  • The anaerobic bacterium Clostridium ljungdahlii was known to produce ethanol from CO, $CO_2$ and $H_2$ In this research the production of ethanol at high concentration from carbon monoxide was studied. ethanol production using fed -batch, ceramic filter supported system, and 2-stage bioreactors were tested and optimized.

  • This paper presents a development of flexible bowl feeder without mechanical tooling for each specific part to be fed. This flexible bowl feeder is capable of recognising different orientations of components using optical fiber sensors and can be adjusted to feed different components by way of a simple programming method. The system would be suitable for feeding and sorting small components prior to auto,ated small batch assembly.

  • Feed rate in the fed-batch reactor is the most important control variable in optimizing the reactor performance. Exact solution can be obtained only for limited cases of simple reactor. The complexity of the model equations makes it extremely difficult to solve fur the general class of system models. Evolutionary programming method is proposed to get the information of the profile types, and the final profile is calculated by that information. The modified evolutionary programming method is used to get the more optimal profiles and it is demonstrated that proposed method can solve a wide range of optimal control problems.

  • Fermentation strategies for recombinant protein production in Pichia pastoris have been investigated and are reviewed here. Characteristics of the expression system, such as phenotypes and carbon utilization, are summarized. Recently reported results such as growth model establishment, app58lication of a methanol sensor, optimization of substrate feeding strategy, DOstat controller design, mixed feed technology, and perfusion and continuous culture are discussed in detail.

Effect of 3,3',4',5-Tetrachlorosalicylanilide on Reduction of Excess Sludge and Nitrogen Removal in Biological Wastewater Treatment Process

  • A metabolic uncoupler, 3,3',4',5-tetrachlorosalicylanilide (TCS), was used to reduce excess sludge production in biological wastewater treatment processes. Batch experiments confirmed that 0.4 mg/l of TCS reduced the aerobic growth yield of activated sludge by over 60%. However, the growth yield remained virtually constant even at the increased concentrations of TCS when cultivations were carried out under the anoxic condition. Reduction of sludge production yield was confirmed in a laboratory-scale anoxic-oxic process operated for 6 months. However, it was found that ammonia oxidation efficiency was reduced by as much as 77% in the presence of 0.8 mg/l of TCS in the batch culture. Similar results were also obtained through batch inhibition tests with activated sludges and by bioluminescence assays using a recombinant Nitrosomonas europaea (pMJ217). Because of this inhibitory effect of TCS on nitrification, the TCS-fed continuous system failed to remove ammonia in the influent. When TCS feeding was stopped, the nitrification yield of the process was resumed. Therefore, it seems to be necessary to assess the nitrogen content of wastewater if TCS is used for reducing sludge generation.

  • 호기성의 Rhizopus japonicus와 혐기성 Zymomonas mobilis로 구성된 혼합고정화 배양계(R-Z계)를 제조하고, 생전분으로부터 에탄올 생산에 응용하였다. R. japonicus를 고정화배양하므로서 액체배양에서 보다 2배 높은 glucose량을 얻었다. R-Z계의 에탄올 생산량은 1.67g/L(Yp/s, 0.094)이 었지만, 배양 24시간째부터 산소공급을 억제한 R-Z 24계에서는 6.54g/L(Yp/s, 0.38)의 에탄올을 얻어 대조구의 약 4배를 향상시켰다. 회분배양에서는 5%의 기질 농도가 적당하였으며, 생산된 에탄올은 15.02g/L(Yp/s, 0.36)이었다. 2% 기질을 5회 첨가한 유가배양에서는 2%기질의 회분배양과 동등한 수율인 0.38을 얻었다.

연속흐름반응조에서 바이오필름형태의 탈염소화 미생물에 의한 TCE분해 모니터링 (Monitoring Anaerobic Reductive Dechlorination of TCE by Biofilm-Type Culture in Continuous-Flow System)

  • A 1.28 L-batch reactor and continuous-flow stirred tank reactor (CFSTR) fed with formate and trichloroethene (TCE) were operated for 120 days and 56 days, respectively, to study the effect of formate as electron donor on anaerobic reductive dechlorination (ARD) of TCE to cis-1,2-dichloroethylene (c-DCE), vinyl chloride (VC), and ethylene (ETH). In batch reactor, injected 60 ${\mu}mol$ TCE was completely degraded in the presence of 20% hydrogen gas ($H_2$) in less than 8 days by anaerobic dechlorination mixed-culture (300 mg-soluble protein), Evanite Culture with ability to completely degrade tetrachloroethene (PCE) and -TCE to ETH under anaerobic conditions. Once the formate was used as electron donor instead of hydrogen gas in batch or chemostat system, the TCE-dechlorination rate decreased and acetate production rate increased. It indicates that the concentration of hydrogen produced in both systems is possibly more close to threshold for homoacetogenesis process. Soluble protein concentration of Evanite culture during the batch test increased from 300 mg to 688 mg for 120 days. Through the protein monitoring, we confirmed an increase of microbial population during the reactor operation. In CFSTR test, TCE was fed continuously at 9.9 ppm (75.38 ${\mu}mol/L$) and the influent formate feed concentration increased stepwise from 1.3 mmol/L to 14.3 mmol/L. Injected TCE was accumulated at 18 days of HRT, but TCE was completely degraded at 36 days of HRT without accumulation of the injected-TCE during the left of experiment period, getting $H_2$ from fermentative hydrogen production of injected formate. Although c-DCE was also accumulated for 23 days after beginning of CFSTR operation, it reached steady-state in the presence of excessive formate. We also evaluated microbial dynamic of the culture at different chemical state in the reactor by DGGE (denaturing gradient gel electrophoresis).

재조합 내열성 트레할로스 합성효소의 생산 (Production of Recombinant Trehalose Synthase from Thermus caldophilus GK24)

  • 트레할로스 합성효소(trehalose synthase)의 효율적인 생산을 위하여, 5 종류의 plasmid를 형질전환 시킨 재조합 E. coli를 이용하여 균체생산량과 효소발현량을 비교하였다. Trehalose synthase의 활성은 fusion partner를 이용한 system 에서는 활성이 나타나지 않았으며, IPTG 유도 발현 시스템보다 항시적 발현 시스템을 사용하는 E. coli K12/pHCETS에서 가장 높은 활성을 나타내었다. 선별된 재조합 E. coli K12/pHCETS를 사용하여 회분식 및 유가배양을 수행하였으며, 유가식 배양의 경우 균체논도는 20 g/L, 최종 trehalose synthase 활성은 13.7 U/ml을 나타내었다. 이러한 결과는 트레할로스 생산을 위한 trehalose synthase가 재조합 E. coli의 발효에 의해 경제적으로 생산되어질 수 있다는 가능성을 보여 주었다.

High-Level Production of Human Papillomavirus (HPV) Type 16 L1 in Escherichia coli

  • Human papillomavirus (HPV), a non-enveloped, double-stranded DNA tumor virus, is a primary etiological agent of cervical cancer development. As a potential tool for prophylactic vaccination, the development of virus-like particles (VLPs) containing the HPV16 L1 capsid protein is highly desired. In this study, we developed a high-level expression system of the HPV16 L1 in Escherichia coli for the purpose of VLP development. The native gene of HPV16 L1 has many rare codons that cause the early termination of translation and result in the production of truncated forms. First, we optimized the codon of the HPV16 L1 gene to the preferable codons of E. coli, and we succeeded in producing the full-size HPV16 L1 protein without early termination. Next, to find the best host for the production of HPV16 L1, we examined a total of eight E. coli strains, and E. coli BL21(DE3) with the highest yield among the strains was selected. With the selected host-vector system, we did a fed-batch cultivation in a lab-scale bioreactor. Two different feeding solutions (complex and defined feeding solutions) were examined and, when the complex feeding solution was used, a 6-fold higher production yield (4.6 g/l) was obtained compared with that with the defined feeding solution.

팽화미분과 zeolite를 이용한 청주 제조공정의 개선 (Improvement of Cheongju Manufacturing Process Using Gelatinized Rice and Zeolite)

  • 공정이 복잡한 청주제조공정을 개선하기 위하여 일반 미분과는 물성이 전혀 다른 팽화미분을 사용하여 새로운 청주제조공정을 개발하였다. 당화액으로부터 고형분을 제거하고 28%(w/v)의 당농도에서 발효를 시작한 경우 효모의 생육이 늦어지며 발효기간이 길어지는 문제점이 발생하였다. 이러한 문제점을 해결하기위하여 20%(w/v)의 당액에 zeolite를 첨가하여 발효를 시작하고, 발효 중 2회에 걸쳐 영양원을 첨가하였다. 이러한 조정으로 효모의 생존율을 증가시켰으며, 총 발효기간을 40%(percent) 감소시킬 수 있었다. 상쾌하고 깊은 맛을 갖으며, 알코올 함량이 18%(v/v)인 청주를 12일 만에 제조할 수 있었다. 발효 침전물로부터 회수한 효모를 4번까지 재사용하여 발효를 실시하였으며, 이 경우 효모의 생존율, 알코올 생성, 향기성분 등의 변화 등이 관찰되지 않아 효모의 재사용이 가능하였다. 따라서 복잡한 방법으로 제조되는 전통적인 청주제조공정을 간단하며 효율적인 방법으로 개선할 수 있었다.
