Isolation of Antioxidative Compound from Scutellaria baicalensis G. (황금으로부터 항산화 활성 성분의 분리)

  • Root of Scutellaria baicalensis G. was extracted with methanol and water to give the yield of 30.0% in order to find the antioxidant substance. The extract was fractionated with diethyl ether, n-butanol and water to test the inhibitive activity against xanthine oxidase. Three fractions inhibited the activities of xanthine oxidase by 48.2%, 10.2% and 2.8%, respectively, at the amount of $0.1\;{\mu}g$. A component that exhibited strong inhibition of xanthine oxidase was isolated from diethyl ether fraction (SE Fr.) by silica gel column chromatography and HPLC, and then identified by $^1H-NMR$, $^{13}C-NMR$ and MS spectrophotometry. EDA (Electron Donating ability) of the compound was 28.5% at the concentration $100\;{\mu}g/3\;ml$. That was identified to be 3,5,7-trihydroxy-2'-methoxyflavanone by spectrophotometric analysis using $^1H-NMR,\;^{13}C-NMR$ and Mass spectrophotometry.

The Anthelmintic Principle of "O-Mae", the Roasted Fruits of Prunus mume, Against Clonorchis sinensis (오매의 간디스토마 살충성 물질에 관한 연구)

  • The anthelmintic substance of the roasted fruits of Prunas mume against Clonorchis sinensis was isolated and its structure was identified by chemical and physical methods. The results obtained from the experiments are as follows: 1) The methanol extract of the roasted fruits of P. mume was fractionated into four parts: petroleum ether, ethyl ether, ethyl acetate and water soluble part. Among these, etherial fraction was found to have strong wormicidal effect on liberated metacercaria of Clonorchis sinensis. 2) From the etherial fraction, the wormicidal substance was isolated by silica gel, polyamide and sephadex column chromatography and identified to be 5-hydroxymethylfurfural (5-HMF) by chemical and spectral data. 3) 5-HMF was synthesized and administered to the rabbits infected with C. sinensis. On the 2nd day after administration, the EPG (eggs per gram in feces) reached to the maximal value. Among the adult worms isolated from the bile duct of the treated animal, 84% of worms were damaged morphologically. 4) The content of 5-HMF in the fruits of P. mume which were roasted in an oven at $90-110^{\circ}C$ for 52 hours and that in the fresh fruits was evaluated by HPLC. The content of 5-HMF was 0.8% in the roasted fruits and 0.02% in the fresh ones.

  • Prunus mume was extracted using various solvents including ethyl ether, ethyl acetate and n-butanol. The extracts were fractionated by column chromatography. Antimicrobial compounds (A, B and C) in fractions showing antimicrobial activity were purified by Sephadex LH 20 column chromatography, and identified by $^{1}H$- and $^{13}C$-NMR analysis as isoeugenol, nomilin and $\beta$-sitosterol, respectively.

A Cytotoxic Activity of Panax Ginseng Extract Against Bome Cancer Cells In Vivo and In Vitro.

  • This study was devised to observe the cytotoxlc activities of petroleum-ether extract of Panax ginseng root(crude Gx) and its partially purified fraction from silicon acid column chromatography(7:3 CX) against sarcoma-180(5-180) and Walker carcinosarcoma 256(Walker 256) in vivo, and murine leukemic lymphocytes(L1210) and human rectal cancer cell(HRT-18) and human colon cancer cells(HT-29 and HCT-48) in vitro . Each cell-line was cultured in medium containing serial concentrations of the crude Gx or 7:3 Gx in vitro. A highly lipid soluble compound in the extract of Panax ginseng root was cytocidal to murine leukemic cells and human colon and rectal cancer cells in vitro In the meantime, ginseng saponin derivatives did not cytotoxic effects at its corresponding concentration. The growth rates of the cancer cells in medium containing ginseng extracts were inhibited gradually to a significant degree roughly in proportion to the increase of the extract concentration. The cytotoxic activity of 7:3 Gx was about 3 times more potent than that of crude Gx, one unit of cytotoxic activity against L121f cells being equivalent to 2.54$\mu\textrm{g}$ and 0.88 $\mu\textrm{g}$ for the crude Gx and 7:3 Gx, respectively. The Rf value of the active compound on silica -gel thin layer chromatography with petroleum-ether/ethyl ether/acetic acid mixture (90:10:1, v/v/v) as a developing solvent was 0.23. The survival times of mice inoculated with S-180 cells were extended about 1.5 to 2 times by the 7:3 Gx treatment compared with their control group. The significantly decreased hemoglobin values of rats after inoculation with Walker 256 were recovered to normal range by oral administration of the crude Gx. The synthetic levels of protein, DNA and RNA in human colon and rectal cancer cells were significantly diminished by treatment with the crude Gx, which can explain a part of the origin of its anticancer activity.

  • Sixty-eight kinds of aliphatic and aromatic substances peaks from ether extract of Codonopsis pilosulae Radix were shown by GC-Mass. Of them, 3 kinds of compounds were identified as ethyl benzene, acetophenone and 1-phenyl-2-butanone, which are compared with authentic specimen, and one compound was assumed as an aliphatic $C_8H_{16}O\;or\;C_9H_{20}$.

Effect of Dandelion Leaf Extracts on Lipid Metabolism in Rats Fed High Cholesterol Diet (민들레잎추출물의 흰쥐 체내 지질대사 개선 효과)

  • The effect of dandelion leaf extracts on lipid metabolism was investigated in high cholesterol-fed rats. Four groups of rats were given a high cholesterol diet containing 10g cholesterol/kg and 2.5 g sodium cholate/kg for 6 weeks. The control group received without dandelion leaf extracts and the other three groups received with one of dandelion leaf extracts (water, ethyl acetate, ether), respectively. Weight gain and feed intake were significantly higher in water group than control group. Serum triglyceride, total cholesterol, cholesteryl ester, chylomicron and VLDL-cholesterol concentratins were lower, whereas HDL-cholesterol and phospholipid concentrations were significantly higher in dandelion leaf extract supplemented-groups than control group. Dandelion extract increased HDL-C/total-C ratio and lowered atherogenic index. Hepatic triglyceride, total cholesterol, free cholesterol and cholesteryl ester concentrations lowered in dandelion leaf extract groups compared to control group. Especially, hepatic triglyceride and free cholesterol levels were lower in ethyl acetate extract group than in water and ether extract groups. Dandelion leaf extract supplements did not affect hepatic phospholipid cencentration. Hepatic HMG-CoA reductase activity was significantly lower in water extract group than the others. ACAT activity was significantly decreased by dandelion extract supplements.

  • Six commercial ginseng products, red ginseng, white ginseng I and II, red ginseng extract, white ginseng extract I and II were extracted with ethyl ether. Total fatty acid composition of the extracted free lipids was analyzed by gas liquid chromatography and digital integrator was used to measure the area of each peak. Crude lipid contents of red and white ginsengs were similiar. Those of red ginseng extract, white ginseng extract I and II were 1.10, 1.13 and 0.40%, respectively. It was found that 22 kinds of fatty acids existed in red and white ginsengs. Among them, 16 kinds of even numbered fatty acids were identified. Linoleic acid in red and white ginsengs was the most abundant. The contents of that in red ginseng, white ginseng I and II were 63.33, 45.55 and 41.06%, respectively. The next most abundant acid was palmitic acid, the contents of which were 11.30, 14.4 and 18.10% for red ginseng, white ginseng I and II, respectively. Major fatty acids for red ginseng extract and white ginseng extract I were linoleic and palmitic acids in the same order of magnitude. Linoleic and palmitic acids for red ginseng extract were 15.93 and 15.71 %, respectively, while linoleic and palmitic acids for white ginseng extract I were 21.94 and 19.15%, respectively. However, white ginseng extract II contained only 9.21% of linoleic acid and 16.13% of palmitic acid which was the major fatty acid.

  • A experiment was performed to investigate the stigmasterol and campesterol in the Panax ginseng. Crude sterol fraction obtained from the ginseng was analysed by TLC and quantitative determination of ${\beta}$-sitosterol was performed with colorimetric method. The results were summarized as follwer, 1. It was observed with ethyl ether-petroleum ether extract of the ginseng that there was no evidence of showing the presence of both free capesterol and stigmasterol but that only ${\beta}$-sitosterol was presence in an significant amount. 2. Average amount of the ${\beta}$-sitosterol was 0.20-0.35mg per gram of the sample.

Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium (오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours.   • As the continued studies for Korean cultivated rhubarb, MeOH extract of the leaves was fractionated with ether, ethylacetate, and n-butanol. From the ethyl acetate fraction of MeOH extract, one flavone glycoside, apigenin-8-${\beta}$-D-glucopyranoside(vitexin, $C_{21}H_{20}O_{10}$) and from the n-BuOH fraction of MeOH extract, two flavonol glycosids, kaempferol-3-O-(2,6-di-O-rhamnopyranosyl)-${\beta}$-D-galactopyranoside$(C_{33}H_{40}O_{19})$and quercetin-3-O-rutinoside(rutin, $C_{27}H_{30}O_{16}$) were isolated and identified through the physico-chemical properties and spectroscopic evidences(UV, IR, NMR, Mass) respectively.
