Effect of Perilla frutescens Extract on The Growth of Food-Borne Pathogens (자소엽 추출물이 식중독 유발세균의 증식에 미치는 영향)

  • In this study, Perilla frutescens wh~ch has been used as a folk remedy was investigated its antimicrobial activity. First, the Perilla frutescens was extracted with methanol at room temperatures, and fractionation of the methanol extracts from Perilla frutescens was carried out by using petroleum ether, chloroform, and ethyl acetate, respectively. The antimicrobial activity. of the. Perilla frutescens extracts was determined using a paper disc method and the growth inhibition curve was determined using methanol extracts of Perilla frutescens against Salmonella enteritidis and Bacillus cereus. The methanol extract of Perilla frutescens showed strong antimicrobial activity against Bacillus cereus at the concentration of 1.000 ppm. The 4,000 ppm of methanol extract from Perilla frutescens. retarded the growth of S. enteritidis and Bacillus cereus up to 72 hours.

Antimicrobial Effect of Sophora angustifolia Extracts on Food-Borne Pathogens

  • This study was performed to investigate the antimicrobial effect of Sophora angustifolia extracts against food-borne pathogens. First, Sophora angustifolia was extracted with methanol at room temperature, and the methanol extracts from Sophora angustifolia were fractionated using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Sophora angustifolia extracts was determined using the paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Sophora angustifolia showed the highest antimicrobial activity against Staphylococcus aureus and Salmonella typhimurium. A synergistic effect was found in the combined extracts of Sophora angustifolia and Portulaca oleracea, compared to the activity of each extract alone. Finally, the growth inhibition curve was determined using the methanol extracts of Sophora angustifolia against Staphylococcus aureus and Salmonella typhimurium. The methanol extract of Sophora angustifolia showed strong antimicrobial activity against Staphylococcus aureus at a concentration of 5,000 ppm. The 5,000 ppm methanol extract from Sophora angustifolia retarded the growth of S. aureus for more than 24 hours and of Salmonella typhimurium for up to 12 hours.

  • As a part of studies on the quality control of crude drug preparation, So-Shi-Ho-Tang, index components of Scutellariae Radix were identified by TLC and quantified by HPLC. Wogonin and baicalein were identified in ethyl ether fraction on silica gel plate with benzene/ethyl acetate (1 : 1, v/v). The content of baicalin was determined by HPLC on ${\mu}-Bondapak\;C_18$ column with 0.5%-phosphoric acid/acetonitrile (73 : 27, v/v). Its recovery rate in the extract granules, compared to the content in the Scutellariae Radix, was $52.1\;{\pm}\;0.7%$.

  • This reserch was carried out to investigate the nitrite scavenging abilities (NSA) of 7 kinds of tea extracts. Nitrites are used as additives of cured meat preperation and exist in plants, foods, and medicines, so we intake it very often easily. Nitrites can act with amines to produce nitrosamine which is known to be a carcinogen. It is known that the pH, concentration of amines, and amounts of nitrite are three important conditions of nitrosamine occurence. 7 kinds of tea used in this experiment were Persimmon tree (Diospyroo kaki Thiunb, Per.), Mulberry tree (Morus alba Linne, Mul.), Rubber tree (eucommia ulmoi-des Oliver, Rub.), Solomon's-seal (Bolygonatu Morr, Som.), Chicory (Cichorrium intybus L, Chi.), Sumach (Rhus javanica L., Sum.), Docwood (Cornus officinale Sieb, Doc.) and they were extracted with methanol (MeOH), ethyl ether (EtEt), ethyl acetate (EtAc), and also the extract existed in the aqueous layer II (Aq L. II) was used.

  • The insecticidal compound priduced by Streptomyces SS-4993 strain was indicated to be a relatively stable small compound by pancreatin treatment. The active compound was adsorbed on HP20 and eluted with 70% acetone. Then, the eluate was extracted with ethyl acetate. The content of the extract was determined by thin layer chromatography (Silica gel 60 F254, hexane-ethyl ether-acetic acid,80 : 20 :1). Three active bands (Rf 0.08, 0.07, 0.88) were identified by dermal application using Bombyx mori larvae. Electrophoretic analysis revealed that the injection of strain SS-4993 culture broth into B. mori larvae induce the reduction of the hemolymph proteins level. Especially, the level of apolipophorin-I was decreased drastically.

  • Inhibition of xanthine oxidase by seaweed extracts obtained from Undaria pinnatifida, Ecklonia stolonifera, Ecklonia cava, Laminaria japonica, Sargassum, Codiumfragile, Enteromorpha compressa and Porphyra tenera were investigated. Extracts of E. stolonifera and E. mua remarkably inhibited xanthine oxidase activity compared to those of other seaweed. The xanthine oxidase inhibitory activity of E. cava was higher than that of E. stolonifera. Diethyl ether extract from E. cava was more effective in the inhibition of xanthine oxidase than other solvent extracts. Two xanthine oxidase inhibitors(A-1 and A-2) from diethyl ether extract were isolated and purified by silica gel column chromatography, thin layer chromatography and high performance liquid chromatography. Xanthine oxidase inhibitory activities of these compounds were 27.8 and 48.1% per 0.4mg, respectively. The active compound A-2 had absorption peak at 420nm, 456nm and 467nm, which can be considered as siphonaxanthine.

Studies on the Cytotoxicity and Antitumor Activity of Perilla frutescens (소엽의 세포독성 및 항암작용에 관한 연구)

  • The cytotoxic and antitumor activity of Perilla frutescens extract on cultured 3T3 fibroblast and skin melanoma cells were evaluated by tetrazolium MTT (MTT) and neutral red (NR) colorimetric assay methods. Lactate dehydrogenase activity was also measured. The light microscopic study was carried out to observe morphological changes of cultured mouse fibroblast and skin melanoma cells. The results were as follows: 1. Water and ether extracts showed a significant cytotoxicity in 3T3 fibroblast and all extracts exhibited a significant anti-tumor activity in skin melanoma cells. Methanol, ethyl acetate and ethanol extracts showed low cytotoxic effects, but exhibited a high anti-tumor activity. 2. The MTT absorbance in 3T3 fibroblast was significantly decreased by treatment with ether, water, chloroform and ethanol extracts and skin melanoma cells was significantly decreased by treatment with all extracts. The difference in MTT absorbance in two cell types was most remarkable when treated with methanol and ethanol extracts. 3. Methanol and ethyl acetate extracts showed the strongest effect in growth inhibition of melanoma cells. These results indicated that methanol extract possessed a low cytotoxicity and a strong anti-tumor activity.

Effects of Cancer Prevention and Immune Stimulation of Fractions from Capsosiphon fulvescens (매생이 추출분획의 암 예방 및 면역증진 효과)

  • The fractions of Capsosiphon fulvescens were studied to verify the anticancer and immunostimulating activity. The fractions from the ethanol extract of C. fulvescens were prepared by the systematic extraction procedure with the solvents such as hexane, ethyl ether, methanol, butanol and H$_2$O. The cytotoxic effects of C. fulvescens fractions against human leukemia cell line U937, mouse neuroblastoma cell line (NB41A3), human hepatoma cell line (HepG2)and rat glioma cell line (C6) were investigated. Ethyl ether fraction of C. fulvescens showed the highest cytotoxicity against all four cell lines tested. In addition, H$_2$O fraction also showed relatively high cytotoxicity. Dose dependent patterns were observed on all four cell lines. The immune-stimulating effects of C. fulvescens fractions on rat macrophage cell line (RAW 264.7) were also investigated. All five fractions of C. fulvescens extract stimulated NO production with concentration dependant manner. These results suggest that C. fulvescens may be a useful candidate for a natural antitumor and immune-stimulating agent.

Characteristics of Nitrite Scavenger Derived from Seeds of Cassia tora L. (결명자 추출물의 아질산염 소거인자의 특성)

  • The present study was conducted to elucidate nitrite scavenger derived from seeds of Cassia tora L. Methanolic extract of Cassia tora L was refractionated into ethyl ether, chloroform, ethyl acetate and water farction, and nitrite scavenging abilities of these fractions were investigated. Among these fractions, ethyl acetate fraction had the strongest nitrite scavenging ability(97%/2 mg). Therefore, to further investigate nitrite scavenger, ethyl acetate fraction was fractionated by silica gel column chromatography with a chloroform-methanol($10:0{\sim}0:10$) and then compound A and B were isolated. Compound A had a stronger nitrite-scavenging effect than compound B. Therefore the further separation of compound A was carried out by HPLC(32% acetonitrile. 1 ml/min) using ${\mu}Bondapak$C18 column$(3.9{\times}300 mm)$ and finally compound A-1 was obtained from compound A. Compound A-1 had by far nitrite-scavenging ability as compared with that of ascorbic acid. Compound A-1 was identified as $nor-rubrofusarin-6-{\beta}-mono-D-glucoside$ from the profiles of UV, IR and $^1H-NMR$.
