Actinomycins에 의한 Adenosine Deaminase의 억제

  • Adenosine deaminase inhibitor was extracted from culture broth of Streptomyces sp. strain V-8 with ethylacetate. The ethylacetate extract showed the characteristic UV absorption spectrum of actinomycins at 440-450 nm. The ethylacetate extract was compared with respect to inhibitory behavior against adenosine deaminase from calf intestinal mucosa with actinomycin D, -C complex and actinomycin V. The Ki values for actnomycin D, -C complex, and actinomycin V against adenosine deaminase were determined to be 9.9 $\times$ 10$^{-6}$ M, 9.6 $\times$ 10$^{-6}$ M and 9.3 $\times$ 10$^{-6}$ M, respectively. The Ki value for the ethylacetate extract of culture broth against adenosine deaminase was determined to be 5.7 $\times$ 10$^{-6}$ M. The kinetic parameters of actinomycin D, -C complex, -V and ethylacetate extract of culture broth for adenosine deaminase were as follows:I$_{50}$ = 1.5 $\times$ 10$^{-5}$ M (actinomycin D), 2.7 $\times$ 10$^{-5}$ M (actinomycin C complex), 3.5 $\times$ 10$^{-5}$ M (actinomycin V), 8.9 $\times$ 10$^{-6}$ M (ethylacetate extract of culture broth). The adenosine deaminase was inhibited noncompetitively by ethylacetate extract of culture broth as well as by actinomycin D, -C complex and actinomycin V.

Antioxidant Effect of Ether and Ethylacetate Fractions of Pueraria thunbergiana Extract on Perilla Oil (들기름에 대한 칡추출물 분획의 항산화 효과)

  • The objective of this study was to determine antioxidant effect of ether and ethylacetate fractions of 70% ethanol extract of some food (acid treated or not) on perilla oil. Each fraction of food extract was added to perilla oil and stored for 0,3,6.9,11 days at 60$^{\circ}C$. Then, the peroxide value (POV) of perilla oil samples were analyzed. Perilla oil contained ${\gamma}$-tocopherol 0.6800 $\mu\textrm{g}$/mg, ${\alpha}$-tocopherol 0.3189 $\mu\textrm{g}$/mg. $\delta$-tocopherol 0.0463 $\mu\textrm{g}$/mg, but it was easily oxidized due to high linolenic acid content. To increase yield of ether and ethylacetate fractions from each food extract, the 70% ethanol extract was treated with 0.2% H$_2$SO$_4$ and fractionized by ether and ethylacetate. Among ether and ethylacetate fractions of 70% ethanol extracts of some food, the yield of ethylacetate fraction of acid treated Pueraria thunbergiana extract was 5 times more than that of ethylacetate fraction untreated with acid. Perilla oil which added 100 ppm ethylacetate fraction of acid treated Pueraria thunbergianan extract showed low POV (44.8 meq/kg) compared to POV (80.0 meq/kg) of control.

Pharmacological Effects of Extract of Bufonis Yenenum (섬수(Bufonis Yenenum) 추출물의 약리작용)

  • Bufonis Venenum is a toad venom and its main components are bufadienolides, namely resibufogenin, bufalin and cinobufagin. The desensitizing effect of Bufonis Venenum is useful for the treatment of the premature ejaculation in Chinese medicine. But, minor components of Bufonis Venenum cause problems such as topical burring, pain, and erectile dysfunction. To clarify and eliminate the components responsible for these side effects, we prepared two extracts of Bufonis Venenum with either 70% ethanol or ethylacetate and tested their pharmacological effects. The extract of Bufonis Venenum with 70% ethanol produced pain response in rat hind paw, and exhibited contraction of rabbit corpus cavernosal muscle in vitro. On the other hand, the ethylacetate extract did not cause pain and smooth muscle contraction. The desensitizing effect of the ethylacetate extract was similar to that of the 70% ethanol extract. In conclusion, these results show that the extract of Bufonis Venenum with ethylacetate does not have the components causing side effects and deserve further study for therapeutic potential in premature ejaculation in men.

Effects of subfractions of Coptidis Rhizoma extract on the nitric oxide production in LPS-stimulated BV2 microglial cells (황련 추출물의 분획화 및 BV2 microglial cells에서 LPS에 의해 유도되는 nitric oxide 생성억제효과 검정)

  • Objectives : Uncontrolled activation of microglia may directly toxic to neurons by releasing various substances such as inflammatory cytokines, nitric oxide(NO), prostaglandin E2 and superoxide. In this study, the effects of the several subfractions isolated from Coptidis Rhizoma extract were investigated on NO production in LPS-stimulated BV2 microglial cells, Methods : Coptidis Rhizoma extract prepared with 80% methanol, and then fractionated with ethylacetate, chloroform, n-butanol and water. BV2 cells were pretreated four subfractions of Coptidis Rhizoma with various concentrations, and then stimulated with LPS. Cytotoxicity of each fraction was measured by MTT assay. NO production was determined in culture surpernatants by Griess reagent. Results : Ethylacetate, chloroform and butanol fractions of Coptidis Rhizoma extract significantly decreased LPS-induced NO production in BV2 cells as a dose-dependent manner without cytotoxicity. Ethylacetate fraction of Coptidis Rhizoma extract was most effective on inhibition of NO production in LPS-stimulated BV2 cells compared with other fractions. Conclusion : This data indicates that Ethylacetate fraction of Coptidis Rhizoma extract shows strong antiinflammatory effects through inhibition of LPS-induced microglial activation.

Antimicrobial Activity and Composition of Extract from Smilax china Root (청미래덩굴 뿌리에서 추출한 순차분획물의 항균활성과 성분 분석)

  • Antimicrobial activities, extraction yields and chemical compositions of the extract from the powder of vacuum dried Smilax china root on various solvents were investigated. Antimicrobial activities against. A rhizogenes, A. tumefaciens, C. utilis, S. cerevisiae, B. megaterium, B. subtilis and E. coli were measured by disc diffusion method. Methanol, chloroform, ethylacetate and butanol extracts exhibited potent antimicrobial activities against B. megaterium and B. subtilis. Also, methanol, ethylacetate and butanol extracts had the activities against A. rhizogenes, and chloroform and ethylacetate extracts against A. tumefacciens. The extraction yields were 13.10%, 8.71%, 2.73%, 0.83%, 0.54% and chemical compositions of their extracts were very different according to the kinds of solvents. The main chemical groups of ethylacetate, methanol, chloroform and butanol extracts were 45.99% phenolics, 36.10% acids, 29.13% phenolics and 18.96% phenolics, respectively.

Screening of Antimicrobial Activity of the Dandelion (Taraxacum platycarpum) Extract (민들레(Taraxacum platycarpum) 추출물의 항균성검색)

  • In ordo. to develop the natural food preservative agent, freeze dried dandelion (Taraxcum platycarpum) was extracted with several solvents, and antimicrobial activity was investigated. The methanol extract obtained from the dandelion exhibited antimicrobial properties against five strains such as Bacillus subtilis, Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, and Vibrio parahaemolyticus. The methanol extract at the concentration of 2000 $\mu\textrm{g}$/ml completely inhibited the growth of B. subtilis, E. coli, L. monocytogenes and V. parahaemolyticus. Antimicrobial activity of the ethylacetate fraction from the methanol extract of dandelion was the strongest fraction compare to those the other solvent fractions such as n-hexane, chloroform, n-butanol and water. The ethylacetate fraction showed the inhibitory effect at the concentration of 0.5 mg/disc on the growth of the food spoilage microorganisms.

Isolation of Antimicrobial Substance from the Korean Traditional Leaf Mustard, Brassica juncea Coss.

  • The antimicrobial effect of each fraction after fractionation of an ethanol extract of leaf mustard was examined in terms of nucleic acid, chloroform, ethylacetate, and butanol. The ethylacetate fraction, which showed the strongest level of antimicrobial effect among the different ethanol extract fractions of leaf mustard, was isolated and purified using silica gel column chromatography and HPLC, respectively, to obtain a single antimicrobial substance called KLM-1. The antimicrobial effect of this substance was 10 times higher than that of the ethylacetate fraction. A further study is on the way to confirm the structure of the antimicrobial substance KLM-1 through LC/Mass and NMR.

Evaluation of Antioxidant Potential of Extract/Fractions of Equisetum arvense (I) (쇠뜨기 추출물의 항산화 작용에 관한 연구(I))

  • In this study, the antioxidative effects of Equisetum arvense extracts were investigated. The free radical (1,1 diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) of extract/fractions of Equisetum arvense was in the order: 50 % ethanol extract ($182.04{\mu}g/mL$) < ethylacetate fraction ($54.50{\mu}g/mL$) < deglycosylated flavonoid aglycone fraction ($14.13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of some Equisetum arvense extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol- dependent chemiluminescence assay. The order of ROS scavenging activity was deglycosylated flavonoid aglycone fraction ($OSC_{50}$, $3.54{\mu}g/mL$) < 50 % ethanol extract ($0.80{\mu}g/mL$) < ethylacetate fraction ($0.006{\mu}g/mL$). Ethylacetate fraction showed the most prominent scavenging activity. The protective effects of extract/fractions of Equisetum arvense on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethanol extract (50%) suppressed photohemolysis in a concentration dependent manner, particularly deglycosylated aglycone extract exhibited the most prominent celluar protective effect ($\tau_{50}$, 161.10 min at $10{\mu}g/mL$). These results indicate that extract/fractions of Equisetum arvense can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS.

Antioxidative Effect of Persimmon Leaves (감잎의 항산화 효과)

  • The production of malondiadehyde(MDA) was significantly decreased when the methoanol extract of persimmon leaves was added to the system. The methanol extract of persimmon leaves was fractionated by using various solvents such as hexane, chloroform, ethylacetate and butanol. Among the above fractions especially the chloroform fraction, ethylacetate fraction revealed the strong antioxidative activities. The hot water extract of the persimmon leaves was less effective than tannin which was extracted from persimmon leaves in antioxidative activity.

Chemical Composition of Pinus koraiensis Seed and Its Biological Activity

  • This study was to investigate the chemical composition and biological activities of Pinus Koraiensis seed. The oil, moisture, ash, crude protein, and carbohydrate contents of P. Koraiensis seed were 58.21, 7.84, 1.56, 14.26, and 18.13%, respectively. The ratios of essential amino acid and nonessential amino acid against total amino acids were 36.6 and 60.3%, respectively. The saponification value of seed oil was 166.8 mg KOH/g oil. Among various fatty acids, the linoleic acid content was the highest, 35.5%, which was approximately 72.6% of polyunsaturated fatty acid. The ethylacetate extract of P. Koraiensis seed had the highest DPPH radical scavenging activity (62.8%) at 7.0 mg/mL, followed by hexane extract, methanol extract, and hot water extract. The maximum nitrite scavenging activity was obtained 59.3% at pH 1.2. The total phenolic concentration of ethylacetate extract was 98.7 mg/g, approximately 4.8 folds higher than that of the hot water extract. The maximum inhibition activities of elastase using ethylacetate extract and collagenase using hexane extract were 58.8 and 40.7%, respectively. These results indicate that P. koraiensis seed extract could be applied to present the possibilities of industrial applications for the developments of cosmetics.
