Development of Microprocessor-based Automatic Storage Controller and Temperature Auto-measurement System for Horticultural Crops (마이크로프로쎄서를 이용한 과채류 자동 저장 제어장치의 제작과 온도 자동계측 관리 시스템)

  • A laboratory scale storage facility consisting of a cold room, sample jars and a ventilation device was designed and built. Storaging sample jars (1.7 l) for fruit were fabricated with transparent acryl and provided with a constant air flow. For the supplying of air to sample jars, the air distributing system was built with solenoid valves, an air precooling coil and a pressure equalizing tank. To provide the programmable storaging environment of the facility a microprocessor-based controller was designed and installed. The controller was built with the 8 bit microprocessor (Z-80), EPROM, RAM, programmable peripheral interface(8255 PPI), and A/D converter. Softwares for the auto-temperature measurement and control of the storage system were developed and systemized in ROM. The automated storage system was applied to citrus storage, and the temperature of the storage facilities was successfully acquisited to the computer and controlled.

Purification and Characterization of a Novel Antifungal Protein from Paenibacillus macerans PM1 Antagonistic to Rice Blast Fungus, Pyricularia oryzae

  • An antifungal protein antagonistic to the rice blast fungus, Pyricularia oryzae was purified from Paenibacillus macerans PM-1 by ammonium sulfate fractionation, Q Sepharose Fast Flow column chromatography, Phenyl Sepharose CL-4B column chromatography and Superose 12 gen filtration. An apparent molecular mass of the purified antifungal protein was determined as 8 kDa by SDS-PAGE and 9 kDa by analytical gel filtration, respectively, suggesting that the purified protein is a monomer. The antifungal protein was stable at pH range from 7-12 and up to $100^{\circ}C$. The protein was also stable at 0.1-1% Tween 20 and Triton X-100. The N-terminal amino acid sequence of the antifungal protein was Thr-Glu-Leu-Pro-Leu-Gly-Ile-Val-Met-Asp-Lys-Tyr-Thr-Asp-Ala-Phe-Lys-Phe-Asp-Met-Phe. Comparison of the determined sequence with other peptide and DNA sequences did not reveal homology at all. Therefore, the purified antifungal protein was speculated to be a novel protein. The condidial germination in vitro of P. oryzae KJ301:93-39 by the purified protein ($5.9{\mu} g/ml$) was limited to $9{\pm}3.2%$ only, compared with $69{\pm}2.4%$ of the control. Ungerminated conidia were swollen at basa and mid cell by the purified protein. In vivo bioassay for inhibition of conidial germination of P. oryzae KJ 301, one of the most predominating racesin Korea. the purified protein ($5.9{\mu} g/ml$)strongly inhibited the conidial germination. The conidia, even though germinated, could not develop any further to produce appressoria efficiently.

  • Semi-active suspension systems are greatly expected to be in the mainstream of future controlled suspensions for passenger cars. In this study, a continuous variable damper for a passenger car suspension is developed, which is controlled actively and exhibits high performance with light weight, low cost, and low energy consumption. To get fast response of the damper, reverse damping mechanism is adapted, and to get small pressure change rate after blow-off, a pilot controlled proportional valve is designed and analyzed. The reverse continuous variable damper is designed as a HS-SH damper that offers good body control with reduced transferred input force from tire, compared with any other type of suspension system. The damper structure is designed, so that rebound and compression damping force can be tuned independently, of which variable valve is placed externally. The rate of pressure change with respect to the flow rate after blow-off becomes smooth when the fixed orifice size increases, which means that the blow-off slope is controllable using the fixed orifice size. The damping force variance is wide and continuous, and is controlled by the spool opening, of which scheme is usually adapted in proportional valves. The reverse continuous variable damper developed in this study is expected to be utilized in the semi-active suspension systems in passenger cars after its performance and simplicity of the design is confirmed through real car test.

  • Watering operation for oyster mushroom growing houses is regarded as drudgery and time consuming farm operation for growers. Most of mushroom growing beds in oyster mushroom growing houses are designed as two-row with four floor beds, therefore the watering and ventilation between the bed floors are much difficult for farmers because of its structural design. The study aimed to reduce the watering operation and improve the mushroom growing environment through the humidification and air supply on mushroom growing beds. Results showed that appropriate size of nozzle is between 0.8~0.5ml/s for the humidification and higher than the 2.0ml/s for the watering. The optimum water supply pressure was regarded as between 1.0~2.0MPa and the uniform distribution of droplet on the bed showed on air flow speed of 14m/s. The prototype was equipped with twin nozzle with. the humidification nozzle of 0.85ml/s and watering nozzle of 5.0ml/s, and the air blast fan with the air speed of 10m/sec in each air spout. In the field test in a practical scale mushroom growing house, it was well operated dependant on the set desire by a electric control unit. The machine can be practically used as air blast watering and air blast humidification for oyster mushroom growing farms without manual.

Performance of a Biofilter for Odor Removal during Manure Composting

  • Odor generated during composting of livestock manure is mainly due to ammonia emission. Biofiltration is a desirable method to control composting odor. This study was conducted to analyze the efficiency of using fresh compost as a biofilter. A mixture of cattle manure and recycled compost was composted in a bin equipped with a suction-type blower. The exhaust gas was filtered through the fresh compost. Residence time was controlled by the flow rate of exhaust gas and the depth of filtering materials. At the aeration rate of 30 L/min(experiment I), ammonia reduction rate varied from 100% to -15% for biofilter A(residence time 56.5 s) and almost 100% for biofilter B(residence time 113 s). At the aeration rate of 30 L/min, the cumulative ammonia reduction rate was 80.5% for biofilter A and 99.9% for biofilter B. At the aeration rate of 50 L/min(experiment II), the lowest reduction rate showed a negative value of -350% on the 8th and 9th day for biofilter A(residence time 33.9 5), and 50% on the loth day for biofilter B(residence time 67.8s). At the aeration rate of 50 L/min, the cumulative ammonia reduction rate was 82.5% fur biofilter A and 97.4% for biofilter B. Filtering efficiency was influenced by residence time. The moisture content(MC) and total nitrogen(T-N) of the filtering material were increased by absorbing moisture and ammonia included in the exhaust gas, while pH was decreased and total carbon(T-C) remained unchanged during the filtering operation.

  • Since the numerical analysis was adopted in the mold design, lots of computational methods have been proposed for the simulations of casting processes for the various shaped molds. Today, it is possible to simulate the filling and solidification processes of most casts using the VOF technique. Though the three-dimensional numerical model based on the Cartesian coordinate system can be applied to any shape of cast, it becomes very inefficient when the three-dimensional model is applied to the cast of axi-symmetrical shape since the control volume includes at least 11 of the physical model. In addition, the more meshes should be distributed along the circumferential boundaries of curved shape in the Cartesian coordinate system fur the better results, while such curved circumferential boundary does not need to be considered in the two-dimensional cylindrical coordinate system. This motivates the present study i.e. developing a two-dimensional numerical model for the axi-symmetrically shaped casts. The SIMPLER algorithm, the VOF method, and the equivalent specific heat method have been adopted in the combined algorithm for the flow calculation, the free surface tracking, and the phase change heat transfer, respectively. The numerical model has been applied to the casting process of a pulley, and it was proven that the mesh and time effective calculation was accomplished comparing to the calculation using three-dimensional model.

Introduction of KIER Pyrolysis Process and 3,000 ton/yr Demonstration Plant (KIER의 열분해유화 공정 기술과 실증플랜트 소개)

  • Since late of 2000, KIER has developed a novel pyrolysis process for production of fuel oils from polymer wastes. It could have been possible due to large-scale funding of the Resource Recycling R&D Center. The target was to develop an uncatalyzed, continuous and automatic process producing oils that can be used as a fuel for small-scale industrial boilers. The process development has proceeded in three stages bench-scale unit, pilot plant and demonstration plant. As a result, the demonstration plant having capacity of 3,000 tons/year has been constructed and is currently under test operation for optimization of operation conditions. The process consisted of four parts ; feeding system, cracking reactor, refining system and others. Raw materials were pretreated via shredding and classifying to remove minerals, water, etc. There were 3 kind of products, oils(80%), gas(15%), carbonic residue(5%). The main products i.e. oils were gasoline and diesel. The calorific value of gas has been found to be about 18,000kcal/$m^3$ which is similar to petroleum gas and shows that it could be used as a process fuel. Key technologies adopted in the process are 1) Recirculation of feed for rapid melting and enhancement of fluidity for automatic control of system, 2) Tubular reactor specially-designed for heavy heat flux and prevention of coking, 3)Recirculation of heavy fraction for prevention of wax formation, and 4) continuous removal & re-reaction of sludge for high yield of main product (oil) and minimization of residue. The advantages of the process are full automation, continuous operation, no requirement of catalyst, minimization of coking and sludge problems, maximizing the product(fuel oil) yield and purity, low initial investment and operation costs and environment- friendly process. In this presentation, background of pyrolysis technology development, the details of KIER pyrolysis process flow, key technologies and the performances of the process will be discussed in detail.

The Effect of Magnolia Bark on the Metabolic Inflammation and Insulin Resistance of ob/ob Mice (후박(厚朴)이 ob/ob 마우스의 대사성 염증과 인슐린 저항성에 미치는 영향 및 관련기전에 대한 연구)

  • Objective: This study was undertaken to investigate how magnolia bark extract affects ob/ob mouse in terms of metabolic inflammation and insulin resistance. Methods: Leptin-deficient ob/ob mice were divided into 2 groups (n=5): a normal saline treatment (=control) and magnolia bark treatment. Wild type mice were the lean group (n=5). After 5 weeks, we measured fasting blood sugar (FBS) and conducted oral glucose tolerance tests (OGTTs) in each group. After 6 weeks, we measured body weight, epididymal fat pad weight, liver weight, serum glucose, serum insulin, and gene expression of tumor necrosis factor-${\alpha}$, interferon-${\gamma}$, and interleukin-6. We characterized the phenotype of adipose tissue macrophages (ATMs) and analyzed fractions of the phenotype in each group by flow cytometry. Results: In the magnolia bark group, fasting blood sugar, oral glucose tolerance levels, and insulin resistance (HOMA-IR) were significantly decreased. The population and proportion of ATMs among leukocytes in adipose tissue were significantly decreased in the magnolia bark group. The population and proportion of M1 type ATMs among ATMs were significantly decreased in the magnolia bark group. Gene expression of tumor necrosis factor-${\alpha}$ was significantly decreased in the magnolia bark group. Conclusions: These results support a positive effect of magnolia bark on metabolic diseases such as insulin resistance and metabolic inflammation in leptin-deficient ob/ob mice.

Cytoprotective Effects of Radix Curcumae Aromaticae in Human Umbilical Vein Endothelial Cells (울금에 의한 혈관내피세포 보호 효과에 대한 연구)

  • In order to validate the use of Radix Curcumae Aromaticae as an anti-inflammatory drug in the traditional Korean medicine, I have investigated the effect of water-soluble extract of Radix Curcumae Aromaticae (ECA) on the expression of inducible heme oxygenase-1 (HO-1), which ha.s anti-inflammatory and cytoprotective effects stimulates, in human umbilical vein endothelial cells (HUVECs) stimulated with a high dose of pro-inflammatory tumor necrosis factor-alpha (TNF-α). The extract protected dose-dependently HUVECs against TNF-α-induced apoptosis, as measured qualitatively by a nuclear staining method using the fluoresoence DAPI and quantitatively by a flow cytometry using fluoresce-enhanced Annexin V antibody, and significantly Increased HO-1 expression, as determined by Western blotting analysis using anti-HO-1 antibody. Biockage of HO-1 activity by a pharmacological inhibitor reversed cytoprotection afforded by the extract, and treatment with carbon monoxide, one of HO-1 metabolites, resulted in cytoprotection comparable to the extract. These results suggest that ECA may have therapeutic potential in the control of endothelial disorders caused by inflammatory cytokines.

Long-term Water Quality Fluctuations in Daechung Reservoir and the Limiting Nutrient Evaluations Using In Situ Enclosure Nutrient Enrichment Bioassays (NEBs) (대청호에서 장기간 수질변동 및 인위적 Enclosure 영양염 투여실험에 따른 제한 영양염류 평가)

  • The objectives of this study were to elucidate spatio-temporal heterogeneity of water chemistry and develop empirical models using trophic variables in Daechung Reservoir during 2005-2010 along with in situ tests of nutrient enrichment bioassays (NEB). The relations of water quality parameters in regard to precipitation showed that seasonal and interannual fluctuations of biological oxygen demand (BOD), total nitrogen (TN) and pH were minor, whereas conductivity, suspended solids (SS), and total phosphorus (TP) were largely varied in response to the magnitude of rainfall. The CHL maxima occurred immediately after the spate of TP during the high flow, indicating that phytoplankton growth was directly controlled by phosphorus. Empirical linear models of CHL-TP indicated that the variation of CHL in premonsoon was accounted 60% ($R^2$ = 0.60, p < 0.05, n = 54) by TP. In the mean time, empirical models of annual CHL-TN showed that the variation of CHL was weakly accounted ($R^2$ = 0.16, p < 0.001) by TN and more strongly ($R^2$ = 0.44, p < 0.001) by TP. Thus, the variation of CHL was more explained by the variation of TP than TN. In situ tests of Nutrient Enrichment Bioassays (NEBs) showed that the growth of CHL was greater in the P-treatments (as $PO_4-P$) than the control and N-treatment (as $NO_3-P$). Overall, our results suggest that phosphorus was aprimary limiting nutrient controlling the seasonal phytoplankton growth, based on the in situ experiments of NEBs.
