This study investigates the antioxidant and anti-adipogeneic effects of fermented Rhus verniciflua., evaluating the total phenol, total flavonoids contents and antioxidant activity of the fermented Rhus verniciflua as well as assessing the lipid accumulation during adipogenesis of 3T3-L1 cells. Our results demonstrate that the total phenolic and flavonoids contents of fermented Rhus verniciflua were $29.2{\pm}0.12$ GAE mg/g and $20.4{\pm}1.52$ RE mg/g, respectively. The antioxidative activities of fermented Rhus verniciflua were significantly increased in a dose dependent manner on DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical scavenging, ABTS(2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging, FRAP(ferric reducing antioxidant power)activity, reducing power. In addition, the fermented Rhus verniciflua did not show any cytotoxicity up to 300 ug/mL. However, the anti-adipogenic effect of fermented Rhus verniciflua extract was barely detectable.
Urushinol, a plant allergen, has significantly restricted the medical application of $Rhus$$verniciflua$, although it has been reported to possess a wide variety of biological activities such as anti-inflammatory, antioxidant, and anti-cancer actions. To reduce the urushinol content while maintaining the beneficial biological activities, mushroom-mediated fermentation of $Rhus$$verniciflua$ was carried out and this method resulted in significantly attenuated allergenicity [1]. In the present study, to examine the neuroprotective properties of mushroom-fermented stem bark of $Rhus$$verniciflua$, two constituents were isolated from mushroom-fermented bark and their neuroprotective properties were examined in a mouse model of kainic acid (KA)-induced excitotoxicity. KA resulted in significant apoptotic neuronal cell death in the CA3 region of mouse hippocampus. However, seven daily administrations of RVH-1 or RVH-2 prior to KA injection significantly attenuated KA-induced pyramidal neuronal cell death in the CA3 region. Furthermore, pretreatment with RVH-1 and RVH-2 also suppressed KA-induced microglial activation in the mouse hippocampus. The present study demonstrates that RVH-1 and RVH-2 isolated from $Rhus$$verniciflua$ and detoxified using mushroom species possess neuroprotective properties against KA-induced excitotoxicity. This leads to the possibility that detoxified $Rhus$$verniciflua$ can be a valuable asset in herbal medicine.

This study was designed to investigate the antihyperlipidemic activity of Fermented Rhus Verniciflua Stokes Extract(FRVSE) on diet-induced hyperlipidemia in rats. The animals were divided into 4 groups of the normal group(fed with normal diet), control group(fed with high fat diet), sample A group(fed with high fat diet and medicated FRVSE 192 ㎎/㎏/day), sample B group(fed with high fat diet and medicated FRVSE 384 ㎎/㎏/day), and received oral administration of each prescription with diet for a period of six weeks. The changes in the body weight, the liver weight, the epididymal fat weight, and the plasma levels of total cholesterol, triglyceride(TG), high density lipoprotein(HDL) cholesterol and low density lipoprotein(LDL) cholesterol were measured. In sample A group, the weight of liver and plasma level of TG decreased significantly. There was a significant decrease in the plasma level of total cholesterol, TG and LDL cholesterol in the sample B group. These results suggest that the FRVSE have a good antihyperlipidemic activity and a potential to treat hyperlipidemia.
To investigate effects of fermentation on volatile components, we analyzed volatile organic compounds of raw and fermented Rhus verniciflua S. bark. A 50%(w/v) sugar solution was used for fermentation. Volatile organic compounds of raw and fermented Rhus verniciflua S. were extracted by the simultaneous steam distillation and extraction(SDE) method, with a mixture of n-pentane and diethylether(1:1, v/v) and analyzed by gas chromatograph-mass spectrometer. A total of 51 and 27 volatile organic compounds were detected in raw and fermented samples, respectively, and were mainly alcohols. Compounds such as ethyl acetate, 2-methyl-3-buten-2-ol, 3-methylbutanal, 1-octen-3-o1, 3-methyl-2-butanone, hexanal and pentanal were detected as the primary compounds in the raw sample. The fermented sample showed sour different volatile compounds, such as ethanol, ethyl acetate, ethyl lactate and 3-methylbutanol. Thus, a number of volatile organic compounds were synthesized after fermentation of Rhus verniciflua S. bark.
Fustin comprised >98 and 73.0-86.7% of the total flavonoid content in the bark, and the stems and lignum of Rhus verniciflua, respectively. The butein, fisetin, and sulfuretin content varied between 0.31-2.17, 0.27-3.32, and 0.15-0.80 mg/g on a dry weight basis, respectively, in different parts of Rhus verniciflua. The urushiol content was 5.09-6.29, 55.05-56.30, and 0.38-0.39 mg/100 g on a dry weight basis in the stems, bark, and lignum, respectively. This showed that the bark of the tree had the highest urushiol content. C15:3 (pentadecatrienyl catechol), C15:1 (pentadecenyl catechol), and C15:2 (pentadecadienyl catechol) comprised 63, 33-35, and 2-3% of urushiol congeners in the tree bark, respectively.

Fermented Rhus verniciflua (FRV) as feed additives act as excellent anti-diarrheal drugs as they increase the intestinal absorption rate therefore being indirectly associated with enhancing growth performance and increasing digestibility in livestock. A total of 80 weaned pigs with an average initial body weight (BW) of $6.82{\pm}1.11kg$ were used to evaluate a diet supplemented with FRV meal in a 6 week feeding trial with two dietary treatments [CON - basal diet; TRT - CON + 0.2% FRV] on growth performance, apparent total tract digestibility (ATTD), blood profiles, fecal microflora, and fecal score. Our results showed that the dietary supplementation of FRV improved (p < 0.05) average daily gain and gain : feed (G : F) ratio during days 15 - 42 and the overall experiment period and also increased the ATTD of dry matter (p < 0.05) at days 14 and 42. On the contrary, there was no effect (p > 0.05) on average daily feed intake, ATTD of nitrogen and energy, and blood profiles during the entire experiment. Moreover, dietary inclusion of FRV significantly increased fecal Lactobacillus (p < 0.05) counts and reduced the diarrhea during days 22 - 42. Thus, the results suggest that FRV can be used as a potential additive to improve growth performance and dry matter and to reduce diarrhea while having beneficial effects on fecal microflora in weanling pigs.
This study was carried out to investigate the quality characteristics of fermented milk containing Rhus verniciflua seed (RVS) extracts (0.0%, 0.1%, 0.3% and 0.5%) and antioxidant activity of RVS extracts. The proximate composition of RVS was moisture (4.76%), crude ash (1.40%), crude fat (5.33%) and crude protein (11.10%). A total polyphenol content of 70% ethanol extract of RVS ($554{\pm}2.64mg/g$) was higher than that of RVS water extract ($145{\pm}3.47mg/g$). 70% ethanol extract ($1103{\pm}6.42mg/g$) of RVS showed higher content in the total flavonoids ($37{\pm}2.30mg/g$) and activities on DPPH free radical scavenging (73.23%) and ABTS free radical scavenging (83.47%) when compared with water extracted of RVS. The quality characteristics, such as pH, titratable acidity, and the number of lactic acid producing bacteria were not remarkably different between the fermented milk samples subjected to treatments with and without the addition of RVS extracts.
In this study, we investigated the biological activities such as anti-tumor, anti-oxidant and antiinflammatory activities as well as single oral dose toxicity of fermented Rhus verniciflua extract (FRVE). In order to examine anti-tumor activity of FRVE, the sarcoma 180 cells were treated with FRVE at various concentrations (0.03, 0.3, 3 and 30 mg/mL) in microtetrazolium (MTT) assay. In MTT assay, all the cells treated with FRVE at various concentrations have shown a significant difference compared with control (p < 0.05). In xanthine oxidase inhibition assay to examine the antioxidant activity, the xanthine oxidase inhibition rate of FRVE at 1.5 mg/mL and 15 mg/mL was $85{\pm}15.01%$ and $99{\pm}16.02%$, respectively. Nitric oxide production in RAW 264.7 cells showed that FRVE showed a significant anti-inflammation effect at 3 mg/mL (p < 0.05). In single oral dose toxicity study, no differences were observed between control and treated groups in clinical signs, body weight gains, feed and water consumptions. The results indicated that lethal dose 50 ($LD_{50}$) of FRVE was found to be higher than 5,000 mg/kg in this experiment. From the above results, we may suggest that FRVE might have useful as a material for functional food and/or animal pharmaceutics.
We studied the effect of fermented Rhus verniciflua stem bark (FRVSB) extract (used in herbal med-icine by Koreans) on the microbial growth and enzyme activity of 12 soybean-fermenting microorganisms, including Bacillus spp., lactic acid bacteria, yeast, and other harmful bacteria. The ethanol and methanol extracts of FRVSB inhibited the growth of Bacillus subtilis, Bacillus licheniformis, Bacillus cereus, and Zygosaccharomyces rouxii, and in the disk diffusion assay, their inhibition zone diameters were 11.06-12.23, 12.32-18.38, 11.47-11.84, and 13.59-14.21 mm, respectively. The water extract did not show any inhibitory effect. In fact, the water extract addition enhanced the growth of B. subtilis and B. licheniformis by 1.3-4.5 fold and that of B. cereus by 1.2-1.4 fold. However, the water extract did not affect the growth of Lactobacillus plantarum, Lactobacillus mesenteroides, Saccharomyces cer-evisiae, and Escherichia coli. The addition of water extract increased the amylase and protease activity of B. subtilis and B. licheniformis.

Rhus verniciflua (RV) Stokes is a herbal medicine that helps improve blood circulation by stimulating digestion, removing extravasated blood, and raising body temperature. The purpose of this research was to study the anti-inflammatory effect of fermented soy products (FSP) containing a fermented RV (FRV) extract on lipopolysaccharides (LPS)-treatedd RAW 264.7 cells. Treatment with FRV extracts (1, 10, $100{\mu}g/mL$) downregulated nitric oxide (NO) and pro-inflammatory cytokines as compared to the LPS-treated group. Besides, the RV extract treatment suppressed the expression of genes related to pro-inflammatory cytokines, matrixins, inflammation, and apoptosis, while increasing the expression of genes involved in the antioxidant system. Furthermore, RVS extract upregulated antioxidant enzymes, such as glutathione, Cu,Zn-SOD, and catalase without changes in the Nrf2-Keap1 pathway. FSP (doenjang, ganjang) containing FRV extracts (0.1, 1, or $10{\mu}g/mL$) significantly decreased the NO and IL-6 levels in an FSP after 8 weeks of fermentation, but not the expression of genes involved in the inflammation and antioxidant system. These result indicate that an FRV extract and FSPs have a potential application in inflammatory conditions.
