Currently, enzymatic synthesis of lactulose, a synthetic prebiotic disaccharide, is commonly performed with glycosyl hydrolases. In this work, a new type of lactulose-producing biocatalyst was developed by displaying β-galactosidase from Bacillus stearothermophilus IAM11001 (Bs-β-Gal) on the surface of Bacillus subtilis 168 spores. Localization of β-Gal on the spore surface as a fusion to CotX was verified by western blot analysis, immunofluorescence microscopy, and flow cytometry. The optimum pH and temperature for the resulting spore-displayed β-Gal was 6.0 and 75℃, respectively. Under optimal conditions, it showed maximum activity of 0.42 U/mg spores (dry weight). Moreover, the spore-displayed CotX-β-Gal was employed as a whole cell biocatalyst to produce lactulose, yielding 8.8 g/l from 200 g/l lactose and 100 g/l fructose. Reusability tests showed that the spore-displayed CotX-β-Gal retained around 30.3% of its initial activity after eight successive conversion cycles. These results suggest that the CotX-mediated spore-displayed β-Gal may provide a promising strategy for lactulose production.

The previous work (Cheong et al., 2006) where the characteristics of acoustic emissions of wind turbines has been investigated according to the methods of power regulation, has showed that the acoustic power of wind turbine using the stall control for power regulation is more correlated with the wind speed than that using the pitch control. In this paper, basically extending this work, the noise generation characteristics of large modern upwind wind turbines are experimentally indentified according to the power regulation methods. To investigate the noise generation mechanisms, the distribution of noise sources in the rotor plane is measured by using the Beam-forming measurement system (B&K 7768, 7752, WA0890) consisting of 48 microphones. The array results for the 660 kW wind turbine show that all noise is produced during the downward movement of the blades. This result show good agreement with the theoretical result using the empirical formula with the parameters: the convective amplification; trailing edge noise directivity; flow-speed dependence. This agreement implies that the trailing edge noise is dominant over the whole frequency range of the noise from the 660 kW wind turbine using the pitch control for power regulation.

In this paper, a moving object detection and face region extraction algorithm which can be used in video monitoring systems is presented. The proposed algorithm is composed of two stages. In the first stage, each frame of an input video sequence is analyzed using three measures which are based on image pixel difference. If the current frame contains moving objects, their skin regions are extracted using color and frame difference information in the second stage. Since the proposed algorithm does not rely on computationally expensive features like optical flow, it is well suited for real-time applications. Experimental results tested on various sequences have shown the robustness of the proposed algorithm.
Modeling of physically non-linear behavior becomes more and more important for the analysis of SFRC structures in practical applications. From this point of view we will present an effective, three-dimensional constitutive model for SFRC, that is also easy to implement in commercial finite element programs. Additionally, the finite element analysis should only require standard material parameters which can be gained easily from conventional experiments or which are specified in appropriate building codes. Another important point is attaining the material parameters from experimental data. The procedures to determine the material parameters proposed in appropriate codes seem to be only approximations and are unsuitable for precise structural analysis. Therefore a finite element analysis of the test itself is used to get the material parameters. This process is also denoted as inverse analysis. The efficiency of the proposed constitutive model is demonstrated on the basis of numerical examples and their comparison to experimental results. In the framework of material parameter identification the idea of a new, indirect tension testing procedure, the "Modified Tension Test", is adopted and extended to an easy-to-carry-out tension test for steel fiber reinforced concrete specimens.

Tidal power is one of new and renewable energy sources. The seawater is stored inside a tidal embankment built at the mouth of a river or bay, where tides ebb and flow. The water turbine-generators produce power by exploiting the gap in the water level between the water outside and inside the embankment. Tidal power plant is a large plant that is installed on the sea. And then, the facility's operations and a separate control system for monitoring and maintenance is required. However, this plant predictive control of building systems and technologies have been avoided the transfer of technology from advanced global companies. Accordingly, the control system for core technology development and localization is urgently needed. This paper presents modeling and simulation using by PSS/E about generator, governor, exciter, and power system stabilizer for control system in Sihwa tidal power plant to improve the efficiency and develope of core technology. And the dynamic characteristics of governor and exciter were analyzed.

It is well known that during tensile testing, a part of the mechanical work done on the specimen is transformed into heat energy. However, the ultimate temperature rise and the rate of temperature rise is related to the nature of the material, conditions of the test and also to the deformation behaviour of the material during loading. The recent advances in infrared sensors and image/data processing techniques enable observation and quantitative analysis of the heat energy dissipated during such tensile tests. In this study, infrared imaging technique has been used to characterise the tensile deformation in AISI type 316 nuclear grade stainless steel. Apart from identifying the different stages during tensile deformation, the technique provided an accurate full-field temperature image by which the point and time of strain localization could be identified. The technique makes it possible to visualise the region of deformation and failure and also predict the exact region of fracture in advance. The effect of thermal gradients on plastic flow in the case of interrupted straining revealed that the interruption of strain and restraining at a lower strain rate not only delays the growth of the temperature gradient, but the temperature rise per unit strain decreases. The technique is a potential NDE tool that can be used for on-line detection of thermal gradients developed during extrusion and metal forming process which can be used for ensuring uniform distribution of plastic strain.
In this study, we used more reliable experimental materials and methods to detect the effects of osteopontin (OPN) on boar sperm in vitro capacitation, acrosome reaction, and fertilization efficiency. We reorganized and obtained the OPN protein of the porcine source. Immunofluorescence and Western blot show the localization and expression of the OPN protein before and after sperm capacitation. To determine whether OPN can affect sperm during sperm capacitation, we examined cyclic adenosine monophosphate (cAMP) concentrations after sperm capacitation, and the results showed that OPN significantly increased the cAMP concentration in sperm (p < 0.05). Flow cytometry showed that 0.1 ㎍/mL OPN-treated sperm had better acrosome reaction ability. In vitro fertilization (IVF) showed that 0.1 ㎍/mL OPN significantly increased the rate of embryo division. In conclusion, this study found that 0.1 ㎍/mL porcine OPN protein can significantly improve porcine capacitated sperm motility, cAMP concentration after capacitation sperm, acrosome reaction ability, and embryo division during IVF and provides new clues to explore the mechanism of OPN's function on sperm.

Unmanned patrol robots are currently being developed for autonomous data survey in construction sites. As the amount of data acquired by robots increases, it is important to utilize proper metadata and system to manage data flow. In this study, we developed three materials, metadata design, robot system and web system, in the purpose of automating construction site data survey using unmanned patrol robots. The metadata was mainly designed to represent when and where raw data was acquired. To identify the location of data acquired, localization data from SLAM algorithm was converted to suit the construction drawings. The robot system and web system were developed to generate, store and parse the raw data and metadata automatically. The materials developed in this study was adopted to Boston Dynamics SPOT, a quadruped robot. Autonomous data survey of 360-picture and environment sensor was tested in two construction sites and the robot worked as intended. As a further study, development on the autonomous data survey to improve the convenience and productivity will be continued.
Trichomonas vaginalis is a pathogen that triggers severe immune responses in hosts. T. vaginalis ${\alpha}$-actinin 2 ($Tv{\alpha}$-actinin 2) has been used to diagnose trichomoniasis. $Tv{\alpha}$-actinin 2 was dissected into three parts; the N-terminal, central, and C-terminal portions of the protein (#1, #2, and #3, respectively). Western blot of these $Tv{\alpha}$-actinin 2 proteins with pooled patients' sera indicated that #2 and #3, but not #1, reacted with those sera. Immunofluorescence assays of two different forms of T. vaginalis (trophozoites and amoeboid forms), using anti-$Tv{\alpha}$- actinin 2 antibodies, showed localization of $Tv{\alpha}$-actinin 2 close to the plasma membranes of the amoeboid form. Fractionation experiments indicated the presence of $Tv{\alpha}$-actinin 2 in cytoplasmic, membrane, and secreted proteins of T. vaginalis. Binding of fluorescence-labeled Trichomonas to vaginal epithelial cells and prostate cells was decreased in the antibody blocking experiment using anti-$Tv{\alpha}$-actinin 2 antibodies. Pretreatment of T. vaginalis with anti-$rTv{\alpha}$-actinin 2 antibodies also resulted in reduction in its cytotoxicity. Flow cytometry, ligand-binding immunoblotting assay, and observation by fluorescence microscopy were used to detect the binding of recombinant $Tv{\alpha}$-actinin 2 to human epithelial cell lines. Specifically, the truncated N-terminal portion of $Tv{\alpha}$-actinin 2, $Tv{\alpha}$-actinin 2 #1, was shown to bind directly to vaginal epithelial cells. These data suggest that ${\alpha}$-actinin 2 is one of the virulence factors responsible for the pathogenesis of T. vaginalis by serving as an adhesin to the host cells.

Myoblast are myogenic precursors that proliferate, activate, and differentiate on muscle injury to sustain the regenerative capacity of skeletal muscle; The neuronal isoform of nitric oxide synthase (nNOS, termed also NOS-I) is expressed in normal adult skeletal muscle, suggesting important functions for Nitric oxide (NO) in muscle biology1,2,3. However, the expression and subcellular localization of NO in muscle development and myoblast differentiation are largely unknown. In this study, we examined effects of the nitric oxide generated by a microwave plasma torch, on proliferation/differentiation of rat myoblastic L6 cells. Experimental data pertaining to nitric oxide production are presented in terms of the oxygen input in units of cubic centimetres per minute. The various levels of nitric oxide are observed depending on the flow rate of nitrogen gas, the ratio of oxygen gas, and the microwave power4. In order to evaluate the potential of nitric oxide as an activator of cell differentiation, we applied nitric oxide generated from the microwave plasma torch to L6 skeletal muscles. Differentiation of L6 cells into myotubes was significantly enhanced the differentiation after nitric oxide treatment. Nitric oxide treatment also increase the expression of myogenesis marker proteins and mRNA level, such as myogenin and myosin heavy chain (MHC), as well as cyclic guanosine monophosphate (cGMP), However during the myotube differentiation we found that NO activate oxidative stress signaling erks expression. Therefore, these results establish a role of NO and cGMP in regulating myoblast differentiation and elucidate their mechanism of action, providing a direct link with oxidative stress signalling, which is a key player in myogenesis. Based on these findings, nitric oxide generated by plasma can be used as a possible activator of cell differentiation and tissue regeneration.
