Molecular characterization of bacterial endosymbionts of Acanthamoeba isolates from infected corneas of Korean patients

  • The endosymbionts of 4 strains of Acanthamoeba(KA/E9, KA/E21, KA/E22, and KA/E23) isolated from the infected corneas of Korean patients were characterized via orcein stain, transmission electron microscopic examination, and 16S rDNA sequence analysis. Double membrane-bound, rod-shaped endosymbionts were distributed randomly throughout both the trophozoites and cysts of each of Acanthamoeba isolates. The endosymbionts of KA/E9, KA/E22, and KA/E23 were surrounded by electron-translucent areas. No lacunae-like structures were observed in the endosymbionts of KA/E21, the bacterial cell walls of which were studded with host ribosomes. Comparative analyses of the 16S rDNA sequences showed that the endosymbionts of KA/E9, KA/E22 and KA/E23 were closely related to Caedibacter caryophilus, whereas the KA/E21 endosymbiont was assigned to the Cytophaga-Flavobacterium-Bacteroides(CFB) phylum. In the 4 strains of Acanthamoeba, the hosts of the endosymbionts were identified as belonging to the Acanthamoeba castellanii complex, which corresponds to the T4 genotype. Acanthamoeba KA/E21 evidenced characteristics almost identical to those of KA/E6, with the exception of the existence of endosymbionts. The discovery of these endosymbionts from Acanthamoeba may prove essential to future studies focusing on interactions between the endosymbionts and the amoebic hosts.

The relationship among self-esteem, life-satisfaction and stress coping style of dental hygiene students (일부 치위생과 학생들의 스트레스 대처방식이 자존감과 삶의 만족도에 미치는 영향)

  • Objectives : This study aimed to determine the effects of stress coping type on self-esteem and satisfaction with life in dental hygiene students. In particular, an attempt is made to determine the effects of sub-factors of stress coping type on self-esteem and satisfaction with life, get a better understanding of self-esteem and satisfaction with life for dental hygiene students, and provide basic data in developing a program that can identify, prevent, and control the problems with stress for dental hygiene students. Methods : This is descriptive correlation research to determine the relations between self-esteem and stress coping type in dental hygiene students. 212 dental hygiene students at colleges in Ulsan Metropolitan City were asked to complete a self-administered questionnaire from June 10 to 20, 2013; then, 209 copies were used in analysis, with the exception of 3 copies which were excluded from recording. Results : Self-esteem and satisfaction with life were positively affected by active stress coping but were negatively affected by passive stress coping. As for such sub-factors of stress coping type as problem-focused coping, pursuit of social support, emotional relief coping, and hopeful thinking, the sub-factors of active coping type also positively affected self-esteem and satisfaction with life. Self-esteem interacted with the sub-factors of stress coping type and satisfaction with life. Conclusions : Since stress coping type was found to be a factor affecting self-esteem and satisfaction with life for students, the results of this study are expected to be used as basic data in developing a program that can help them cope actively with stress.

An Analysis on Rural Housing Standard Plans for the Elderly based on Case Studies of Elderly Housing in Rural Area (농촌 고령자주택 사례조사에 근거한 농어촌주택 표준설계도 분석)

  • The objective of this paper is to investigate problems associated with Rural Housing Standard Plans for the Elderly based on the current status of 12 houses belonging to elderly people (60's and over) living in rural areas and engaged in farming activities, and to suggest improvements. The methods employed were a field survey with interviews and literature review. The results are as follows. First, the Plans were deemed inappropriate to prevent the possible accidents in entrance area such as slippiness and falls because the eaves are not long enough. Second, the area of foyer and the volume of foyer storage in the Plans are not enough to accommodate the subject's behaviors and their rural life. Third, rather than using the sink, many subjects preferred to wash simply in the shower while sitting, so the planning of a bathroom can't meet the subject's needs and behaviors. Fourth, the subjects prefer L + DK to LDK but the Plans were designed in the manner of LDK only. Fifth, the subjects have needs for bathrooms belong to the master bedrooms, but only one case of Plans satisfied their needs. Six, bedrooms with the exception of the master bedroom are used as storage in order to make up for a lack of storage space, but many storage space such as dressing rooms and pantries was not taken into consideration in the Plans. Lastly, the planning of storehouses does not meet the subjects' using behaviors for storehouses and the level of desirable capacity of goods.

N Screen Service Users' Motivations for Use and Dissatisfying Factors (N 스크린 서비스 이용자의 이용 동기와 불만족 요인에 관한 연구)

  • This research attempts to identify N-screen users' motivational and dissatisfying factors in relation to their willingness for continued future use of the service. An on-line survey was conducted (N=558) to reveal five major factors for N-screen use: Medium usability (interface usability and daily usefulness), Socialness (potential for social use with family and friends), Mobility (use without restraints of time and space), Contents (diversity of contents catalogue and related information search), and Habitual use. With exception of Contents factor, four of those factors were proven to be meaningful predictors of the users' willingness for continued future use of N-screen service. Meanwhile, two factors were extracted as the major source for N-screen users' dissatisfaction: Customer service (dissatisfaction associated with the service itself) and Contents (dissatisfaction relating to contents use). The effect of those two dissatisfaction factors was examined. The results show that Contents dissatisfaction, in comparison with Customer service dissatisfaction, was a more powerful predictor for discontinuing of N-screen service use.

  • Personal Information Protection Act was first enacted in March, 2011, amended in September, 2011 and became effective. Nevertheless, the risk of interfering with personal information protection still remains, associated with collection, plagiarism, leakage, and even sales of personal information. The public libraries are not an exception. Accordingly, this study investigates the status of personal information protection in public libraries of Seoul and Gyeonggi province. Also, it tries to discover problems associated with the act and presents a model scheme to improve this situation. The scheme relates to (1) securing of a sufficient budget; (2) reinforcing professional education related to personal information; (3) developing detailed guidelines for public libraries.

The Role of Thallium-201 Scintigraphy in Bone and Soft Tissue Tumor (근골격계 종양에서 탈륨 스캔의 역할)

  • Thallium-201 scintigraphy is used to discriminate the malignant bone tumor from the benign by qualitatively and quantitatively, and to predict the response of preoperative chemotherapy in osteosarcoma, by comparing the changes of thallium uptake ratio after chemotherapy to the tumor necrosis ratio. Thallium-201 scintigraphy scan should be done prior to surgical biopsy. PICKER Prism 2000 gamma camera with high resolution parallel hole collimator is usually used for scanning. The patient is injected with 2-3mCi of Tl-201 and the early phase is checked in 30 minutes and delayed phase in 3 hours. The scan images are visually evaluated by a blinded nuclear medicine physician. We could evaluate true positive, true negative, false positive and false negative by the comparison of results with those of biopsy, and calculate positive and negative predictive value(%), sensitivity(%), specificity(%) and diagnostic accuracy(%). For the quantitative analysis of thallium uptake, we drew the region of interest on the tumor side and contralateral normal side as mirror image, and calculated the uptake ratio with dividing the amount of gamma count in tumor side by normal side. We could calculate the percent changes of thallium uptake ratio in early and delayed phase, and compare them to the ratio of tumor necrosis. Thallium-201 scintigraphy proved as useful imaging study to discriminate malignant bone tumor from benign, but had exception in giant cell tumor and low grade malignant bone tumors. We can use T1-201 scan to differentiate the benign from the malignant tumor, and to evaluate the response of preoperative chemotherapy or radiotherapy, and to determine the residual tumor or local recurrence. For the better result, we need to have a more detail information about false positive cases and a more objective and quantitative reading technique.

Non-Benzoquinone Geldanamycin Analog, WK-88-1, Induces Apoptosis in Human Breast Cancer Cell Lines

  • Heat shock protein 90 (Hsp90) is treated as a molecular therapeutic target for the prevention and treatment of cancer. Geldanamycin (GA) was the first identified natural Hsp90 inhibitor, but hepatotoxicity has limited its clinical application. Nevertheless, a new GA analog (WK-88-1) with the non-benzoquinone skeleton, obtained from genetically engineered Streptomyces hygroscopicus, was found to have anticancer activity against two human breast cancer cell lines. WK-88-1 produced concentration-dependent inhibition of cell proliferation, cell cycle arrest, and apoptosis in estrogen receptor (ER)-positive MCF-7 and ER-negative MDA-MB-231 cell lines. Detailed analysis showed that WK-88-1 downregulated some key cell cycle molecules (CDK1 and cyclin B1) and lead to $G_2/M$ cell cycle arrest. Further studies also showed that WK-88-1 could induce human breast cancer cell apoptosis by downregulating Hsp90 client proteins (Akt, p-Akt, IKK, c-Raf, and Bcl-2), decreasing the ATP level, increasing reactive oxygen species production, and lowering the mitochondrial membrane potential. Meanwhile, we discovered that WK-88-1 significantly decreased the levels of Her-2 and $ER-{\alpha}$ in MCF-7 cells but not in MDA-MB-231 cells. In addition, WK-88-1 significantly increased caspase-3, -8, and -9 activities and the cleavage of PARP in a concentration-dependent manner (with the exception of caspase-3 and PARP in MCF-7 cells). Taken together, our preliminary results suggest that WK-88-1 has the potential to play a role in breast cancer therapy.

Appropriate In Vitro Methods for Genotoxicity Testing of Silver Nanoparticles

  • Objectives We investigated the genotoxic effects of 40-59 nm silver nanoparticles (Ag-NPs) by bacterial reverse mutation assay (Ames test), in vitro comet assay and micronucleus (MN) assay. In particular, we directly compared the effect of cytochalasin B (cytoB) and rat liver homogenate (S9 mix) in the formation of MN by Ag-NPs. Methods Before testing, we confirmed that Ag-NPs were completely dispersed in the experimental medium by sonication (three times in 1 minute) and filtration ($0.2{\mu}m$ pore size filter), and then we measured their size in a zeta potential analyzer. After that the genotoxicity were measured and especially, S9 mix and with and without cytoB were compared one another in MN assay. Results Ames test using Salmonella typhimurium TA98, TA100, TA1535 and TA1537 strains revealed that Ag-NPs with or without S9 mix did not display a mutagenic effect. The genotoxicity of Ag-NPs was also evaluated in a mammalian cell system using Chinese hamster ovary cells. The results revealed that Ag-NPs stimulated DNA breakage and MN formation with or without S9 mix in a dose-dependent manner (from $0.01{\mu}g/mL$ to $10{\mu}g/mL$). In particular, MN induction was affected by cytoB. Conclusions All of our findings, with the exception of the Ames test results, indicate that Ag-NPs show genotoxic effects in mammalian cell system. In addition, present study suggests the potential error due to use of cytoB in genotoxic test of nanoparticles.

Development and Evaluation of Action Learning in Clinical Practice of Nursing Management (간호관리학 임상실습에서 액션러닝의 개발 및 평가)

  • The aim of this study is to find the effect of action learning on the problem solving process of nursing students during the clinical practice of nursing management. A total of 99 senior nursing students participated in this study. Data was collected from May 2006 to October 2007 and statistical analysis for paired t-test was performed on the data using SPSS/WIN 14.0. The results of the data show that there was a significant increase in the problem solving process for nursing students after the implementation of action learning(t=-4.718, p=.000). In the problem solving process, there was a significant increase in definition of problem(t=-4.123, p=.004), design of problem solution(t=-2.973, p=.002), execution of problem solution(t=-3.264, p=.000) and investigation of problem solving(t=-3.677, p=.000). The only exception in the problem solving process was detection of problem(t=-1.858, p=.066). Therefore, action learning provides nursing students a new alternative for improving the problem solving process and clinical adaptability after graduating from nursing school.

A Comparative Analysis of the Library Exceptions in Copyright Laws in Major Countries (주요국 저작권법의 도서관 예외 규정 비교 분석)

  • The purpose of this study is to enhance the understanding of the provisions for library exceptions in copyright laws and to suggest the direction for the revision of provision in the Copyright Act of Korea. This study compares and analyzes the provisions for library exceptions in several countries' copyright law including Korea, Japan, UK, Australia and USA. This study found that the most common issues in library exception provisions are providing reproduction for users, reproduction for preservation, reproduction for interlibrary loan and digitization of library collections but the conditions for each issue are quite different. The suggestions for the revision of the library exceptions in the Copyright Act of Korea are as follows: changing the amount of reproduction of unpublished works, of works which cannot be obtained at a fair price and of periodicals for users, the transmitting digital reproduction to the user under limited conditions, preparing conditions for reproduction for preservation in libraries, preparing the applicable provisions for reproductions for interlibrary loans and for the limitation of works to be digitized and extending their use at the same time.
