Pregnancy block by male pheromones in mice differs in incidence depending on the combination of strains. Female mice of BALB/cA strain mated with BALB/cA males show a 100% pregnancy block when exposed to males of inbred strain DDK shortly after copulation (Chung et al., Biol Reprod 1997). In the present study, BALB/cA females mated with the males of other strains (CBA/J, C3H/HeN, C57BL/6Cr, and IXBL) showed higher pregnancy rates (66.6-87.5%) even when they were exposed to DDK males. In the pharmacological induction of pregnancy block with dopamine agonist (Bromocriptine, 4mg/kg BW), BALB/cA females mated with BALB/cA males showed a 100% pregnancy block. In contrast, BALB/cA females mated with CBA/J, C3H/HeN, and C57BL/6Cr males showed higher pregnancy rates (40-70%). These results suggest that the better pregnancy rate of BALB/cA females mated with alien males may be due to the stronger viability of F 1 embryos. This interpretation was confirmed by an embryo transfer experiment in which a higher implantation rate was observed when BALB/cA embryos grown in BALB/cA females exposed to BALB/cA males were transferred into recipient BALB/cA females exposed to DDK males. These results suggest that the embryonic genotype or viability of the embryo is one factor contributing to the occurrence of pregnancy block by male pheromones in mice.
1bis study investigated the antioxidative effect in kidney of senescence-accelerated prone SAMP8 mice with calorie restriction. 4-weeks-old SAMP8 female mice were divided into 4 groups according to the experimental feeding period: for 4, 8, 12 month, and at natural death. Each group was subdivided into 2 groups, with thirteen mice each one, as ad libitum group and as dietary restriction group (60% of ad libitum feeding amount). After feeding for a given period, the mice were sacrificed to get the following results: among the experimental groups, there wereno significant differences in xanthine oxidase (XOD) activity in their kidney tissues. The contents of cytochrome $P_{450}$ decreased in ad libitum group and dietary restriction group by age. The activity of NADPH-cytochrome $P_{450}$ reductase showed a trend similar to cytochrome $P_{450}$. Superoxide radical content increased with age. At the 4th, 8th and 12 months of the experimental period, the activity in the dietary restriction group was less than that of ad libitum group by as much as 17% 14% and 14% respectively. For hydrogen peroxide, the contents were increased in the ad libitum group with age, while no correlation between content and age was observed in the dietary restriction group. In the 8th and 12th months of the experimental period, the were in the dietary restriction group less than that of ad libitum group counterpart as much as 17% and 20o/c, respectively. For the cellular membrane stability of the kidney, no significant correlation with age was observed in either the dietary restriction group or the ad libitum group. However at the 12th month of the experiment, however, the stability in the dietary restriction group was 11 % higher than that in the ad libitum group. In conclusion, with these results obtained from the SAMP8 mouse model, we demonstrate that dietary restriction has the effects of anti-oxidation and anti-senescence in the kidney.
Objective: Photodynamic therapy (PDT) is an emerging therapeutic procedure suitable for the treatment of cervical cancer. However, the side effects of PDT are severe, including skin ulceration, so we designed an experiment to examine the effects of multiple low-dose photodynamic therapy of 5, 10, 15, 20-tetrakis(1-methylpyridinium-4-yl) porphyrin (Tmpyp4) on tumour growth by utilizing a model in nude mice implanted with Hela cervical cancer cells. Materials and Methods: Female BALB/c nude mice (aged 5-6 weeks, weighing 18-20 g) were used. Hela cervical cancer cells were injected subcutaneously ($1{\times}10^7cells/200{\mu}L$). Ten days after injection, the mice were divided into three groups (n=6), the A group of controls without any treatment, the B group receiving a single-treatment with Tmpyp4 (10 mg/kg, intratumor injection) and irradiation (blue laser, $108J/cm^2$), and the C group given three-treatments with Tmpyp4 (10 mg/kg, intratumor injection) and irradiation at intervals of two days. After starting treatment, tumours were measured every two days, to assess growth. At 2 weeks after the last treatment of C group, tumour tissue and organs were collected from each mouse to evaluate tumor histology and organ damage. Results: Tumour growth in C group was significantly inhibited compared with A and B groups (P<0.05), without any injury to the skin and internal organs. Conclusion: Our novel findings demonstrated that multiple low-dose photodynamic therapy of Tmpyp4 could inhibit cervical cancer growth significantly with no apparent side effects.

The present study was carried out to investigate the potential acute toxicity of Leuconostoc citreum GJ7 (Leu. citreum GJ7), a lactic acid bacterium isolated from Kimchi, in ICR male and female mice. The test article was administered to the mice orally or intraperitoneally. Mortality rates, clinical findings, and body weight changes were monitored for the 2 weeks following administration. The results showed that in 50% of the cases, lethal doses ($LD_{50}$) of Leu. citreum GJ7 were determined as >5, 000 mg/kg (p.o.) and >2500 mg/kg (i.p.) in both sexes. There were no significant changes in general conditions, body weights clinical signs and any gross lesions between vehicle control and Leu. citreum GJ7-treated groups. Hence, it is suggested that Leu. citreum GJ7 does not induce any significant acute toxicity in ICR mice.

This study was performed to evaluate hepatoprotective effect of Daewhang-whangryunhaedok-Tang(DWT) on liver injured rats induced by $CCl_4$ and the acute oral toxicity of it in mice. The activities of serum transaminase(ALT/AST), alkaline phosphatase(ALP) and lactic dehydrogenase (LDH), the levels of serum total cholesterol(TC) and triglyceride(TG), change of liver enlargement, and inhibitory activities of lipid peroxidation, catalase and glutathione-S-transferase(GST) in liver microsome were determined in hepatotoxic rats induced by $CCl_4$ DWT DWT was significantly reduced the serum ALT, AST, ALP, LDH, TC and TG levels. And, the increase of lipid peroxidation, decrease of catalase and GST activities in the liver microsome of $CCl_4$-intoxicated rat were significantly improved by the treatment of DWT. Male and female mice were administered maximum dosages of 5,000 mg/kg b.w. of DWT. After single oral administration of DWT to mice, we observed them daily for 2 weeks. DWT did not induce any toxic signs in the mortalities, clinical signs, body weight changes, and gross necropsy findings of mice. Based on these results, it is concluded that DWT may have the hepatoprotective effect on $CCl_4$ induced hepatotoxicity in rats. Also, DWT may have no side effect and its $LD_{50}$ value may be over 5,000 mg/kg b.w. in mice.
Purpose : The Purpose of this research was to investigate the effects of Cheongyeolhawlhyeoltanggagyehyeoldeung (CYHHT) on anti-inflammatory effects. Methods : As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators were determined in mouse lung fibroblast cells(mLFC) and RAW264.7 cells. Also, changes in pathological features by drug treatment were investigated in the in vivo edema-induced rats by carrageenin /arachidonic acid or in the colitis-induced mice by DSS treatment. Results : The cytotoxicity of CYHHT on mLFC and RAW264.7 cells was not observed at 100, 50, 10, and $1{\mu}g/ml$ of CYHHT treatments. $IL-1{\beta}$, IL-6 and NOS-IImRNA expression of RAW264.7 cells was inhibited by CYHHT treatments in a dose-dependent manner. CYHHT treatment of RAW264.7 cells inhibited $TNF-{\alpha}$ and COX-2 mRNA expression. CYHHT treatment of RAW264.7 cells significantly inhibited IL-6 and NO production. CYHHT treatment of RAW264.7 cells inhibited ROS production. CYHHT inhibited rat's paw edema induced by carrageenin or arachidonate treatment in all concentrations examined. The body weight and colon length of colitis-induced mice were recovered to a normal level by DSS treatment. Clinical disease levels were significantly improved compared to the control animals. CYHHT treatment of colitis-induced mice significantly increased hematological values such as WBC and RBC counts, Hgb and HCT levels, but decreased PLT values. CYHHT treatment of colitis-induced mice decreased IL-6 and $TNF-{\alpha}$ production significantly CYHHT treatment of colitis-induced mice significantly increased CD3+(T) cell counts. In contrast, CYHHT treatment decreased CD19+ B cell counts and CD3+/CD69+ significantly, and also decreased B/T ratio (%) though not significant. Conclusion : These results indicated that CYHHT could be used for treating diverse female diseases caused by the inflammation.
The use of therapeutic ultrasound(US) in humans with malignant neoplasms has been contraindicated in physical therapy practice. Some studies have shown that results after application of US differ according to tumor type and penetration depth. The purposes of this study were to determine the effects of US on melanoma in mice and to determine treatment dosage. Twenty-four female C57BL/6 mice, age 8 weeks. The right flank of all mice was shaved, and a 0.1 ml suspension of cells was injected subcutaneously into the animals' right flank. In this study, 24 subjects were randomly divided into three groups: experimental group 1(n=8), experimental group 2(n=8), control group(n=8). In the experimental group 1, animals received continuous 3 MHZ US treatment, administered at $2.0W/cm^2$ for five minutes. In experimental group 2, animals received continuous 3 MHz US treatment, administered at $1.0W/cm^2$ for 5 minutes. The control group received the same handling as other experimental groups, including rodent chow, water, US gel application but US head pressure without the power turned on. After 10 days treatment, all mice were killed with a potassium solution. Tumors were excised and weighed on an electrical balance and fixed in a 10% neutral buffered formalin solution. Tumor weights were smaller in experimental group 2(0.3838 g) than in the control group(0.6275 g). Tumor weights of the experimental group 1(0.015 g) were smaller than those of experimental group 2. Continuous therapeutic US decreased the weight of subcutaneous melanoma tumors in mice. The treatment dosage($2.0W/cm^2$) we suggest was more effective than earlier studies on decreasing tumor size with ultrasound.
Repeated injections of low-doses of mercuric chloride in rats or mice induce polyclonal activation which includes the induction of anti-glomerular basement membrane (GBM) antibodies and circulating immune complex and it results in nephritis. Because this disease is autoimmune mediated disease resulted from immune dysfunction, immunomodulators are used to control the symptoms or to cure the disease. Irpex lacteus Fr. is a kind of new medicinal fungus. The polysaccharide fraction extracted from submerged fermentation of Irpex lacteus Fr. decreased the serum agglutinin, serolysin and IgM plaque forming cells in normal mice. The hitherto obtained clinical results suggested that it significantly improved the oligourea, edema, and hypertension in patients who have nephritis. To elucidate the action-mechanisms of Irpex lacteus Fr., we established the experimental model of HgCl$_2$induced polyclonal activation by intraperitoneal administrations of HgCl$_2$to mice. To assess the immunomodulating effect of Irpex lacteus fraction, we Investigated its effects on the mitogen induced proliferation and IgM PFC counts of splenic lymphocytes in mice during the treatment of HgCl$_2$. The Irpex lacteus polysaccharide reduced the abnormally increased mitogen induced Iymphocyte proliferation and IgM PFCs to almost normal levels. And the Irpex lacteus polysaccharides prevented the increasement of serum immunoglobulin level induced by HgCl$_2$. These data suggested that the Irpex lacteus polysaccharides might have the immunomodulating activity to prevent and /or improve the HgCl$_2$ induced autoimmune disease.
Carcinogenicity of di(2-ethylhexyl)phthalate(DEHP) to the mose forestomach and its inhibitor activity for the initiation of Benzo[a]pyrene(BP)-induced mouse forestomach neoplasia were studied on the mouse forestomach carcinogenesis regimen. One hundred female ICR mice(6~7 weeks of age) were hosed in a poly-carbonate cage (4 mice/cage) in a humidity- and temperature-controlled room subjected to a semipurified diet for a week. Mice were divided into 4 treatment groups (25 mice/treatment): Basal diet, DEHP, BP, and BP+DEHP. On Monday and wednesday, 0.1ML DEHP mixed with 0.1ml olive oil (for DEHP and DEHP+BP treatment groups) or 0.1ml saline+0.1ml olive oil (for basal diet group) was intubated, p.o., and on Friday, 2mg BP dissolved in 0.2ml olive oil (for BP and BP+DEHP treatment groups) was intubated, p.o. This cycle was repeated for 4 weeks. Beginning with the first intubation of BP an continuing thereafter, body weight and food intake were recorded once and twice weekly, respectively. All surviving mice were sacrificed 22 weeks after the first dose of BP intubation and countered forestomach tumor. No tumor was formed by DEHP treatment. 5.75 tumors per mouse was formed by BP treatment, whereas its number was reduced to 4.53 by BP+DEHP treatment. Similar results were seen in the tumor incidence. Body weight gain was not affected by DEHP treatment, when compared to that b basal diet treatment. The body weight was significantly reduced by BP treatment, but its reduction was recovered to the level of the basal diet group by BP+DEHP treatment. No significant difference was seen in food intake among all treatment groups. These results indicate that DEHP lacks carcinogenic activity to the mose forestomach and rather inhibits the initiation of BP-induced mose forestomach neoplasia.

Objective: Exercise is a risk factor for infertility in women. However, research on the effects of different intensities of exercise on folliculogenesis has not yielded clear results. This study was conducted to analyze the effects of differences in the intensity of exercise on folliculogenesis in mice. Methods: Nineteen female BALB/c mice (age, 3-4 months; weight, 13-25 g) were randomly divided into four groups: control, mild exercise, moderate exercise, and high-intensity exercise. The mice in the exercise groups engaged in swimming, with additional loads of 3%, 6%, or 9% of body weight, respectively. There were five swimming sessions per week for 4 weeks, with a gradually increasing duration every week. At the end of the treatment, ovarian extraction was carried out and hematoxylin and eosin staining was performed to identify folliculogenesis. Results: There were significant differences in the number of total follicles between the control and moderate-exercise groups (p=0.036) and between the mild- and moderate-exercise groups (p=0.005). The mean number of primary follicles was higher in the moderate-exercise group than in the mild-exercise group (p=0.006). The mean number of secondary, tertiary, and Graafian follicles did not differ significantly among groups (p≥0.05). However, the number of total follicles and follicles in each phase tended to increase after exercise, especially moderate-intensity exercise. Conclusion: Exercise of different intensities affected the total number of follicles and primary follicles. The number of follicles of each phase tended to increase after exercise. Moderate-intensity exercise had better effects than other intensities of exercise.
