Nutritional Chemical Composition in the Different Parts of Artemisia argyi H. (섬애쑥(Artemisia argyi H.)의 부위별 영양화학성분)

  • The chemical components in different parts of Artemisia argyi was investigated to provide industrial possibilities as functional foods The analysis result of proximate composition in leaves, stems and roots of Artenisia argyi was substantially as follows. The crude protein contents were 19.87, 6.14 and 5.68%, the crude lipid contents were 4.56, 1.30 and 1.20%, the crude fiber contents were 16.80, 29.70 and 29.45%, respectively. The major mineral components in Artemisia argyi were potassium, calcium and magnesium. Contents of potassium and calcium in leaves were 4,270.24 and 617.64 mg/100 g, respectively, they were more than double the contents of root. Sucrose and glucose as main free sugars were detected in the leaves and roots. However, glucose and fructose were identified in the stem. Total amino acids showed 17 amino acids. Contents of total amino acid in the leaves was the highest as 4,864.11mg/100g, and the stems and roots showed 1,953.99 and 1,601.73mg/100g, respectively. The major amino acids in the leaves and stems were proline(963.91 and 407.52mg/100g) and aspartic acid(577.38 and 299.17mg/100g), respectively. Glutamic acid(206.34mg/100g) and arginine(193.23mg/100g) were main amino acids in the roots. The major fatty acids in all parts were linoleic acid($C_{18:2}$), behenic acid($C_{22:0}$), and palmitic acid($C_{16:0}$). Eupatilin(35.0mg/100g) and jaceosidin (107.63mg/100g) as physiological compounds contents were higher in leaves than other parts.

Physicochemical Characteristics of Ethanol Extract from Artemisia Argyi H. Using Different Preparation Methods (전처리 방법을 달리한 섬애약쑥 에탄올 추출물의 이화학적 특성)

  • This study was performed to determine the ethanol ratios (30%, 50%, 70%, and 90%) of extraction solvent of Seomaeyakssuk (Artemisia Argyi H.) prepared using different methods (drying at room temperature [D], aging at $60^{\circ}C$ for 7 days after drying [AD], and roasting for 30 min at $160^{\circ}C$ after drying [RD]). The extract yield of the D extracts was lower than that of the AD and RD extracts, but the ethanol concentration of extract solvent did not affect. The L, a, and b values of the D extracts were highest, whereas those of the AD extracts were lowest. No clear trend was observed in the ethanol ratios. The soluble solids, total phenol, total flavonoid, jaceosidin, and eupatilin contents of each extract varied significantly, with RD > AD > D. The soluble solids significantly increased by ethanol ratio of extraction solvent, but other phytochemicals contents of 50% and 70% ethanol extracts were higher than others without affecting the processing methods. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was highest (77.71%) in the 70% ethanol extract obtained from RD. 2,2-azinbis-(3-ethylbenzo-thiazoline-6-sulphonate(ABTS) radical scavenging activity was significantly higher in 30-70% ethanol extract than 90% ethanol extract from RD. The results suggest that the contents of active ingredients and radical scavenging activity of ethanol extracts from Seomaeyakssuk were highest in the RD extract using 50-70% ethanol.

The Study For Changes In Components And Activities Of Artemisiae Herba. At Various Processing Temperature (가열온도(加熱溫度)에 따른 애엽(艾葉)의 성분변화(成分變化) 및 생리활성(生理活性)에 관(關)한 연구(硏究))

  • Purpose : The study for changes in components and acitvities of Artemisiae Herba at various processing temperature is generally regarded as a foundation in setting the optimum heat-processing temperature and for getting the maximum activities for medical usage of this herb. Methods: Therefore some experiments were performed either in vitro or in vivo and various changes were observed - the changes in the weitht of Artemisiae Herba, the changes in the relative amount of three kinds of extracts from Artemisiae Herba ( diluted ethanol extract, water extract, ether extract ), the TLC pattern of essential oil at various processing temperature, the existance of inhibitory effects both on ${\beta}$-Glucuronidase activities, and on heat-induced hemolysis, the effects on increased vascular permeability. The valid results derived from the experiments are as follows. Results: 1. The weight of Artemisiae Herba prominently decreased at 240^{\circ}C$. 2. The contents of diluted ether extract were maximum in the unprocessing condition. Those of water extract were maximum at 180^{\circ}C$ and at 210^{\circ}C$. and the changes of diluted ethanol extract at 150^{\circ}C$. 3. The TLC pattern of essential oil in Artemisiae Herba at various processing temperature showed that a component began to increase at Rf 0.56 and another component began to decrease at Rf 0.86. 4. The contents of Eupatilin in Artemisiae Herba at various processing temperature continued to decreased in proportion to the temperature rise, the extent of which was prominent at 210^{\circ}C$, and was unnoticeable at 270^{\circ}C$. 5. Inhibitory effects on ${\beta}$-Glucuronidase activities, trypsin activities and heat-induced hemolysis increased in proportion to the density of Artemisiae Herba. Inhibitory effects on ${\beta}$-Glucuronidase activities and trypsin activities were relatively high at 180^{\circ}C$ while on the writhing syndrome and inhibitory effects on increased vascular permeability induced by acetic acid were maximum at 240^{\circ}C$. those on heat-induced hemolysis were relatively high at 240^{\circ}C$. 6. In vivo, both analgesic effects Conclusions: To maximize of the effectiveness of Artemmisiae Herba, the ideal heating temperature is in the range of 180^{\circ}C{\sim}240^{\circ}C$.

DA-6034 Induces $[Ca^{2+}]_i$ Increase in Epithelial Cells

  • DA-6034, a eupatilin derivative of flavonoid, has shown potent effects on the protection of gastric mucosa and induced the increases in fluid and glycoprotein secretion in human and rat corneal and conjunctival cells, suggesting that it might be considered as a drug for the treatment of dry eye. However, whether DA-6034 induces $Ca^{2+}$ signaling and its underlying mechanism in epithelial cells are not known. In the present study, we investigated the mechanism for actions of DA-6034 in $Ca^{2+}$ signaling pathways of the epithelial cells (conjunctival and corneal cells) from human donor eyes and mouse salivary gland epithelial cells. DA-6034 activated $Ca^{2+}$-activated $Cl^-$ channels (CaCCs) and increased intracellular calcium concentrations ($[Ca^{2+}]_i$) in primary cultured human conjunctival cells. DA-6034 also increased $[Ca^{2+}]_i$ in mouse salivary gland cells and human corneal epithelial cells. $[Ca^{2+}]_i$ increase of DA-6034 was dependent on the $Ca^{2+}$ entry from extracellular and $Ca^{2+}$ release from internal $Ca^{2+}$ stores. Interestingly, these effects of DA-6034 were related to ryanodine receptors (RyRs) but not phospholipase C/inositol 1,4,5-triphosphate ($IP_3$) pathway and lysosomal $Ca^{2+}$ stores. These results suggest that DA-6034 induces $Ca^{2+}$ signaling via extracellular $Ca^{2+}$ entry and RyRs-sensitive $Ca^{2+}$ release from internal $Ca^{2+}$ stores in epithelial cells.

  • 황해쑥(Artemisia argyi Lev. et. Vnt)은 국화과의 다년생 초본 식물로서 쑥, 산쑥과 함께 건조한 것이 애엽이라는 생약재로 쓰이며 2016년 기준 전국적으로 81 ha에서 871톤이 생산되고 있다. 주요 유효성분으로 Eupatilin, Jaceosidin 등이 있으며 위궤양에 효과가 있다고 알려져 있어 최근에는 스티렌정 등 신약원료로 각광 받고 있다. 황해쑥은 종자번식 뿐만 아니라 포복경을 이용한 영양번식이 가능하며 현재까지 '섬애' 등 1품종이 개발되어 있으나 농가와 산업체가 요구하고 있는 수량성 또는 Eupatilin 등 성분함량이 높은 품종개발이 필요하다. 국내 연구 결과를 보면 대부분 년 1회 수확을 기준으로 연구되어 왔으나 실제로는 년 2회 수확하는 경우가 더 많다. 따라서 본 실험에서는 년 1회 뿐만 아니라 년 2회로도 수확하여 조사하였다. 시험재료는 AA1801 등 7계통 이며, 시비량은 10 a 당 질소 15.3 kg 인산 20 kg 가리 5 kg이고, 흑색 비닐을 피복한 후 조간 30 cm 주간 15 cm로 하였다. 시험구배치는 난괴법 3반복으로 하였으며 포복경을 5 cm로 잘라 4월 19일에 정식하였다. 1회 수확 시에는 8월 12일, 2회 수확 시에는 6월 17일, 9월 19일에 수확 하였다. 년 1회 수확 시 계통 별 생육특성을 보면, 초장은 202.4~231.5 cm로 평균 213.6 cm 였으며 경경은 10~12.14 mm로 평균 10.9 mm 였다. 분지수는 32.6~37.3개로 평균 35.2개 였으며 엽병장, 엽장, 엽폭은 2.4~4.2 cm, 9.3~12 cm 9.2~12.2 cm로 평균 3.3 cm, 10.7 cm 그리고 10.9 cm 였다. 전초의 생중량과 건중량은 각각 4.9~6.9 kg, 1.8~2.7 kg로 평균 5.5 kg, 2.1 kg 이였다. 특히 AA1902는 다른 계통에 비해 분지수는 적었으나 초장이 크고 경경이 굵었으며 생중량과 건중량도 가장 무거웠다. 년 2회 수확 시 전초의 생중량과 건중량은 각각 5.1~6.2 kg, 1.7~2.0 kg로 평균 5.6 kg, 1.8 kg 이였다. 수량성으로 봤을때는 AA1902는 1회 수확에 적합하며 AA1801, AA1802는 2회 수확 시 생중량과 건중량이 무거웠다. 또한 AA1903의 경우 수확 회수와 상관없이 수량성이 높은 계통으로 판단되었다. 본 자료는 황해쑥 품종육성의 기초자료로 활용하고자 한다.

Antiallergic Effect of Two Variants of Artemisia princeps Pampanini

  • Antiallergic activities of two variants of Artemisia princeps Pampanini SJ-1 (named as Sajabalssuk) and SS-1 (named as Sajuarissuk) cultivated in Ganghwado, which contain high content of eupatilin compared to those cultured by other places, were investigated to evaluate the possibility as inhibitors against allergic diseases. Ethanol and supercritical fluid extracts of SJ-1 and SS-1 inhibited the release of ${\beta}-hexosaminidase$ from RBL-2H3 cells, although their water extracts were inactive. These extracts potently inhibited lipopolysaccharide-induced NO production of RAW264.7. However, these extracts almost did not scavenge free radicals. Oral administration of these extracts to mice inhibited passive cutaneous anaphylaxis reaction induced by IgE, and acute dermatitis induced by 12-O-tetradecanoylphorbol-13-acetate. However, these extracts did not inhibit chronic dermatitis. Scratching behaviors, vascular permeability, and writhing syndromes were weakly inhibited by these extract at a dose of 50 mg/kg. Based on these findings, we believe that SJ-1 and SS-1 can improve IgE-induced allergic diseases such as rhinitis and asthma.

Artemisia argyi H.Lev. & Vaniot Inhibits Matrix metalloproteinases in the Interleukin-1β-stimulated Primary Chondrocytes and Attenuates Osteoarthritis Progression in Mice (황해쑥의 Interleukin-1β 유도 연골세포에서 Matrix metalloproteinase들의 억제효과 및 마우스에서 관절염 진행 감소 효과)

  • 황해쑥(Artemisia argyi H.Lev. & Vaniot)은 우수한 항염증 활성을 지닌 것으로 다양하게 보고 되었다. 그러나, 대표적인 염증 질환 중 하나인 골관절염에 미치는 영향은 현재까지 알려져 있지 않다. 따라서, 본 연구에서는 염증 유발 연골 세포 및 골관절염 유발 동물 모델에 미치는 황해쑥 효과에 대해 조사하였다. 첫째, interleukin 1 beta를 투여한 관절 연골 세포에 황해쑥 물 추출물을 투여한 후 metalloporeinase (MMP) -3 및 MMP-13의 발현을 mRNA 및 단백질 수준에서 분석 하였다. 또한, 내측 반월상 연골의 불안정화에 의해 유도 된 골관절염 마우스 모델을 사용하여 황해쑥 물 추출물의 골관절염 억제 효과를 분석 하였다. 세포 실험에서, 본 황해쑥은 MMP-3와 MMP-13의 mRNA 및 단백질 발현을 유의하게 억제하였다. 또한. 황해쑥 물 추출물을 투여한 실험 동물의 관절 조직을 Safranin O 염색을 실시하여 분석한 결과 연골 하 골판 두께의 감소 및 활막 염증 개선 효과가 관찰 되었다. HPLC를 이용한 성분 분석 결과, 황해쑥 물 추출물은 항염증 및 항관절염 활성을 가진 jaceosidin과 eupatilin을 함유하는 것으로 나타났다. 본 연구결과로부터 황해쑥은 골관절염의 치료 또는 예방에 유망한 소재로 개발될 수 있음을 시사한다.

Effects of Activated Carbon on the Reduction of Benzo(a)pyrene in Artemisia argyi Extract

  • Background and objective: Artemisia argyi has a long history as an effective treatment for various diseases. The detection of environmental pollutant benzo(a)pyrene, a known human carcinogen, in the leaves of Artemisia argyi is cause for concern. For medicinal plant extracts, both a reduction of benzo(a)pyrene as well as the maintained effectiveness of the compound are important. Therefore, in this study, we propose an optimized process for the addition and filtration of activated carbon to reduce benzo(a)pyrene and change the contents of the indicating substance(jaceosidine and eupatilin). Methods: Artemisia argyi EtOH extract containing 36 ppb of benzo(a)pyrene was added to 0.1, 0.5, 1.0, and 1.5% (w/w) of activated carbon for 120 min and filtered using an activated carbon filter 1, 2, 3, and 5 times respectively. The content of benzo(a)pyrene and indicating substances in Artemisia argyi extract were then measured with high performance liquid chromatography (fluorescence and UV detectors). Results: As the amounts of activated carbon powder and filtering cycles increased, the content of benzo(a)pyrene in the Artemisia argyi extract decreased. However, when activated carbon powder 1.5% was added to the extract, and when the activated carbon filter was filtered five times, the results were reduced by 15% and 30~40% respectively. The optimal extraction condition for reducing benzo(a)pyrene was adding 1.5% of activated carbon powder. This resulted in reducing benzo(a)pyrene by 83% and indicating substances by about 4%. Conclusions: Here we present a process for reducing benzo(a)pyrene in Artemisia argyi extract using activated carbon to reduce toxicity and minimize the loss of active ingredients. This approach has potential application within a manufacturing process of various medicinal plant extracts.

DA-6034 ameliorates hepatic steatosis and inflammation in high fat diet-induced obese mice

  • Background: Nonalcoholic fatty liver disease (NAFLD) is characterized by an increase in hepatic triglyceride content and increased inflammatory macrophage infiltration through the C-C motif chemokine receptor (CCR) 5 pathway in the liver. DA-6034 (7-carboxymethyloxy-3',4',5-trimethoxy flavone), is a synthetic derivative of eupatilin that exhibits anti-inflammatory activity in inflammatory bowel disease. However, the effect of DA-6034 on the inflammatory response in NAFLD is not well elucidated. Therefore, we aimed to determine the effect of DA-6034 on hepatic steatosis and inflammation. Methods: Forty male C57BL/6J mice were divided into the following four groups: (1) regular diet (RD), (2) RD with DA-6034, (3) high fat diet (HFD), and (4) HFD with DA-6034. All mice were sacrificed 12 weeks after the start of the experiment. The effects of DA-6034 on macrophages were assessed using RAW 264.7 cells. Results: DA-6034 not only reduced hepatic triglyceride levels and lipid accumulation but also macrophage infiltration and proinflammatory cytokines in HFD-fed mice. According to fluorescence-activated cell sorter analysis, DA-6034 reduced the CD8+ T cell fraction in the liver of HFD-fed mice. DA-6034 also reduced CCR5 expression and the migration of liver macrophages in HFD-fed mice and inhibited CCR2 ligand and CCR4 ligand, which stimulated the migration of macrophages. Conclusion: Overall, DA-6034 attenuates hepatic steatosis and inflammation in obesity by regulating CCR5 expression in macrophages.

Antioxidative and Antibacterial Activities of Artemisia princeps Pampanini Extracts (사자발쑥 추출물의 항산화 및 항균 활성)

  • In the present study, the antioxidative and antibacterial activities of Artemisia princeps Pampanini (A. princeps Pamp.) extract were investigated. The ethyl acetate fraction of A. princeps Pamp. showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}=12.27{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of A. princeps Pamp. extract on $Fe^{3+}-EDTA/H_2O_2$ systems were investigated using a luminol-dependent chemiluminescence assay. The ethyl acetate fraction of the extract ($OSC_{50}=0.33{\mu}g/mL$) had a 5 times greater ROS scavenging activity than L-ascorbic acid ($1.50{\mu}g/mL$), known as a water soluble antioxidant. The cellular protective effects of fractions of A. princeps Pamp. on the rose-bengal sensitized photohemolysis of human erythrocytes were examined. The aglycone fraction of extracts suppressed photohemolysis in a concentration dependent manner. The inhibitory effects of A. princeps Pamp. extract on tyrosinase were investigated to assess their whitening efficiency. The ethyl acetate fraction demonstrated a 7 times higher tyrosinase inhibitory effect ($IC_{50}=29.20{\mu}g/mL$) than albutin, known as a whitening agent. The antibacterial activity of ethyl acetate fractions against various normal skin flora were measured. The results showed that the antibacterial activity of the fraction was the highest on Staphylococcus aureus, Bacillus subtilis, and Propionibacterium acnes. Antioxidant substances were isolated and purified from the ethyl acetate fractions. Eupatilin and jaceosidin were identified. These results indicate that the extract/fractions of A. princeps Pamp. can function as antioxidant and/or antibacterial agents for the skin.
