Objective: To explore the influence of different ways of blood transfusion on the expression levels of interleukins (IL) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) inperi-operative patients with esophageal cancer. Materials and Methods: A total of 80 patients with esophageal cancer who underwent radical operations were selected as study patients and randomly divided into an observation group (treated with autologous blood transfusion) and control group (with homologous blood transfusion). Changes of intra-operative indexes and peri-operative blood indexes, from hemoglobin (Hb) and hematocrit value (Hct), to levels of inflammatory factors like interleukins-6 (IL-6), IL-8, IL-10 and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) were compared. Results: Operations for patients in both groups were successfully conducted, and no significant differences in mean surgical duration and intra-operative hemorrhage volume, fluid infusion volume and blood transfusion volume were detected (p>0.05). Compared with values before surgery, Hb and Hct levels decreased significantly while white blood cell count (WBC) increased 1, 5 and 7 d after operation (p<0.05, p<0.01). In addition, WBC was apparently higher in observation group than in control group 5 and 7 d after operation (p<0.01). Compared with before surgery, in the observation group, levels of IL-6, IL-8 and IL-10 had no significant differences after operation (P>0.05), but TNF-${\alpha}$ level increased y (p<0.01), whereas in control group, IL-6 level had no significant difference (p>0.05), IL-8 level decreased obviously (p<0.05), IL-10 level increased markedly first and then decreased gradually as time passed but its level remained elevated (p<0.01), and TNF-${\alpha}$ level increased first and then decreased, and there was no significant difference 7 d after operation (p>0.05). Conclusions: Decreased IL-8 and increased IL-10 levels are two important reasons for immunosuppression after homologous blood transfusion, whereas autologous blood transfusion can alleviate this while increasing the TNF-${\alpha}$ level, which also has potential to improve anti-tumor immunity in the human body.
연구배경 : 결핵성 흉막염의 감별 진단 목적으로 널리 이용되고 있는 검사는 adenosine deaminase와 INF-$\gamma$를 세포성 면역에 또다른 중요한 매개체로 알려진 TNF-$\alpha$의 진단적 의의를 조사하고, 현재 임상에서 유용한 지표로 사용되고 있는 adenosine deaminase와의 감별력을 비교하고자 본 연구를 시행하였다. 방 법 : 삼출성 흉막염 80예(결핵성 : 39예, 암성 : 31예, 부폐렴흉막염 : 10예)를 대상으로 흉수의 기본적인 세포조성, 화학 검사와 더불어 ADA와 TNF-$\alpha$(Medgenix IRMA kit)를 측정하였다. 결 과 : 흉수내 ADA농도는 결핵성 흉막염에서 $48.7{\pm}32.7U/L$로 비결핵성 흉막염 $26.0{\pm}41.3U/L$에 비해 유의한 차이로 높았고(p < 0.05), TNF-$\alpha$치 또한 결핵성 흉막염에서 $184.1{\pm}214.2pg/mL$로 비결핵성 흉막삼출액 $44.1{\pm}114.2pg/mL$에 비해 유의하게 높았다(p < 0.01). ROC 곡선을 이용하여 ADA와 TNF-$\alpha$의 감별력을 가장 높일 수 있는 기준치를 정하였을 때, ADA는 30U/ml, TNF-$\alpha$는 15pg/ml로 측정되었고, 각각의 기준치를 이용하여 감수성과 특이도를 구하였을때, ADA는 감수성 66.7%, 특이도 85.0%, TNF-$\alpha$는 감수성 69.2% 특이도 87.1%를 보였다. 두 검사의 민감도와 특이도를 비교하기 위한 ROC 곡선에서, ROC 곡선아래의 면적(area under curve)은 ADA와 TNF-$\alpha$사이에 유의한 차이가 없었다(ADA 0.83, TNF-$\alpha$ 0.82). 다중회귀분석(multiple stepwise regression)에서 ADA가 가장 유용한 지표로 계산되었으나 TNF-$\alpha$의 추가가 분별력에 더 이상의 기여함은 없었다. 결 론 : 결핵성 흉막염과 비결핵성 흉막염을 감별 진단하는데 ADA와 함께 TNF-$\alpha$도 유용한 지표로 이용될 수 있으나 두가지 검사를 함께 시행함에 따른 진단에 있어서의 잇점은 없었다.
Sargassum serratifolium ethanolic extract has been known for strong antioxidant and anti-inflammatory properties. We prepared hexane fraction from the ethanolic extract of S. serratifolium (HSS) to improve biological activities. In this study, we investigated the effects of HSS on the inhibition of tumor necrosis factor (TNF)-${\alpha}$-induced monocyte adhesion to human umbilical vein endothelial cells (HUVECs). We found that HSS suppressed the production of cell adhesion molecules such as intracellular adhesion molecule-1 and vascular cell adhesion molecule-1 in TNF-${\alpha}$-induced HUVECs. Moreover, TNF-${\alpha}$-induced production of monocyte chemoattractant protein 1 and keratinocyte chemoattractant was inhibited by HSS treatment. HSS suppressed TNF-${\alpha}$-induced nuclear factor kappa B ($NF-{\kappa}B$) activation via preventing proteolytic degradation of inhibitor ${\kappa}B-{\alpha}$. HSS induced the production of heme oxygenase 1 via translocation of Nrf2 into the nucleus in TNF-${\alpha}$-treated HUVECs. Overall, HSS alleviated vascular inflammation through the downregulation of $NF-{\kappa}B$ activation and the upregulation of Nrf2 activation in TNF-${\alpha}$-induced HUVECs. These results indicate that HSS may be used as therapeutic agents for vascular inflammatory disorders.
연구목적: 본 연구는 in vitro 상에서 치수, 치은, 치주인대 세포를 neuropeptide (substance P, Calcitonin gene related peptides (CGRP))및 inflammatory cytokine (TNF-$\alpha$)으로 자극 시 matrix metalloproteinase (MMPs)의 생성 및 발현을 관찰한 것으로 치아와 치아 주변 조직에 염증이 존재할 경우 neuropeptide 및 inflammatory cytokine과 치아 경조직 혹은 치조골의 remodeling에 중요한 역할을 하는 matrix metalloproteinase (MMPs)와의 관계를 규명하고자 하였다. 연구 재료 및 방법: 시편으로는 우식이 없는 건전한 제3대구치(n = 10)를 사용하였으며, 발거 후 즉시 Phosphate buffered saline (PBS)에 보관하고 치아에서 치은과 치주인대 조직을 채취하였다. 치아를 종축으로 절단하고 치수 조직을 채취하여 조각으로 분리한 후 시편을 PBS에서 세 번 세척하였다. Plate에 치수, 치은, 치주인대 시편 조각을 위치시켜 Dulbecco's Modified Eagle Medium (DMEM)을 첨가하여 세포를 배양하였다. 치수, 치은, 치주인대 세포를 culture dish에서 confluence에 도달할 때 까지 배양하여 Fetal Bovine Serum (FBS)가 포함되지 않은 배지로 교환하여, $37^{\circ}C$에서 24시간 동안 배양한 후 Phosphate buffered saline (PBS)로 1회 세척하고 Substance P ($10^{-8}\;M$, $10^{-5}\;M$)가 포함된 배양액과 Mock (배양액만 포함됨)으로 4시간, 24시간동안 자극하였다. CGRP ($10^{-6}\;M$)을 함유한 배양액 및 TNF-$\alpha$(2 ng/mL)를 포함한 배양액으로 각각 24시간동안 세포를 자극하였다. 각각 다른 농도의 TNF-$\alpha$(2 ng/mL, 10 ng/mL, 100 ng/mL)를 포함한 배양액으로 24시간 동안 세포를 자극 한 후 RNase protection assay 및 Enzyme linked immunosorbent assay를 시행하였다. 결과: SP와 CGRP는 치수, 치은, 치주인대 세포의 MMPs발현에 관여 하지 않았다. TNF-$\alpha$로 24시간 자극 시 치수, 치은, 치주인대 세포에서 MMP-1,-12, -13의 발현을 증가시켰다. 반면, TNF-$\alpha$는 치수, 치은, 치주인대 세포들에서 TIMP-3의 발현을 감소시켰다. 서로 다른 농도의 TNF-$\alpha$(2 ng/mL, 10 ng/mL, 100 ng/mL)로 24시간 자극 시 MMP-1과 MMP-13의 발현이 증가하였다. 결론: 치아와 치아 주변 조직에 염증이 존재 시 TNF-$\alpha$가 증가함에 따라 치수, 치은, 치주인대로부터의 치아의 경조직 혹은 치조골의 remodeling에 관여하는 matrix metalloproteinase (MMPs)가 중요한 역할을 하는 것으로 사료된다.
Objectives : Tumor necrosis factor-${\alpha}$(TNF ${\alpha}$) is a proinflammatory cytokine involved in the pathogenesis of rheumatoid arthritis. This study was designed to investigate the relation between TNF-${\alpha}$ gene polymorphism and rheumatoid arthritis in Korean population. Methods : This study was carried out on 103 rheumatoid arthritis patients who fulfilled the American College of Rheumatology 1987 revised criteria for rheumatoid arthritis and 208 healthy control subjects. Blood samples from all subjects were obtained for DNA extraction. The extracted DNA was amplified by polymerse chain reaction(PCR). PCR products were visualized by 2% agarose gel electrophoresis. We investigated the genotyping of TNF-${\alpha}$ by using Pyrosequencing. Results: The genotypes of TNF-${\alpha}$ gene were GG, AG and AA. While the distribution of TNF-${\alpha}$ polymorphism in control subjects was 92.31%, 7.21%, 0.48% respectively, in rheumatoid arthritis patients was 93.20%, 6.80%, 0.00%(GG, AG, AA). There was no statistical significant allelic frequency difference between control and rheumatoid arthritis groups. Conclusions : We concluded that there was no significant association between TNF-${\alpha}$ gene polymorphism and rheumatoid arthritis. However, the findings of this study need to be confirmed in more patients and further studies.
Substance P can stimulate secretion of tumor necrosis $factor-\;{\alpha}\;(TNF-\;{\alpha}\;)$ from astrocytes stimulated with lipopolysaccharide (LPS). Here I report that Chilbogeum can modulate cytokines secretion from primary cultures of rat astrocytes. Chilbogeum $(10\;{\mu}g/ml)$ significantly inhibited the $TNF-\;{\alpha}$ secretion by astrocytes stimulated with LPS and Substance P. Interleukin-1 (IL-1) has been shown to elevate $TNF-\;{\alpha}$ secretion from LPS-stimulated astrocytes while having no effect on astrocytes in the absence of LPS. Treatment of Chilbogeum $(10,\;100\;{\mu}g/ml)$ to astrocytes stimulated with both LPS and Substance P decreased IL-1 secretion significantly. The secretion of $TNF-\;{\alpha}$ by LPS and Substance P in astrocytes was progressively inhibited with increasing amount of IL-1 neutralizing antibody. Upon stimulation from various agents, these cells adopt a reactive phenotype, a morphological hallmark in Alzheimer's disease (AD) pathology, during which they themselves may produce still more inflammatory cytokines. Chilbogeum $(10,\;100\;{\mu}g/ml)$ significantly inhibited the $TNF-\;{\alpha}$ secretion by CCF-STTG1 astrocytoma cells stimulated with $A\;{\beta}$ and IL-1. These results suggest that Chilbogeum may inhibit $TNF-\;{\alpha}$ secretion by inhibiting IL-1 secretion and that Chilbogeum has an antiinflammatory activity in AD brain.

Upon stimulation from various agents, these cells adopt a reactive phenotype, a morphological hallmark in AD pathology, during which they themselves may produce still more inflammatory cytokines. Substance P (SP) can stimulate secretion of tumor necrosis $factor-\;{\alpha}$$(TNF-\;{\alpha})$ from astrocytes stimulated with lipopolysaccharide (LPS). Sgp $(10\;to\;1000\;{\mu}g/ml)$ significantly inhibited the $TNF-\;{\alpha}$ secretion by astrocytes stimulated with $A-{\beta}-$and IL-1. These results suggest that Sgp may inhibit $TNF-\;{\alpha}$ secretion by inhibiting IL-1 secretion and that Sgp has an antiinflammatory activity in AD brain
Interactions of the cell adhesion molecules are known to play important roles in mediating inflammation. The proinflammatory cytokine, tumor necrosis factor-${\alpha}$(TNF-${\alpha}$), activates the NF-kB signaling pathway, which induces the expression of various genes, such as intercellular adhesion molecule-1 (ICAM-1). In this study, the effect of vitamin C on the ICAM-1 expression induced by TNF-${\alpha}$ in a human neuroblastoma cell line, SK-N-SH was investigated. Treatment with vitamin C resulted in the downregulation of the TNF-${\alpha}$-induced surface expression and ICAM-1 mRNA levels in a concentration-dependent manner. Moreover, a gel shift analysis indicated that vitamin C dose-dependently inhibited the NF-kB activation and IkB${\alpha}$ degradation induced by TNF-${\alpha}$. Taken together, these results suggest that vitamin C downregulates TNF-${\alpha}$- induced ICAM-1 expression via the inhibition of NF-kB activation.
본 연구에서 돼지 PBMC에 t10c12-CLA 처리는 TNF-${\alpha}$생산을 증가시켰으나, LPS 자극 PBMC에서는 TNF-${\alpha}$생산을 감소시켰다. t10c12-CLA 처리는 PBMC의 inhibitory ${\kappa}B$ ($I{\kappa}B$)-${\alpha}$ 단백질 분해를 증가시키고 NF-${\kappa}B$ p65 활성 수준을 증가시켰다. 그러나 LPS 자극 PBMC에서는 상반되는 효과가 관찰되었다. 특히, LPS 비자극 PBMC에서 t10c12-CLA는 NF-${\kappa}B$ 저해제인 caffeic acid phenethyl ester (CAPE)를 처리한 경우 NF-${\kappa}B$ p65 활성 수준을 증가시켰으나 반대로 LPS로 자극한 CAPE 처리 PBMC에서는 NF-${\kappa}B$ p65 활성 수준을 억제시켰다. 이상의 결과는 t10c12-CLA가 돼지 PBMC에 있어 LPS 자극 유무에 따라 다른 효과를 가질 수 있으며, 이는 NF-${\kappa}B$ p65 활성도의 변화와 관련성이 있음을 보여주고 있다.
목적: 기관지폐이형성증(bronchopulmonary dysplasia,BPD)의 발생에는 폐의 염증반응이 중요하게 작용한다. Tumor necrosis factor-$\alpha$ (TNF-$\alpha$)는 대표적인 proinflammatory cytokine으로, TNF-$\alpha$ 고생산 부위의 단일염기다형성과 BPD발생 사이의 연관성을 알아보고자 하였다. 방법: 2006년 1월부터 12월까지 서울대학교병원에서 출생하여 신생아중환자실에 입원한 재태주령 32주 미만이면서 출생체중 1,500 g 미만의 미숙아 93예를 대상으로, 실시간 중합효소연쇄반응을 이용하여, TNF-$\alpha$ 유전자 촉진자 5개 부위(-1031T/C,-863C/A, -857C/T, -308G/A, -238G/A)의 단일염기다형성을 분석하였다. 결과: BPD 군은 전체연구대상93예 중에서 38예로40.9%에 해당하였다. BPD 군에서 대조군보다 재태연령($27^{+5}{\pm}2^{+0}$ wk vs. $29^{+2}{\pm}1^{+4}$ wk, P<0.0001)과 출생체중(990${\pm}$270 g vs. 1,220${\pm}$230 g, P<0.0001)이 낮고, 신생아 호흡곤란증후군(71.1% vs. 49.1%, P=0.035) 과 동맥관개존(71.1% vs. 50.9%, P=0.052) 의 발생빈도가 높아서 두 군간의 임상적 특성에는 차이가 있었다. 그러나 TNF-$\alpha$ 유전자 단일염기다형성 분석에서는, 각각의 TNF-$\alpha$ 대립유전자의 빈도와 일배체형의 분포 모두가 BPD 군과 대조군 사이에 차이를 보이지 않았다. 결론: 한국인 조산아에서는, TNF-$\alpha$ 유전자 다형성보다는 임상적인 위험인자가 BPD 발생에 중요하게 작용하였다.

② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
