  • On-line efficiency optimization control of an induction motor drive using neural network is important from the v viewpoints of energy saving and controlling a nonlinear system whose charact81istics are not fully known. This paper p presents a neural networklongleftarrowbased on-line efficiency optimization control for an induction motor drive, which adopts an optimal slip an밍J.lar frequency control. In the proposed scheme, a neuro-controller provides minimal loss operating point i in the whole range of the measured input power. Both simulation and experimental results show that a considerable e energy saving is achieved compared with the conventional constant vlf ratio operation.

A New CW CO2 Laser with Precise Output and Minimal Fluctuation by Adopting a High-frequency LCC Resonant Converter

  • The current study proposes the design of a hybrid series-parallel resonant converter (SPRC) and a three-stage Cockcroft-Walton voltage multiplier for precisely adjusting the power generated by a continuous wave (CW) $CO_2$ laser. The design of a hybrid SPRC, called LCC resonant converter, is described, and the fundamental approximation of a high-voltage and high-frequency (HVHF) transformer with a resonant tank is discussed. The results of the current study show that the voltage drop and ripple of a three-stage Cockcroft-Walton voltage multiplier depend on frequency. The power generated by a CW $CO_2$ laser can be precisely adjusted by a variable-frequency controller using a DSP (TMS320F2812) microprocessor. The proposed LCC converter could be used to obtain a maximum laser output power of 23 W. Moreover, it could precisely adjust the laser output power within 4.3 to 23 W at an operating frequency range of 187.5 to 370 kHz. The maximum efficiency of the $CO_2$ laser system is approximately 16.5%, and the minimum ripple of output voltage is about 1.62%.

  • Yeast expression plasmids containing an appropriate leader sequence and bovine $\beta$-casein cDNA were constructed to produce $\beta$-casein for the study of its functional characteristics. Two kinds of expression systems for $\beta$-casein were constructed using pCGYl444 as a precursor plasmid. This plasmid is a yeast-E. coli shuttle vector which contains the chelatin promoter. The plasmid pISB202 contains the invertase leader sequence and $\beta$-casein gene. The plasmid pDEB303 contains the original bovine $\beta$-casein leader sequence gene. These two plasmids were introduced into S. cerevisiae AB116 which is a strain deficient in the major yeast proteinases. Each clone was grown in minimal media for 24 h before induction by $CuSO_4$. The cells were thus grown under expression conditions. Both strains harbouring pISB202 and pDEB303 expressed bovine $\beta$-casein. The $\beta$-casein was detected using immunochemical staining after western blot. Secretion of $\beta$-casein was detected in the culture broth. The estimated amount of secreted $\beta$-casein was approximately 50 ${\MU}g$/l.

A Green Fluorescent Protein-based Whole-Cell Bioreporter for the Detection of Phenylacetic Acid

  • Phenylacetic acid (PAA) is produced by many bacteria as an antifungal agent and also appears to be an environmentally toxic chemical. The object of this study was to detect PAA using Pseudomonas putida harboring a reporter plasmid that has a PAA-inducible promoter fused to a green fluorescent protein (GFP) gene. Pseudomonas putida KT2440 was used to construct a green fluorescent protein-based reporter fusion using the paaA promoter region to detect the presence of PAA. The reporter strain exhibited a high level of gfp expression in minimal medium containing PAA; however, the level of GFP expression diminished when glucose was added to the medium, whereas other carbon sources, such as succinate and pyruvate, showed no catabolic repression. Interestingly, overexpression of a paaF gene encoding PAA-CoA ligase minimized catabolic repression. The reporter strain could also successfully detect PAA produced by other PAA-producing bacteria. This GFP-based bioreporter provides a useful tool for detecting bacteria producing PAA.

  • F. velutipes Leu 2 gene (${\beta}-isopropylmalate$ dehydrogenase gene) was used for transformation of P. florida leucine requiring auxotrophic mutant P101. Transformation frequency was very low but the transformed colony can grow on minimal medium very slowly. Transformation was identified by Southern hybridization and reverse transformation into E. coli using chromosome DNA isolated from transformed P. florida.

The Influence of Water Temperature and Food Concentration on the Filtration Rates of the Asiatic Clam, Corbicula fluminea (수온과 먹이생물의 농도 변화에 따른 재첩의 여과율 변동)

  • This study was undertaken with the intent to describe the influence of water temperature and food concentration on the filtration rates of the Asiatic clam, Corbicula fluminea. The clams were collected at Lake Geumho near Yeongsan river, during March 2001. Food organism, Scenedesmus sp. (KMCC FC-34), was indoor-cultured in f/2 medium, and was used to measure the filtration rate of the clams. Filtration rate of the clams was measured by indirect method. Cell concentrations of food organisms were determined by direct counting cells using the hemacytometer under the light microscope. The filtration rate of the clams increased with water temperature up to circa $25^{\circ}C$. Above this temperature, the filtration rate decreased rapidly. The minimal filtration rate of the clams was recorded at $5^{\circ}C$. Thermal coefficient, $Q_10$ values at low temperature range were much higher than those at high temperature range. These results indicate the asiatic clam is more sensitive in cold water like most of marine bivalves. There was a strong reversed correlation between filtration rate and food concentration. Filtration rate of the clams was reduced as food concentration was increased.

  • A highly specific antigenic protein of 31 kDa from plerocercoid of Spirometra mansoni (sparganum) was obtained by gelatin affinity and Mono Q anion-exchange column chromatography. The purified 31 kDa protein was subjected to N-glycan enzymatic digestion for structural analysis. The relative electrophoretic mobility was analyzed by SDS-PAGE, before and after digestion. On SDS-PAGE after enzymatic digestion, the 31 kDa protein showed a molecular shift of approximately 2 kDa, which indicated the possession of complex N-linked oligosaccharides (N-glycosidase F sensitive) but not of high-mannose oligosaccharides (endo-beta-N-acetylglucosaminidase H, non-sensitive). Chemically periodated 31 kDa protein showed statistically non-significant changes with human sparganosis sera by enzyme linked immunosorbent assay (ELISA). Therefore, the dominant epitopes of the 31 kDa molecule in human sparganosis were found to be mainly polypeptide, while N-glycans of the antigenic molecule in sparganum was minimal in anti-carbohydrate antibody production.

Studies on the Biological and Chemical Properties of Musty Ginseng Root and its Causal Mechanism (적변삼의 생물.화학적 특성과 그 발생원인에 관하여)

  • Rusty root of ginseng has been known as one of the limiting factors in ginseng production in Korea. An attempt was, therefore, made to elucidate biological and chemical natures of the rusty root, and the redox Potential of the ginseng cultivated soils were measured and compared with diseased and non-diseased soils. Reddish discoloration was most frequently observed on the epidermis of ginseng root and the pigments were accumulated in all epidermal cells of the diseased lesions. The lower the redox potential of the ginseng cultivated soil was, the more severe the rusty root was observed. Fe content in the diseased epidermis was 3 times higher than that of healthy one. Organic acids such as oxalic, malonic, succinic, and citric acids were also higher in the mss root than in the healthy one. Thin layer chromatogram of phenolic acid fractions obtained from the epidermal cells of the rusty root of ginseng exhibited 3 to 4 unidentified substances not found in the healthy root. Also lignification of the epidermal cells and the activity of phenylalanine ammonia lyase were greater in the rusty root than the healthy root. Colony formation and conidia production of F. solani, And mycelial growth and sclerotium formation of Sclerotinia sp. isolated from ginseng root were suppressed in a nutritionally minimal medium supplemented with water extract of rusty ginseng root epidermis. It is, therefore, suggested that rusty root of ginseng is caused by unfavorable rhizosphere environmental stress or stresses resulting abnormal metabolism in the root as a selfdefence mechanism of non-specific resistance responses.

  • Advanced imaging approaches (computed tomography, CT; magnetic resonance imaging, MRI; $^{18}F$-fluorodeoxyglucose positron emission tomography, FDG PET) have increased roles in cervical cancer staging and management. The recent FIGO (International Federation of Gynecology and Obstetrics) recommendations encouraged applications to assess the clinical extension of tumors rather than relying on clinical examinations and traditional non-cross sectional investigations. MRI appears to be better than CT for primary tumors and adjacent soft tissue involvement in the pelvis. FDG-PET/CT has increased in usage with a particular benefit for whole body evaluation of tumor metabolic activity. The potential benefits of advanced imaging are assisting selection of treatment based upon actual disease extent, to adequately treat a tumor with minimal normal tissue complications, and to predict the treatment outcomes. Furthermore, sophisticated external radiation treatment and brachytherapy absolutely require advanced imaging for target localization and radiation dose calculation.

Joint and Link Module Geometric Shapes of Modular Manipulator for Various Joint Configurations (다양한 관절 구성을 위한 모듈라 매니퓰레이터의 관절 및 링크 모듈 형상 도출)

  • A modular manipulator in serial-chain structure usually consists of a series of modularized revolute joint and link modules. The geometric shapes of these modules affect the number of possible configurations of modular manipulator after assembly. Therefore, it is important to design the geometry of the joint and link modules that allow various configurations of the manipulators with minimal set of modules. In this paper, a new 1-DoF(degree of freedom) joint module and simple link modules are designed based on a methodology of joint configurations using a series of Rotational(type-R) and Twist(type-T) joints. Two of the joint modules can be directly connected so that two types of 2-DoFs joints could be assembled without a link module between them. The proposed geometries of joint and link modules expand the possible configurations of assembled modular manipulators compared to existing ones. Modular manipulator system of this research can be a cornerstone of user-centered markets with various solution but low-cost, compared to conventional manipulators of fixed-configurations determined by the provider.
