In order to find out superior corn line which has a strong antioxidant activity in the corn silk, antioxidant compounds and antioxidant activity were measured from various inbred and F1 lines. As a result, the contents of phenolic and flavonoid compounds range from 532 mg/100 g to 3,274 mg/100 g and from 980 mg/100 g to 2,420 mg/100 g respectively. Absorbance at 517 nm for contents of anthocyanins ranges from 0.05 to 0.76. Correlation coefficients between antioxidant compounds and various antioxidant assays such as DPPH, ABTS and FRAP were analyzed. Correlation coefficients between antioxidant activity and phenolic compounds and anthocyanin were significantly high in ABTS and FRAP assays. Considering acidic extraction condition and intervention between anthocyanins and DPPH solution, ABTS assay and FRAP assay are more suitable methods to evaluate antioxidant activity of corn silks. Especially, ABTS assay is thought to be the best method among three assays because the antioxidant activity in ABTS assay showed high correlation with phenol, flavonoid and anthocyanin compounds respectively. Among the samples, S15 which showed the highest contents of total phenolic compounds and the most potent antioxidant activity in ABTS and FRAP assay will be a good source for functional material.

This study was investigated to analyze the contents of flavonoid and polyphenol compounds, and inhibitory activities of tyrosinase and antioxidation to measure physiological effect of reflux water extraction (WE), reflux ethanol extraction (EE) and hot water extract under high pressure (HWE) of Torreya nucifera seed. HWE yields the highest contents of flavonoid compounds (176.34 mg/g) and polyphenol compounds (112.95 mg/g). The tyrosinase inhibitory rates were $5.62{\sim}28.71%$ at 2.0 mg/mL and HWE showed the highest inhibition rate. The nitrite scavenging abilities of all extracts were over 90% at pH 1.2 and 3.0 at the concentration of 2.0 mg/mL. The superoxide dismutase (SOD)-like activities of HWE was the highest value of 33.58%. The electron donating abilities (EDA) were $66.46{\sim}89.72%$ and HWE was the highest when the extracts were tested at 0.1 mg/mL. The EDA of all extracts were decreased with an increment of the extracts concentrations. The xanthine oxidase inhibitory rate of HWE was the highest value of 89.29% at the concentration of 2.0 mg/mL and the WE and HWE were over 75% rate of xanthine oxidase inhibition at 0.5 mg/mL.

This study was investigated the effects of heat treatment on polyphenolic compounds and antioxidant activities of yacon. Raw yacon was heated at $100^{\circ}C$ and $121^{\circ}C$ for 15, 30, or 60 min by using an autoclave. The browning intensity, levels of free and bound phenolic and flavonoid compounds, and 1,1-diphenyl-2-picrylhydrazyl(DPPH) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) radical-scavenging activity of yacon extracts following heat treatment were measured. The browning index, free polyphenolic content, and antioxidant activities in the extracts were significantly increased (p<0.05) with both increased heating temperature and time, while bound polyphenolic levels were decreased. The levels of free phenolic and flavonoid compounds in the yacon extract heated to $121^{\circ}C$ for 60 min were increased by 1.2 and 1.1 folds compared to raw yacon respectively. Moreover, DPPH and ABTS radical-scavenging activities of the yacon heated to $121^{\circ}C$ for 60 min were 1.7 fold and 2.0 fold higher, respectively, than those of raw yacon. A significant (p<0.01) correlation was observed among browning intensity, free and bound phenolic and flavonoid levels, and DPPH and ABTS radical-scavenging activities in heated yacon extract. Thus, heat treatment can be used as a method to enhance the antioxidant compound content in and the antioxidant activity of yacon.

Nelnumbo nucifera Gaerth. (Nymphaeaceae) has been used in a korean traditional medicine to treat fever, sunstroke and dizziness. The receptaculums of this plant were refluxed with MeOH, and then fractionated with organic solvents ($CH_2Cl_2$, EtOAc and n-BuOH) to screen whitening activity using tyrosinase inhibitory activity. EtOAc ($IC_{50}$, 45.23 ${\mu}g/ml$) fractions showed a good tyrosinase inhibitory activity. Column chromatographic separation of $CH_2Cl_2$ and EtOAc fractions of Receptaculum nelumbinis led to the isolation 3 compounds. Their chemical structures were characterized as ${\beta}$-sitosterol (1), quercetin 3-O-${\beta}$-D-galactopyranoside (2) and kaempferol 3-O-${\beta}$-D-glucopyranoside (3) by comparison NMR spectral data and with those in references, respectively. Isolated compounds 1 and 3 were firstly isolated from Receptaculums nelumbinis. Compounds 2 and 3 showed potent whitening activities.
In this inverstigation, the various flavonoid aglycones were evaluated for their inhibitory activities against croton-oil or arachidonic acid induced mouse ear edema by oral or topical administration. The compounds tested were thirteen derivatives of flavan-3-ol(catechin and epicatechin), flavanone (flavanone and naringenin), flavone (flavone, chrysin and apigenin), flavonol(favonol, galangin, quercetin and morin) and isoflavone (biochanin A and 2-carbethoxy-5,7-dihydroxy-4'-methoxyisoflavone), along with hydrocortisone, indomethacin, 4-bormophenacyl bromide, nordihydroguaiaretic acid and phenidone as positive controls. A(isoflavone) were found to show broad inhibitoty activities (14-52%) against croton-oil or arachidonic acid induced ear edema by oral or topical application at the dose of 2 mg/mouse, although they showed less activity than hydrocortisone (26-88%) or indomethacin (36-80%). Flavonoid agtlycones tested showed higher activity when aplied topically than by the oral administration. It was also found that they inhibited arachidonic acid induced edema more profoundly than croton-oil induced edema by topical application. In arachidonic acid induced edema when applied topically, flavone derivatives such as flavone, chrysin and apigenin were revealed to be the good inhibitory agents in addition to flavonols and isoflavones. When quercetin and biochanin. A were selected for evaluating in carrageenan induced rat pleurisy and biochanin both flavonoids showed antiinflammatory activity at the dose of 70 mg/kg by the oral adminis-tration. All of these results revealed that flavonoid aglycones, especially 5,7-dihydroxy-flavonols having hydroxyl group(s) in B-ring and biochanin A (isoflavone) possessed in vivo antiinflammatory activity.

In this study, we extracted active components from thistle, persimmon leaf, and new green which are known to have a high content of antioxidants and also analyzed the 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavening activity and flavonoid content. Both ultrapure water and alcohol were used as extraction solvents and the ratio of both solvents, sample, amunts extraction time, and extraction temperature were varied. The optimal extraction condition of each natural compounds were 2.5~3.5 h of the extraction time and 50 g/L of the sample amount. The optimal ratio of ultrapure water and alcohol and extraction temperature were as follows; persimmon leaf (55~65 vol%, $50{\sim}60^{\circ}C$), thistle (40~50 vol%, $55{\sim}65^{\circ}C$) and new green (55~65 vol%, $50{\sim}60^{\circ}C$). In addition, the antioxidant capacity and flavonoid content of the extract increased in the order of persimmon leaf, thistle, and new green.

Matrix metalloproteinases (MMPs) are a family of structurally and functionally related zinc-dependent enzymes responsible for proteolytic degradation of extracellular matrix components such as base membrane or interstitial stroma. MMPs play an important role in a variety of physiological and pathological tissue remodeling processes, including wound healing, embryo implantation, tumor invasion and metastasis. Since MMP-9 (gelatinase B) has unique ability to cleave type IV collagen, gene expression of MMP-9 has been focused on as a pharmacological target. Flavonoids are a class of compounds that are widely spread in plants. In the coures of screening for the suppressors of MMP-9 gene expression from natural products, Metasequoia glyptostroboides was selected. Six flavonoids, sciadopitysin, isoginkgetin, bilobetin, 2,3-dihydrohinokiflavone, luteolin and apigenin were purified as suppressors of MMP-9 gene expression from M. glyptostroboides. The suppressing activity of the isolated flavinoids on the MMP-9 gene expression was measured by gelatin zymography and Nothern blot analysis.
Objective: The aim of this study is to optimize the extraction of beneficial substance from Cudrania tricuspidata leaves grown at Jirisan Mountain in South Korea by three different solvents depending on extraction time and at different temperature. Methods: The total phenolic contents were determined by the method reported by $S{\acute{a}}nchez$-Moreno et al. The total flavonoid contents were analyzed by Slinkard and Singleton. The DPPH radical scavenging activity was determined according to the method reported by Blois Results: The extraction yield for each solvent is 9.05-14.1%, 2.17-5.67%, and 2.3-3.9% for D.W., ethanol, and hexane, respectively. The overall results were maximized for the extract obtained with D.W. for 5 min at $100^{\circ}C$. The average phenol contents were 77.11, 45.64, and 0.343 mg/g at $100^{\circ}C$ in water, $78^{\circ}C$ in ethanol, and $68^{\circ}C$ in hexane, respectively. The flavonoid contents were the highest in the materials extracted with D.W., and were increased with increasing temperature, regardless of the extraction solvents, whether water (green), polar organic ethanol, or nonpolar organic hexane. In the ethanol extract, the flavonoid contents are increased gradually from 5.66 mg/g to 7.73 mg/g. The total flavonoid contents were proportional to the concentrations of the water extracts, ranging from 4.14 mg/g to 48.89 mg/g. The antioxidative activities of the water-extracted compounds are generally increased with increasing temperature from 42.5% to 85.5%. Those of the hexane extracts are increased slowly from 3.79% to 8.8%, while those of ethanol extracts are increased from 29.8% to 47.4%. Conclusion: The extraction yields were dependent upon solvents for extraction as well as extraction time and the temperature. The optimal extraction time was 5 min and the extraction yields were increased with increasing temperature excepted hexane. Of the three tested extraction solvents, the greenest solvent of water shows excellent results, suggesting that water is among the most effective solvents for natural sample extractions for general medicinal, pharmaceutical, and food applications.

We examined the lung anti-fibrotic properties of flavanones and flavones, which are flavonoid compounds, in bleomycin- and TGF-β1-stimulated A549 cells. Taken together, treatment with Bleomycin and TGF-β1 increased intracellular ROS by increasing the expression of various NOX families in A549 cells; further, the increased ROS levels resulted in increased fibrosis markers and induced pulmonary fibrosis. Flavonoid treatment has been demonstrated to alleviate or inhibit pulmonary fibrosis by modulating Smad-dependent and -non-dependent TGF-β mechanisms by modulating intracellular NOX expression.

Purpose: Ixeris strigosa (IS) is a perennial plant that commonly grows in meadows. The leaves and roots of IS have been used in medicine as a sedative. This study evaluated the antioxidant and carbohydrate-digestive-enzyme inhibitory effects of IS to determine its potential as an essential antioxidant and glycemic inhibitor for type 2 diabetics. Methods: The antioxidative and α-amylase and α-glucosidase inhibitory activities were examined using the water extracts (ISW), ethanol extracts (ISE), and solvent fractions from IS. The antioxidative activities were measured using in vitro methods by measuring the 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity. Results: Investigations of the total polyphenol, flavonoid content, in vitro antioxidant activity, and α-amylase and α-glucosidase inhibitory activities of the IS extract showed that the ISE had higher total phenolic and flavonoid contents than the ISW, as well as high antioxidant activity. The ethanolic extracts of IS (70%) had an α-amylase inhibitory activity of 78.55%. The ethyl acetate fraction (90.56%) showed higher α-glucosidase inhibitory activity than the positive control, acarbose (83.01%). Conclusion: Among the ISE fractions, the ethyl acetate and butanol fractions showed the best digestive enzyme inhibitory activity. Moreover, the antioxidant activity of the extract and the carbohydrate, α-amylase, and α-glucosidase inhibitory effects showed a stronger correlation with the total phenol and flavonoid contents compared to the ISW. As a result, the antioxidant and digestive enzyme inhibitory activities of high ISE are due to the phenolic compounds, particularly the flavonoid compounds. Therefore, ethyl acetate and butanol fractions of the 70% ethanol extract are excellent anti-diabetic functional materials.
