealed that loliolide induced AKT phosphorylation, and this effect led to stabilization of β-catenin, which plays a crucial role in the hair-inductive properties of DP cells. Further experiments showed that loliolide increased the expression levels of the DP signature genes, ALP, BMP2, VCAN, and HEY1. Furthermore, conditioned media from loliolide-treated DP spheroids significantly enhanced proliferation and the expression of hair growth regulatory genes in keratinocytes. These results suggested that loliolide could function in the hair growth inductivity of DP cells via the AKT/β-catenin signaling pathway.

Coping with large litters: the management of neonatal piglets and sow reproduction

  • As a result of intensive breeding, litter size has considerably increased in pig production over the last three decades. This has resulted in an increase in farrowing complications. Prolonged farrowing will shorten the window for suckling colostrum and reduce the chances for high-quality colostrum intake. Studies also agree that increasing litter sizes concomitantly resulted in decreased piglet birth weight and increased within-litter birth weight variations. Birth weight, however, is one of the critical factors affecting the prognosis of colostrum intake, and piglet growth, welfare, and survival. Litters of uneven birth weight distribution will suffer and lead to increased piglet mortality before weaning. The proper management is key to handle the situation. Feeding strategies before farrowing, management routines during parturition (e.g., drying and moving piglets to the udder and cross-fostering) and feeding an energy source to piglets after birth may be beneficial management tools with large litters. Insulin-like growth factor 1 (IGF-1)-driven recovery from energy losses during lactation appears critical for supporting follicle development, the viability of oocytes and embryos, and, eventually, litter uniformity. This paper explores certain management routines for neonatal piglets that can lead to the optimization of their colostrum intake and thereby their survival in large litters. In addition, this paper reviews the evidence concerning nutritional factors, particularly lactation feeding that may reduce the loss of sow body reserves, affecting the growth of the next oocyte generation. In conclusion, decreasing birth weight and compromised immunity are subjects warranting investigation in the search for novel management tools. Furthermore, to increase litter uniformity, more focus should be placed on nutritional factors that affect IGF-1-driven follicle development before ovulation.

Characterization of Genes Related to the Cell Size Growth and CCN Family According to the Early Folliculogenesis in the Mouse (쥐의 초기 난포 발달에 관여하는 Cell Size Growth 및 CCN Family 유전자에 관한 연구)

  • Objectives: Previously, we sought to compile a list of genes expressed during early folliculogenesis by using cDNA microarray to investigate follicular gene expression and changes during primordialprimary follicle transition and development of secondary follicles (Yoon et al., 2005). Among those genes, a group of genes related to the cell size growth was characterized during the ovarian development in the present study. Methods: We determined ovarian expression pattern of six genes related to the cell size growth (cyr61, emp1, fhl1, socs2, wig1 and wisp1) and extended into CCN family (${\underline{c}}onnective$ tissue growth factor/${\underline{c}}ysteine$-rich 61/${\underline{n}}ephroblastoma$-overexpressed), ctgf, nov, wisp2, wisp3, including cyr61 and wisp1 genes. Expression of mRNA and protein according to the ovarian developmental stage was evaluated by in situ hybridization, and/or semiquantitative reverse transcriptase polymerase chain reaction (RT-PCR), and immunohistochemistry, respectively. Results: Among 6 genes related to the cell size growth, cyr61 and wisp1 mRNA was detected only in oocytes in the postnatal day5 mouse ovaries. cyr61 mRNA expression was limited to the nucleolus of oocytes, while wisp1 was expressed in the cytoplasm and nucleolus of oocytes, except nucleus. cyr61 mRNA expression, however, was found in granulosa cells from secondary follicles. The rest 4 genes in the cell size growth group were detected in oocytes, granulosa and theca cells. Cyr61 and Wisp1 proteins were expressed in the oocyte cytoplasm from primordial follicle stage. Especially, Cyr61 protein was detected in pre-granulosa cells, Wisp1 protein was not. By using RT-PCR, we evaluated and decided that Cyr61 protein is produced by their own mRNA in pre-granulosa cells that was not detected by in situ hybridization. cyr61 and wisp1 genes are happen to be the CCN family members. The other members of CCN family were also studied, but their expression was detected in oocytes, granulose and theca cells. Conclusions: We firstly characterized the ovarian expression of genes related to the cell size growth and CCN family according to the early folliculogenesis. Cyr61 protein expression in the pre-granulosa cells is profound in meaning. Further functional analysis for cyr61 in early folliculogenesis is under investigation.

A Comparative Analysis of Basal Body Temperature to Ultrasound, as a Method of Ovulation Detection in Induced Ovulatory Menstrual Cycles (배란유도주기에 따른 초음파검사와 기초체온표의 비교분석)

  • Four points on the basal body temperature (B.B.T.) curve was correlated with the estimated time of ovulation, as determined by serial ultrasound in 50 induced menstrual cycles from 22 subjects. The time of ovulation was estimated by measuring the maximal diameter of follicles and observing the morphologic changes within the ovary from follicle to corpus luteum. The results were as following; 1. The diameter of the follicle measured at the day before disappearance was 21.1 mm on an average (S.D.: 2.14). The average follicular growth for 4 days before ovulation was measured at a rate of 2.8 mm/day, and rapid growth of follicle was observed 3.1 mm/day at the day before. 2. The changes associated with rupture of the follicles were the followings, in order of frequency; decrease in size(94%), disappearance of follicles(64%), fluid in the Cul-de-Sac(26%) and increased internal echoes(16%). 3. Only 20 of 50 cycles, exhibited a BBT dip and correlated with the estimated time of ovulation by ultrasound in 2 of which cases(10%). BBT nadir, 30 of 50 cycles, correlated in 5(16.7%). The first day of hyperthermic plateau(FDHP) and BBT coverline was exhibited in all cycles, correlated in 41(82%) and 35(70%) cases. 4. The relationship between the diameter of dominant dominant follicle, measured by ultrasound, and the basal body temperature curve were as following. During cycles in which dip was observed on the BBT curve, the follicular diameter were 10.5${\pm}$2.12 mm on 4 days prior to the point (D-4), and 12.5${\pm}$2.12 mm (D-3), 15.5${\pm$2.12 mm (D-2), 17.0${\pm}$1.41 mm (D-1) and 21.5${\pm}$2.12 mm just prior to the dip (D-0). In the nadir; 9.6${\pm}$1.67 mm (N-4), 12.8${\pm}$1.79 mm (N-3), 16.2${\pm}$1.92 mm (N-2), 18.2${\pm}$2.17 mm (N-1) and 21.4${\pm}$2.61 mm (N-0). In the First day of Hyperthemic Plateau (FDHP); 9.8${\pm}$1.36 mm (F-4), 12.4${\pm}$1.41 mm (F-3),15.1${\pm}$1.57 mm (F-2), 18.1${\pm}$1.67 mm (F-1) and 21.2${\pm}$2.25 mm (F-0). In the BBT coverline endopint; 9.9${\pm}$.39 mm (C-4), 12.5 ${\pm}$1.44 mm (C-3), 15.2${\pm}$1.64 mm (C-2), 18.0 ${\pm}$1.69 mm (C-1), and 21.2${\pm}$2.31 mm (C-0). 5. The relationship between the ultrasonographic signs of ovulation and the basal body temperature curve were as following. The BBT dip correlated with the ovulation in 2 cases, which revealed decrease in follicular diameter (100%), fluid pattem in the Cul-de-Sac (1 case, 50%) and complete disappearance of follicle (1 case, 50%). In the nadir (5 cases); the ultrasonographic signs of ovulation were decrease in follicular diameter (5 cases, 100%), fluid pattern in the Cul-de-Sac (1 case, 20%) and complete disappearance of follicle (3 cases, 60%). In the First day of Hyperthermic Plateau (41 cases); decrease in follicular diameter (40 cases, 97.6%), fluid pattern in the Cul-de-Sac (11 cases, 26.8%), appearance of internal echo and thickening of the wall (6 cases, 14.6%) and com plete disappearance of follicle (28 cases, 68.3%). In the BBT coverline endpoint (35 cases); decrease in follicular diameter (33 cases, 94.3%), fluid pattern in the Cul-de Sac (9 cases, 25.7%), appearance of internal echo and thickening of the wall (5 cases 14.3%) and complete disappearance of follicle (20 cases, 57.1%).

Effect of FSH Pretreatment on Follicle Development and Oocyte Recovery by Ultrasound-guided Aspiration in Holstein Heifers (젖소의 초음파 유도 채란율에 대한 FSH 전처리 효과의 비교)

  • Ultrasound-guided follicular aspiration was performed in Holstein heifers once weekly with or without pretreatment of single or multiple decreasing doses using a total of 400 mg FSH. Oocytes were aspirated with a 6.5 MHz convex-array ultrasound trasducer designed for intravaginal use. All the visible follicles larger than 4 mm in diameter were punctured with a 17 gauge, 55 cm needle at each aspiration session and the follicular fluids containing oocytes were obtained by vacuum suction. The results obtained were as follows: As a preliminary experiment, the recovery rates of folicular oocytes by ultrasound-guided aspiration from the isolated ovaries of Korean native cows were compared between suction methods using manual syringe or vacuum pump. The recovery rate of oocytes using vacuum pump (80.7%) was significantly (P<0.05) higher than that using manual syringe (47.1%). The follicles were counted by their size in diameter with ultrasound image, and recovery rates and grades of follicular oocytes collected by ultrasound-guided aspiration were investigated in Holstein heifers pretreated with or without FSH. A group of heifiers were injected with multiple decreasing doses (twice a day for 3 days) of a total of 400 mg FSH. The other 2 groups were injected with a single dose of 400 mg FSH mixed with 25% PVP. Ultrasound observation of follicle population and/or ultrasound-guided transvaginal oocyte aspiration were performed 12 hrs following the last FSH injection in the multiple dose group, and 48 or 60 hrs after FSH injection in the single dose groups. Most of the visible follicles had small size of less than 3 mm in diameter in unstimulated heifers (71.0%), but medium size in all the heifers treated with FSH. (70.5 to 92.8%). The number of OPU follicles per session (4.6$\pm$1.9) were much less, compared to the vilsible follicle counts (9.7$\pm$2.2), in the nustimulated heifers due to the small dominant follicles. Among 4 goups of heifers the most visible as well as OPU follicles were observed in the heifers at 60 hrs following treatment of a single dose of 400 mg FSH (21.2$\pm$2.3 and 21.0$\pm$2.0), and the differences in both the follicle counts between the groups was found significant (P<0.05) The rates of oocyte recovery from the follicles by ultrasound-guilded aspiration were varied 46.3 to 75.0% in the heifers unstimulated and treated with a single dose of 400 mg FSH, but the group difference was not significant. The number of recovered oocytes per session a, pp.ared to be highest at aspiration at 60 hrs following single FSH (10.6$\pm$2.2) than at aspiration at 48 hrs after single FSH (7.8$\pm$2.7) or in the unstimulated heifers (3.4$\pm$3.0). The proportion of grade I and II oocytes to all oocytes collected was varied 31.8 to 64.0% between the groups. However, there was found no significant difference in both the number of oocytes recovered per session and the percentage and the percentage of grade I and II oocytes. From the above results it was concluded that the more oocytes of superior quality might be recovered economically by ultrasound-guided aspiration at 60 hrs following the pretreatment of a single dose of 400 mg FSH and by suction using a vacuum pump system of about negative pressure of 75 to 85 mmHg.

  • This study was carried out to investigate the ultrastructural changes of ooplasm and follicular membrane of oocytes, obtained from 150 of 3-year-old female rainbow trout(Oncorhynchus mykiss). All data were collected from March in 1989 to February in 1990, and from August to October in 1991. The size of the nucleoli and number of the yolk granules increased as the oocyte growed. Yolk granules were deposited in the oocyte as crystalline granules. Due to the presence of large early and late maturing oocytes, their ovaires were enlarged, transparent and granular. The lattice was broken down at hydration, leaving the egg transparent. As thepercentages of fish in LPO and EMO stage increased from September to October, Mean GSI values increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in bothgonadosomatic index and macroscopic apearance. Under the natural conditions,t he ovarian follicle influences the histological development and periodical secretion of the hormones, sufficient for a oogenesis and gonadal steroid production. The electrophoretic pattern of major band in mature stage was much thicker(70∼110k dalton) than that in previtellogenic phase.
