We have now constructed a novel recombinant baculovirus Autographa californica nuclear polyhedrosis virus (AcNPV) producing polyhedra with Bacillus thuringiensis (Bt) CryIAc crystal protein. The recombinant polyhedra produced by the recombinant baculovirus, Btrus, in insect cells was characterized. The recombinant baculovirus has two independent transcription units in opposite orientations with two promoters, p10 or polyhedrin gene promoter each initiating transcription of either native polyhedrin or fusion protein with polyhedrin and Bt Cry1Ac crystal protein. Surprisingly, this recombinant baculovirus stably produced recombinant polyhedra which were nearly similar to those of wild-type AcNPV. The immunogold staining experiment showed that the recombinant polyhedra were assembled with polyhedrin and Bt Cry1Ac crystal protein, and contained virus particles. Insecticidal toxicity of recombinant polyhedra of Btrus to the fall webworm, Hyphantria cunea, was strikingly improved in comparison with the wild-type AcNPV.
Dissolution and degradation of the parasporal crystal proteins produced from B. thuringiensis var. kurstaki, B. thuringiensis var. dendrolimus and B. thuringiensis var. aizawai were investigated. SDS-polyacrylamide gel electrophoresis analysis showed that the crystals contained major protein with molecular weight of approximately 134 kDa for B. thuringiensis var. aizawai 143 kDa for B. thuringiensis var. kurstaki and 149 kDa for B. thuringiensis var. dendrolimus, respectively. Crystals of three other strains were incubated alkali solutions at various pH or gut juice of Silkwarm Bombyx mori, Fall webworm Hyphantria cunea, and Common Cabbage worm Pieris rapae. When crystals of these strains were solubilized by alkali solutions, no major differences among strains B. thuringiensis could be detected. Among the strains studied, crystal protins (130-66 kDa) consist of protease resistant polypeptides in the 45-66 kDa size range when treated with gut juice of three insect species.
The work on the 'Studies of radiosensitivity' in Korea has been initiated since 1959 by the author at various institutions in home and foreign countries, and more than 20000 of plant materials in 73 varieties and 41 kinds are treated under varied conditions for the studies of radiosensitivity in plants. However, a study of the susceptibility to radiation in insect pests as well as their biological effects has not been previously reported in this country. During the year of 1964 and 1965, near 50000 of insects at the stages of adult, egg, larva and pupa in 14 different kinds are irradiated under acute X and gamma rays to investigate the behavior of growth and lethality after irradiation, and thrir general tolerances to radiations are compared by the kind of insects, stage of growth, radiation sources and exposured dosages. Through the experiments, it is shown that the lethality in most of insects are high as several kilo roentgens above and no significant effects on pupation. emergence, and oviposition in the treated generation at the dose of 3 Kr. below, but in the following generation , the rice stem borer for instance, no oviposition or decreased laying is observed from the dosages at 3 Kr. to 1 Kr., and the hatching was possible from 1.5 Kr. below of irradiated larvae although laying and hatching are seemed in normal. In general, the lethal dose $50\%$ of tested infect materials at the larval stage can he classified some what 60-80 Kr. for Pine caterpillar, Fall webworm, Camphor silk moth and their relatives, at 40-60 Kr. for Gypsy moth, Monley prominent and some relatives and at 20-30 Kr. for House fly, Rice weevil, Pine sawfly etc. Plant parasitic nematodes such as Hirschmannia and Paratylenchus shown very high resistance to radiation and the LD-50 is observed from 100 Kr. or above. A concept to evaluate the radiosensitivity in insects by taxological family line as a practical measure is not answerable yet, but it ran he solved within the near future through the continuing experiments.
The study has been carried to investigate a possibility to control several major insect pest of forest by microbial pathogens existing in nature as one of the biological control measure. Microorganisms including polyhedral virus isolated from diseased fall webworm were total of 4 kinds pathogenic microbes among these 4 kinds were polyhedral virus and Bacillus .species. Control effect of these two pathogens appeared to be $70.6\%$ and $49.5\%$, respectively, when they were compared with those of control plot that was $27.8\%$. Each one of bacterium species and fungus species were isolated from diseased Japanese alder leaf beetle. Pathogenisity to the healthy beetle was recognized by the fungus species, while the bacterium showed none of pathogenisity. The fungus was identified as Beauveria sp. and its effect on the beetle control was $96.2\%$ while untreated plot showed $49.2\%$ of dead beetles in the same period. Fifteen species of microbes were isolated from diseased larvae of pine gall midge. Six species out of 15 showed certain level of insecticidal effect to the larvae of the insects. The highest efficiency was showed by a fungus species, Fusarium sp. and was followed by Bacillus SP. I, Spicaria sp. pathogens isolated from larvae of pine gall midge did not affected to both of Japanese alder leaf beetles and fall webworms in any means.

An inclusion forming virus isolated from a fan webworm, Hyphantria cunea, in 1975 was identified as a nuclear polyhedrosis virus. With the virus isolated in Korea, it was considered that the virus would be one of the valuable microorganism in microbial control. In this connection, 1) the shape and size of the virus for identification, 2) susceptibility of the various instar larvae to the virus, 3) the effects of storage condition on the pathogenicity and the cross infection of the virus to the larvae of Bombyx mori were examined. The results are summarized as follows; 1. The polyhedron was tetrahedron or hexahedron of $2\mu$ in size and the rod-shaped virus particles consisting of $2\~14$ rods in a bundle were $330m{\mu}\times35m{\mu}$ in size. 2. The hexagonal nuclear polyhedra were found only in the nucleus of the midgut cells but were variable in size. 3. The $LD_{50}$ values for the various instar larvae of H. cunea were $8.377\times10^4\;PIBs/ml$ for the second, $4.974\times10^5\;PIBs/ml$ for the fifth instar larvae. The $LT_{50}values$ for $10^6\;PIBs/ml$ were 9.6 days for the second, 11.5 days for the third, 12.0 days for the fourth and 17 days for the fifth instar larvae. 4. The susceptibility of H. cunea to the nuclear polyhedrosis virus was greater in the first generation than in the second generation. 5. The effect of the storage conditions on the pathogenicity of the nuclear polyhedra was less in refrigerator $(5^{\circ}C)$ and in freezing $(-80^{\circ}C)$ than in room temperature $(18.5^{\circ}C)$, especially as air-dried polyhedra than as suspension. The pathogenicity of the polyhedra seemed to decrease by sunlight during storage as cadavers, since rather greater decrease in pathogenicity was found in sunny condition than in shady condition. 6. The effective spray concentration was $6.4\times10^7\;PIBs/ml$ in the field and its $LT_50$ values for the third and the fifth instar larvae were 4.8 days and 14.2 days, respectively. 7. No cross infections were found in the nuclear polyhedrosis virus between H. cunea and B. mori. larvae.
The intent of this research is to acquire some basic informations about formulation of the viral pesticide, Hyphantria cunea nuclear polyhedrosis virus and its virulence under field condition. The nuclear polyhedrosis virus was formulated as wettable powder using spreader, sticker and U.V. protector. The formulated product and aqueous virus were diluted with water at the concentration of 1${\times}$106PIB/ml and sprayed on mulberry leaves in the field. The leaves were fed with 3rd instar larvae of H. cunea to determine the inactivation period of the viral pesticides. The aqueous virus was completely inactivated on 5th day after spray, while the formulated one showed a spare mortality to the larvae even on 20th day after spray. In field application test, The fromulated and aqueous virus were sprayed on individual mulberry tree and 3rd instar laevae of H. cunea were fed on the trees. The mortality of the larvae one day after spray of the formulated and aqueous virus were about 50% and 40%, respectively. The formulated virus exhibited a persistent virulence to the larvae up to 9th day after spray, which the mortality was approximately 30%. The residual virulence of the formulated and aqueous virus was extended up to 4th day and 2nd day after spray, respectively.
An endoparasitoid wasp, Cotesia plutellae, has been used for a biological control agent against the diamondback moth, Plutellae xylostella. It has a symbiotic polydnavirus in their reproductive tract, which is required for its successful parasitization. Here, we measured a specific replication time of the polydnavirus during female development of C. plutellae. We, also, analyzed the reproductive potentials of female C. plutellae under mating or different host conditions. At $25^{\circ}C$, pupal C. plutellae began to develop adult tissues such as compound eyes and wings since day 2. At day 5, all adult tissues including antennae were developed and were ready to emerge. With polyclonal antibody raised against C. plutellae polydnavirus, an immunobloting could confirm virus replication at day 4 during pupal stage. Virus particles could be visualized by transmission electron microscope in the oviduct lumen of day 5 pupae. After adult eclosion, venom gland and ovarian calyx increased in size, though ovarioles did not. Mated females layed large number of eggs (over $60\%$) at first 4 days during their mean longevity of ca. 8 days at $25^{\circ}C$. Unmated females showed less active ovipositional behavior, where all the eggs developed into males. C. Plutellae parasitized both P. xylostella and fall webworm, Hyphantria cunea. However, C. Plutellae developed faster and showed higher successful paarasitization in P. xylostella than in H. cunea.
This study was conducted to investigate the effect of 20-hydroxyecdysone on diapausing pupae of Hyphantna cunea. Treatment of 20-HE at the dosage of 5 $\mu$ g or higher/g live weight terminated diapause in about 50% of Hyphantna cunea pupae treated, and some malformed adult emerged at dosages of 10 or 12.5 $\mu$g of 20-hydroxyecdysone/g live wt. The maximum concentration of 20-hydroxyecdysone in the whole body of normally developing pupae was about 3.2 $\mu$g/g on the 6th day after pupation, and in that of diapausing pupae treated with 20 -HE about 4.1 $\mu$g/g on the 6th day after the treatment. But diapausing pupae showed a low level( 1.7 $\mu$g/g) of maximum 20-HE concentration. In diapausing pupae treated with 20- HE, glycongen content was lower than in normally developing pupae, but the changing pattern was similar to that observed in normally developing pupae. Glucose(and/or sorbitol) and trehalose content of 20-HE-injected pupae reached the maximum value on the 6th day after the injection, which were higher than those of normally developing pupae. The amount of soluble haemolymph proteins was lower but the total soluble protein content of haemolymph-rernoved whole body was higher in 20-HE-injected pupae than that in normally developing pupae.
