Performance of Cone Type Tower Fermentor for Ethanol Production (탑형발효기에 의한 에탄올 생산)

  • A cone type of tower fermentor loaded with flocculating Sacchromyces uvarum was used to study the fermentor performance. The performance of cone type fermentor was compared with those of other fermentors. The maximum yeast concentration in the cone type of tower fermentor was 35.9-43.0g/1.hr and the maximum ethanol productivity was 14.75g/1.hr at the dilution rate 0.26 $hr^{-1}$. The ethanol yield was 0.446-0.472g ETOH/g Glucose. It was concluded that a cone type of tower fermentor might offer better perspectives for continous etanol fermentation.

  • The production of laccase by Fomitella fraxinea was studied. The addition of minerals were necessary far laccase production by Fomitella fraxinea. Jar fermentor and Air-sparging fermentor performed high productivity In laccase activity by F. fraxinea. Laccase activity reached 3,540 in 8 days (Jar fermentor) and 3,100 in 6 days (Air-sparging fermentor) respectively.

  • In an attempt to develop a novel process for ethanol production from starch, a simultaneous saccharification and fermentation (SSF) process using Zymomonas mobilis and amyloglucosidase (AMG) was studied in continuous modes. Compared with a conventional cylindrical column type of fermentor, the tapered column type of fermentor was found to be superior in terms of reactor performance for ethanol fermentation. The tapered columm fermentor packed with coimmobilized Z. mobilis and AMG alleviated the problems which were associated with CO2 evolution and provided a significantly better flow pattern for both liquid and gas phases in the fermentor without channelling. However, the fluidized bed type of tapered column fermentor using flocculent strain of Z. mobiles and immobilized AMG showed lower productivity (5.2g/1/h) than that of packed bed type of tapered column fermentor(9.2g/l/h).

  • The effects of fermentor types on biopolymer production with Cordyceps militaris has been studied in this articles. Since the shear stresses of stirred tank ferementor and bubble column fermentor were different, the mycelium growth and biopolymer production were different. The mycerial growth was high with the bubble column fermentor for the early stage but biopolymer production was lower than that with stirred tank fermentor. The production of biopolymer was closely related to shear stress to the cells and biopolymer production was increased with increased shear stress.

Comparison of Rheological Properties of Powder Chlorella sp. Cultivated in Fermentor and Pond

  • The current study was conducted to identify the differences in the rheological properties of Chlorella sp. powder cultured in a fermentor and in a pond-like environment. Cells. cultured in the same media were harvested and spray dried. The biomass yield from the fermentor culture was 4.7% (dry basis), while that from the pond was 4.3% (dry basis). Measurements of the loose bulk density, tapping test, Hausner's ratio, and compressibility test all revealed differences between the rheological properties of the Chlorella sp. from the two cultivation systems. Although both the fermentor and pond cultured Chlorella sp. showed the same angle of repose, the mean size of the cells was 2.26 $\mu\textrm{m}$ and 2.89 $\mu\textrm{m}$, respectively. The weight of the Chlorella sp. tablets cultured in the fermentor and pond was 0.663 g/tablet and 0.593 g/tablet, respectively, while the friability of the tablets was 21% and 41%, respectively. Observation by Transmission Electron Microscope (TEM) showed that the cell wall of the Chlorella sp. cultured in the fermentor was thinner and more spherical than that cultured in the pond, thereby providing the main characteristic rheological properties of the powder.

  • The productivity of alkaloid in the airlift fermentor operation was less than that of suspension coltures of Eschscholtzia californica cells in the shake flask. To overcome the productivity reduction, a gas recycle airlift fermentor was developed because the gas-stripping in normal airlift fermentor was believed to play a significant role for productivity reduction. The alkaloid content in the gas recycle system with Eschscholtzia californica suspension cells was 2.7 times higher than that of normal airlift fermentor. The productivity of alkaloids and $CO_2$ concentration were affected by the volume of gas reservoir in the gas recycle airlift fermentor.

  • In order to predict gas hold-up and oxygen transfer rate during sisomicin fermentation employing air lift fermentor, simulated media similar to fermentation broths in rheological proper- ties were prepared and used. Rheological properties of fermentation broths from 40 hours and 60 hours of cultivation were analyzed by applying to Power's Law equation. Regardless of addition and no addition of MgSO$_{4}$, the tendencies, that n value was decreased and K value was increased as aeration rate was increased, were shown. Simulated media of twelve different fermentation broths were formulated in a range of 0.7 to 2.1% CMC, and the values of gas hold-up and k$_{L}$a depending on superficial air velocity were measured using these simulated media. And the relation- ships, $\varepsilon$=$\alpha$U$_{Gr}$$\beta$, K$_{L}$a=$\gamma$U$_{Gr}$$\delta$ were obtained, and these equations are thought to be used to predict the values of gas hold-up and k$_{L}$a during fermentation.

  • A cone-type tower fermentor packed with Sacchromyces uvarum was employed to examine the continuous ethanol fermentation process. The maximum yeast concentration in the cone-type tower fermentor was 37.5-39.5g/$\ell$, the maximum ethanol productivity at the dilution rate of $hr^{-1}$ was 16.3g/$\ell$ . hr and the average ethanol yield was 0.48g EtOH/g glucose, which was 94% of the maximum theoretical yield. It was concluded that a cone-type tower fermentor might offer better perspectives for continuous ethanol fermentation.

Application of Scale-Up Criterion of Constant Oxygen Mass Transfer Coefficient ($k_La$) for Production of Itaconic Acid in a 50 L Pilot-Scale Fermentor by Fungal Cells of Aspergillus terreus

  • The scale-up criterion of constant oxygen mass transfer coefficient ($k_La$) was applied for the production of itaconic acid (IA) in a 50 L pilot-scale fermentor by the fungal cells of Aspergillus terreus. Various operating conditions were examined to collect as many $k_La$ data as possible by adjusting the stirring speed and aeration rate in both 5 L and 50 L fermentor systems. In the fermentations performed with the 5 L fermentor, the highest IA production was obtained under the operating conditions of 200 rpm and 1.5 vvm. Accordingly, we intended to find out parallel agitation and aeration rates in the 50 L fermentor system, under which the $k_La$ value measured was almost identical to that ($0.02sec^{-1}$) of the 5 L system. The conditions of 180 rpm and 0.5 vvm in the 50 L system turned out to be optimal for providing almost the same volumetric amount of dissolved oxygen (DO) into the fermentor, without causing shear damage to the producing cells due to excessive agitation. Practically identical fermentation physiologies were observed in both fermentations performed under those respective operating conditions, as demonstrated by nearly the same values of volumetric ($Q_p$) and specific ($q_p$) IA production rates, IA production yield ($Y_{p/s}$), and specific growth rate (${\mu}$). Specifically, the negligible difference of the specific growth rate (${\mu}$) between the two cultures (i.e., $0.029h^{-1}$ vs. $0.031h^{-1}$) was notable, considering the fact that ${\mu}$ normally has a significant influence on $q_p$ in the biosynthesis of secondary metabolites such as itaconic acid.

Characterization and Stability of Gardenia Jasminoides Biotransformed Pigment Produced in Jar Fermentor (Jar Fermentor에서 생산된 치자 생물변환 색소의 특성 및 안정성)

  • Yellow pigment of Gardenia jasminoides was converted into new pigment by whole-cell biotransformation of thirteen different microbial species. The color value of the biotransformed pigment, which was produced by Streptococcus mutans MK-34, was higher than those of other biotransformed pigments. The biotransformed pigment produced by S. mutans MK-34 dispalyed an characteristic absorption peak at 588 nm and the absorption value increased during the incubation in a jar fermentor. The effects of light and temperature $(60^{\circ}C)$ on storage stability of the biotransformed pigment were investigated. As a result, the biotransformed pigments produced by Streptococcus mutans and Bacillus subtilis were more stable than Gardenia jasminoides yellow pigment during storage.
