Quantification of γ-Oryzanol Components and Comparison Its Biological Activity in Brown Rice (현미(추청벼, 흑진주벼, 신동진벼)에 함유된 γ-Oryzanol의 함량 분석 및 생리활성 효과 비교)

  • We analyzed the contents of ${\gamma}$-oryzanol, which is contained in brown rice of the nation rice varieties Chucheong, Heukjinju and Sindongjin, by HPLC. Furthermore we also performed experiments on its biological activity, to prove the effectiveness of rice bran. The contents of ${\gamma}$-oryzanol contained in hulled rice showed 1,587 ppm for Heukjinju, followed by Chucheong(1,038 ppm), and by Sindongjin(472 ppm). In anti-oxidative activity, we performed an experiment, by measuring the radical scavenging activity of DPPH. Heukjinju showed the best effect, and Chucheong showed the worst effect. In cholesterol lowering activity, Heukjinju showed the best activity and Sindongjin showed the worst effect. In anti-bacterial activity as well, Heukjinju showed the best activity, and Sindongjin showed the worst effect. Through these experiments, we compared the contents of ${\gamma}$-oryzanol, which is contained in hulled rice(Chucheong, Heukjinju, Sindongjin). Also, we found the anti-oxidation effect, cholesterol lowering effect, and anti-bacterial activity of the ${\gamma}$-oryzanol extracts. Based on our research, we expect that ${\gamma}$-oryzanol can work as a new drug, or nutritional supplement.

  • Whitening agents are eagerly demanded especially by oriental women who often suffers from the pigmentary disorders such as melasma and solar lentigines. As these pigmentary disorders are exacerbated by ultraviolet (UV), the whitening agents could exert its effect not only by inhibiting melanin synthesis but also by inhibiting UV activated signals. Eumelanin protects UV-induced DNA damages so that the chemicals which could reduce UV-induced DNA damages might be the ideal lightening agents. The effect of newly synthesized antioxidants, a-tocopheryl ferulate, on protective effect for UV-induced DNA damages as well as inhibiting melanin synthesis are briefly shown. For clinical evaluation, our results of the efficacy of lightening agents on treating pigment macules in combination with chemical peeling are shown. Furthermore, newly developed facial image analyzers to quantitatively evaluate the improvement of pigment macules are introduced.

The Darkening Effect of Phytosterylferulate on the Rice Bran Oil (Phytosterylferulate가 米糠油 暗色化에 미치는 影響에 關한 硏究)

  • An attempt has been made to investigate the nature of the characteristic darkening reaction occurring in the rice bran oil during the storage. Several pigments were separated by using column and thin layer chromatography of the pigments was made in the light of the knowledge of absorption spectral behavior. The presence of trace amount of iron and phytosteryl esters of ferulic acid (3-methoxy-4-hydroxy-cinnamic acid) was found to be responsible for the development of the characteristic color in rice bran oil. The model reactions with the ferulate, iron and fatty acids could be suggested the mechanism.

Isolation of Chemical Compounds from xBrassicoraphanus (배무채(xBrassicoraphanus)의 화학성분 분리)

  • xBrassicoraphanus is an intergenic breed crossed between Brassica campetris L. ssp. pekinensis and Raphanus sativus L. that have been daily consumed. xBrassicoraphanus was known to have good tastes and biological activities. Nevertheless, its constituetnts were not elucidated yet. Thus, in the present study, to indirectly evaluate the biological activity of xBrassicoraphanus, 12 compounds were isolated from leaves and roots of xBrassicoraphanus. On the basis of spectroscopic evidences, the structures of these compounds isolated from leaves of xBrassicoraphanus. were identified as ${\beta}-sitosterol$, indole-3-acetonitrile, ferulic acid, methyl ferulate, linolenic acid methyl ester, linolenic acid and coniferyl alcohol, while the chemical structures of compounds isolated from the roots of were xBrassicoraphanus were characterized as ${\beta}-sitosterol$, indole-3-acetonitrile, ferulic acid, methyl ferulate, linolenic acid methyl ester, 1-methoxyindole-3-acetonitrile, goitrin, 4-hydroxycinnamyl alcohol, coniferyl alcohol, palmitic acid and daucosterol. These can be classified as three steroids, two indole cyanides, two cinnamic acid derivatives, one cinnamyl alcohol derivative, three fatty acid derivatives one isothiocyanate. These results suggest that the compounds isolated from xBrassicoraphanus were almost identical with known components of Brassica campetris L. ssp pekinensis or Raphanus sativus L. However, it is necessary to investigate more about the difference of amounts of constituents according to harvest time and variant species amounts.

Antioxidant Constituents from the Stem of Sorghum bicolor

  • The EtOAc soluble fraction from the stem of Sorghum bicolor showed a strong free radical scavenging activity. Five major compounds were isolated from this fraction. They were identified by spectral data as methyl ferulate (1), methyl p-hydroxycinnamate (2), p-hydroxybenzaldehyde (3), tricin (4), and quercetin 3,4 -dimethyl ether (5). Among these compounds, 1 exhibited a strong, free radical scavenging activity on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) with an $IC_50$ value of 0.7 $\mu$M. We further studied the effects of these isolated compounds on the lipid peroxidation in rat liver microsomes induced by non-enzymatic method. All five compounds showed anti-lipid peroxidation activity ($IC_50$ values of 0.5, 0.4, 0.3 and 0.3 $\mu$ M, respectively).

  • Seven compounds, scopoletin (1), p-hydroxyphenethyl trans-ferulate (2), $1-({\alpha}-L-rhamnosyl(1\;{\rightarrow}\;6)-{\beta}-D-glucopyranosyloxy)-3,4,5-trimethoxybenzene$ (3), benzyl ${\alpha}-L-rhamnopyranosyl(1\;{\rightarrow}\;6)-{\beta}-D-glucopyranoside$ (4), suberic acid (5), sebacic acid (6) and scopolin (7) were isolated from the methanol extract of the roots of Althaea rosea Canival. Compounds $1\;{\sim}\;7$ were first isolated from this plant. The isolated compounds were tested for their cytotoxicity against human cancer cell lines using a SRB method in vitro.

Regulation of Proopiomelanocortin and Melanocortin 1 Receptor by UVB: Inhibitory Effect of Antioxidants

  • Epidermal cells produce a panel of antioxidants as well as cytokines after UVB irradiation, which counteract reactive oxygen species, however, how these antioxidants might regulate melanogenesis is unclear. An important constituent of the cellular antioxidant buffering system which controls the redox state of proteins is thioredoxin (TRX), a 13-kD protein that catalyzes thiol-disulfide exchange reactions, regulates activation of transcription factors, and possesses several other biological functions similar to cytokines. TRX suppressed the UVB-induced production and secretion of $\alpha$-melanocyte stimulating hormone ($\alpha$-MSH) and of adrenocorticotropic hormone (ACTH), and also suppressed proopiomelanocortin (POMC) mRNA expression by normal human keratinocyte (KC)s. Further, L-cysteine, N-acetyl-cysteine, $\alpha$-tocopheryl ferulate showed suppressive effect on UVB-induced POMC mRNA expression. However, TRX released from UVB-irradiated KCs stimulated melanogenesis by up-regulating MSH receptor expression and its binding activity in melanocyte (MC)s. UVB-induced KC derived cytokines such as IL1, IL6, and ET1 upregulated MSH-receptor binding ability as well as MCl-R mRNA expression in cultured normal human MCs. MCl-R has a tendency to be upregulated by UVB-induced KC-derived cytokines as well as by direct UVB irradiation. These results suggest that antioxidants such as TRX suppresses UVB induction of POMC, but in the case of MCl-R, this gene can be mainly in the trend of upregulation by UVB-induced KC-derived factors including TRX.
