  • The traditional feedstock for dimethyl ether (DME) has been natural gas obtained by pipeline from a nearby natural gas or oil field. This report focuses on other feedstock: Coal bed methane (CBM). The resource availability and suitability of CBM for DME manufacturing have been investigated. CBM in a short time has become an important industry, providing an abundant clean-burning fuel and also suggesting as a feedstock for gas industry. The use of CBM will have very little impact on the KOGAS' DME process design and economics up to 50 vol% of $CO_2$ in the CBM source. Many of the CBM sources in Asia are high in $CO_2$, but pose no difficulties for the KOGAS' DME plant. Since tri-reformer requires substantial $CO_2$ in its feed, no $CO_2$ removal from the CBM feed is needed. The $CO_2$ in the CBM means that less $CO_2$ needs to be recycled from the downstream in the process.

Separation of Nattokinase from Bacillus subtilis Fermentation Broth by Expanded Bed Adsorption with Mixed-mode Adsorbent

  • Mixed-mode hydrophobic/ionic matrices exhibit a salt-tolerant property for adsorbing target protein from high-ionic strength feedstock, which allows the application of undiluted feedstock via an expanded bed process. In the present work, a new type of mixed-mode adsorbent designed for expanded bed adsorption, Fastline $PRO^{\circledR}$, was challenged for the capture of nattokinase from the high ionic fermentation broth of Bacillus subtilis. Two important factors, pH and ion concentration, were investigated with regard to the performance of nattokinase ad-sorption. Under initial fermentation broth conditions (pH 6.6 and conductivity of 10 mS/cm) the adsorption capacity of nattokinase with Fastline PRO was high, with a maximum capacity of 5,350 U/mL adsorbent. The elution behaviors were investigated using packed bed adsorption experiments, which demonstrated that the effective desorption of nattokinase could be achieved by effecting a pH of 9.5. The biomass pulse response experiments were carried out in order to evaluate the biomass/adsorbent interactions between Bacillus subtilis cells and Fastline PRO, and to demonstrate a stable expanded bed in the feedstock containing Bacillus subtilis cells. Finally, an EBA process, utilizing mixed-mode Fastline PRO adsorbent, was optimized to capture nattokinase directly from the fermentation broth. The purification factor reached 12.3, thereby demonstrating the advantages of the mixed-mode EBA in enzyme separation.

Optimization of Phototrophic Growth and Lipid Production of a Newly Isolated Microalga, Desmodesmus sp. KAERI-NJ5

  • In this study, a novel microalgal strain, Desmodesmus sp. KAERI-NJ5, was isolated, identified, and evaluated as a candidate for biodiesel feedstock. In a preliminary study, the effects of four general microalgal growth factors, including temperature, pH, light intensity, and concentration of nitrogen source ($KNO_3$), on the microalgal photoautotrophic growth were evaluated. With the exception of light intensity, the growth factors needed to be optimized for the microalgal biomass production. Optimization was done using response surface methodology. The optimal conditions for biomass production were pH 6.54, $27.66^{\circ}C$, and 0.52 g/l $KNO_3$. The biomass production at the optimal conditions was 1.55 g/l, which correlated well with the predicted value of 1.5 g/l. The total lipid and fatty acid methyl ester contents of the cells grown at the optimal conditions were 49% and 21.2% of cell dry weight, respectively. To increase the lipid content of the biomass, microalgae were challenged by nitrogen starvation. Enhancement of total lipid and fatty acid content up to 52.02% and 49%, respectively, were observed. Lipid analysis of the nitrogen-starved cells revealed that C16 and C18 species accounted for 95.9% of the total fatty acids. Among them, palmitic acid (46.17%) and oleic acid (39.43%) dominantly constituted the algal fatty acids. These results suggest Desmodesmus sp. KAERI-NJ5 as a promising feedstock for biodiesel production.

  • 그린뉴딜정책의 실현은 석탄을 연료에서 원료로 활용분야로의 전환을 촉진시킬 것이다. 석탄은 수소의 생산, 인조 흑연 및 활성탄의 제조 원료로 활용될 수 있다. 석탄은 Steam carbon(SC) 반응과 Water-Gas Shift(WGS) 반응 및 탄산화 반응을 통하여 수소를 생산할 수 있으며, CO2격리기술과 연동되어 사용되어야 한다. 인조흑연은 실리콘이나 철 등의 무기촉매의 존재하에서 탄화도가 높은 무연탄 등을 2400~2800℃의 흑연화 온도까지 열처리함으로서 제조될 수 있기 때문에 무연탄은 석유계 피치에 비해 원료 가격경쟁력 측면에서 잠재성이 있다. 한편, 최근 목질기원의 활성탄에 필적하는 넓은 비표면적 혹은 많은 양의 미세기공을 가진 석탄기원의 활성탄이 제조될 수 있음을 여러 연구를 통해 확인되었다. 따라서 석탄기원의 활성탄은 목질기원의 활성탄을 대체할 수 있을 것으로 기대된다.

Sustainable anaerobic digestion of euphorbiaceae waste for biogas production: Effects of feedstock variation

  • Anaerobic digestion (AD) refers to the biological process which can convert organic substrates to biogas in the absence of oxygen. The aim of this study was to determine the capability of feedstock to produce biogas and to quantify the biogas yield from different feedstocks. A co-digestion approach was carried out in a continuous stirred tank reactor operated under mesophilic conditions and at a constant organic loading rate of 0.0756 g COD/ L.day, with a hydraulic retention time of 25 days. For comparison, mono-digestion was also included in the experimental work. 2 L working volumes were used throughout the experimental work. The seed culture was obtained from composting as substrate digestion. When the feedstock was added to seeding, the biogas started to emit after three days of retention time. The highest volume of biogas was observed when the seeding volume used for 1000mL. However, the lowest volume of biogas yield was obtained from both co-digestion reactors, with a value of 340 mL. For methane yield, the highest methane production rate was 0.16 L CH4/mg. The COD with yield was at 8.6% and the lowest was at 0.5%. The highest quantity of methane was obtained from a reactor of Euphorbiaceae peel with added seeding, while the lowest methane yield came from a reactor of Euphorbiaceae stems with added seeding. In this study, sodium bicarbonate (NaHCO3) was used as a buffering solution to correct the pH in the reactor if the reactor condition was found to be in a souring or acidic condition.

Characterization of physiochemical and nutrient profiles in canola feedstocks and co-products from bio-oil processing: impacted by source origin

  • Objective: The objective of this study was to characterize physiochemical and nutrient profiles of feedstock and co-products from canola bio-oil processing that were impacted by source origin. The feedstocks and co-products (mash, pellet) were randomly collected from five different bio-oil processing plants with five different batches of samples in each bio-processing plant in Canada (CA) and China (CH). Methods: The detailed chemical composition, energy profile, total digestible nutrient (TDN), protein and carbohydrate subfractions, and their degradation and digestion (CNCPS6.5) were determined. Results: The results showed that TDN1x was similar in meals between CA and CH. CH meals and feedstock had higher, truly digestible crude protein (tdCP) and neutral detergent fiber (tdNDF) than CA while CA had higher truly digestible non-fiber carbohydrate (tdNFC). The metabolizable energy (ME3x), net energy (NELp3x, NEm3x, and NEg3x) were similar in meals between CA and CH. No differences were observed in energy profile of seeds between CA and CH. The protein and carbohydrate subfractions of seeds within CH were similar. The results also showed that pelleting of meals affected protein sub-fractionation of CA meals, except rapidly degradable fractions (PB1), rumen degradable (RDPB1) and undegrdable PB1 (RUPB1), and intestinal digestible PB1 (DIGPB1). Canola meals were different in the soluble (PA2) and slowly degradable fractions (PB2) between CA and CH. The carbohydrate fractions of intermediately degradable fraction (CB2), slowly degradable fraction (CB3), and undegradable fraction (CC) were different among CH meals. CH presented higher soluble carbohydrate (CA4) and lower CB2, and CC than CA meals. Conclusion: The results indicated that although the seeds were similar within and between CA and CH, either oil-extraction process or meal pelleting seemed to have generated significantly different aspects in physiochemical and nutrient profiles in the meals. Nutritionists and producers need to regularly check nutritional value of meal mash and pellets for precision feeding.

Quantitation of relationship and development of nutrient prediction with vibrational molecular structure spectral profiles of feedstocks and co-products from canola bio-oil processing

  • Objective: This program aimed to reveal the association of feed intrinsic molecular structure with nutrient supply to animals from canola feedstocks and co-products from bio-oil processing. The special objective of this study was to quantify the relationship between molecular spectral feature and nutrient availability and develop nutrient prediction equation with vibrational molecular structure spectral profiles. Methods: The samples of feedstock (canola oil seeds) and co-products (meals and pellets) from different bio-oil processing plants in Canada (CA) and China (CH) were submitted to this molecular spectroscopic technique and their protein and carbohydrate related molecular spectral features were associated with the nutritional results obtained through the conventional methods of analyses for chemical and nutrient profiles, rumen degradable and intestinal digestible parameters. Results: The results showed that the spectral structural carbohydrates spectral peak area (ca. 1,487.8 to 1,190.8 cm-1) was the carbohydrate structure that was most significant when related to various carbohydrate parameters of canola meals (p<0.05, r>0.50). And spectral total carbohydrate area (ca. 1,198.5 to 934.3 cm-1) was most significant when studying the various carbohydrate parameters of canola seeds (p<0.05, r>0.50). The spectral amide structures (ca. 1,721.2 to 1,480.1 cm-1) were related to a few chemical and nutrient profiles, Cornell Net Carbohydrate and Protein System (CNCPS) fractions, truly absorbable nutrient supply based on the Dutch protein system (DVE/OEB), and NRC systems, and intestinal in vitro protein-related parameters in co-products (canola meals). Besides the spectral amide structures, α-helix height (ca. 1,650.8 to 1,643.1 cm-1) and β-sheet height (ca. 1,633.4 to 1,625.7 cm-1), and the ratio between them have shown to be related to many protein-related parameters in feedstock (canola oil seeds). Multi-regression analysis resulted in moderate to high R2 values for some protein related equations for feedstock (canola seeds). Protein related equations for canola meals and carbohydrate related equations for canola meals and seeds resulted in weak R2 and low p values (p<0.05). Conclusion: In conclusion, the attenuated total reflectance Fourier transform infrared spectroscopy vibrational molecular spectroscopy can be a useful resource to predict carbohydrate and protein-relates nutritional aspects of canola seeds and meals.

  • Semi-solid Al-Zn-Mg alloys were produced using a cooling plate method in order to investigate the extrudability. Al melt was poured on cooling plate which was adjusted at $60^{\circ}$ with respect to the horizontal plane, and the melt was cooled by water circulation underneath. Obtained Semi-solid feedstock has globular microstructure but also contains considerable amount of gas pore. Due to the pore, tensile elongation of the semi-solid feedstock was very low and it doesn't show yield point phenomenon. Isothermal hot extrusion was carried out using at $400^{\circ}C$ with a ram speed of 1mm/sec and an extrusion ratio of 25:1. The extruded bar show noticeably improved tensile ductility and strength because pore volume fraction decreased from 5% to 0.8% after extrusion. Mechanical properties of the semi-solid extruded bar were compared with that of commercial casting alloy..
