Effects of $O_2$ saturation and pH on Gas-bubble disease of blue crab, Portunus trituberculatus (꽃게, Portunus trituberculatus 유생의 가스병 발생에 미치는 산소포화도 및 pH의 영향)

  • In order to examine the effects of $O_2$ saturation and pH on gas-bubble disease of blue crab, portunus trituberculatus, zoeal larvae were reared under 5 different concentrations (6.0, 4.0, 3.1, 2.2, and 0.7million cell/cc) of Chlamydomones sp. under natural light for 4days. Observed pH and $O_2$ saturation in the rearing chamber increased drastically from 12 to 18 o'clock with the increasing of Chlamydomones sp. concentrations, but rapidly decreased from 2 to 6 o'clock in the rearing chamber. Gas-bubble disease of zoeal larvae was observed 4hours after starting of experiment at the pH 8.5 and 118% $O_2$ saturated conditions in rearing chamber during the day, and recovered rapidly with the decreasing of pH and $O_2$ saturation at night. Thus, gas-bubble disease of zoeal larvae was repeated with the increased pH($\geq$8.5) and $O_2$ saturation ($\geq$118%) almost every days. Mortality of zoeal larvae was observed when pH and $O_2$ saturations were reached 8.5 and 118% levels respectively, and deteriorated with the increasing of water temperature, pH and $O_2$ saturation.

  • A diagnostic test for viral hemorrhagic septicemia virus (VHSV), which infects more than 80 species of freshwater and marine fish at home and abroad, causing mass mortality, was conducted to provide quantitative data on the amount of virus expression in various tissues of flounder in chronological order. The tissues were collected in chronological order after the intraperitoneal injection of 3.0E+07 tissue culture infective dose50 (TCID50) per 0.1mL per fish of VHSV to randomly selected flounder. As a result of relative quantification through real-time PCR, the highest levels of virus expression were found in the spleen, kidney, gill, and liver on day 5. This study proved that the spleen was an appropriate site for the final diagnosis of VHSV in the early stages of infection and will provide important information for the diagnosis of legal infectious diseases in Korea.

Characterization of Photobacterium sp. YW2207 isolated from rainbow trout (Oncorhynchus mykiss) raised in a fresh water farm in South Korea (국내 양식 무지개송어(Oncorhynchus mykiss)에서 분리된 Photobacterium sp. YW2207의 특성)

  • Photobacterium sp. YW2207 was isolated from rainbow trout raised in a fish farm located in Yeongwol-gun, Gangwon Province, South Korea. Based on 16S rRNA sequence analysis and phylogenetic analysis, it was confirmed that Photobacterium sp. YW2207 showed 100% similarity with Photobacterium piscicola and Photobacterium phosphoreum, and 94.6% similarity with P. damselae subsp. damselae. Biochemical analysis revealed that Photobacterium sp. YW2207 is a Gram-negative, motile bacterium with a cell size of 1.5~3×3~5 ㎛. The bacteria were cultured on nutrient agar, brain heart infusion agar, Muller-Hinton agar, tryptic soy agar, and thiosulfate citrate bile sucrose agar with NaCl concentrations ranging from 0 to 2.5%. The API50CHE and API20E tests indicated lower utilization capabilities compared to the P. damselae strains provided in the API database. Furthermore, unlike most Photobacterium species, Photobacterium sp. YW2207 presented negative for catalase test. Results from the flow cytometric measurement indicated that Photobacterium sp. YW2207 exhibited a more diverse distribution of cell sizes and had larger cell sizes compared with P. damselae subsp. damselae. Minimum inhibitory concentration tests showed that Photobacterium sp. YW2207 had low susceptibility to β-Lactam and aminoglycoside antibiotics, while having high susceptibility to tetracycline, doxycycline, and quinolone antibiotics. Pathogenicity on rainbow trout revealed that an immersion of 1×105 CFU/ml did not cause mortality or clinical symptoms.

An outbreak of Lactococcus garvieae Infection in Cage-cultured Red Lip Mullet Chelon haematocheilus with Green Liver Syndrome

  • Red lip mullet Chelon haematocheilus (body weight = $468{\pm}91g$) which became sick during an outbreak of disease at mariculture facilities at Cheonsu Bay, Korea, during July-August 2013, were examined to identify the cause of the disease. Diseased mullets displayed green liver syndrome, and Lactococcus garvieae were isolated from their internal organs. Argulus sp., Trichodina sp., and/or Vibrio spp. were also discovered in some infected fish. Histopathological examination revealed that fatty liver syndrome with hepatocyte degeneration, reflected in heterokaryons, inflammatory lesions, and melanomacrophage centers ($MMC_S$), had caused fibrosis around the kidney, spleen, and blood vessels. After the outbreak, visceral fat and green liver syndrome in the mullets were consistently observed throughout the year in the same mariculture facilities, indicating that the cultured mullets suffered a chronic metabolic disorder. Although Vibrio spp. were also isolated from some individuals, L. garvieae, which is known to be a causative agent of red lip mullet mortality, was isolated from all diseased individuals. This is the first report of L. garvieae infection in cultured red lip mullet.

Effects of Water Temperature Change on the Hematological Responses and Plasma Cortisol Levels in Growing of Red Spotted Grouper, Epinephelus akaara

  • This study was conducted to determine the stress response [ethological (operculum movement number (OMN)), hematological (hematocrit and hemoglobin), biochemical (glucose, cortisol and glutamic oxaloacetic transaminase (GOT))] in red spotted grouper, Epinephelus akaara during exposure of different water temperature in winter season. This species (Total length, $18.56{\pm}0.34cm$) previously maintained in water temperature of $15^{\circ}C$ were transferred to 15, 20 and $25^{\circ}C$. During experimental period (7 days), OMN, hematocrit (Ht), glucose and GOT values were significantly high in $15^{\circ}C$ when compared to 20 and $25^{\circ}C$. Hemoglobin value was also increased at $15^{\circ}C$, but no significant differences. There was no differences in cortisol levels among the temperature groups. No fish mortality was observed during the experimental period. From these results, $15^{\circ}C$ is likely more stressful to red spotted grouper than $20^{\circ}C$ and $25^{\circ}C$. These observations confirm that red spotted grouper adapts better to temperatures between 20 and $25^{\circ}C$ during the winter season.

  • Soil, ground water, and sea bed are exposed to a continuous accumulation of polluted materials, causing serious environmental damage. It has been reported that such pollution causes a massive mortality of fish stock in rivers due to the resuspension of toxic chemicals, occurring during strong wind conditions. Therefore, it becomes apparent that there is an immediate demand for the restoration treatment of polluted river bed (or sea bed) sediment layers. Pollution levels of major rivers and ports, such as Paldang, Kyungan rivers, and Masan port, are becoming of great public concern, and are posing a serious environmental threat. In particular, the pollution of the Shi-hwa river has become a nation wide issue for the last few years. In spite of such public concern, the pollution levels of such rivers or ports are worsening everyday. In this study, an environmentally sound engineering package is introduced that helps to restore the polluted river bed or sea bed sediments. This engineering package consists of a suction facility, followed by a series of mechanical, chemical, and biological treatment units. The suction facility is designed to minimize secondary pollution that occurs from the resuspension of toxic materials during suction. The sea bed cleaning engineering package is designed to be installed on the top of a floating barge. Such a combination of environmental plant and shipbuilding technology provides a cost-effective solution, minimizing the transportation between suction and treatment facilities.

Sensitive, Accurate PCR Assays for Detecting Harmful Dinoflagellate Cochlodinium polykrikoides Using a Specific Oligonucleotide Primer Set

  • Harmful Cochlodinium polykrikoides is a notorious harmful algal bloom (HAB) species that is causing mass mortality of farmed fish along the Korean coast with increasing frequency. We analyzed the sequence of the large subunit (LSD) rDNA D1-D3 region of C. polykrikoides and conducted phylogenetic analyses using Bayesian inference of phylogeny and the maximum likelihood method. The molecular phylogeny showed that C. polykrikoides had the genetic relationship to Amphidinium and Gymnodinium species supported only by the relatively high posterior probabilities of Bayesian inference. Based on the LSU rDNA sequence data of diverse dinoflagellate taxa, we designed the C. polykrikoides-specific PCR primer set, CPOLY01 and CPOLY02 and developed PCR detection assays for its sensitive, accurate HAB monitoring. CPOLY01 and CPOLY02 specifically amplified C. polykrikoides and did not cross-react with any dinoflagellates tested in this study or environmental water samples. The effective annealing temperature $(T_{p})$ of CPOLY01 and CPOLY02 was $67^{\circ}C$. At this temperature, the conventional and nested PCR assays were sensitive over a wide range of C. polykrikoides cell numbers with detection limits of 0.05 and 0.0001 cells/reaction, respectively.

A Pragmatic Approach for Determining Overfishing and Overfished Condition for Assessing Data-deficient Fisheries (평가자료가 결핍된 어업의 과도어획상태와 자원의 남획여부를 결정하는 실용적인 방법)

  • 본 연구는 어업자원이 건강한 상태에 있는지 아니면 남획된 상태에 있는지를 판단하고, 현재 어업활동이 과도한지 아닌지를 판단하기 위한 간단하고 실용적인 방법을 개발하였다. 자원평가를 수행시에는 고가의 자료와 고도의 기술력이 요구되는 어려움이 있는데, 특히 소규모 어업과 다종어업에서 어려움이 더욱 심하다. 본 연구에서 개발된 방법은 어업자원의 구체적인 상태와 어업활동의 객관적인 상태를 고려해서 어업자원을 보존하고 어업활동을 규제할 수 있는 실용적이고 신뢰할 수 있는 방법이다. 예를 들어, 자원량과 어획사망률을 추정하는데 필요한 자료가 없거나, 추정할 수있는 과학적 능력이 부족한 경우에는, 자원량을 대용해서 단위노력당어획량을 사용하고 어획사망률을 대용해서 어획노력량 자료를 사용할 수 있다. 본 연구에서 제시한 방법의 실현가능성과 유용성을 입증하기 위하여, 복수어종으로 집계된 어획량과 어획노력량 만으로 된 경우나 또는 개체군 특성치 정보와 체장자료 만이 이용 가능한 경우에 대해 적용하였다. 이 방법은 수산자원의 보존이라는 전지구적 목표를 달성하는데 있어서, 필요한 자원상태와 어업상태를 판단해서 어획활동을 규제하고 조정할 수 있는 유용한 방법이 될 수 있을 것으로 보인다.

Effects of Ammonia Concentration on Histological and Physiological Status in Black Seabream (Acanthopagrus schlegeli)

  • The histological changes of gill, liver, spleen and muscle, and respiration and blood variables and liver glycogen content were examined in black seabream, Acanthopagrus schlegeli. Fish were exposed to a high level of total ammonia nitrogen (10.4 mg/l) and recovered from exposure $(0.4{\pm}0.2mg/l)$ in a closed recirculating seawater system. In the process of exposure, mortality was $9\%$, and hyperplasia, necrosis or inflammation appeared in all tissues except for muscle. Oxygen consumption was decreased by $49\%$, and red blood cell (RBC) number, hematocrit and hemoglobin concentration were significantly decreased, while plasma glucose contents, activities of glutamate-oxaloacetate transaminase (GOT) and glutamate-pyruvate transaminase (GPT) increased. Liver glycogen content significantly increased from $6.6\%\;to\;10.4\%$. A large amount of hemosiderin was observed in the splenic tissue. During the recovery period, RBC number, hematocrit, hemoglobin concentration, GOT and GPT activities were returned to the normal status. Histological status of liver tissue was returned to the normal, but liver glycogen content was not recovered. During the recovery period, spleen melanin-macrophages temporarily increased, but subsequently decreased to the normal status.

Dietary Guidelines for the Elderly

  • Dietary guidelines are a distillation of dietary advice from health professionals to the general public. They are based upon current scientific knowledge about the relationships between diet and disease, nutrients available in the food supply of a country, and the profile of morbidity and mortality in that country. With two different sets of dietary guidelines used for more than an decade in Korea. the necessity of revising dietary guidelines has been raised continuously from academia and research. Funded by a grant from the Health Technology Planing and Evaluation Board. Dietary guidelines for each age group were drafted as a research project and the one for the Korean elderly is as follows: Dietary Guidelines for the korean elderly(draft) - Have a variety of easily digestible foods on time; at least 3 meals a day and some snakes. - Be physically active to maintain appetite and/or ideal body weight. ; maintain a balance between activity and what you eat. -Increase consumption of bean-and dairy-and dairy-products. - Consume enough amounts of fresh dark-green and yellow vegetables and fresh fruits. - Consume adequate amounts of assorted kind of animal foods including fish, meat and poultry. - If you drink alcoholic beverages, limit your intake and, drink enough water and other averages; alcohol may interact with your medication and affect your appetite. Aforementioned draft and related contents are expected to be utilized as a neat base in formulating(or revising) dietary guidelines for Korean by the Government in near future.
