Antimicrobial Activities of Organic Extracts from Fruit of Thuja orientalis L. (측백나무 열매 추출물의 항균활성)

  • This study was carried out to investigate the antimicrobial activities of organic extracts obtained from the fruit of Thuja orientalis L. The native fruits in Korea were collected and extracted by 80% ethanol, and the extract was sequentially fractionated with n-hexane, chloroform, ethylacetate, and butanol. The fraction yields of n-hexane, chloroform, ethylacetate, butanol and water of ethanol extract were 10.15%, 10.05%, 1.45%, 45.35% and 27.55%, respectively. n-Hexane-soluble fraction showed the highest antibacterial activity against gram positive bacteria, while the chloroform, ethylacetate, butanol and aqueous fractions did not show any antibacterial activity. Minimum inhibitory concentration (MICs) on Staphyloycoccus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction were $100\;{\mu}g$, $500\;{\mu}g$ and $50\;{\mu}g$/disc, respectively. The antibacterial activity was not destroyed by heating at 80, 100, $120^{\circ}C$ for 30 min and was not affected by pH. In the inhibitory test against the Staphylococcus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction showed potent growth inhibition at the concentration of 0.1 and $0.5\;{\mu}g/mL$ for 12~24 hours and n-hexane-soluble fraction did not show any mutagenic activity.

Screening of Antioxidative Compounds toward Human Low Density Lipoprotein (LDL) from Useful Plants (유용식물로부터 Human Low Density Lipoprotein(LDL)에 대한 항산화제의 탐색)

  • This study was designed to investigate antioxidative activity of substances isolated from 25 kinds of useful plants resources toward free radical and human low density lipoprotein(LDL). Methanol extracts of Oenothers odorate had the highest antioxidative activity similar with ${\alpha}$-tocopherol. Methanol extracts of Oenothers odorate was extracted again by the ethylacetate. The ethylacetate soluble acidic fraction obtained from methanol extract of Oenothers odorate showed highest activity toward human LDL. Each fraction was purified through Sepadex LH-20 chromatography by elution of chloroform-methanol mixture (90:10 v/v). Fraction, F-2 obtained from Oenothers odorate showed at highest levels of electron donating activity. Fraction, F-2 was identified as 3,4-dihydroxybenzoic acid and 3-hydroxycinnamic acid.

Anthraquinones with Immunostimulating Activity from Cassia tora L.

  • Many of plants had been reported having immunostimulating activity. This study reports the immunostimulating activity of Cassia tora L. (Leguminosae) seed, by means of solvent extraction method. Ethanol extract and solvent fractions, n-hexane, chloroform, ethylacetate, n-butanol and aqueous layer of Cassia tora L. seed were tested for immunostimulating activity in vitro. The ethylacetate-soluble fraction caused significant inhibition on the production of nitric oxide by murine macrophages (RAW 264.7), and mouse splenocytes were also stimulated at the concentration of 10 pgfmL. Three anthraquinones, chrysophanol (1), isochrysophanol (2) and aloe-emodin (3) with immunostimulating activity were isolated from the ethylacetate-soluble fraction of Cassia tora L. seed through activity-monitored fractionation and isolation method. These results permit Cassia tora L. to be useful as one the of natural immunostimulating crops.

Extraction and Bioassay of Allelochemicals in Jerusalem Artichoke

  • Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.

Phenolic Compounds from Japanese Anise (Illicium anisatum L.) Leaves

  • Japanese anise (Illicium anisatum L.) leaves were collected and ground after drying, then immersed with 50 % aqueous acetone for 3 days. After filtration, the extracts were fractionated with n-hexane, chloroform ($CHCl_3$), ethylacetate (EtOAc) and $H_2O$, and then freeze dried after concentration. A portion of EtOAc (3.12 g) and $H_2O$ (6.08 g) soluble fractions were chromatographed on a Sephadex LH-20 column with various aqueous MeOH solution to isolate the compounds. Compound 1 ((+)-catechin) was isolated from EtOAc soluble fraction. Compounds 2 (quercetin), 3 (quercitrin) and 4 (2''-O-rhamnosylvitexin) were isolated from $H_2O$ soluble fraction. For the first time, quercitrin (3) and 2''-O-rhamnosylvitexin (4) of the isolated compounds were obtained from the extracts of japanese anise leaves.

Functional and Volatile Flavor Compounds in Traditional Kyungsando Squid sikhe (경상도 전통마른오징어 식해의 향기성분 및 기능성)

  • The volatile compounds of traditional Kyungsando squid sikhe were identified by GC-MS. The amount of ${\alpha}-zingibirene$ among identified volatile compounds was 19.73 mg/kg. The major volatile compounds of sikhe were (Z)-Di-2-propenyl disulfide, ${\alpha}-curcumene$, methyl allyl disulfide, (E, E)-a-farnesene, pentanol, z-citral, 3-ethyl-1,2-dithi-5-ene-${\beta}-elemene$, ${\beta}-elemene$, acetic acid, and ${\beta}-phellandrene$. The volatile compounds of sikhe were compose of 49 including hydrocarbone groups, 15 aldehydes groups, 33 alcohol groups kinds, 11 ketone and ester groups. The fraction obtained from sikhe were tested for electron donating ability, angiotensin converting enzyme inhibitory activity and xanthine oxidase inhibitory activity. There were no electron donating abilities$(SC_{50})$ in hexane and water soluble fractions. On the other hand, the angiotensin converting enzyme abilities of ethylacetate and butanol soluble fractions were $310.64\;{\mu}g/mL$ and $1096.49\;{\mu}g/mL$, respectively. Angiotensin converting enzyme inhibitory activities$(IC_{50})$ of ethylacetate butanol soluble fractions were 1.623 mg/mL and 1.303 mg/mL, respectively. Xanthine oxidase inhibitory activities$(IC_{50})$ of ethylacetate fraction and butanol soluble fractions were 3.591 mg/mL and 2.083 mg/mL, respectively.

Hematopoietic Characteristics of the Ethylacetate Fraction of Panax ginseng and Identification of the Chemical Constituents (인삼의 에칠아세테이트 추출분획의 조혈활성 특성 및 구성 성분 동정)

  • The ethylacetate soluble fraction of Panax ginseng extract, which has been reported to exhibit hematopoietic effect, was divided into three subfractions, i.e., Fr. I, Fr. II and Fr. III by silica gel column chromatography. The hematopoietic effect of each subfraction was examined in rats treated with anticancer agent, Ara-C($1-{\beta}-D-arabinofuranosylcytosine)$). Among them, Fr. III showed 90% recovery of leucocyte and erythrocyte counts in bonemarrow cell depleted rats, suggesting it to be hematopoietic fraction. From Fr. III, two major compounds were isolated and identified as ginsenoside $Rh_1$ and $Rg_2$ by $^{13}C-NMR$ and degradation method.

Evaluation of Biological Activity on Hawthorn Tree (Crataegus pinnatifida) Extracts (산사나무 추출물의 생리활성 평가)

  • The wood and bark of Hawthorn tree (Crataegus pinnatifida Bunge) were immersed with 70% aqueous acetone for 3 days. After filtering, the wood extracts were fractionated with n-hexane, chloroform ($CHCl_3$), n-butanol (n-BuOH) and $H_2O$, and the bark extracts were fractionated with n-hexane, $CHCl_3$, ethylacetate (EtOAc) and $H_2O$. Then antioxidative and anti-inflammatory activities were evaluated on each fraction. Antioxidative activity indicated high activity in the n-butanol soluble fraction of wood and in the EtOAc soluble fraction of bark. Especially, the EtOAc soluble fraction of bark showed much higher antioxidative value compared to the controls, buthylated hydroxytoluene (BHT) and ${\alpha}$-tocopherol. In the anti-inflammatory activity, all of the tested fractions except the $H_2O$ soluble fraction of wood showed high inhibitory effect on nitric oxide (NO) production. Based on the above results, the extracts of hawthorn tree may be applied for one of the natural biomass sources that can be used as an antioxidant and an anti-inflammatory substance.

Phenolic compounds from the leaves of Paulownia Coreana Uyeki (오동나무 잎의 페놀성 화합물)

  • The leaves of Paulownia Coreana Uyeki were collected, extracted with acetone-$H_2O$(7:3, v/v), fractionated with n-hexane, methylene chloride and ethylacetate, and freeze dried to give some dark brown powder. The ethylacetate soluble mixture was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. Spectrometric analysis such as NMR and MS including TLC were performed to characterize the structures of the isolated compounds. From the ethylacetate fraction, five flavonoides and three phenolic acids were isolated and determined.

Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Fritillaria thunbergii Miquel (패모(Fritillaria thunbergii Miquel)의 항균,항혈전 및 항산화 활성 평가)

  • Beimu (Fritillaria thunbergii Miquel), a bulbous plant of Liliaceae found in Korea, Japan and China, has been used as an antitussive and expectorant agent, and is also useful in alleviating stonsillitis and bronchiolitis. Most researches have been focused on micro-propagation and plant regeneration, component analysis, and dormancy relieving of beimu. Reports regarding the biological activity of beimu, such as anti-Helicobacter pyroli or platelet aggregation inhibition activity, are few and not widely available. In this study, methanol extract and its organic solvent fractions were prepared from Fritillaria thunbergii Miquel and their antimicrobial, antithrombin, and antioxidant activities were evaluated, respectively. The methanol extract contained lots of water-soluble materials (58.98%) and hexane-soluble oils (14.85%). The ethylacetate and butanol fraction at $500{\mu}g$/disc concentration showed strong antibacterial activity against tested bacteria, except Escherichia coli. Antifungal activity was not observed in methanol extract and its fractions. The hexane, ethylacetate and butanol fractions showed strong antithrombin activity at 4.8 mg/ml concentration. Especially, the ethylacetate fraction showed 95.4 sec of thrombin time at a concentration of 1.2 mg/ml, which is comparable to aspirin, a widely used antithrombosis agent. For antioxidation activity, the ethylacetate and butanol fraction showed good 1,1-diphenyl-2-picryl hydrazyl scavenging activity ($IC_{50}$ of $344{\sim}368{\mu}g$/ml). In superoxide dismutase-like activity and reducing power, the fractions showed $20{\sim}25%$ of vitamin C, and $51{\sim}54%$ of butyl hydroxytoluene, respectively.
