BACKGROUND/OBJECTIVES: Different fatty acids exert different health benefits. This study investigated the potential protective effects of perilla, olive, and safflower oils on high-fat diet-induced obesity and colon inflammation. MATERIALS/METHODS: Five-week old, C57BL/6J mice were assigned to 5 groups: low-fat diet (LFD), high-fat diet (HFD) and high-fat diet supplemented with-perilla oil (HPO), olive oil (HOO), and safflower oil (HSO). After 16 weeks of the experimental period, the mice were sacrificed, and blood and tissues were collected. The serum was analyzed for obesity- and inflammation-related biomarkers. Gene expression of the biomarkers in the liver, adipose tissue, and colon tissue was analyzed. Micro-computed tomography (CT) analysis was performed one week before sacrifice. RESULTS: Treatment with all the three oils significantly improved obesity-induced increases in body weight, liver weight, and epididymal fat weight as well as serum triglyceride and leptin levels. Treatment with perilla oil (PO) and safflower oil (SO) increased adiponectin levels. The micro-CT analysis revealed that PO and SO reduced abdominal fat volume considerably. The mRNA expression of lipogenic genes was reduced in all the three oilsupplemented groups and PO upregulated lipid oxidation in the liver. Supplementation of oils improved macroscopic score, increased colon length, and decreased serum endotoxin and proinflammatory cytokine levels in the colon. The abundance of Bifidobacteria was increased and that of Enterobacteriaceae was reduced in the PO-supplemented group. All three oils reduced proinflammatory cytokine levels, as indicated by the mRNA expression. In addition, PO increased the expression of tight junction proteins. CONCLUSIONS: Taken together, our data indicate that the three oils exert similar anti-obesity effects. Interestingly, compared with olive oil and SO, PO provides better protection against high-fat diet-induced colon inflammation, suggesting that PO consumption helps manage inflammation-related diseases and provides omega-3 fatty acids needed by the body.
This study was performed to examine the feeding effects of $Angelica$$keiskei$$Koidz$ (AK) and its processed products on serum, liver, and body fat content and the expression of antioxidant genes in rats fed a high fat diet. AK and its processed products were added at 3-5% to a high fat diet and fed to adult rats for 6 weeks. In experiment 1 (EXP 1), the rats were fed with one of six diets including a control diet (normal fat), high fat diet (HF), and HF + AK additives groups (four groups). In experiment 2 (EXP 2), the rats were separated into three groups of HF, HF + AK whole leaves, and HF + fermented juice (FS) + squeeze (SA). Body weight was not different among the groups in either experiment. The liver weight was lower in the FS and SA groups compared to that in the other groups (P<0.05). Serum luteolin was higher in the AK and processed products groups compared to that in the HF group (P<0.05). Gene expression of the antioxidative enzymes catalase and glutathione-s-reductase in the liver was higher in the AK processed products group than that in the other groups (P<0.05). The results suggest that the intake of AK and its processed products increased the expression of antioxidant enzymes in animals fed a high fat diet, reduced hepatic cholesterol content, and increased the effective absorption of luteolin.
This study was performed to investigate the effect of therapeutic exercise on brain-derived neurotrophic factor manifestation after global brain ischemia in rats. Nine rats with global ischemia were divided at random into two group. In the control group, three rats remained in cage. But, in the end, two rats were alive. In the therapeutic exercise group, six rats remained. The five rats of this group was swam for 30 minutes everyday for a week. The brain-derived neurotrophic factor expression was identified from immunohistochemistry. The results of this study were as follows : 1. In the control group, a little expression of brain-derived neurotrophic factor was observed at cortex and hippocampus layer, but cell body and axon was observed obscurely. 2. In the experimental group, a much expression of brain-derived neurotrophic factor was observed at cortex and hippocampus layer, and cell body and axon was observed clearly. In the neurological examination(beam-walking test). experimental group was obtained higher 1.4 points than control group. BDNF expression was increased by swimming for 30 minutes everyday for a week. Therefore, therapeutic exercise contribute to brain plasticity after brain ischemia.
오존은 수돗물 정수장에서 이용되는 소독 물질로 미세오염 물질들을 비롯해서 박테리아나 병원성 미생물체를 효과적으로 제거하는 것으로 많은 연구가 보고되어 있다. 본 연구에서는 실내 사육 중인 붉은 체색을 지닌 Glyptotendipes tokunagai를 대상으로 서로 다른 농도의 오존 노출에 따른 영향을 파악하기 위해 치사율, 체색 변화와 heat shock protein 70 (HSP70) 유전자 발현을 측정하였다. 오존에 노출된 G. tokunagai에서 농도-시간 의존적으로 치사율 증가가 관찰되었다. 또한 체색 변화는 오존 농도에 따라 붉은색의 체색이 체절마다 엷어지며 탈색되고 경직되는 현상이 보였다. HSP70 유전자 발현은 저농도인 0.2~0.5 ppm에서 노출 10분과 20분에 유의한 수준으로 높게 나타났으나(P<0.05), 30분 노출 후에는 발현량이 감소하는 경향을 보였다. 생리적으로 저산소층에 대해 적응능력이 뛰어난 깔따구 경우에도 오존은 매우 강력한 치사 효과를 유발하여 30분 노출 후 경직과 헤모글로빈 파괴로 인한 탈색이 유발되는 것을 보여주었다. 따라서 본 결과는 수돗물 정수장에서 병원성 미생물을 제거하는 데 사용되는 오존이 수생물에 주는 영향성을 파악하는 기초자료로서 활용될 수 있을 것이다.
Background: Oral carcinoma (OC) remains one of the most difficult malignancies to cure. Hesa-A is an Iranian herbal-marine compound that has shown promising anti-tumor properties against various human tumors. However, its mechanism of action remains to be addressed. The present study was conducted to evaluate the effect of two doses of Hesa-A on mRNA expression of erb$\backslash$b2 as a main prognosticator tumor marker for OC in an animal model. Materials and Methods: A total of 60 rats were randomly divided into 5 groups of 12 animals each. Rats in carcinoma groups received 0, 250 and 500mg/kg body weight doses of Hesa-A 3 times a day. The other two groups were considered as treated and untreated control groups. At the end of the experiment, animals were sacrificed and tongue tissues subjected to H and E staining and real time PCR. Results: Our results showed that compared to the control group, erb$\backslash$b2 was over-expressed ~ 30% in the carcinoma group. After treatment with 250mg/kg and 500mg/kg body weight of Hesa-A, erb$\backslash$b2 levels dropped by 24.1% and 3.4 % respectively compared to the control carcinoma group (p<0.01, p<0.0001). Moreover, there was a significant relation between erb$\backslash$b2 mRNA content and observed pathological changes in studied groups (p<0.05). Conclusions: These data provide insight into mechanism(s) by which Hesa-A may improve clinical outcome of oral carcinoma by affecting oncogene erb$\backslash$b2 expression and suggest Hesa-A as an effective chemotherapeutic agent in treatment of HER+tumors.
디지털 콘텐츠에서 HCI로 활용되는 가상 인간은 얼굴 표정과 신체자세와 같은 모달을 이용하여 다양한 감정을 표현하지만 비언어적 다중모달의 조합에 대한 연구는 많지 않다. 감정을 표현하는 가상 인간을 제작하려면 계산 엔진 모델은 얼굴 표정과 신체자세와 같은 비언어적 모달의 조합이 사용자에 의해 어떻게 인식되는지를 고려해야 하기 때문에 본 연구는 가상 인간의 감정 표현 디자인에 필요한 비언어적 다중모달의 영향을 분석하여 제시한다. 먼저 가상 인간에 대한 다중모달 별 감정 인식을 평가하여 다른 모달간의 상대적 영향성을 분석하였다. 그리고 일치하는 얼굴과 자세 모달을 통해 기본 감정 및 정서가와 활성화 인식에 대한 영향을 평가하며 감정이 불일치하는 다중모달을 통해 일상생활에서 빈번하게 드러나는 중첩된 감정의 인식 정도를 관측하였다. 실험 결과, 가상 인간의 얼굴과 신체자세의 표정이 일치하면 감정 인식이 용이하며, 얼굴 표정으로 감정 카테고리를 판별하지만 감정의 활성화 차원 판단에는 자세 모달리티가 선호됨을 확인하였다. 본 연구 결과는 감정을 드러내는 가상 인간의 행동 동기화 및 애니메이션 엔진 시스템 구현에 활용할 수 있다.
탈놀이는 무언극 형태와 유언극 형태가 혼합되어 있음에도 불구하고 기존의 연구에서 연극적·미학적 논의가 유언탈놀이 중심으로 전개된 사실을 반성하고, 무언극을 본격적으로 논의하여 유언극과 변별되는 연극미학을 정립하려고 하였다. 먼저 텍스트의 문제로 기존의 채록본을 검토하여 무언탈놀이를 기록함에 있어서 개괄적·설명적·해석적 기록을 한 오류를 지적하고, 객관적·묘사적 기록방법을 취하고, 무보(舞譜)를 작성하고, 동선(動線)까지 표시할 것을 주장하였다. 그리고 영상기록의 필요성도 재확인하였다. 다음으로 무언극을 '인상의 미믹(mimic)적 묘사와 몸짓에 의해 모방하는 표현예술'로 규정하고 물리적 신체동작과 기호화된 신체언어에 의한 미믹에 대하여 하회별신굿탈놀이의 무동춤·걸립놀이, 봉산탈춤의 노장·소무놀이와 사상좌놀이, 수영들놀음의 사자춤놀이를 집중적으로 분석하였다. 마지막으로 등장인물이 동일한 무언극과 유언극, 동일한 마당의 무언탈과 유언탈을 대조하여 유언극이 음성언어매체를 이용하여 무언극보다 표현영역을 확장하고 표현력을 강화한 사실을 구명하였다. 그리고 탈놀이가 무언극으로 시작하여 유언극 시대로 이행하였지만 무언극이 여전히 존재하여 발달 단계가 상이한 두 연극양식이 적층·혼합되어 있는 연극사적 사실을 확인하였다.
본 연구는 수영운동이 백색지방조직의 혈관신생을 조절함으로써 비만을 개선하는지를 조사하였다. 고지방식이를 섭취한 암컷 쥐는 모의 수술 군(Sham), 난소절제 수술 군(OVX) 및 수영운동을 실시한 난소절제 군(OVX + Swim)으로 나뉘었다. Sham에 비해 OVX는 몸무게, 지방조직무게 및 지방세포의 크기가 증가 되었다. 그러나 OVX + Swim의 이러한 요소들(: 몸무게, 지방조직무게 및 지방세포의 크기)은 OVX에 비해 감소 되었다. Sham에 비해 OVX는 백색지방조직에서 혈관신생 촉진인자와 MMPs의 유전자 발현이 증가하였고, 혈관신생 억제인자들의 유전자 발현은 감소하였다. 그러나 OVX + Swim은 OVX에 비해 백색지방조직에서 혈관신생 촉진인자와 MMPs의 유전자 발현이 감소하였고, 혈관신생 억제인자들의 유전자 발현은 증가하였다. 이러한 연구결과들은 고지방식이를 섭취한 난소절제 암컷 쥐에서 수영운동이 백색지방조직의 혈관신생을 억제함으로써 비만을 개선한다는 것을 제시하였다.
단백질 인산화를 통한 세포내 신호전달기구 중 serine/threonine kinase에 속하는 Akt/PKB는 세포 생존과 사멸, 당대사 등을 조절하는 것으로 알려져 있다. 이러한 이유로, Akt/PKB 단백질은 천연물질들로부터 항암제를 탐색하기 위한 한 가지 target으로 사용되어 왔다. 본 연구에서는 Akt/PKB 단백질을 대량으로 생산하기 위하여 대장균의 단백질 발현 시스템을 이용하여 human Akt/PKB 단백질을 발현시켰다. 대장균에서 대량 발현된 Akt는 일반적인 조건에서는 inclusion body를 형성하였다. 배양온도 $27^{\circ}C$에서 0.01-0.09 mM IPTG로 발현 유도 시 발현된 human Akt/PKB 단백질 상당 부분이 가용화 되었다. 발현된 Akt kinase를 $Ni^{2+}$-NTA agarose column으로 정제하고, anti-Akt antibody를 이용하여 정제된 단백질이 Akt kinase 임을 확인하였다. 정제된 human Akt/PKB 단백질은 세포추출물에 존재하는 인산화 단백질을 이용하여 in vitro에서 인산화 되었으며, 인산화된 활성형 human Akt/PKB protein kinase는 human Akt/PKB protein kinase 특이 형광 peptide를 특이적으로 인산화하였다.
As a $\beta_2$-adrenergic agonist, clenbuterol decreases body fat, but the molecular mechanism underlying this process is unclear. In the present study, we treated 293T and L-02 cells with clenbuterol and found that clenbuterol downregulates SREBP-1c expression and upregulates CREB1 expression. Considering SREBP-1c has the function of regulating the transcription of several lipogenic enzymes, we considered that the downregulation of SREBP-1c is responsible for body fat reduction by clenbuterol. Many previous studies have found that clenbuterol markedly increases intracellular cAMP levels, therefore, we also investigated whether CREB1 is involved in this process. The data from our experiments indicate that CREB1 overexpression inhibits SREBP-1c transcription, and that this action is antagonized by CREB2, a competitive inhibitor of CREB1. Furthermore, since PPARs are able to repress SREBP-1c transcription, we investigated whether clenbuterol and CREB1 function via a pathway involving PPAR activation. However, our results showed that clenbuterol or CREB1 overexpression suppressed PPARs transcription in 293T and L-02 cells, which suggested that they impair SREBP-1c expression in other ways.
