  • This experiment was carried out to elucidate the sugar composition of polysaccharides and the structural features of water-soluble polysaccharides(WSP) isolated from Korean oak mistletoe, Loranthus yadoriki Sieb. The 48-hours ball-milled meals of extractive-free dried mistletoe sawdusts were extracted with distilled water for $24hrs{\times}2$ at room temperature. The extracts poured into 95% ethyl alcohol to precipitate. The separated precipitate of WSP, in form of yellowish white powder by lyophilization, was fractionated into four subfractions of WSP-1, WSP-2, WSP-3 and WSP-4 by anion exchange chromatography on DEAE-cellulose column. The sugar composition of WSPs was analyzed by GLC in form of their glycitol acetates, and the structure of polysaccharides in Fractions WSP-1 and WSP-2 was determined by FT-IR and GC-MS after methylation through and acetylation. The sugars of WSPs from Korean oak mistletoe, Loranthus yadoriki, are majorly arabinose and galactose in stem, galactose in leaves very high in content and showed difference in composition and monomeric units between stems and leaves. D-galactose, D-glucose and L-arabinose are the simple sugars consisting of polysaccharides in WSP-1. ($1{\rightarrow}3$)-Linked galactan is the bakcbone with side chain of ($1{\rightarrow}5$)- -L-arabinofuranosyl residues and ($1{\rightarrow}6$)- -D-galactopyranosyl residues, and ($1{\rightarrow}4$)-linked glucan also presents. ($1{\rightarrow}4$)-Linked rhamnogalacturonan and ($1{\rightarrow}4$)- and ($1{\rightarrow}3$)-linked galactan present in WSP-2.

  • The chemical compositions of the root bark of Ulmus species and mucilage were investigated. The methanol extractives were fractionated with n-hexane, ethyl ether, ethyl acetate and water and antibacterial activities of each fraction were investigated. Although differences of chemical compositions of the Ulmus species were not noticeable, nitrogen and lignin contents of xylem were higher than those of phloem. However, ash and extractive contents showed adverse results. The mucilage of root bark was slowly gushing out by water and similar to pectin substance with high acidic sugar content. In fractionation experiment, the yield of methanol extraction was about 7% and the portion of water-soluble materials in methanol extractives was 78%. These water- soluble materials had the most high antibacterial activity on Staphylococcus aureus and Salmonella typhimurium.

  • Swietenia mahagoni is one of the commercial timbers in Indonesia. Mahogany heartwood is an important characteristic as it relates to the natural durability and aesthetics of the wood. Lipophilic extractives are known to be involved in the heartwood formation process. Therefore, this study aims to determine the lipophilic compounds associated with heartwood formation. The n-hexane extract from sapwood and heartwood samples (1 to 5 years) was analyzed by gas chromatography-mass spectrometry. The results showed that the content of n-hexane extract ranged from 0.76% to 2.45% based on dry wood. The main group of compounds identified in the lipophilic fraction consisted of sterols (β-sitosterol, stigmasterol, campasterol, and cyclolaudenol), fatty acids (palmitic, oleic, linoleic, and stearic acid), and hydrocarbons (pentadecane, 1-octadecane, hexadecane, cyclotetracosane, cycloeicosane, and cyclooctacosane) after heartwood formation. In addition, the hydrocarbon fraction was the largest, followed by sterols, fatty acids, and 1-heneicosanol. In the radial variation, the distribution of fatty acids was greater in the sapwood than in the heartwood (4-year-old). However, the reverse pattern was found at the age of 5 years. The lipophilic fraction was generally more abundant in the heartwood compared to the sapwood, especially at 5 years of age, with much higher levels than when the heartwood was forming (4 years). These findings show that when the heartwood formation begins, the lipid composition was not fully metabolized at the beginning of heartwood formation compared to 5-year-old trees.

Sitological Quality Evaluation of Cultured and Wild River Puffer, Takifugu obscurus (Abe) (양식산과 자연산 황복, Takifugu obscurus (Abe)의 식품학적 품질평가)

  • The present study was performed to evaluate sitological quality of the cultured and wild river puffer, Takifugu obscurus. Proximate composition, the content of extractive nitrogen, the content of nucleotides and their related compounds, total and free amino acid, and fatty acids were analysed and sensory evaluation in the muscle of the river puffer were compared. The cultured river puffer had a higher moisture content compared to the wild fish, while there was no significant difference in crude lipid, crude protein and ash contents. Nucleotides and their related compounds including ATP, ADP, AMP, IMP, HxR and Hx were detected. The result from analyzing ATP-related compound showed difference in total content by wild and cultured river puffer, and IMP content that had largest influence upon the savory taste of sliced raw fish, was higher in the cultured fishes than wild ones. Breaking strength level of the wild river puffer was higher than that of the cultured fish. High levels of C16:0 and C18:0 were shown in all samples and -3 polyunsaturated fatty acid content were not different between the cultured and wild river puffers (P>0.05). Total 17 amino acids were detected in the samples, and most of the samples had high contents of glutamic acid, aspartic acid, lysine and leucine and low contents of cystine, histidine, methionine and tyrosine. The result from surveying free amino acid content of wild and cultured river puffer showed difference in content, but generally taurine and lysine content for the whole free amino acid held the most part. There was no significant difference in texture, flavor and overall acceptance score between the cultured and wild fishes (P>0.05).

Studies on the changes of main components during the fermentation of Anchvy sauce (멸치젓 발효숙성중 주요성분의 변화)

  • This research aimed at investigating the changes of volatile basic nitrogen, amino nitrogen and lipids during the fermentation of 6 month Anchovy cured under room temperature with various treatments(20, 30 and 40% salted) and examing the optimum condition of Anchovy sauce. The results are summerized as the V.B.N which increased with the curing period of anchovy from 14 mg% to 90~107mg% in 180 days curing at 20% salt level. Amino nitrogen in minced anchovy was higher than in whole anchovy during fermentation and the content of Extractive Nitrogen in the curing anchovy containing 20% of salt, kept the highest amount in 60 curing days. As a rule, minced anchovy showed more rapidly increased than whole anchovy. The lipid in curing anchovy containing 20% and 30% of salt has already been oxidized in 30 days while the lipid of anchovy cured with 40% salt prolonged the initial stage to 45 days. During fermentation, peroxide value and acid value showed constant increasing, while thiobarbituric acid began to decrease after 120 days curing. Among the non-polar lipids, linolenic acid, linoleic acid and erucic acid was decomposed by 24.5%, 22.2%, and 20.0%, respectively. It was noticed that the decomposition of polar lipid was retarded by higher salt content.

Comparison of the Antioxidative Abilities of Commercial Gal Geun Tang (시판 갈근탕의 항산화능의 비교)

  • Each 3 g of five different commercial Gal Geun Tang extractive granules was mixed with 97 $m\ell$ of methanol or water for 1.5 hour at room temperature and filtered through a Whatman No. 1 filter paper, and the filtrate was used to determine antioxidant activity. Antioxidative abilities of each extracts were compared by measuring total phenol content (TPC), electron donating ability (EDA), inhibition of lipid peroxidation, and reducing power Average TPC of methanol extracts from five samples was 0.14 mM within the range of 0.07~0.23 mM, while that of water extracts was 0.23 mM, higher than that of methanol extracts. Average EDA of the methanol extracts was 85.54%, higher than that of the 0.1% BHT (72.26%). Average EDA values of the water extracts were 69.7%. Average value of inhibition against lipid peroxidation of the methanol extracts was 26.95%, and the range of five samples was 17.02~37.36%. Inhibition against lipid peroxidation of the water extracts showed relatively low value, 18.62%. Reducing power of the methanol extracts was 0.77 unit, which was 73.1% of 0.1% BHT, and that of water extracts was 0.88 unit. These results indicate that commercial Gal Geun Tang shows high antioxidant ability, though there are some differences depending on extract solvent and manufacturing company.

  • In order to elucidate the taste compounds of wild eel, Anguilla japonica, free amino acids, nucleotides and their related compounds, organic bases, sugars, organic acids and minerals were analyzed, and then followed by organoleptic test of synthetic extract prepared on the basis of the analytical data. Taste panel assessments of synthetic extracts prepared with each extractive component omitted were carried out by a triangle difference test, and changes in profile were assessed. In free amino acid composition, lysine was dominant occupying $25\%$ of total free amino acid. The other abundant amino acids were glycine, arginine, alanine and histidine. From the results of analysis of nucleotides, IMP was dominant showing about $70\%$ of total nucleotides while ATP, ADP, AMP, inosine and hypoxanthine were low in content. Among organic bases total creatinine was abundant. The amount of betaine was 24 mg/100 g. The main organic acid were butyric acid, valeric acid and succinic acid. As for the sugars, glucose and inositol were 1-2 mg/ 100g in content. $K^+,\;Na^+,\;PO_{4}^{3-}\;and\;Cl^-$ were found to be the major ions. From the results of omission test the major components which contribute to produce the taste were glycine, serine, glutamic acid, IMP, $Na^+,\;K^+,\;Cl^-,\;PO_{4}^{3-}$, lysine, alanine, isoleucine, aspartic acid and creatinine.

Food Component Characteristics of Tang from Conger Eel By-products (붕장어 부산물로 제조한 붕장어탕의 식품학적 특성)

  • For the effective use of the conger eel by-products, such as head and frame, Tang, which is the Korean-type soup, from conger eel by-products (TCEB) was developed and its food component characterization was compared with that of commercial Chueotang, loach Tang. According to the results of viable cells and coliform group of TCEB heated at $115^{\circ}C$ for various times, the reasonable $F_0$ value was 8 min. The proximate composition of TCEB was 90.7% for the moisture, 4.8% for the protein, 2.6% for the lipid, and 1.5% for the ash. The extractive-nitrogen content of TCEB was 243.1 mg/100 g, which was higher than that of commercial Chueotang, 208.0 mg/100 g. The total amino acid content of TCEB was 4,310 mg/100 g and its major amino acids were glutamic acid (637.3 mg/100 g, 14.8%), glycine (409.1 mg/100 g, 9.5%) and alanine (404.4 mg/100 g, 9.3%). TCEB was not felt in the sensual fish odor and its sensual taste was good. The health functional properties for health of TCEB were 1.29 as a PF (protection factor) for antioxidative activity and 39.4% for angiotensin Ⅰ converting enzyme (ACE) inhibiting activity.

A Study on the Lipid Components of Hazel Nut Oil (개암종실(種實)의 지질성분(脂質成分)에 관한 연구)

  • Physico-chemical characteristics of crude oil extracted from Korean Hazel nut were determined and its proximate composition was also analyzed. The proximate composition of Hazel nut was shown to be moisture 4.0%, crude protein 15.5%, crude fat 64%, nitrogen free extractive 11.7%, crude fiber 2.0% and crude ash 2.5%. The content of crude fat in Corylus sieboldiana was about 3% higher than in Corylus mandshurica.. Physico-chemical characteristics of crude oil found were: specific gravity, $0.916(15/15^{\circ}C)$; refractive index, $1.468(15^{\circ}C)$; saponification value, 184; iodine value, 94.5: acid value, 0.2; and unsaponifiable content, 0.25%. The lipid fractions in the crude oil obtained by silicic acid column chromatography were found to be composed of about 97% neutral lipids and about 3% compound lipids. Among the neutral lipids by TLC, triglycerides were 98% as the major components, free fatty acids and free strols were 0.5% and 1.3%, respectively. Esterified sterols were not found. The predominant fatty acids were oleic $(76{\sim}80%)$, linoleic (15%) and palmitic (5.0%), and the P/S ratio was $1.8{\sim}2.8$.

Age Variation in Extractive Nitrogenous Constituents of the Cultured Ascidian, Halocynthia roretzi Muscle (양식산 우렁쉥이, Halocynthia roretzi 함질소 엑스성분의 연령차)

  • The muscle extracts of the ascidian, Halocynthia roretzi cultured for two and three years old on the southern coast near Chungmu and the eastern coast near Pohang of Korea, were analyzed for extractive nitrogen (EN), free amino acids (FAA), combined amino acids (CAA), nucleotides and related compounds (NRC), quaternary ammonium bases and guanidino compounds using specimens collected in February 1989 and in April 1989, and compared for those contents with each other. As for the amount of EN, no remarkable difference was found between two- and three-year-old samples collected at St. 1 in the spring and winter seasons, while at St. 2 in the spring season the two-year-old sample was distinctly lower than the three-year-old one. Taurine, proline, glutamic acid, glycine and alanine were the major FAA in every sample. The amount of taurine, the most prominent FAA, was higher in three-year-old sample than in two-year-old one regardless of sampling station and season. Most of the other major FAA showed a similar tendency to EN at both sampling stations in both seasons. Adenosine 5'-triphosphate (ATP), adenosine 5'-diphosphate (ADP), adenosine 5'-monophosphate (AMP), inosine 5'-monophosphate (IMP), inosine (Ino) and hypoxanthine (Hyp) were detected in all the samples and ATP, ADP and AMP were the major ingredients. The amounts of total NRC were in parallel with those of EN and total FAA. As for the contents of betaines, two- and three-year-old samples collected in the winter season exhibited a great discrepancy each other, the former being clearly lower than the latter, but no remarkable difference was observed between two samples of two groups in the spring season. In proximate composition of the muscles, the two-year-old sample was considerably higher in moisture content and lower in protein and glycogen contents than the three-year-old one at St. 2 in the spring season. The large discrepancies observed between two- and three-year-old samples from St. 2 seems to be attributable to the difference in size of samples rather than to the difference in age.
