Nucleic Acid Aptamers: New Methods for Selection, Stabilization, and Application in Biomedical Science

  • The adoption of oligonucleotide aptamer is well on the rise, serving an ever increasing demand for versatility in biomedical field. Through the SELEX (Systematic Evolution of Ligands by EXponential enrichment), aptamer that can bind to specific target with high affinity and specificity can be obtained. Aptamers are single-stranded nucleic acid molecules that can fold into complex three-dimensional structures, forming binding pockets and clefts for the specific recognition and tight binding of any given molecular target. Recently, aptamers have attracted much attention because they not only have all of the advantages of antibodies, but also have unique merits such as thermal stability, ease of synthesis, reversibility, and little immunogenicity. The advent of novel technologies is revolutionizing aptamer applications. Aptamers can be easily modified by various chemical reactions to introduce functional groups and/or nucleotide extensions. They can also be conjugated to therapeutic molecules such as drugs, drug containing carriers, toxins, or photosensitizers. Here, we discuss new SELEX strategies and stabilization methods as well as applications in drug delivery and molecular imaging.

Repeated Loading Tests of Reinforced Concrete Beams Containing Headed Shear Reinforcement (Headed Shear Bar를 사용한 콘크리트 보의 반복 하중 실험)

  • The repeated loading responses of four shear-critical reinforced concrete beams, with two different shear span-to-depth ratios, were studied. One series of beams was reinforced using pairs of bundled stirrups with $90^{\circ}C$ standard hooks, having free end extensions of $6d_b$. The companion beams contained shear reinforcement made with larger diameter headed bars anchored with 50mm diameter circular heads. A single headed bar had the same area as a pair of bundled stirrups and hence the two series were comparable. The test results indicate that beams containing headed bar stirrups have a superior performance to companion beams containing bundled standard stirrups, with improved ductility, larger energy adsorption and enhanced post-peak load carrying capability. Due to splitting of the concrete cover and local crushing, the hooks of the standard stirrups opened, resulting in loss of anchorage. In contrast, the headed bar stirrups did not lose their anchorage and hence were able to develop strain hardening and also served to delay buckling of the flexural compression steel. Excellent load-deflection predictions were obtained by reducing the tension stiffening to account for repeated load effects.

Fabrication of p-type FinFETs with a 20 nm Gate Length using Boron Solid Phase Diffusion Process

  • A simple doping method to fabricate a very thin channel body of the p-type FinFETs with a 20 nm gate length by solid-phase-diffusion (SPD) process was developed. Using the poly-boron-films (PBF) as a novel diffusion source of boron and the rapid thermal annealing (RTA), the p-type sourcedrain extensions of the FinFET devices with a threedimensional structure were doped. The junction properties of boron doped regions were investigated by using the $p^+-n$ junction diodes which showed excellent electrical characteristics. Single channel and multi-channel p-type FinFET devices with a gate length of 20-100 nm was fabricated by boron diffusion process using PBF and revealed superior device scalability.

  • One of the important topics concerning the safety of electrical and electronic system is the reliability of the wiring system. The Time-Frequency Domain Reflectometry(TFDR) is a state-of-the-art system for detection and estimation of the fault on a wiring/cable. The purpose of this paper is to implement a Labview based TFDR Real Time system though the instruments of PCI extensions for Instrumentation(PXI). The TFDR Real Time system consists of the five parts: Reference signal design, signal generation, signal acquisition, algorithm execution, results diplay part. In the signal generation and acquisition parts we adopt the Arbitrary Waveform Generator(AWG) and Digital Storage Oscilloscope(DSO) PXI modules which offer commonality, compatibility and easy integration at low cost. And execution of the PXI modules not only is controlled by the Labview programing but also the total system process is executed by the Labview application software.

On-off Regulation of 3' Exonuclease Excision to DNA Polymerization by Exo+ Polymerase

  • The role of 3' exonuclease excision in DNA polymerization was evaluated in primer extensions using 3' allele-specific primers that had exonuclease-digestible and exonuclease-resistant 3' termini. With exonuclease-digestible unmodified 3' mismatched primers, the exo+ polymerase yielded template-dependent products. Using exonuclease-resistant 3' mismatched primers, no primer-extended product resulted from exo+ polymerase. As a control, polymerase without proofreading activity yielded primer-dependent products from 3' mismatched primers. These data indicated that a successful removal of the mismatch is required for DNA polymerization from the 3' mismatched primers by exo+ polymerase. In addition to the well-known proofreading from this mismatch removal, the premature termination in DNA polymerization, due to the failure of the efficient removal of the mismatched nucleotides, worked as an off-switch in maintaining the high fidelity in DNA replication from exo+ polymerase.

  • In this note we establish a new non-convex hybrid iteration algorithm corresponding to Khan iterative process [4] and prove strong convergence theorems of common fixed points for a uniformly closed asymptotically family of countable quasi-Lipschitz mappings in Hilbert spaces. Moreover, the main results are applied to get the common fixed points of finite family of quasi-asymptotically nonexpansive mappings. The results presented in this article are interesting extensions of some current results.

Numerical analysis of RC hammer head pier cap beams extended and reinforced with CFRP plates

  • This paper presents a numerical study on structural behavior of hammer head pier cap beams, extended on verges and reinforced with carbon fiber reinforced polymer (CFRP) plates. A 3-D finite element (FE) model along with a simplified analytical model are presented. Concrete damage plasticity (CDP) was adapted in the FE model and an analytical approach predicting the CFRP anchor strength was adapted in both FE and analytical model. Total five quarter-scaled pier cap beams with various CFRP reinforcing schemes were experimentally tested and analyzed with numerical approaches. Comparison between experimental results, FE results, analytical results and current ACI guideline predictions was presented. The FE results showed good agreement with experimental results in terms of failure mode, ultimate capacity, load-displacement response and strain distribution. In addition, the proposed strut-and-tie based analytical model provides the most accurate prediction of ultimate strength of extended cap beams among the three numerical approaches.

Implementation of a Human Body Motion Pattern Classifier using Extensions of Primitive Pattern Sequences (프리미티브 패턴 나열의 확장에 의한 사람 몸 동작 패턴 분류기의 구현)

  • 사람의 몸 동작을 인식해야하는 여러 응용분야에서의 필요성이 대두되면서 이 분야로의 연구가 활발해지고 있다. 이 논문은 사람의 비언어적 행동을 자동적으로 분석할 수 있는 인식기 개발에 관한 것으로 실세계 3 차원 좌표값을 입력으로 하는 사람 몸 동작 패턴 분류기의 구현방법을 소개한 것이다. 하나의 사람 몸 동작은 각 몸 구성 성분(손, 아래팔, 위팔, 어깨, 머리, 몸통 등)의 움직임을 조합해서 정의한 수가 있기 때문에 개별적인 각 몸 구성성분의 움직임을 인식하여 조합해서 임의의 동작을 판별하려는 방법을 적용한다. 사람 몸 동작 패턴 분류기는 측정된 실세계 3 차원 좌표 자료를 양자화한 후 xy, zy 평면에 투영한 값을 자자 구한다. 이 결과를 각각 8 방향 체인 코드로 바꾸고 2 단계 체인 코드 평활화 사업을 하여, 4 방향 코드 체적화 및 대표 코드로의 압축단계를 거친다. 이로서 생성된 프리미티브 패턴나열들을 동작 클래스별로 분류하여 프리미티브 패턴나열의 확장으로 각각의 식별기를 구축하여 각 몸 구성 성분별 동작들을 분류한다. 일련의 실험이 행해져 그 타당성을 확인하였으며, 차후에 이 분류기는 비언어적 행동 분석을 위한 사람 몸 동작 인식기의 전처리 단계로 사용되어진 것이다.

ON COMMUTATIVITY OF NILPOTENT ELEMENTS AT ZERO

  • The reversible property of rings was initially introduced by Habeb and plays a role in noncommutative ring theory. In this note we study the reversible ring property on nilpotent elements, introducing the concept of commutativity of nilpotent elements at zero (simply, a CNZ ring) as a generalization of reversible rings. We first find the CNZ property of 2 by 2 full matrix rings over fields, which provides a basis for studying the structure of CNZ rings. We next observe various kinds of CNZ rings including ordinary ring extensions.

  • In the theory of hypergeometric and generalized hypergeometric series, classical summation theorems have been found interesting applications in obtaining various series identities for ${\Pi}$, ${\Pi}^2$ and ${\frac{1}{\Pi}}$. The aim of this research paper is to provide twelve general formulas for ${\frac{1}{\Pi}}$. On specializing the parameters, a large number of very interesting series identities for ${\frac{1}{\Pi}}$ not previously appeared in the literature have been obtained. Also, several other results for multiples of ${\Pi}$, ${\Pi}^2$, ${\frac{1}{{\Pi}^2}}$, ${\frac{1}{{\Pi}^3}}$ and ${\frac{1}{\sqrt{\Pi}}}$ have been obtained. The results are established with the help of the extensions of classical Gauss's summation theorem available in the literature.
