Comparison of DNA isolation methods for detection of foodborne pathogens by real-time PCR from foods (식품으로부터 식중독 세균 검출을 위한 Real-time PCR에 적합한 DNA 추출 방법 비교)

  • This study was conducted to find out the most suitable DNA isolation methods for PCR detection of foodborne pathogens. Four DNA isolation methods including Universal Genomic DNA Extraction Kit (TaKaRa), PrepMan Ultra (Applied Biosystems), boiling method and alkaline lysis method (w/PEG) were tested and compared. The Universal Genomic DNA Extraction kit (TaKaRa) was considered as the more efficient isolation method for Escherichia coli O157:H7 and Staphylococcus aureus in lettuce, fish and beef. Meanwhile to detect the foodborne pathogens directly from foods without enrichment, the four different buffers such as double-distilled water, saline, glycine-saline, glycine-saline with Tween-20 and beef extract were also evaluated. As a result, saline was more suitable buffer for E. coli O157:H7. And double-distilled water was more suitable buffer than saline for S. aureus, respectively

  • Background: Butaphosphan (BTP) has recently been introduced into the Korean aquaculture sector as a stressattenuating agent. In this study, a sensitive chemical analytical method was established for the detection of BTP in the olive flounder (Paralichthys olivaceus) tissues. Methods: Utilizing a method employing liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS), detection sensitivity, specificity, and precision were satisfactorily established. Temporal changes in the BTP plasma and muscle concentrations were assessed after a single intramuscular injection of BTP (50 and 150 mg/kg) to the olive flounder maintained at 13 ℃ or 22 ℃. Results: High BTP plasma levels were achieved immediately after the injection, and the drug was rapidly eliminated. Additionally, plasma BTP levels were markedly dependent on the elimination rate, which, in turn, seemed dependent on the water temperature, with the drug elimination half-life and mean residence time significantly shorter at 22 ℃ than 13 ℃. Overall, muscle BTP levels were markedly lower than the plasma levels. Notably, muscle levels were not influenced by water temperatures. Muscle BTP concentrations were used to estimate the necessary withdrawal period for drugs used in food fish, with BTP levels maintained far below the possible hazardous limit. Conclusions: In conclusion, the established LC-MS/MS method can be used for BTP residue detection with high sensitivity and reproducibility.

Detection of Carp Vitellogenin with Piezoelectric Immunosensor (압전류적 면역센서에 의한 잉어 Vitellogenin 검출)

  • Vitellogenin has been known as a potent biomarker protein for the estrogenic activity in fish exposed to endocrine disruptors. In this study, a piezoelectric immunosensor making use of an anticarp vitellogenin antibody and an AT-cut quartz crystal microbalance as the biological component and transducer was prepared, followed by its application to the analysis of carp vitellogenin as follows. Antibody immobilization was conducted by chemisorption of a thiolated antibody with a heterobifunctional thiolation cross-linker, sulfosuccinimidyl 6-[3-(2-pyridyldithio)propionamido]hexanoate. The reaction buffer for the immunosensor system was optimized as 0.1 M sodium phosphate (pH 7.4). Concentration-dependent sensor responses were obtained in the vitellogenin concentrations ranging from 0.4864 to 486.4000 nM, with a linear correlation between vitellogenin concentration and frequency shift in double-logarithmic scale. The limit of detection of the immunosensor for carp vitellogenin was presumed as 0.4864 nM.

Real-time Voltammetric Assay of Cadmium Ions in Plant Tissue and Fish Brain Core

  • Optimum analytical conditions for cyclic voltammetry (CV) and square wave (SW) stripping voltammetry were determined using mercury-mixed carbon nanotube paste electrode (PE). The results approached the microgram working ranges of SW: 10.0-80.0 $ugL^{-1}$ and CV: 100-700 $ugL^{-1}$ Cd (II); working conditions of 300-Hz frequency, 100 mV amplitude, 1.6 V accumulation potential, 400 sec accumulation time, and 40 mV increment potential. First, analysis was performed through direct assay of cadmium ions deep into the fishs brain core and plant tissue in real time with a preconcentration time of 400 sec. The relative standard deviation of 10.0 $mgL^{-1}$ Cd (II) observed was 0.064 (n = 12) at optimum conditions. The low detection limit (S/N) was set at 0.6 $ugL^{-1}$ ($5.33{\times}10^{-9}$ M). The methods can be used in direct analysis in vivo or in real-time monitoring of plant tissue.

  • The thermal effluent discharged from nuclear power plants can affect the offshore ecosystem change. The ability of measuring sea surface temperature in high resolution with Airborne MSS thermal spectral band(8.5 $\mu$m ~ 12.5 $\mu$m) and Landsat ETM+(10.4$\mu$m ~ 12.5 $\mu$m) gives us an information of spread range of thermal effluent. This information can be used as one of major factors fur analyzing the impact of the fish farm damage around the nuclear power plants. Every season from November 1999, this research has been conducted to investigate the extent of diffusion of thermal effluent discharged from KoRi, UlJin and WolSung Nuclear Power Plant located at the coastline of the East Sea of Korea.

Paralytic Shellfish Poisoning Toxicity of Shellfishes, Sold at Fish Markets in Seoul

  • In order to investigate the distribution of paralytic shellfish poison, we examined the toxicity during from February to October in 2000. Of 591 shellfish samples, $17(2.88\%)$ samples were detected. Scapgarca broughtonii was highest collected $14.29\%(2/14)$. In the monthly detection rate of PSP, April was highest $13.3\%(8/60)$, in the regional collecting rate, Cheon-nam coastal area was highest $3.82\%(10/262)$, and in cases of imported area, China was $8.3\%(1/12)$. Imported area as well as domestic area samples should be strengthen to examine enduringly.

  • The possibilities for HPLC analysis of lipids have been revolutionised by the availability of evaporative light-scattering detectors, with which the response is independent of the nature of the mobile phase and does not depend On the presence of specific chromophores in the lipids. It was thus possible to develop an HPLC procedure, involving ternary gradient elution, for separating all the lipid classes in animal tissues in a single step. Although reversed-phase HPLC has been widely used for the analysis of molecular species of lipids, sliver ion chromatography can be a valuable alternative. For example, a stable silver ion column for HPLC was developed which permitted resolution of molecular species of triacylglycerols, even from such complex samples as fish oils, again With light-scattering detection and gradient elution. The capacity for HPLC resolution of diastereomeric diacyl-sn-glycerol derivatives, prepared from triacylglycerols. has lead to a new simple method for stereospecific analysis of the latter.

  • Methyl tert-butyl ether (MTBE), an octane booster that is added to the reformulated gasoline, has been a widespread contaminant in aquatic ecosystem. MTBE is a recalcitrant pollutant having low biodegradability. Due to its higher water solubility and low octanol-water partition coefficient, it can be rapidly transported to the surrounding water environment. Also, MTBE is a known animal carcinogen, and is classified as a possible human carcinogen by U. S. Environmental Protection Agency. The adverse effect of MTBE to aquatic biota was widely reported. In Korea, the recent detection of MTBE in groundwater near gasoline filling stations has drawn concern to public health and ecosystem. To address this concern, the effect of MTBE to human health and ecosystem was discussed in this review. Also, ecotoxicity data of MTBE for fish, invertebrates, and algae were extensively compared to estimate the hazard concentration 5($HC_5$) of MTBE as a screening level.

  • Recently, aquaculture farm sites have been increased with demand of the expensive fish species and sea food like as seaweed, laver and oyster. Therefore coastal water quality have been deteriorated by organic contamination from marine aquaculture farm sites. For protecting of coastal environment, we need to control the location of aquaculture sites. The purpose of this study is to detect the laver aquaculture sites using multispectral remotely sensed data with autodetection algorithm. In order to detect the aquaculture sites, density slice and contour and vegetation index methods were applied with SPOT and IKONOS data of Shinan area. The marine aquaculture farm sites were extracted by density slice and contour methods with one band digital number(DN) carrying 65% accuracy. However, vegetation index algorithm carried out 75% accuracy using near-infra red and red bands. Extraction of the laver aquaculture site using remotely sensed data will provide the efficient digital map for coastal water management strategies and red tide GIS management system.

  • This paper describes the development of an automated, hand-held sensor for the fast assessment of seafood freshness. The sensor developed here combined: an array-based chemical sensor, composed of incrementally different conducting polymer elements deposited on a small chip; a highly sensitive, custom-made electronics for the detection of very small signal changes; precise temperature control of the sensor chamber; and an on-board microcontroller for data collection, storage, automation, and analysis. The instrument was used to successfully test seafood samples with different degree of freshness and spoilage. A linear relationship between microbiological count and e-Nose signal for three different fish fillet was developed. Once the linear relationship is included into the hand-held unit software, the e-Nose signal can be used for assessment of seafood freshness without performing the microbiological count technique.
