Preparation of $Gal^3Man_4(6^3-mono-{\alpha}-D-galacto-pyranosyl-{\beta}-mannotetraose)$ by Bacillus sp. ${\beta}-mannanase$ and Growth Activity to Intestinal Bacteria (Bacillus sp.유래 ${\beta}-mannanase$에 의한 $Gal^3Man_4(6^3-mono-{\alpha}-D-galacto-pyranosyl-{\beta}-mannotetraose)$ 조제 및 장내세균에 대한 생육활성)

  • For the elucidation of substrate specificity to the brown copra meal by Bacillus sp. ${\beta}-mannanase.$, the enzymatic hydrolysate after 24 hr of reaction was heated in a boiling water bath for 10 min, and then centrifuged to remove the insoluble materials from hydrolysates. The major hydrolysates composed of D.P 5 and 7 galactosyl mannooligosaccharides. For the separate of galactosyl mannooligosaccharides, the supernatant solution of 150 ml was put on a first activated carbon column. The column was then washed with 5 l of water to remove mannose and salts. The oligosaccharides in the column were eluted by a liner gradient of $0{\sim}30%$ ethanol, at the flow rate of 250 ml per hour. The sugar composition in each fraction tubes was examined by TLC and FACE analysis. The combined fraction from F3 was concentrated to 30 ml by vacuum evaporator. Then put on a second activated carbon column. The oligosaccharides in the column were eluted by a liner gradient of $0{\sim}30%$ ethanol (total volume: 5 l), at the flow rate of 250 ml per hour. The eluent was collected in 8 ml fraction tubes, and the total sugar concentration was measured by method of phenol-sulfuric acid. The major component of F2 separated by 2nd activated carbon column chromatography were identified $Gal^3Man_4(6^3-mono-{\alpha}-D-galactopyranosyl-{\beta}-mannotetraose)$. To investigate the effects of brown copra meal galactomannooligosaccharides on growth of Bifidobacterium longum, B. bifidum were cultivated individually on the modified-MRS medium containing carbon source such as $Gal^3Man_4$, compared to those of standard MRS medium.

수처리 목적의 대기압플라즈마를 이용한 유사 폴리도파민 필름 증착

  • Polydopamine은 수중 접착력, 친환경 접착제, nanoparticle absorption 등 다양한 특성으로 많이 연구되고 있는 소재이다. 본 연구에서는 dopamine을 이용하여 수중 금속을 흡착시키는 thin film을 제작하였다. 종래의 Polydopamine coating 방법으로 wet coating 이 사용되고 있다. 하지만 wet 방식의 경우 시간이 오래 걸릴 뿐만 아니라 in-line, roll to roll 방식을 적용하는 것이 어렵기 때문에 생산적이지 않다. 이에 본 연구에서는 Atmospheric Pressure Plasma(APP)를 이용 하여 Polydopamine-like film을 coating 하였다. APP의 경우 vacuum system, solution tank가 필요 없고 in-line, roll to roll 방식을 적용 할 수 있기 때문에 더 경제적이고 생산적인 공정이다. 또한 기존의 Plasma polymerization 방법은 Plasma energy가 높기 때문에 source의 분자구조가 바뀌거나 atom 단위로 분해된다. source의 분자구조가 바뀌는 "Atomic polymerization", Neiswender-Rosskamp Mechanism이 적용되면 wet 방식 coating한 film과는 다른 특성을 갖게 된다. 하지만 APP polymerization은 Plasma energy가 vacuum plasma 보다 매우 낮기 때문에 stile polymerization mechanism을 구현 하는데 적합 하다. stile polymerization mechanism은 Plasma 내부에서 polymer source를 분해 성장 시켜서 Polymer film 얻는 것이 아닌 source의 분자구조가 깨지지 않으면서 polymer growing 시키는 방법이다. dopamine source의 분자구조를 최대한 유지하려고 하는 이유는 metal absorption과 같은 특성이 dopamine chemical structure에 영향을 받기 때문이다. 많은 논문들에서 dopamine의 catechol group이 metal absorption, adhesion force에 영향을 주는 주요 인자라고 주장하고 있기 때문이다. 그래서 본 논문에서는 Dopamine source의 형태를 보존하면서 Polymerization 하는 방법으로 APP process를 사용 하여 낮은 전압에서 Polydopamine-like film을 제작 하였다. APP system 의 Plasma 방전부 에 Dopamine source를 유입하기 위하여 본 논문에서는 Piezo Spray 방식을 사용 하였다. Dopamine을 evaporator 하는 것이 어렵고 chemical composition이 유사한 monomer를 사용해서 Plasma Polymerization으로 Dopamine 분자 구조를 재현하는 것도 어렵다. 그래서 본 연구에서는 Dopamine을 water에 immerse 하고 Dopamine solution을 mist 상태로 만들어서 Plasma discharge area에 유입하였다. 이러한 방법으로 만들어진 film은 Polydopamine film은 아니지만 Polydopamine film과 유사한 Chemical composition, chemical structure, metal absorption을 갖는 것을 FT-IR, SEM, XPS을 이용 하여 확인 하였다. Dopamine source의 보존에 대하여 명확하게 확인하기 위하여 FT-IR을 측정 하였다. 전압에 따른 Benzene ring, hydroxyl group의 비율을 확인 하였다. 낮은 전압으로 coating 된 Polydopamine-like film 일수록 hydroxyl group peak($3400{\sim}3000cm^{-1}$)과 비교하여 Benzene ring peak($1600{\sim}1580cm^{-1}$ and $1510{\sim}500cm^{-1}$)이 흡수를 더 많이 하는 것을 확인 할 수 있다. 이것은 Benzene ring이 파괴되지 않고 보존되는 것을 보여준다. Dopamine에서 Benzene ring은 absorption main factor인 catechol에 있는 chemical structure이다. 즉 Benzene ring peak이 높을수록 Catechol이 잘 보존 되었다는 의미 이다. Catechol의 보존은 absorption main factor가 보존 된다는 의미 이다. 이러한 Polydopamine-like film으로 As, Cr, Mg, Cu 200ppm solution에 대한 filtration 능력을 확인 하였다. As, Cr, Cu, Mg 의 제거율이 각각 약25%, 35%, 45%, 65%인 것을 확인 하였다. 이 수치는 시중에 판매되는 제품들과 비교했을 때 300%~500% 향상된 수치 이다.

  • 금속재료 중 알루미늄(Al)은 일반적으로 많이 사용되는 철강재에 비해 그 비중이 약 $1/3(2.7g/cm^3)$인 경량이고 열전도율이 약 3배($196kcal/^{\circ}C$, $20^{\circ}C$)로 높은 특성 등을 갖고 있다. 또한 대량생산에 의한 경제성을 가지기 때문에 건축 구조재, 전기 및 가전 등 다양한 산업 분야에서 널리 사용되고 있다. 특히, 위와 같은 특성으로 인해 열교환기의 종류에서 응축기(Condenser) 및 증발기(Evaporator)는 알루미늄(Al)을 널리 사용하고 있다. 하지만 단일 응축기 부품에도 이종 알루미늄 소재가 사용됨에 따라 갈바닉 부식이 발생할 수 있다는 단점이 있다. 따라서 본 연구에서는 현재 상용되고 있는 열교환기 중 응축기에서의 이종 알루미늄 재료로 인해 나타나는 갈바닉 부식 특성을 관찰-분석-연구 하였다. 본 연구에 사용된 알루미늄 재료는 현재 응축기 재료 중에서 각각 Tube와 Fin으로 널리 상용되고 있는 Al 1100과 Al 3003을 사용하였다. 표면 모폴로지는 SEM을 통해 관찰하였고 EDS를 통해 조성원소를 분석하였다. 또한 내식성 평가를 위해 5% NaCl 환경에서의 SST(Salt spray test, 염수분무시험) 시험과 3% NaCl 용액 내 자연침지 시험 및 탈기된 3% NaCl 용액 내 전기화학적 동전위 양극 분극 시험을 진행하였으며 더불어 갈바닉 부식 시험에 따른 혼합 전위 측정 및 외관 관찰을 하였다. 각 재료는 실험에 대해 동일한 표면적을 노출시켜 시험하였다. 5% NaCl 환경에서의 염수분무 시험 결과 Fin(Al 3003)의 경우에는 Tube(Al 1100) 보다 빠른 부식거동을 보이며 국부적인 공식부식(Pitting corrosion)이 촉진되었다. Tube(Al 1100)의 경우에는 치밀한 Al2O3 형성과 부식에 따른 Al(OH)3를 생성함에 따라 Fin(Al 3003)에 비해 느린 부식거동을 보였다. 3% NaCl 용액 내 자연 침지 및 전위 측정 결과 초기 전위는 Fin(Al 3003, 약 -0.85V/SCE)이 Tube(Al 1100, 약 -1.05V/SCE)에 비해 더 높은 값을 가지며 약 72시간 이후 Tube(Al 1100) 시편이 더 안정적인 전위 값을 나타냈다. 이는 안정적인 Al(OH)3 피막 형성에 기인한 것으로 사료된다. 탈기된 3% NaCl 용액 내 전기화학적 동전위 양극 분극 시험 결과 Fin(Al 3003) 시편에서 더 귀한 부식 전위 값을 나타냈지만 부식 전류는 더 낮은 값을 나타냈다. 상기 시험 결과를 바탕으로 Fin-Tube 간 장기간 접촉 시에는 갈바닉 부식이 발생할 수 있을 것으로 사료된다. 갈바닉 부식 시험 결과 초기 혼합 전위는 약 -1.05 V/SCE를 나타냈으며 약 288시간 경과 후 약 -0.85 V/SCE 값을 나타냈다. 이는 자연 전위 측정 및 동전위 양극 분극 시험에서의 부식 전위 값에서 알 수 있듯이 더 비한 전위인 Tube(Al 1100) 시편이 Fin(Al 3003) 시편에 대해 희생양극적(Sacrificial anode)인 역할을 한 것을 확인할 수 있었다. 또한 갈바닉 부식 전위 측정 간 외관 관찰에서도 Tube(Al 1100) 시편은 빠르게 흑변 하는 것을 확인할 수 있었으며 Fin(Al 3003) 시편은 침지 300시간 이후에도 초기와 유사한 표면 상태 및 광택을 유지하였다. 이상의 SST 시험, 자연 침지 시험, 전기화학적 양극 분극 시험 및 갈바닉 부식 시험 결과를 바탕으로 단일 부품 내 이종 알루미늄 소재 간 접촉 및 그에 따른 갈바닉 부식 발생을 확인할 수 있었다. 따라서 동종 성분이라고 할지라도 단일 부품 제작 시에는 그 사용 환경에 따라 이종 금속 재료의 사용에 대한 재고가 필요할 것으로 사료된다.

Fabrication of $TiO_{2}$ In-line Reflection Mirror and Its Characteristics for Fiber Optic Fabry-Perot Interferometric Sensor (광섬유 Fabry-Perot 간섭형 센서 제조를 위한 $TiO_{2}$ 반사막의 형성 및 그 특성)

  • For the fabrication of high sensitive intrinsic fiber optic Fabry-Perot interferometeric sensor, the deposition conditions of $TiO_{2}$ thin film used to the internal mirrors of the sensor were investigated. The $TiO_{2}$ film deposited by RF magnetron sputter had higher refractive index ($2.36{\sim}2.48$) and better stoiciometry (O/Ti = 2) than that deposited bye-beam evaporator. In the case of forming $TiO_{2}$ internal mirror by using fusion splicing technique, the $TiO_{2}$ reflection mirror deposited by RF magnetron sputter in the condition of 120W RF power showed high. reflectance and excellent controllability of reflection power. The fabricated intrinsic fiber optic Fabry-Perot interferometer with two $TiO_{2}$ internal mirrors deposited under the condition showed very stable fringe patterns. It is, therefore, expected that the interferometer will be applicable to various high precision sensors.

Effects of Sodium Metabisulfite and Adipic Acid on Browning of Garlic Juice Concentrate during Storage (Sodium metabisulfite와 adipic acid가 마늘 농축액의 저장 중 갈변현상에 미치는 영향)

  • The effect of sodium metabisulfite and adipic acid on the color changes of garlic juice was evaluated. The garlic(Uisung variety) juice was extracted and 1% sodium metabisulfite and 1% adipic arid were added to the garlic juice before or after concentration. Garlic juice was concentrated by heating at 90$\^{C}$, by heating using a rotary vacuum evaporator at 45$\^{C}$, or by freeze-drying at -50$\^{C}$ until the volume was reduced to 70% of the original volume. The garlic juice concentrate was kept at 4$\^{C}$ and 25$\^{C}$ for 60 days and evaluated for the color change and the relation between the additives and browning. Browning of the garlic juice concentrate containing adipic acid and sodium metabisulfite was effectively inhibited compared with the control. Inhibition effect of sodium metabisulfite on the browning of the concentrate was stronger than that of adipic acid. The addition of sodium metabisulfite afrer concentration of garlic juice was the most effective on browning inhibition in the 9$\^{C}$-heated concentrate, but the addition of sodium metabisulfite before concentration was the most effective in the 45$\^{C}$-heated concentrate and the -50$\^{C}$-freeze-dried concentrate. In the color change of the garlic juice concentrate during the storage, a and b values of the concentrate with sodium metabisulfite were lower than those of the control, which means that sodium metabisulfite inhibited the browning of the garlic juice concentrate.

Purification and Characterization of Biosurfactant from Bacillus sp. DYL130 (Bacillus sp. DYL130 균주의 Biosurfactant의 정제 및 특성)

  • Bacillus sp. DYL130 producing biosurfactant was isolated from soil samples in the Duck-yu mountain and identified as Bacillus sp. by analysis of 16S rDNA sequence. Purification of the biosurfactant was performed by using affinity chromatography and TLC. The biosurfactant of culture medium from Bacillus sp. DYL130 was eluted with 100% methanol using affinity chromatography. To remove methanol, a rotary evaporator was used and enrichment sample was dissolved in alkaline water(pH 10). The purified biosurfactant was identified by TLC. It was confirmed that the Rf value of the biosurfactant was 0.78. Antifungal activity against Botrytis cineria was showed the strongly activity as active antagonist. Maximum emulsification activity and stability were obtained from soybean oil. The critical micelle concentration (CMC) of purified biosurfactant was 35mg/l and the purified biosurfactant inhibited biofilm forming by Bacillus sp..

  • The performance and applicability to food concentration of heat pump were investigated. During heating the water of spa from $43^{\circ}C$ to $51^{\circ}C$, COP's of heat pump (R-12, 150 HP) were 4.03 at heating part and 3.5 at cooling part. And, the efficiency of compressor (${\alpha}$) was 0.477 While the city water was heated to $39^{\circ}C$ by heat pump (R-22, 10 HP), its COP's were 3.0 at heating part and 1.87 at cooling part. During concentrations sucrose solution by centrifugal evaporator (ALFA-LAVAL CO, CTIB) with heat pump, heat capacity for condensating water vapor was required greater 15% than the latent heat for concentrating and then the overall heat transfer coefficient was $1196\;Kcal/m^{2}.\;h.\;^{\circ}C$. When low temperature concentration ($30-35^{\circ}C$, 28-40 Torr) of garlic extract was carried out by the water of $60^{\circ}C$ and $15^{\circ}C$ adjusted by heat pump, the ratio of heat capacity for concentrating vs. that for condensating of water vapor was 0.961.

Field Cooling Tests of Paddy Stored in Steel Bins with a Grain Cooler (곡물냉각기를 이용한 철제 원형빈에서 벼 냉각)

  • Two field cooling tests were conducted to evaluate the cooling characteristic of paddy with a prototype grain cooler. The first test was carried out during summer season in a steel bin with 180.3ton of paddy at Sunchon. And the second test was carried out during harvesting season in a steel bin with 272.2ton of paddy at Ulsan. At the first test, initial paddy temperature of 23.6$^{\circ}C$ was dropped to 14$^{\circ}C$, and initial moisture content of 19.9% was dropped to 19.3% after 52.5 hours of cooling. At the second test, initial paddy temperature of 16.1$^{\circ}C$ dropped to 5.5$^{\circ}C$ after 78.0 hours of cooling. And, at the first test, the average air flow rates of chilled air leaving the grain cooler and penetrating the grain layer were 77.5 ㎥/min and 42.5 ㎥/min, respectively. To prevent leakage of chilled air from plenum chamber of steel bin, which was about 45% of the average air flow rates of chilled air leaving the grain cooler, a proper method was required. The average total power consumption at the first test during summer was 22.1 ㎾ with control of fan damper. At the second test, it was 17.4 ㎾ due to controlling the capacity of compressor with unloading solenoid valve and changing the flow rates of hot refrigerant gas flowing into evaporator and reheater from compressor, resulting in 27% reduction of energy consumption.

Development of a New Commercial Grain Cooler (곡물냉각기의 개발)

  • The objectives of this study were to develop a new commercial grain cooler suited to domestic weather and post-harvesting conditions for paddy, and to evaluate the performance. A prototype grain cooler capable of cooling paddy of 200 tons within 24 hours was developed. The grain cooler was designed to control the refrigeration capacity from 0 to 100% by controlling the capacity of compressor with unloading solenoid valve and by changing the flow rates of hot refrigerant gas flowing into reheater and evaporator from compressor. And a controller with one chip microprocessor was developed to control temperature and relative humidity of cooling air. The maximum cooling capacity of the grain cooler was 35,284㎉/hr at condensing/evaporating pressure of 16.5/3.6 kgf/$\textrm{cm}^2$. Maximum flow rate of cooling air was 120 ㎥/min at static pressure of 279 mmAq. The total maximum required power was 22.8㎾, and total required energy was saved from 26.7 to 33.3% of maximum power depending on operating conditions. The coefficient of performance of refrigeration devices and total coefficient of performance of the grain cooler were 4.71 and 1.8, respectively.

Effects of Iksujisundan on Renal Function, Peroxynitrite Scavenging Activity and Polyol Pathway in Streptozotocin-induced Diabetic Rats (익수지선단(益壽地仙丹)이 Streptozotocin으로 유발된 당뇨병 흰쥐의 신기능, 활성산소, 활성질소 및 Polyol Pathway에 미치는 영향)

  • Objectives : Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Iksujisundan has been known to be effective for the treatment of diabetes. The present study was carried out to investigate the effect of Iksujisundan on renal function, peroxynitrite(ONOO-) scavenging activity and polyol pathway in streptozotocin-induced diabetic rats. Methods : The crushed Iksujisundan was extracted 3 times, each time with 3 volumes of methyl alcohol at 60$^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 87.8g. Iksujisundan extract was orally administreted at 100 mg per 1 kg of body weight for 20 days to the diabetic rats induced by streptozotocin(60mg/kg). The effects of Iksujisundan extract on the streptozotocin-induced diabetic rats were observed by measuring the serum level of glucose, insulin, lipid components, creatinine and BUN, and also the kidney levels of superoxide anion radical(${\cdot}$O2-), nitric oxide(NO) and ONOO-, and also the enzyme activities involved in the polyol pathway. Results : The effects of Iksujisundan on the streptozotocin-induced diabetic rats with regards to body weight, blood glucose and indulin levels, creatinine and BUN levels, total cholesterol and triglyceride lavels, and HDL-cholesterol levels were all shown to be good enough to prevent and cure the diabetes and its complications. Iksujisundan inhibited the generation of ${\cdot}$O2-,NO and ONOO- in the kidney of streptozotocin-induced diabetic rats. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the streptozotocin-induced diabetic rats were reversed toward natural activities. Conclusions : Iksujisundan might inhibit the development of diabetes and its complications by scavenging reactive oxygen and nitrogen species, thereby by reducing oxidative stresses and also by regulating the activities of polyol pathway enzymes, all of which could help to recover kidney function.
