These experiments were carried out to develop a novel, simple, and rapid method to determine urinary sulfonamides using fluorescamine by spectrofluorometry. To get optimal conditions for the sulfonamide-fluorescamine reaction, sulfonamides such as sulfamethazine, sulfamerazine, sulfadimethoxine and sulfamonomethoxine, dissolved in buffers with various pH ranges were reacted with various concentrations of fluorescamine. and then, the fluorescence intensity and stability of the fluorophore were measured. To eliminate the interfering substances in urine, the fluorophore in buffers and urine with a definite pH range was extracted with some organic solvents. After then the fluorescence intensity was measured in organic and aquous phases. The results obtained were summarized as follows: 1. The maximal fluorescence of sulfonamides was presented in acidic state, pH 4.5~5.0, at 30 minutes after reaction. 2. The optimal concentration ratio of sulfamethazine and ffuorescamine was more than 1 : 40 in mole. 3. In pH 4.0, the intensity was maximal but was time-dependent, whereas in pH 8.0, the intensity was time-independent. 4. Sulfamethazine-fluorescamine conjugate could be dissolved in some of organic solvents in acidic state such as chloroform, n-butanol, and ethylacetate. 5. Sulfamethazine-flnorescamine conjugate in swine urine coule be extracted with ethylacetate in acidic state, pH 4.0~5.0.
Recent social interests on mad cow disease and Creutzfelt-Jacob disease lead researcher' focus onto pathogenic prion proteins causing those diseases. The purpose of this study is to introduce a novel method for micromolecular level quantification of PrP 106-126 peptide, which is a part of prion protein. Fluorescamine has been chosen due to its fluorescence emission characteristics upon reaction with primary amines and Alpha-imager is used to detect the intensity of fluorescence. We succeeded in setting optimal conditions for quantification of PrP 106-206, amyloidogenic prion peptide, at micromolecular level. This study will contribute to identify prion protein aggregation inhibitor and develop new prion protein drug.
Hair is made of proteins containing various amino acids. Ultraviolet (UV) radiation is believed to be responsible for the most damaging effects of sunlight, and also plays an important role in hair aging. The purpose of this study was to investigate the changes in morphological and chemical structures after ultraviolet B (UVB) irradiation of human hair. The UVB-irradiated hair showed characteristic morphological and structural changes, compared to those of the normal hair. The result from a scanning electron microscope (SEM) equipped with an energy dispersive X-ray diffractometer (EDX) showed that the scale of UV-irradiated hair appeared to be rough and the amount of oxygen element was higher than that of the normal hair. Fluorescence and three dimensional (3D) topographical images were obtained by a confocal laser scanning microscope (CLSM). In 3D images, the green emission intensity of normal hair was much higher than that of fluorescing UVB-irradiated hair. The intensity of green emission reflects the intrinsic fluorescence of hair protein. Also, a fluorescent imaging method using fluorescamine reagent was used to identify the free amino groups resulting from a peptide bond breakage in UVB-irradiated hair. Strong blue fluorescence of UVB-irradiated hair, which indicates a very high level of amino groups, was observed by CLSM. Therefore, the fluorescamine as an extrinsic fluorescence could provide a useful tool to identify the peptide bond breakage in UVB-irradiated hair. Infrared image mapping was also employed to assess the cross-sections of normal and UVB-irradiated specimens to examine the oxidation of disulfide bonds. The degree of peak areas with strong absorbance for the disulfide mono-oxide was spread from the outside to the inside of hair. The spectroscopic techniques used alone, or in combination, launch new possibilities in the field of hair cosmetics.

Copolymers containing oxime-urethane groups, which led to the formation of pendant amino groups photochemically, were applied to a fluorescent image-recording material through the reaction with fluorescamine, a prefluorescent dye for amino groups. This imaging method permits the fluorescent image to be erased or restored by treatment with base or acid. Copolymers containing phthalimide carbamate groups were applied to a bicolor fluorescent imaging material through the consecutive reaction with fluorescamine and rhodamine. A various colored fluorescent micropattern (green, red or red-yellow) was observed through the changes of excitation wavelength by using a conforcal microscope.
A fluorometric method is described which permits the assay of trypsin inhibitor contained in a sample in an extremely small amount, utilizing a novel reagent generally called fluorescamine. The fluorometric assay with an enzyme kinetic approach has been found to be at least 100 times more sensitive than the well-known Kunitz's spectrophotometric method, considerably taster and less complicated, when it was demonstrated with the anti tryptic activities of very dilute extracts from soybean, red-bean and mung bean. Details of experimental procedure as well as theoretical considerations will be discussed.
Novel chitosan beads containing a series of bisamino-terminated polyethylene glycol (Bisamino-PEG, $Jeffamine^{(R)}ED$ series) have been prepared via capillary extrusion method using an alkaline solution. The results of swelling kinetics of chitosan beads showed that as the chain length of PEG in chitosan beads increased, the swelling process of the beads proceeded slowly. In order to study the releasing kinetics quantitatively, fluorescamine was coupled to the pendant amino groups and the releasing processes were followed by UV spectra. The results revealed that the releasing process was retarded in the order of $Jeffamine^{(R)}ED$-600 < $Jeffamine^{(R)}ED$-900 < $Jeffamine^{(R)}ED$-2001 as the chain length of PEG was increased. The slow release of PEG from the beads is considered to be governed by the chain length of PEG and interpolymeric hydrogen bonding between PEG and the chitosan molecule.
Several chitosan beads were prepared by W/O emulsion and capillary extrusion methods. The surface structures were observed through SEM pictures, the surface areas were determined by BET method, the available amine contents were measured by picric acid titration method, and the results were correlated each other. The beads from W/O emulsion method were smaller in size but had higher hardness, surface area, and available amine contents than the ones from capillary extrusion method. Moreover, the surface area, available amine contents, apparent density and the hardness showed large differences according to the drying processes. As compared between the solvent dehydration and the freeze drying technique in W/O emulsion method, the former showed higher surface area, higher available amine contents and hardness than the latter. SEM pictures revealed that in case of solvent dehydration method, very small pores existed, whereas in case of freeze drying method, cell-like pores existed on the surface. The results of picric acid titration showed that the beads dried by solvent dehydration method were 10 times higher in amine contents than those from freeze drying method, and the amount of amine contents were increased as the surface area was increased. After fluorescamine was reacted with chitosan beads, the fluorescence labeled beads were observed through fluorescence microscope. The results showed that most of primary amine existed on the surface of beads and pores.
Sulfonamides are widely used to treat mastitis of cattle in field. The study was carried out to analyze sulfonamid residues In raw milk from south Kyeonggi area. The milk sample was deproteinated with acetone and defatted with hexane. The residual sulfonamides were extracted with ethylacetate, concentrated under vaccum, reconstituted with the acetate buffer-methanol mixture, reacted with fluorescamine, and then analyzed by HPLC-fluorescence detector(EX. 39nm, Em. 495nm) with methanol : acetate buffer system(3/2, v /v) as a mobile phase. The results analyzed by Thin layer chromatography and High performance liquid chromatography were summarized as follows. 1. A total of 24 cases out of 478 raw milk samples(5.0%) collected during April and May showed positive reaction to sulfonamide residues. Among 24 positive reactors, 9 cases were positive to sulfanilamides(32.1%), 8 cases were positive to sulfathiazoles(28.6%) and 5 cases were positive to sulfamonomthoxines (7.9%) , respectively. 2. During July and August, 31 cases out of 464 raw milk samples(6.7%) showed positive re action to sulfonamide residues. Among them S cases were positive to sulfanilamides, 5 cases sulfathiazoles and 5 cases sulfamonomethoxines(16.1%), respectively.
