Genetic Stability of Magnaporthe oryzae during Successive Passages through Rice Plants and on Artificial Medium

  • Genetic instability of the rice blast fungus Magnaporthe oryzae has been suggested as a major factor underlying the rapid breakdown of host resistance in the field. However, little information is available on the mechanism of genetic instability. In this study, we assessed the stability of repetitive DNA elements and several key phenotypic traits important for pathogenesis after serially transferring two isolates though rice plants and an artificial medium. Using isolate 70-15, we obtained a total of 176 single-spore isolates from 10 successive rounds of culturing on artificial medium. Another 20 isolates were obtained from germ tubes formed at the basal and apical cells of 10 three-celled conidia. Additionally, 60 isolates were obtained from isolate KJ201 after serial transfers through rice plants and an artificial medium. No apparent differences in phenotypes, including mycelial growth, conidial morphologies, conidiation, conidial germination, appressorium formation, and virulence, or in DNA fingerprints using MGR586, MAGGY, Pot2, LINE, MG-SINE and PWL2 as probes were observed among isolates from the same parent isolate. Southern hybridization and sequence analysis of two avirulence genes, AVR-Pita1 and AVR-Pikm, showed that both genes were also maintained stably during 10 successive generations on medium and plants. However, one reversible loss of restriction fragments was found in the telomere-linked helicase gene (TLH1) family, suggesting some telomere regions may be more unstable than the rest of the genome. Taken together, our results suggest that phenotype and genotype of M. oryzae isolates do not noticeably change, at least up to 10 successive generations on a cultural medium and in host plants.

Efficacy of Antagonistic Bacteria for Biological Control of Rhizoctonia Blight (Large patch) on Zoysiagrass (잔디 갈색퍼짐병(Large patch)의 생물학적 방제를 위한 길항 미생물의 선발과 효력 검정)

  • Rhizoctonia blight (large patch) caused by Rhizoctonia solani AG2-2 is one of the major diseases on zoysiagrass in golf courses. In this study, anatgonistic bacteria to R. solani AG2-2 were selected in vitro tests using confrontation bioassay and triple layer agar diffusion method. The most active bacteria, Bacillus subtilis CJ-9 were tested for controlling large patch in pots. Relative Performance Indies (RPI) was used as a criterion for the selection of potential biocontrol agent. B. subtilis CJ-9 showed resistance to major synthetic agrochemicals used in golf course. In field tests at golf course, B. subtilis CJ-9 was more effective in suppression of large patch severity and population development of R. solani AG2-2 in soil than chemical fungicides. B. subtilis CJ-9 could be an alternative to chemical fungicides for eco-friendly management of large patch on zoysiagrass.

  • For enhancing productivity of glulam, high frequency (HF) curing technique was researched in this study. Heat energy is generated by electromagnetic energy dissipation when HF wave is applied to a dielectric material. Because both lamina and adhesives have dielectric property, internal heat generation would be occurred when HF wave is applied to glulam. Most room temperature setting adhesives such as phenol-resorcinol-formaldehyde (PRF) resin, which is popularly used for manufacturing glulam, can be cured more quickly as temperature of adhesives increases. In this study, dielectric properties of larch wood and PRF adhesives were experimentally evaluated, and the mechanism of HF heating, which induced the fast curing of glue layer in glulam, was theoretically analyzed. Result of our experiments showed relative loss factor of PRF resin, which leads temperature increase, was higher than that of larch wood. Also, it showed density and specific heat of PRF, which are resistance factors of temperature increase, were higher than those of wood. It was expected that the heat generation in PRF resin by HF heating would occur greater than in larch wood, because the ratio of relative loss factor to density and specific heat of PRF resin was greater than that of larch wood. Through theoretical approach with the experimental results, the relative strengths of ISM band HF electric fields to achieve a target heating rate were estimated.

Deterioration of Agronomic Characteristics of Drought-Resistant GM Rice (CaMsrB2-8) (가뭄저항성 GM벼(CaMsrB2-8)의 농업적 특성의 퇴화 가능성 검정)

  • The purpose of this study is to observe the growth habit and investigate a possibility of cultivating the GM rice (CaMsrB2-8) as a rice cultivar having drought resistance. Germination viability test showed that there was no significant difference between the drought-resistant GM(CaMsrB2-8) and non-GM (Ilmi) rice which was the parent variety at the GM rice. All the seeds of CaMsrB2-8 and Ilmi germinated after 6 days. Viviparous germination was not found in CaMsrB2-8 and Ilmi that was grown in greenhouse at $23{\pm}2^{\circ}C$ with water spraying for 40 days. Ratooning of CaMsrB2-8 and Ilmi was observed in 7-14 days and found uniform in field condition. CaMsrB2-8 seemed to grow faster than Ilmi. But CaMsrB2-8 and Ilmi were similar in 14-21 days. Both CaMsrB2-8 and Ilmi showed low seed shattering and more than 90% grains were ripened. All the seeds scattered in the paddy soil surface were not germinated after passing the winter. This study suggests that the drought-resistant GM rice was not significantly different with the parent variety of Ilmi in many agronomic characteristics such as wildness traits.

  • An analysis of recent studies and a field survey were conducted to investigate the distribution of the exotic species Ophraella communa LeSage (Coleoptera, Chrysomelidae) and the results' applicability for biological control against ragweed, which disturbs the island's ecosystem. The ragweed beetle (O. communa) can be found anywhere on the host plant Ambrosia artemisiifolia L., which grows in Jeju Island. Moreover, ragweed beetles possess the following characteristics: a high host plant specificity, a high-temperature resistance, an excellent mobility, and multivoltinism. Ragweed is extensively distributed and gradually expands its habitat further on Jeju Island. For this reason, it is recommended to take advantage of O. communa for the biological control of ragweed instead of pesticides or direct removal, considering that Jeju is endowed with many natural reserves and unique insular traits.

  • Shear wave velocity($V_S$), which can be obtained using various seismic tests, has been emphasized as representative geotechnical dynamic characteristic mainly for seismic design and seismic performance evaluation in the engineering field. For the application of conventional geotechnical site investigation techniques to geotechnical earthquake engineering, standard penetration tests(SPT) and piezocone penetration tests(CPTu) together with a variety of borehole seismic tests were performed at many sites in Korea. Through statistical modeling of the in-situ testing data, in this study, the correlations between $V_S$ and geotechnical in-situ penetrating data such as blow counts(N value) from SPT and piezocone penetrating data such as tip resistance ($q_t$), sleevefriction($f_s$), and pore pressure ratio($B_q$) were deduced and were suggested as an empirical method to determine $V_S$. Despite the incompatible strain levels of the conventional geotechnical penetration tests and the borehole seismic tests, it is shown that the suggested correlations in this study are applicable to the preliminary estimation of $V_S$ for Korean soil layers.

A study on the relationship between Schmidt Hammer's 'R' and bedrock microforms (기반암 하상 미지형과 슈미트 해머 반발 값과의 관계에 대한 연구)

  • Physical strength of the rock is the most important factor of resistance to erosion and has been measured through various way. Bedrock microforms, like potholes and grooves, are the forms sculpted by the erosional processes of flow and the location and morphology are strongly affected by the differential erosion. It also assumed that the physical strength of the rock controls the erosion rate and mode of erosion. The schmidt hammer has been used to measure the rock strength in the field for the geomorphological research. To find the relationship between the rock strength and microforms, Schmidt hammer's R(rebound) were measured in the Baeksuktan, middle reach of Gilancheon, Cheongsong, Gyungsangbuk do. The overall values of rebound of the local sandstone showed over 65 in most cases, so it can be regarded as 'very strong'. It is found that the rebound values of the rock surface decreased towards current water level. It also, however, found that there was no systematic differences in rebound values among the topographically high and lows in the bedrock surface. There was no statistically significant difference in rebound values of the area with well developed microforms and others. The values of R from the exposed faces and inside of the microforms are similar. In the case of conglomerate, the part with the gravel showed higher values that the parts with sands. The rebound values are decreased near of(<1cm) the geological discontinuities(including joint and faults), so this line of weakness could be the point of initiation of active erosion to form microforms. However there is large variations in rebound values within this part. It also should be mentioned that topological relation between the strike of the geologic discontinuities and flow direction looks control the mode of erosional processes.

Giant Magnetoresistance of Antiferromagnetic Cr-Al based Multilayer Spin-Valve with Anti-Corrosion and Thermal Stability (내열 내식용 Cr-Al반강자성계 스핀밸브막의 거대자기저항 효과)

  • The magnetic properties, thermal stability and anti-corrosion properties of $Cr_86Al_14$ spin valves multilayers were studied. It was found that the magnetic properties of $Cr_86Al_14$ spin valves depend on the thickness of antiferromagnetic, ferromagnetic and non-ferromagnetic layers. Exchange coupled field ($H_{ex}$) and magnetoresistance ratio (%) showed the largest value of 20 Oe, 2 % in $glass/Cr_{86}Al_{14}(600 $\AA$)/Ni_{81}Fe_{19}(50$\AA$)/Cu(40 $\AA$)/Ni_{81}Fe_{19}(40 $\AA$)$ spin valves. The $H_{ex}$ MR ratios (%) of CrAl and FeMn spin valves were decreased with increasing annealing temperatures and were lost at 150 $^{\circ}C$, 250 $^{\circ}C$ respectively. Based on these result, it was elucidated that CrAl is more thermally stable than FeMn. It was also shown that there was no change of $H_{ex}$ MR ratios in CrAl, while FeMn was changed and lost 15 days later in corrosion resistance test under 35 $^{\circ}C$, 90 % humidity condition. FeMn was found to be pitted and peeled off 15 days later by SEM micrographic analysis.

Development of embedded type antenna structure with NFC and WPC complex function (NFC 와 WPC 복합기능의 삽입형 안테나 복합체 개발)

  • The objective of this study is to develop an embedded antenna structure with NFC and WPC composite functions. By selecting stable materials, the optimal component ratio of the polymer sheet was determined. The low cost embedded winding method compared to the existing FPCB was devised. During the winding process, characterization and process technology were developed. We also fabricated a ferrite mold to process the WPC grooves and developed the process technology for optimizing the WPC antenna. The following conclusions were obtained. (1) Optimum composition ratio was derived as Fe 87.5%, Si 7%, Al 5.5% and selected as the final material. (2) Optimal sheet conditions were derived from the experimental evaluation method and the experimental design method through the combination test of the optimized sheet and the conventional mass production FPCB. (3) According to coil diameter and inner diameter, Q value fluctuation, resistance value and efficiency fluctuation are obtained. Therefore, the most suitable coil condition is selected and Rx matching is performed. (4) The EMV load modulation test and the cognitive distance test of the polymer sheet and the ferrite sheet showed that the recognition distance of the polymer sheet at 1k and 4K was 32-33 mm and the recognition distance of the ferrite sheet at the same condition was 30-31 mm.

  • Axial compressive failure loads ($P_n$) of diameter 500 mm and diameter 600 mm A type PHC pile were calculated as 7.7 MN and 10.6 MN, respectively. In the static pile load tests, the maximum axial compressive loads of the above 2 kinds of A type pile were measured as 6.9 MN and 8.8 MN respectively, therefore these measured maximum loads were at the level of 90% and 83% of $P_n$ respectively. Long-term allowable axial compressive loads ($P_a$) of the above 2 kinds of A type pile were 1.7 MN and 2.3 MN respectively. From the bi-directional pile load test data on the prebored PHC piles, it was confirmed that the allowable axial compressive bearing resistance was estimated as 131% of the long-term allowable compressive load of the PHC pile and showed higher than the allowable bearing capacity calculated by the current design method. Therefore, it has been verified that the PHC pile can be used up to the maximum long-term allowable compressive load, and it is suggested that the ultimate pile capacity formula used in the current design for prebored PHC piles should be improved to accommodate the actual capacity.
