Detection and Analysis of the Potential Risk of VHSV in Bivalves in Korea (한국 이매패류 내 VHSV 검출 및 잠재적 위험성 분석)

  • VHSV is a major viral agent that affects freshwater and marine fish, causing serious economic losses in aquaculture in the world. Due to their filter-feeding activity, bivalve mollusks may act as viral transmitters after accumulation of the fish viruses released into seawater from infected fish. Amplification by RT-PCR was carried out to investigate the presence of VHSV in pacific oysters (Crassotrea gigas) and blue mussels (Mytilus edulis), inhabiting regions around aquatic farms in Korea. Primers designed from conserved regions of VHSVs allowed us to detect four different types of VHSV in a single PCR. Twenty two of the eighty four samples showed positive results of VHSV in a 2-step RT-PCR. Using six positive samples from three different regions in Korea, we cloned and sequenced the glycoprotein (G) gene (467-bp long) of VHSVs. Genetic analysis of the VHSVs detected in shellfish in various geographical areas of Korea showed highly restricted results to VHSV type Iva. This was in agreement with the reports showing only a single genotype of VHSV (Iva genotype) in outbreaks in cultured or wild fish in Korea. Consequently, we investigated VHSVs carried by bivalve mollusks inhabiting the vicinity of aquatic farms, and revealed correlationship between the type of viral accumulated in shellfish by filter-feeding, and those detected in disease outbreaks in fish.

  • Arowana (Scleropages formosus) is the most valuable group of ornamental fishes and very much in demand in the ornamental fish trade and commands high price ranging from hundreds to thousands of dollars per fish. In this paper, we described a case of mortality of arowana from a private aquarium in Korea. A bacterial pathogen from fish organs (brain, kidney, liver) was cultured, identified and confirmed using Vitek System 2, API 20E test, multiplex PCR and 16S rRNA gene sequencing. The morphological and biochemical properties of the bacterium isolated from the brain, kidney and liver of the fish were similar to Aeromonas sobria. Positive amplification products using the multiplex PCR assay for detection of A. sobria were obtained from these organs. The 16S rRNA gene of the isolates from fish was identical and exhibited 100% sequence similarity with A. sobria (AY987762.1) strain available from GenBank. This bacterium contained hemolysin gene, a virulence factor that plays an important role in outbreaks of disease and is pathogenic to humans as well as in fish. Although this opportunistic bacterium was isolated from a fish without any external symptoms, this pathogen may act as a reservoir and enhance chances of zoonosis to human such as during handling.

  • The epidemiological study was performed to survey the prevalence of fish pathogens in cultured olive flounder, Paralichthys olivaceus from 2005 to 2007. In this study, the fish pathogens were detected from 1,528 among 2,238 fish samples collected yearly in 5 sites from February, May, August and November. Annual incidences for three years show a yearly increase and there were 60.6% in 2005, 66.7% in 2006 and 72.3% in 2007, respectively. Seasonal prevalence was 63.5% in February, 67.4% in May, 75.1% in August and 64.4% in November for three years. The detection rates of 6 viral pathogens were 35.6% in 2005, 44.6% in 2006 and 24.4% in 2007 and the peak rate was 55.4% at adult size group (above 41cm). Viral nervous necrosis virus (24.7%) has been the most predominant virus in this investigation, while much lower rates were noted in viral haemorrhagic septicemia virus (10.6%) and red sea bream iridovirus (0.9%).

  • In the past, fish diseases were bacterial in aqua farms, but in recent years, the frequency of fish diseases has increased as they have become viral and mixed. Viral diseases in an enclosed space called a aqua farm have a high spread rate, so it is very likely to lead to mass death. Fast identification of fish diseases is important to prevent group death. However, diagnosis of fish diseases requires a high level of expertise and it is difficult to visually check the condition of fish every time. In order to prevent the spread of the disease, an automatic identification system of diseases or fish is needed. In this paper, in order to improve the performance of the disease identification system of Paralichthys olivaceus based on deep learning, the existing pre-processing method is compared and tested. Target diseases were selected from three most frequent diseases such as Scutica, Vibrio, and Lymphocystis in Paralichthys olivaceus. The RGB, HLS, HSV, LAB, LUV, XYZ, and YCRCV were used as image pre-processing methods. As a result of the experiment, HLS was able to get the best results than using general RGB. It is expected that the fish disease identification system can be advanced by improving the recognition rate of diseases in a simple way.

Epidemiological Studios on Clonorchis sinensis Infection along the Nam-river in Gyeongnam Province, Korea (남강유역 간흡충감염의 역학적 조사연구)

  • An epidemiological study on Clonorchis sinensis infection along the Nam-river (total length ; 186km) flowing in Gyeongsang-nam-do, southern part of Korea, was carried out. Formalin-ether concentration technique and Stoll's egg counting method were employed to figure out the prevalence of C. sinensis infection. For the detection of cercariae from Parafossarulus sp., the snail host of C. sinensis, each snail was placed in aerated tap water, and examined for expelled cercariae. For observing the metacercariae the fresh water fish favorably eaten in raw conditions wore prepared by means of pressing the muscles between two slide glasses and/or by digesting them with artificial gastric juice. The fresh water fish were fed to the rabbits to get the worms and to identify the morphology of adult C. sinensis. The results are summarized as follows: 1. Overall Prevalence of C. sinensis infection was 38.75 from 5,291 examinees; 44.15 (1,408 out of 3,196) in male and 30.4% (637 out of 2,095) in female. 2. The prevalence rates were 42.0% at the upper stream, 41.2% around the vicinity of Jinyanglake, 34.2% at Jinju city, 34.2% at middle stream and 40.3% at down stream regions, respectively. 3. By age, the highest positive rate (53.4∼54.3%) was observed in 30 to 59 years of age. In this age group, the rate in males was 59.7∼62.2%, and in females 42.2∼44.4%, In the age group of less than 19 years it was 7.5∼20.9%. 4. By social strata, the positive rate was 16.5% in the primary school children, 22.6% in school students, 46.2% in teachers and local officers and 49.6% in the general inhabitants. 5. The quantitative examinations with the stool collected from clonorchiasis cases revealed that the light infection (less than 4,000/EPG) was 53.6%, moderate infection (4,001∼10,000 /EPG) 30.3% and heavy infection (more than 10,001/EPG) 16.1%, respectively. More than half of total cases examined were light infection, and 73.2% of female examinees were lightly infected with this cuke. 6. The average value of EPG was 4,963 (male, 6,057; female, 2,557 and the highest value was obtained from the age group of 30 to 59 years (5,240∼6,454). 7. The prevalence of Metagonimus yokogawai infection in humans was 5.5%, and 89.8% of metagonimiasis cases wore double-infected arith C. sinensis. The highest prevalence rate was observed in Jinju city (11.2%). 8. Total of 5,005 Parafossarulus sp., the snail intermediate host of C. sinensis, were examined for the detection of cercariae. The cercarial expulsion rate was 0.34%, and the snails collected in Jin-yang-lake side and in the down stream expelled mainly the cercariae of C, sinensis. 9. About 788 cercariaejday (range: 127∼1,503) were expelled daily from a snail naturally infected with C. sinensis. The snails which released more than 1,000 cercariae/day were 30.8% out of total collected. A snail uniquely released 5,840 cercariae/day in this study. 10. The other trematode cercariae besides C. sinensis were also detected, and the rates out of total snails were the cercariae of Losogenes liberum 6.71%, Cyathocotyle orientalis 0.26%, Notocotylus attenuatus 2.52% and Mucobucaris 0.54%, 11. Ten out of 18 species of fresh water fish caught along the river harbored the metacercariae of C, sinensis. The highest rate of metacercarial infection in fish was detected in Pseudorasbora larva (85.9%). The fish mainly eaten by the inhabitants along the Nam-river, and the metacercarial infection rates were: Zacco platypus 8.0%, Hemibarbus sp. 18.2∼26.7%, Gnathepcgcn sp. 37.5%, Ischikauia steenackeri 42.9% and Pseudogobio esocinus 16.7%. 12. Out of 36 P. larva, the number of metacercariae were about 109 (ranEe; 18∼446) per fish and 27 per gram of flesh. The fish caught in spring harbored the highest number of metacercariae. As indicated above, the prevalence of C. sinensis infection in the inhabitants residing around the Nam-river was relatively high. The farther toward the upper stream areas, the higher was the positive rate of C. sinensis infection, and most of clonorchiasis cases were lightly infected. The snail hosts of C. sinensis distributed all around the Nam-river. Several species of freshwater fish were infected with the metacercariae of C. sinensis and the infection rates were relatively high. To prevent C. sinensis infection in the endemic areas, the effective health education system is suggested as a control measure, although mass treatment is also expected to be useful, using chemotherapeutic agents such as "praziquantel", a recently developed anthelminthic for C. sinensis inftraction.

DIFFERENCES IN VIBRIO PARAHAEMOLYTICUS DETECTION RATIO BY ANALYTICAL METHODS (측정방법에 따른 장염비브리오균의 검출율의 비교)

  • The differences in Vibrio parahaemolyticus detection ratio were compared between the isolation methods, the Most Probable Number technique and single dilution tube method. During the period from February to October in 1976, 298 samples of sea water, 112 of bottom deposit, 169 of shellfish, and 80 of fish samples collected along the south coastal area of Korea were examined to determine the detection ratio of Vibrio parahaemolyticus. It was often observed that Vibrio parahaemolyticus was detected in higher diluted samples even through negative in lower dilution. Three hundred and forty three samples out of 659 samples submitted to the test by MPN procedure appeared positive for Vibrio parahaemolyticus showing $52\%$ detection ratio. Whereas only 149 samples, corresponding $22.5\%$, were positive for Vibrio parahaemolyticus in the lowest dilution grade. The positive result was $24.5\%$ in the lowest dilution grade and $50\%$ by MPN Procedure in sea water samples, $28.6\%$ and $65.2\%$ in bottom deposit, $22.5\%$ and $56.2\%$ in shellfish and $7.5\%$ and $32.5\%$ fish samples. When tested by triplicate tubes, $61.7\%$ of 149 Vibrio parahaemolyticus positive samples appeared positive in one tube, $28.9\%$ of them were positive in two tubes and $9.4\%$ of them were positive in all three tubes. The detection ratio determined by MPN procedure was more than two times higher than that of single dilution in triplicate tubes.

Diagnosis of Trace Toxic Uranium Ions in Organic Liver Cell

  • Uranium is toxic and radioactive traces of it can be found in natural water and soils. High concentrations of it in biological systems cause genetic disorders and diseases. For the in vivo diagnosis, micro and nano range detection limits are required. Here, an electrochemical assay for trace toxic uranium was searched using stripping voltammetry. Renewable and simplified graphite pencils electrode (PE) was used in a three-electrode cell system. Seawater was used instead of an electrolyte solution. This setup can yield good results and the detection limit was attained to be at $10{\mu}gL^{-1}$. The developed skill can be applied to organic liver cell.

  • Heterotrophic dinoflagellate Pfiesteria piscicida (Dinophyceae) has been claimed to produce potent ichthyotoxins that cause disorientation and eventually death of fish and other marine animals. A real-time PCR probe targeting for SSU rRNA gene was used for detection of P. piscicida in Chinhae Bay, Korea. PCR inhibitors were successfully removed by dilution of template DNA. Positive detections were shown from surface water samples indicating the presence of P. piscicida in Chinhae Bay.

  • Purpose : The purpose of this study was to evaluate the clinical utility of rapid detection of Down syndrome and Edward syndrome by Interphase Fluorescence in Situ Hybridization (FISH) analysis. Methods : Aretrospective study in 309 cases of amniotic fluid samples, analysed by interphase FISH with DNA probes specific to chromosome 18 and 21, was performed. All FISH results w ere compared with conventional cytogenetic karyotypings. Results : The results were considered as informative and they were obtained within 48 hrs. A case of Down syndrome and a case of Edward syndrome were diagnosed by FISH and confirmed by subsequent cytogenetic analysis. In 12 cases with normal FISH results, the cytogenetic analysis showed a case of partial trisomy 22, three cases of sex chromosomal aneuploidy, two cases of mosaicism, two cases of microdeletion, and four cases of structural rearrangement. Conclusion : FISH is a rapid and effective diagnostic method, which can be used as an adjunctive test to cytogenetic analysis, for prenatal identification of chromosome aneuploidies. For the more genome-wide screening with variety of probes, the technique of FISH is both expensive and labor-intensive.

  • Viral hemorrhagic septicemia (VHS) is one of the most serious viral disease of farmed rainbow trout and some marine fishes in Europe and North America. It has been reported in various marine fish species of Asian countries and induced cause mass mortality in Japanese flounder (Paralichthys olivaceus) culturing in Korea. The aims of this study were to monitor VHSV in wild marine fishes and to give critical information for controling the disease through prophylactic methods. Prevalence of the viral disease, geological distribution and reservoir of the virus were investigated using wild marine fishes captured in southern coast and east china sea for two years. (Reverse Transcriptase Polymerase Chain Reaction) RT-PCR results showed that VHSV were detected in 17 (10.6%) out of 160 fish. G gene sequences of viral strains isolated in this study were closely related to that of a reference strain, KVHS01-1, belonging to VHSV genotype Ⅰ. The results suggest that some of wild marine fishes are VHSV carriers and may spread the pathogen directly to fish farmed in coastal area.
