발효 금은화 및 황련의 유해균 억제 효과 (Antimicrobial Effects of Fermented Coptidis rhizoma and Lonicerae Flos against pathogen)

  • Objectives This study was designed to examine antimicrobial effects of Fermented Coptidis rhizoma and Lonicerae Flos against pathogens. Methods Lactobacilli MRS broth was added to 200mL glass bottle containing 20% herb powder(w/v) followed by 30 minute sonication and then shaking at 70 rpm in $70^{\circ}C$ water bath for 3 hours in order to extract fermented herb. Fermented herb extract was autoclaved at $121^{\circ}$ for 15minutes. $2{\times}10^7$ CFU/mL subcultured bacteria was inoculated and cultured for 24 hours and centrifuged at 3000 rpm for 5 minutes. After transferring to 15 mL conical tube, the viable cells were counted. Results and Conclusion Fermented Coptidis rhizoma and Lonicerae Flos both showed antimicrobial effect on pathogens especially when Fermented Coptidis rhizoma was experimented against Staphylococcus aureus.

배독환발효방(排毒丸醱酵方)의 면역조절작용을 통한 항아토피 효능 (Effects of Baedokhwanbalhyobang (BDHBH) on Immune Modulation in Dermatitis Model of NC/Nga Mice)

  • Herbal medicine has a high body absorption rate when it ferments. Biological and clinical research on the fermented herb gradually increases because it has effective materials for the treatment of a disease and it is a little bitter. In this study, we investigated the effect of fermented Baedokhwanbalhyobang (BDHBH) on attenuation of the development of atopic dermatitis in NC/Nga mice by evaluating the cytokine level in serum, the mRNA expression of cytokine and histological alteration of the skin, and the skin severity. We have come to the following conclusion. BDHBH led to a significant decrease in the skin severity score (63.1%) as compared to the control group. CD4+/CD45+, CD4+, B220+/CD23+, and CD11b+/Gr-1+cells of peripheral mononuclear cells (PBMCs) in the BDHBH-treated group were decreased to 6.7%, 31.1%, 22.4%, 36.6%, respectively. CD3+and CD11b+/Gr-1+immune cells in dorsal skin of the BDHBH-treated group were decreased to 52.9% and 28.0%. The levels of IL-5 and IL-13 in serum of the BDHBH-treated group were inhibited to 18.8% and 5.1%. The mRNA expressions of IL-5 and IL-13 in dorsal skin were also decreased to 30.6% and 27.8% after the treatment of BDHBH. BDHBH inhibited the proliferation and differentiation of eosinophils. In histological examination, BDHBH decreased the thickness of epidermis and dermis, and infilatration of mast cells as compared to the control group. These results indicate that BDHBH inhibits the pathogenic development of atopic dermatitis in NC/Nga mice. These results may indicate that BDHBH attenuates the development of atopic dermatitis-like lesions by lowering immune cells and inflammatory cytokine levels, and that it is valuable in drug development for the treatment of atopic dermatitis. Further experiments on the components of BDHBH will be needed to better understand the effect of a fermented herb as compared to a herb.

4종의 발효균주가 당귀의 nodakenin과 decursin의 함량에 미치는 변화 (Nodakenin and Decursin Contents of Fermented Angelicae Gigantis Radix by 4 Species Strain)

  • Objectives : The purpose of this study was to investigate the nodakenin and decursin contents in each fermented Angelicae gigantis Radix by 4 species of ferment strains. Methods : The strains of fermented Angelicae gigantis Radix were Lactobacillus plantarum(SE1), L. acidophilus(AC), Bacillus subtilis(B2) and B. licheniformis(BL2). The fermentative changes of nodakenin and decursin were analyzed using HPLC. Results : All of 4 species strains reduced nodakenin and decursin concentration in Angelicae gigantis Radix by fermentation. But fermentability were difference between strains. The nodakenin concentration of AC was lower than B2. The decursin concentrations of SE1 and AC were lower than B2 and BL2. Conclusions : 4 species of fermental strain were difference decomposable rate with nodakenin and decursin in fermented Angelicae gigantis Radix.

천연발효 경과에 따른 삼정환의 미생물 변화 및 발효특성 (Microbial Change and Fermentation Characteristics during Samjung-Hwan Natural Fermentation)

  • Objectives: Samjung-hwan (SJH), a well-known traditional fermented herb formula recorded in Dongui Bogam, has been commonly used for prolonging life for four hundred years in Eastern Asia. However, fermented SJH has not been investigated in terms of microbial ecology until present time. Methods: SJH was fermented for five weeks and fermentation characteristics during SJH fermentation were performed including pH, acidity and microbial profiling. Also, we measured total polyphenol and total flavonoid contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. In order to select starter candidate, several lactic acid bacteria were isolated from fermented SJH. Results: pH of fermented SJH was decreased from 4.7 to 3.0 and acidity was increased from 0.45% to 1.72%. Also, fermented SJH increased antioxidant indicator such as total polyphenol and total flavonoid as well as DPPH free radical scavenging activity. Lactobacillus brevis was increased, Pseudanabaena sp. was decreased, and Lactococcus lactis subsp. lactis was stable during 5-week fermentation of SJH. L. brevis and L. plantarum were isolated from fermented SJH. Conclusions: Fermented SJH for four weeks had optimal effect on antioxidant and fermentation characteristics such as pH, acidity and microbial profile. Further studies are required to develop starter and analyze functional compounds in oder to produce standardized SJH.

A Fermented Ginseng Extract, BST204, Inhibits Proliferation and Motility of Human Colon Cancer Cells

  • Panax ginseng CA Meyer, a herb from the Araliaceae, has traditionally been used as a medicinal plant in Asian countries. Ginseng extract fermented by ginsenoside-${\beta}$-glucosidase treatment is enriched in ginsenosides such as Rh2 and Rg3. Here we show that a fermented ginseng extract, BST204, has anti-proliferative and anti-invasive effects on HT-29 human colon cancer cells. Treatment of HT-29 cells with BST204 induced cell cycle arrest at $G_1$ phase without progression to apoptosis. This cell cycle arrest was accompanied by up-regulation of tumor suppressor proteins, p53 and p21$^{WAF1/Cip1}$, down-regulation of the cyclin-dependent kinase/cyclins, Cdk2, cyclin E, and cyclin D1 involved in $G_1$ or $G_1/S$ transition, and decrease in the phosphorylated form of retinoblastoma protein. In addition, BST204 suppressed the migration of HT-29 cells induced by 12-O-tetradecanoylphorbol-13-acetate, which correlated with the inhibition of metalloproteinase-9 activity and extracellular signal-regulated kinase activity. The effects of BST204 on the proliferation and the invasiveness of HT-29 cells were similar to those of Rh2. Taken together, the results suggest that fermentation of ginseng extract with ginsenoside-${\beta}$-glucosidase enhanced the anti-proliferative and the anti-invasive activity against human colon cancer cells and these anti-tumor effects of BST204 might be mediated in part by enriched Rh2.

황련해독탕 및 발효황련해독탕의 항염증 효과 (Anti-inflammatory effects of Hwangnyeonhaedok-tang and Fermented Hwangnyeonhaedok-tang)

  • Objective : Recently interest of fermented herb medicine has increased in Oriental medicine. Herb medicine fermented in a proper condition resultingly has better or new virtue by microbe. This study was designed to compare the anti-inflammatory effects of herbal decoction and fermented herbal decoction in Hwangnyeonhaedok-tang (Huanglian fiedu-tang; HHT). Methods : The effects of HHT and FHHT on anti-inflammation were measured by production of NO and pro-inflammatory cytokine productions including TNF-${\alpha}$ and IL-1${\beta}$, IL-6 in LPS induced RAW 264.7 cells. Results : 1. Cell viability in three doses of HHT extract (0.01, 0.05 and 0.10 mg/ml) and two doses of FHHT extract (0.01, 0.05 mg/ml) was more than 70% and 80% each. 2. In RAW 264.7 cells, LPS displayed significant stimulatory effects on the production of NO. However, both HHT, FHHT showed significant inhibitory effects on the production of NO. 3. Pre-treatment by HHT in a various concentrations (0.01, 0.05, 0.10, 0.50mg/ml) reduced the elevated production of TNF-${\alpha}$, IL-1${\beta}$, IL-1${\beta}$ mRNA, IL-6 mRNA. More than 0.05mg/ml concentrations of HHT reduced the production of IL-6. And TNF-${\alpha}$, IL-1${\beta}$ level of mouse serum reduced in a various dose(0.1, 0.5, 1.0g/kg). 4. Pre-treatment by FHHT in a various concentrations (0.01, 0.05, 0.10, 0.50mg/ml) reduced the elevated production of TNF-${\alpha}$. More than 0.05mg/ml concentrations of FHHT reduced the expression of TNF-${\alpha}$ mRNA. 0.1, 0.50mg/ml concentrations of FHHT reduced the production of IL-6. And IL-1${\beta}$ level of mouse serum reduced in a various dose(0.1, 0.5, 1.0g/kg). Conclusions : These results indicated that both HHT and FHHT could have anti-inflammatory capacity by inhibiting the production of NO and cytokines in vitro. But HHT has more strong anti-inflammatory effects than that of FHHT in broad outlines.

The Screening of Fermented Medicinal Herbs to Identify Those with Anti-inflammatory Properties

  • Objectives : Consumption of fermented foods has been known to alleviate some of the symptoms of atopy and may limit allergy development, while there are also many medicinal herbs proved to be effective for immunologically-mediated diseases. In this study, we introduced modern zymology to ferment some herbs to see if fermentation has the possibility of increasing the anti-inflammatory effects of medicinal herbs. Interleukin-4 (IL-4) and interferon-gamma $(INF-\gamma)$ have been demonstrated to be the main factors in the pathology of allergic diseases. Methods : We measured the levels of IL-4 and $INF-\gamma$ on concanavalin A-induced BALB/c mice spleen cells, which were subsequently treated with fermented and unfermented herbs. We then compared the fermented groups with unfermented groups to see if the anti-inflammatory effects of the herbs were influenced by fermentation. Results and Conclusions : Our results showed that fermentation had the potential to increase the anti-inflammatory effects of some medicinal herbs, and Astragalus membranaceus and Salvia miltiorrhiza would be the most suitable medicinal herbs for fermentation among the herbs in this study.

Effects of fermented black ginseng on wound healing mediated by angiogenesis through the mitogen-activated protein kinase pathway in human umbilical vein endothelial cells

  • Background: Fermented black ginseng (FBG) is produced through several cycles of steam treatment of raw ginseng, at which point its color turns black. During this process, the original ginsenoside components of raw ginseng (e.g., Re, Rg1, Rb1, Rc, and Rb2) are altered, and less-polar ginsenosides are generated (e.g., Rg3, Rg5, Rk1, and Rh4). The aim of this study was to determine the effect of FBG on wound healing. Methods: The effects of FBG on tube formation and on scratch wound healing were measured using human umbilical vein endothelial cells (HUVECs) and HaCaT cells, respectively. Protein phosphorylation of mitogen-activated protein kinase was evaluated via Western blotting. Finally, the wound-healing effects of FBG were assessed using an experimental cutaneous wounds model in mice. Results and Conclusion: The results showed that FBG enhanced the tube formation in HUVECs and migration in HaCaT cells. Western blot analysis revealed that FBG stimulated the phosphorylation of p38 and extracellular signal-regulated kinase in HaCaT cells. Moreover, mice treated with $25{\mu}g/mL$ of FBG exhibited faster wound closure than the control mice did in the experimental cutaneous wounds model in mice.

백출 및 발효백출의 장점막 투과성 개선 효과 및 항염증효과 (Effect of Unfermented and Fermented Atractylodes macrocephalae on Gut Permeability and Lipopolysaccharide-Induced Inflammation)

  • Objectives: The aim of this study is to investigate anti-imflammatory and protective effect for intestinal epithelial cells with Atractylodes macrocephae (AM), a traditional Korean Herbal medicine and fermented Atractylodes macrocephae (FAM) with Lactobacillus plantarum. Methods: HCT-116 and Raw 264.7 cells were used in this study. Using NO assay, we measured lipopolysaccharide (LPS)-induced anti-inflammatory effect. We measured permeability of intestinal epithelial cells with transepithelial electrical resistance and horseradish peroxide flux assay. Water soluble tetrazolium salt assay was used to see cell proliferation. All the results were presented in mean and standard deviation. We used Student's t-test for analyzing significance of results. Results: In Raw 264.7 cells NO production decreased 22.4% with pre-treatment of AM and FAM, especially with FAM in high concentration. In HCT-116 cells LPS-induced intestinal permeability had a protective effect with both AM and FAM, which was also tend to be proportional to the concentration. Cell viability increased up to 135.52% after treatment of high concentration of FAM in HCT-116, while there was no significant change in Raw 264.7 cells with herb treatments. Conclusions: These results show evidence that AM, especially fermented ones, significantly reduced intestinal membrane permeability. They also had a protective effect as well as an anti-inflammation effect for HCT-116 and Raw 264.7 cells. This suggest that FAM may be a therapeutic agent for Leaky gut syndrome by reducing intestinal permeability.

마우스 악성흑색종세포에서 울릉도 곰취, 미역취, 삼나물 추출물의 멜라닌 생성 억제 효과 (Antimelanogenic Effect of Ligularia fischeri, Solidago virga-aurea, Aruncus dioicus Extracts from Ullung Island in Murine Melanoma Cells)

  • 현재까지 많은 연구들은 멜라닌 합성과정의 기전을 규명하는데 초점이 맞춰졌으며 이러한 이유로 tyrosinase 저해제 개발에 집중되어 왔다. 이에 본 연구는 울릉도 곰취, 미역취, 삼나물 추출물을 이용하여 미백효과를 검증하였다. tyrosinase 저해 활성을 측정한 결과 울릉도 곰취, 미역취, 삼나물 발효 전 추출물의 경우 33%, 발효 후 추출물의 경우 45% 저해율을 세포 내 tyrosinase 활성 확인 결과 울릉도 곰취, 미역취, 삼나물 발효 전 추출물이 41%, 발효 후 추출물은 56.5%의 활성 저해율을 관찰 할 수 있었으며, 발효 후 추출물이 발효를 하지 않은 추출물보다 미백 효과가 더 뛰어 남을 확인할 수 있었다. 미백 관련 전사 인자인 MITF, TRP-1, TRP-2, tyrosinase의 단백질 발현을 측정한 결과 발효 전 후 추출물은 B16F10 melanoma 세포에서 tyrosinase, TRP-1과 TRP-2의 단백질 생합성을 두 추출물 모두 억제하였지만, 특히 발효 후 추출물의 경우 단백질 발현량이 더 감소하였다. Tyrosinase, TRP-1과 TRP-2의 발현을 조절하는 전사인자로는 MITF가 관여하는 것으로 알려져 있으며, 실제로 MITF는 melanin생성과 관련된 여러 유전자의 발현을 조절하는데 중요한 작용을 하고 있다. 따라서 울릉도 발효 후 추출물이 melanin 생성과 관련된 중요한 세 가지 단백질의 생합성을 전사단계에 서 조절 전사인자인 MITF의 작용에 대한 효과를 확인한 결과 울릉도 발효 후 추출물이 MITF의 단백질 발현을 울릉도 발효 전 추출물보다 생성을 억제하는 효과가 더 높음을 확인하였다. 이러한 결과를 토대로 미백 생합성에 있어서 상위 신호단계에 있는 전사인자 MITF의 활성을 억제함으로써 하위 신호전달 과정을 억제하는 것임을 알 수 있었다. 이상의 결과로 미루어 보아 발효 후 추출물의 미백효과가 있음을 확인 할 수 있었으며 앞으로의 연구를 뒷받침 하여 기능성 소재 개발가능성을 제시하고 있다.
