Comparative Efficacy of Antifungal Agents for Aquaculture Fish and their Eggs (양식 어류와 이들 난에 대한 항곰팡이성 약물들의 효과 비교)

  • In fresh water fish hatcheries and farms, Saprolegniales often cause serious mortality to the fish and their eggs. Malachite green is an effective antifungal agent, but is carcinogenic to fish and humans. Alternative antifungal agents are needed. Presently, we tested various concentrations of MBT-01108 (Opuntia ficus-indica extracts) alone and in combination with bronopol, formalin and sodium chloride (MBT-01108 mixture) on in vitro mycelial growth and in vivo remediation of adult eel, Anguilla japonica, infected with Saprolegnia sp. and fertilized eggs of chum salmon, Oncorhynchus keta, to evaluate the compounds' antifungal efficacy on eyed-egg and hatching rates. MBT-Oll08 mixtures incorporating bronopol and formalin at respective concentrations of 50 and 30 parts per million (ppm), and 100 and 20 ppm were most effective in controlling Saprolegnia in vitro and in vivo (P<0.05). Repeated daily exposures to 50 ppm and 100 ppm MBT-01108 were more effective than exposure every 2-3 days post-fertilization for the inhibition of Saprolegnia infection of rainbow trout, Oncorhynchus mykiss eggs as compared with control (0 ppm).

Experimental infection of atypical Aeromonas salmonicida in Nile tilapia Oreochromis niloticus and its treatment with carvacrol and cymene mixture

  • The pathogenecity of atypical Aeromonas salmonicida was studied in healthy Oreochromis niloticus. Inoculum at concentration of $1.5{\times}10^8CFU/ml$ and $3{\times}10^8CFU/ml$ was injected into healthy fish through intramuscular and intraperitoneal injections. Experimentally infected Oreochromis niloticus showed ulceration at the dorsal musculature and trunk region in addition to black coloration, congested gills, exophthalmia, and ocular hemorrhage. Congested liver and kidney were recorded in post-mortem examination. Mortality of the experimentally infected Oreochromis niloticus reached 100% after intramuscular injection at concentration of $3{\times}10^8CFU/ml$. Histopathological investigation of infected organs was also performed. There was a focal area of bundles of skeletal musculature showing hyalinization. In addition, hyperplasia, congestion, and fusion were noticed in the gill lamellae. There was also congestion in the blood vessels in the ocular chamber. Severe congestion was also noticed in the central vein of liver associated with focal aggregation of the melanin pigmented cells in the parenchyma. Degenerative changes were noticed in the epithelial cells lining of kidney tubules. Plant extracts carvacrol and its biological precursor cymene were found to be effective in treating experimentally infected Oreochromis niloticus at concentration of 100 or 200 ppm.

Evaluation of a Non-destructive Diagnostic Test for Kudoa septempunctata in Farmed Olive Flounder Paralichthys olivaceus (넙치(Paralichthys olivaceus) 근육 채취 방법에 따른 Kudoa septempunctata 진단 효율 비교)

  • Kudoa septempunctata, a myxosporean parasite that infects olive flounder Paralichthys olivaceus is known to cause Kudoa food poisoning. Entire trunk muscle (ETM) is used for diagnosis of the parasite in fish and this method demands sacrifice of the host, causing a loss of commercial value. We developed a non-destructive method that uses a plastic syringe-style implanter to draw the sample, called the part-point muscle (PPM) sampling technique. We validated the PPM method in fish infected with K. septempunctata at the level detectable by the ETM method. We confirmed that the PPM method is equally sensitive in comparison to the ETM method for diagnosing K. septempunctata spores in olive flounder muscle. Our study also confirmed that the parasite is uniformly distributed in the dorsal muscle of infected fish. Over a period of 1 month, we observed no mortality of the host fish used for sampling by the PPM method. Thus, our studies demonstrate that the PPM sampling technique is an efficient, non-destructive method for diagnosing K. septempunctata in olive flounder.

Selective non digestion of yellow mealworm Tenebrio molitor larvae by arowana

  • This study reveals the unusual case reported for the first time on the selective non digestion of yellow mealworm (Tenebrio molitor) larvae by arowana fish. In January 2005, an Asian arowana (Scleropages formosus) (red variety), from Daesang Tropical Fish Corporation, Seoul, Korea, mortality was observed due to unknown cause. No putative causal factors were suggested by bacteriological and parasitological examinations. Internal examination of the dissected stomach showed some undigested debris with mandible parts of mealworms attached to the mucosal lining of stomach wall. Feeding experiment of yellow mealworm (Tenebrio molitor) was conducted on the silver arowana (Osteoglossum bicirrhosum). Result showed that on the test group, fish released vomitous material containing undigested mandibles of mealworm was observed on the 24th day of the feeding experiment. Histopathological examination of the cross section of the stomach wall layers of the test groups, showed detached parts of the mucosal layer and gastric pits around the damaged area with intact mucularis tissues. While the control group fed with mealworm larvae without head part showed intact stomach wall layers consisting of gastric pit, mucosa and mucularis tissues. Fish on both treatments survived until the termination of experiment. The removal of mealworm larvae head before feeding probably help or aid in the fast digestion of these insect larvae.

Identification of sex-specific SNPS in burbot Lota lota using RAD sequencing: conservation and management applications

  • The development of sex-specific genetic assays in a species provides both a method for identifying the system of sex determination and a valuable tool to address questions of conservation and management importance. In this study, we focused on the identification of single nucleotide polymorphisms (SNPs) that differentiate genetic sex in burbot Lota lota. Burbot are the only true freshwater representative of the cod family and a species of conservation and management importance throughout Eurasia and North America. To identify sex-specific SNPs, we utilized restriction site-associated DNA sequencing (RADseq) to interrogate thousands of SNPs in burbot samples of known phenotypic sex. We discovered 170,569 biallelic SNPs, none of which fit the pattern expected under female heterogamety. However, we identified 22 SNPs that fit the pattern expected under male heterogamety (males heterozygous XY, females fixed XX) and, from these, developed two genetic assays that robustly (~ 97% genotyping success) and accurately (> 99% correct) sexed burbot samples. These sex-specific genetic assays will benefit growing conservation aquaculture programs for this species and allow future assessments of sex-specific migration, growth, and mortality.

BREEDING OF THE PUFFER FUGU RUBRIPES (자주복 Fugu rubripes (Temminck et Schlegel)의 종묘 생산에 관한 연구)

  • Fingerlings hatched from the eggs of the puffer, Fugu rubripes, which were spawned on May 21, 1969, and were cultivated. The results of their growth during 150 days, until October 25th, are summarized as follows: 1. The eggs began to hatch after 163 hours, at a water temperature of 15.9 to $17.4^{\circ}C$, and the hatching rate was $61.56\%$. 2. They reached the post-larval stage 6 to 7 days after hatching, and at this time a high mortality occurred. The mortality rate was 57.26 to $68.0\%$. 3. Sixteen days after hatching some of the larger fingerlings (6.7mm in total length) began attacking some of the smaller ones (4.6mm in to total length). 4. Twenty five to twenty eight days after hatching, the fish changed their food, and at this time a second high mortality occured. The total mortality rate amounted to 90.7 to $90.9\%$ of the total hatch. 5. After the fingerling stage. cannibalism occurs. The fish usually attack the caudal part of other fingerlings. It occurs regardless of body length and when the food supply is short. 6. The food coeffiicient at the age of 46 days (when body length is 53 to 68 mm) was 5.5 for short-necked clams, 8.5 for earth-worms, and 8.7 for fishes. 7. A third hish mortality occurred 53 to 63 days after hatching, the total mortality amounting to 95.76 to $97.34\%$, and the main cause of the mortality was found to be rickets resulting from nutritional deficiency 8. The growth rates were as follows: 2.68mm just after hatching, 3.84mm at the age of 10 days; 7.96mm after 25 days; 20.96mm or 130mg after 40 days; 73.68mm or 9.06g after 80 days: and 123mm or 31.8g after 150 days. 9. The water temperature during the above period was 15.7 to $28.4^{\circ}C$ with an average of $22.10^{\circ}C$ and the salinity was 25.53 to $34.50\%$ with an average of $32.07\%$, 10. The young of this species could endure well a wide range and sudden rise in salinity, and could survive easily when the salinity suddenly fell to $5\%$, but a considerable mortality occured when it fell to $3\%$. 11. When the fish were tranferred directly to fresh water from normal sea water they died out in 9 hours and 40minutes. However, when transferred from water of $5\%$ salinity at which they were reared for 54 days, they survived for 60 hours and 40 mimutes longer than in the former case.

Effect of High Dietary Carbohydrate on the Growth Performance and Physiological Responses of Juvenile Wuchang Bream, Megalobrama amblycephala

  • An optimum dietary carbohydrate content is important for maximum fish growth. In this study, we fed Wuchang bream (Megalobrama amblycephala) with either control diet (30.42%) or high carbohydrate diet (52.92%) for 90 d. Fish were fed to apparent satiation three times daily in an aquarium with automatic temperature control and circulated water. Growth performance, plasma biochemical parameters, hepatic morphology and enzyme activities were determined. It was shown that compared to fish fed control diet, fish fed high carbohydrate diet had higher plasma triglyceride and cortisol levels for d 90, and lower alkaline phosphatase level for d 45, lower hepatic superoxide dismutase and total antioxidative capacity for d 90, higher malondialdehyde for d 45 and glycogen content for d 45 and 90 (p<0.05). Histological and transmission electron microscopy studies showed that hepatocytes of fish fed high carbohydrate diet contained large lipid droplets, causing displacement of cellular organelles to periphery of hepatocytes. The relative level of hepatic heat shock protein 70 (HSP70) mRNA of Wuchang bream fed high carbohydrate diet was significantly higher than that of fish fed the control diet for 90 d (p<0.05). These changes led to decreased specific growth rate and increased feed conversion ratio (p<0.05). Upon hypoxia challenge, fish fed high carbohydrate diet had higher cumulative mortality than those fed the control diet (p<0.05). These results suggested that high dietary carbohydrate (52.92%) was detrimental to the growth performance and health of Wuchang bream.

  • A new bacterial infection occurred among the cultured Korean catfish, Silurus asotus, in Chunbuk prefecture, Korea, 1993. This disease produced about 30% mortality in the fish for 4 months. The diseased fish was swimming listless at the water surface with head up and tail down, sometimes spinnig in circles. The most outstanding clinical sign was ulceration on the skull and at the base of the pectoral fins. The causative organism was isolated from the brain, kidney, spleen and liver of diseased fish, and identified as Edwardsiella icaluri by the biochemical and biophysical characteristics. After intraperitoneal innoculaton of the isolate, the pathogenicity was proved positive for Korean catfish, S. asotus, and channel catfish, Ictalurus punctatus, but negative for carp, Cyprinus carpio.

Comparison of Growth Parameters in Selected and Unselected Strains of Olive Flounder Paralichthys olivaceus (선발 육종넙치 Paralichthys olivaceus 및 일반넙치의 성장비교)

  • To estimate the effect of selective breeding on the improvement of growth in olive flounder Paralichthys olivaceus, we compared the growth of a strain selected for rapid growth to that of an unselected strain from a commercial hatchery. The fish strains were fed with either moist pellets (MP) (dry matter, 59.8% crude protein; 14.1% lipid) or extruded pellets (EP) (dry matter, 50.4% crude protein; 13.8% lipid) for 190 days and were reared under similar conditions. The mortality rates were less than 2% and were not significantly different among the experimental groups. The growth rate of the selected fish was significantly greater than that of the unselected fish regardless of the diet type, and both strains fed MP grew significantly faster than those fed EP. The selected fish consumed more feed than the unselected fish. However, there was no significant difference in the feed efficiency between the selected and unselected fish. These results demonstrate that the selected fish exhibited superior growth rates, and that this was associated with a greater intake of food. Thus, selective breeding may be useful for improving the growth of commercial olive flounder.

Replacement of Dietary Vitamin C with Citrus and Fermented Citrus By-products in Korean Rockfish Sebastes schlegelii (조피볼락(Sebastes schlegelii) 사료 내 합성 비타민C 대체원으로써의 감귤부산물과 발효 감귤부산물의 첨가 효과)

  • The study was conducted to investigate the dietary supplementation of citrus by-product (CBP) and CBP fermented with Bacillus subtilis (BS) or B. pumilus (BP) on growth performance, feed utilization, innate immune responses and disease resistance of Korean rockfish Sebastes schlegelii. Triplicate groups of juvenile fish (initial weight of 22.9 g) were fed one of five experimental diets which were formulated to replace a synthetic vitamin C (L-ascorbyl-2-polyphosphate, LAPP) with CBP or fermented CBP. A basal diet without LAPP and CBP was used as the control diet (Con). At the end of the 13 weeks feeding trial, growth performance, feed utilization and survival of fish fed the Con were significantly lower than those of fish fed the LAPP, CBP and fermented CBP diets. Fish fed the Con were significantly lower in lysozyme activity than fish fed LAPP, CBP and fermented CBP diets. In a challenge test against Edwardsiella tarda, cumulative mortality of fish fed LAPP, CBP and fermented CBP diets was significantly lower (20-30%) than that of the Con (60%). The findings in this study indicate that the CBP and/or fermented CBP can be a promising vitamin C source for LAPP replacement in diet for Korean rockfish.
